ABSTRACT

@#Introduction: Acts of criminal behaviour that occur have various modes and motives. Also, criminals always try to hide or eliminate evidence at the crime scene. In most cases, police or forensic experts often find DNA on items at the crime scene. One of these items is a ring, which is an item that humans often wear. Methods: This study used 24 samples of rings that had been worn for 8 hours and were incubated at room temperature. All these 24 samples then were distinguished into 4 groups, in which each group was consisting of 6 samples and incubated for 0, 1, 3, and 7-days. DNA identification was then carried out using UV spectrometer for DNA quantification and DNAzol method for DNA extraction. Results: The mean result of DNA quantification on day 0 (control) was 1020,833 ± 0.28903 ng/μL, day 1 was 546 ± 0.093569 ng/μL, day 3 was 1066.333 ± 0.117372 ng/μL, and day 7 was 1054.083 ± 0.070733 ng/μL. PCR process used STR primers with loci vWA, FGA, and TH01 and visualization used the silver nitrate method. Conclusion: The final results showed that all samples could be amplified using 3 STR loci, namely vWA, FGA, and TH01.

ABSTRACT

Fibrinogen alpha chain (FGA), a subunit of fibrinogen, might be a potential player for type 2 diabetes mellitus (T2DM), since the plasma levels of fibrinogen is known to be related to the incidence of T2DM. To elucidate the potential role of FGA in T2DM, we investigated whether FGA genetic variations are relevant in T2DM in the Korean population. Seven FGA single nucleotide polymorphisms (SNPs) were genotyped in Ansung and Ansan cohorts (474 T2DM subjects and 470 normal controls) in Korea. The association between SNPs and T2DM was determined by logistic regression analysis. Genetic relevance of SNPs to T2DM-related phenotypes was investigated by multiple linear regression analysis. Statistical analysis revealed that among seven FGA SNPs, significant associations with T2DM were observed in FGA rs2070011 (p=0.013-0.034, OR=0.72~ 0.79), rs6050 (p=0.026~0.048, OR=1.24~1.37), and rs2070022 (p=0.016~0.039, OR=0.70~0.72). Two SNPs, rs2070011 and rs6050, also showed significant association with T2DM-related phenotypes such as triglyceride (p=0.005~0.011 for rs2070011 and p=0.003~0.008 for rs6050), total cholesterol (p=0.01 for rs2070011 and p=0.024 for rs6050) and fasting glucose (p=0.035~ 0.036 for rs2070011 and p=0.048 for rs6050) in 470 normal controls. Our association study implies that FGA might be an important genetic factor in T2DM pathogenesis in the Korean population by affecting plasma lipid and glucose levels.

ABSTRACT

Research to build database of Nineplexll allelic frequency of Vietnam population applied for gene forensic purposes. The results as followed: there were 6 alleles in D3S1358; 19 alleles in FGA. In Viet (Kinh) population, polymorphism of vWA, especially of FGA was higher than in some other foreign population. For examples, the number of vWA alleles in Viet population is 9, but in Japanese and Caucasian population is 8; FGA alleles in Viet population is 19, but in Japanese is 15, and in Caucasian is 12. Two alleles of FGA are 14 and 32 were found out. There are many ethnics living in Vietnam, so the authors would like to survey allelic frequency of other ethnics.
