ABSTRACT

Quality spawns are an important factor to support the success of button mushroom cultivation. The quality of mushroom spawn is in?uenced by the nutritional composition of the media. The purpose of this study was to determine the best media for the growth of F0 and F1 button mushroom. The research was carried out in March-May 2021 at the Biology Laboratory, Faculty of Agroindustry, University of Mercu Buana Yogyakarta. The study consisted of 2 series of experiments. This study consisted of 2 series of experiments. The ?rst experiment was to test the growth quality of F0 champignon on 3 types media, that is potato dextrose agar (PDA),mung been sprout extract media, and white sweet potato media. The second experiment tested the growth quality of F1 button mushroom grown on three types of media, that is corn, barley and sorghum. Both experiment were single factor trials arranged in a completely randomized design (CRD) with three replications, and each replication consisted of 5 units. The results showed that the growth of F0 button mushroom grown on PDA media and mung bean sprout extract was better than white sweet potato extract media. For the growth of F1 button mushroom, the best media was corn, the second was sorghum and the third was barley

ABSTRACT

Objective:To investigate the expression and clinical significance of F0 ATP synthase C subunit (Csub) in patients with ischemic heart disease (IHD).Methods:The 101 patients with chest pain admitted to the department of emergency of the People's Hospital of Yuhuan from May 2019 to December 2020 were enrolled, including 59 patients with acute myocardial infarction (AMI) and 42 patients with unstable angina pectoris (UAP). At the same time, 50 age-matched healthy subjects in the health examination center were selected as the healthy control (HC). All patients had completed blood sampling before the intervention of drugs or other intervention measures in the emergency room. The content of serum Csub was detected by enzyme linked immunosorbent assay (ELISA), and the relationship between Csub and clinical characteristics was analyzed. At the same time, the contents of hypersensitivity cardiac troponin T (hs-cTnT) and MB isoenzyme of creatine kinase (CK-MB) in blood were detected by electrochemical luminescence. The receiver operator characteristic curve (ROC curve) was drawn to evaluate the value of Csub, hs-cTnT, and CK-MB in the early diagnosis of IHD.Results:The baseline data such as age, gender, and history of the three groups were balanced. There were significant differences in low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), CK-MB, hs-cTnT and B-type natriuretic peptide (BNP), but there were no significant differences in other biochemical indexes. The Csub content in the AMI group and the UAP group were significantly higher than those in the HC group [8.96% (6.37%, 11.53%), 4.27% (3.23%, 6.49%) vs. 1.56% (1.07%, 2.33%), both P < 0.01]. Moreover, the Csub in the AMI group with more severe myocardial ischemia was higher than UAP group [8.96% (6.37%, 11.53%) vs. 4.27% (3.23%, 6.49%), P < 0.01]. A total of 59 patients with AMI were treated with percutaneous coronary intervention (PCI). According to the median of Csub, AMI patients were subdivided into above-median group (29 cases) and below-median group (30 cases). The results showed that there were no significant differences in the number of coronary artery lesion branches, the number of stent implantation and postoperative medication between the two groups. ROC curve analysis showed that the area under the curve (AUC) and 95% confidence interval (95% CI) of Csub, hs-cTnT and CK-MB in the diagnosis of IHD were 0.98 (0.95-1.00), 0.99 (0.99-1.00), 0.94 (0.89-0.99), respectively. The diagnostic efficacy of Csub was slightly lower than that of hs-cTnT but higher than that of CK-MB. When the cut-off value of Csub was 4.74%, the sensitivity and specificity for the diagnosis of IHD were 100% and 87.0%, respectively. Conclusions:Csub increased significantly in the serum of IHD patients, and further increased with the severity of ischemia. It can be used as a new diagnostic biomarker for the diagnosis and evaluation of the development of myocardial ischemia.

ABSTRACT

Solid phase microextraction(SPME)in combination with high-resolution mass spectrometry was employed for the determination of metabolomic profile of mouse melanoma growth within in vitro 2D,in vitro 3D,and in vivo models.Such multi-model approach had never been investigated before.Due to the low-invasiveness of SPME,it was possible to perform time-course analysis,which allowed building time profile of biochemical reactions in the studied material.Such approach does not require the multiplication of samples as subsequent analyses are performed from the very same cell culture or from the same individual.SPME already reduces the number of animals required for experiment;therefore,it is with good concordance with the 3Rs rule(replacement,reduction,and refinement).Among tested models,the largest number of compounds was found within the in vitro 2D cell culture model,while in vivo and in vitro 3D models had the lowest number of detected compounds.These results may be connected with a higher metabolic rate,as well as lower integrity of the in vitro 2D model compared to the in vitro 3D model resulting in a lower number of compounds released into medium in the latter model.In terms of in vitro-in vivo extrapolation,the in vitro 2D model performed more similar to in vivo model compared to in vitro 3D model;however,it might have been due to the fact that only compounds secreted to medium were investigated.Thus,in further experiments to obtain full metabolome infor-mation,the intraspheroidal assessment or spheroid dissociation would be necessary.

ABSTRACT

Malignant cell transformation could be considered as a series of cell reprogramming events driven by oncogenic transcription factors and upstream signalling pathways. Chromatin plasticity and dynamics are critical determinants in the control of cell reprograming. An increase in chromatin dynamics could therefore constitute an essential step in driving oncogenesis and in generating tumour cell heterogeneity, which is indispensable for the selection of aggressive properties, including the ability of cells to disseminate and acquire resistance to treatments. Histone supply and dosage, as well as histone variants, are the best-known regulators of chromatin dynamics. By facilitating cell reprogramming, histone under-dosage and histone variants should also be crucial in cell transformation and tumour metastasis. Here we summarize and discuss our knowledge of the role of histone supply and histone variants in chromatin dynamics and their ability to enhance oncogenic cell reprogramming and tumour heterogeneity.

ABSTRACT

BACKGROUND AND OBJECTIVES: Numerous studies have indicated deterioration of speech perception in noisy conditions among the elderly even those with normal hearing capabilities. The aim of this study was to investigate the effects of age on the speech-in-noise identification by speech-in-noise (SIN) test, subjective ratings of hearing difficulties by speech, spatial, and qualities of hearing scale (SSQ) questionnaire and encoding of fundamental frequency (F0) by Speech auditory brainstem response (ABR) in the elderly and comparing the results with young people. SUBJECTS AND METHODS: The present study was conducted on 32 elderly people aged over 60 years old (17 male and 15 female) with the mean age of 68.9 (standard deviation=6.33) possessing normal peripheral hearing and 32 young subjects (16 male and 16 female) aged 18-25 years old. RESULTS: Findings showed that the score of SIN test is lower among the elderly people as compared with young people in signal-to-noise ratios of 0 and -10 based on Iranian version of SSQ questionnaire (p < 0.001). The range of F0 amplitude in the elderly people is also lower than young people (p < 0.001) in Speech ABR. CONCLUSIONS: It seems that speech processing in older people is deteriorated comparing with young people regardless of their normal peripheral auditory thresholds. This decrease will result in weaker perception and improper segregation of speech from other competing sources.

ABSTRACT

Objetivou-se estudar a dinâmica dos carboidratos e a fluorescência da clorofila a em folhas de limeiras ácidas 'Tahiti', submetidas a épocas diferentes de anelamento e incisão anelar de ramos. Foram estudadas quatro épocas de realização de anelamento e incisão anelar, com um tratamento adicional, sem interrupção do floema. O anelamento, independentemente da época de realização, aumentou os teores de amido (55 a 90%) e açúcares solúveis totais (8 a 18%) nas folhas do 'Tahiti' até por volta dos 50 aos 60 dias. A incisão anelar, independentemente da época de realização, aumentou a concentração de amido (10 a 35%) em relação à testemunha, porém, o aumento foi inferior ao que ocorreu nas plantas aneladas (55 a 90%). O anelamento promoveu aumentos de F0 e redução da relação Fv/Fm, sendo os valores máximos (350) e mínimos (0,78), respectivamente, atingidos por volta dos 60 dias. Para as plantas da testemunha e incindidas, houve aumento de F0, sendo o valor máximo atingido aos 50 dias (320), porém inferior aos observados para o anelamento, e não houve alteração da relação Fv/Fm .


The aimed to study the dynamics of carbohydrates and chlorophyll fluorescence a in leaves of acid lime trees 'Tahiti' subjected to different times girdling and ringing of branches. It was studied four times the realization of girdling and ringing, with an additional treatment, without phloem interruption. The girdling, regardless of the time of realization, increased starch (55 to 90%) and soluble sugars levels (8 to 18%) in 'Tahiti' leaves until around 50 to 60 days. The ringing, regardless of achievement time, increased starch content (10 to 35%) to the control compared, but increase was less than to that which occurred in girdled plants (55 to 90%). The girdling promoted F0 increase and Fv/Fm ratio reduced, being the maximum (350) and minimum (0.78), respectively, were reached at about 60 days. For control and ringing plants, there was F0 increase, being the maximum value was achieved in 50 days (320), but lower than those observed for girdling, and there was no change in Fv/Fm ratio.

ABSTRACT

A common complaint of older listeners is that they can hear speech, yet cannot understand it, especially when listening to speech in a background noise. When target and competing speech signals are concurrently presented, a difference in the fundamental frequency (DeltaF0) between competing speech signals, which determines the pitch of voice, can be an important and commonly occurring cue to facilitate the separation of the target message from the interfering message, consequently improving intelligibility of the target message. To address the question of whether the older listeners have reduced ability to use DeltaF0 and how the age-related deficits in the processing of DeltaF0 are theoretically explained, this paper is divided into three parts. The first part of this article summarizes how the speech-communication difficulties that older listeners have are theoretically explained. In the second part, literatures on the perceptual benefits from DeltaF0 and the age-related deficits on the use of DeltaF0 are reviewed. As a final part, three theoretical models explaining the general processing of DeltaF0 are compared to discuss which better explains the age-related deficits in the processing of DeltaF0.

ABSTRACT

Objective To evaluate the effect of sustained silencing Forkhead box Ml (F0XM1) gene by short-hairpin RNA (shRNA) expression vector on cell growth of hepatocelluar carcinoma (HCC) in vitro.Methods Four shRNA expression vectors targeting different sequences of human F0XM1 mRNA were constructed.The expression vector with the best interfering effect and the negative control plasmid were used to transfect HCC cell line QGY-7703, stably transfected cell clones were selected by neomycin (G418).Cell growth was evaluated by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay and colony formation was assessed by clonogenic assay.Cell apoptosis was detected by double staining with APC conjugated Annexin V and PI.Results F0XM1 protein was detected with different levels in all these studied human cell lines.The expression vector shRNA-1026 exhibited excellent interference effect after transient transfection, which showed 38.5% and 53.2% reduction of FOXM1 mRNA and protein level respectively.The growth of QGY-7703 cells was inhibited after stable inhibition of FOXM1 expression by shRNA-1026, which was indicated by decreased absorbance value of the test group after culture for 48, 72 and 96 h compared to control group (t = 10.830,3.578 and 5.734 respectively, P < 0.05).Stable inhibition of F0XM1 also led to reduced colony formation ( t = 5.336, P < 0.05 ) and increased apoptosis of QGY-7703 cells in comparison to control cells (t = 6.827, P < 0.05 ).Conclusions Stable silencing F0XM1 gene by shRNA suppresses the growth of HCC cells in vitro.
