ABSTRACT

In recent years, the study suggests that Mori Folium has an effect of hypoglycemic, and its main active hypoglycemic effect components are alkaloids, flavonoid, polysaccharides, etc.To comprehensively understand the active components and the mechanism of Mori Folium for hypoglycemic, the active components and mechanism of Mori Folium for prevention and treatment of diabetes were reviewed.

ABSTRACT

OBJECTIVE:To virtually screen potential α-glycosidase inhibitor ingredients from C. mori and F. mori,and to pro-vide reference for finding out new typeα-glycosidase inhibitor ingredient. METHODS:Surflex-Dock module of Sybyl-x 2.0 molecu-lar simulation software was used to perform the docking of small molecule compound,which was from the ingredients of C. mori and F. mori as ligand stated in literatures,with α-glycosidase. Total score of affinity scoring function was equal to 7 as the thresh-old value,to judge potential α-glycosidase inhibitor ingredient in C. mori and F. mori. RESULTS:After 70 small molecule com-pounds docked with α-glycosidase, 10 compounds showed binding activity (Total score≥7.00). Among them, moracin M-3′-O-β-D-glucopyranoside,5,7,2′-trihydroxyflavanone-4′-O-β-D-glucoside,mulberroside A,resveratrol-4,3′-di-O-β-D-gluco-pyranoside and 1,4-dideoxy-1,4-imino-(2-O-β-D-glucopyranosyl)-D-arabinitol had higher binding activity with α-glycosidase(Total score>8.00). CONCLUSIONS:Multi-constituents of C. mori and F. Mori show potential α-glycosidase inhibitory activity. The method is a kind of highly targeted,rapid and efficient approach to discover α-glycosidase inhibitor from traditional Chinese medi-cine.

ABSTRACT

OBJECTIVE:To establish a quantitative analysis method to determine the contents of 1-deoxynojirimycin in mulberry resources such as Morus alba,Excrementum bombycis,Folium mori.METHODS:1-deoxynojirimycin in mulberry resources were determined by RP-HPLC method and UV detector on Hypersil C18(250 mm?4.6 mm,5 ?m)column, using acetonitrile-0.1% glacial acetic acid(50 ∶ 50)as moble phase. The flow rate was 1.0 mL?min-1 and the detection wavelength were set at 265 nm with column temperature at 35 ℃. RESULTS:The linear range of 1-deoxynojirimycin were from 5.0?10-4 mg?mL-1 to 1.0?10-3 mg?mL-1 (r=0.999 9)with recoveries of 97.7%(RSD=2.0%,n=6).CONCLUSION:The method is simple, accurate and reproducible for the quality control of Mulberry resources.

ABSTRACT

Objective To observe the hypoglycemic effect of total flavonoids from mulberry tree leaf (MTF) on diabetic rats and the effect of MTF on disaccharidases from rats. Methods Diabetic rats were treated with MTF, and then the change of blood glucose of these rats was observed; the brush border membrane from the small intestine of rats was stripped and homogenized, which was incubated with MTF and disaccharides, and the inhibitory rate of MTF on disaccharidases was determined. Blood samples from portal and peripheral veins were separately collected at 30, 60, 120 min after maltose solution was infused into small intestine in vivo, then the difference of blood glucose concentration between portal and peripheral veins was determined. Results MTF exerted a hypoglycemic effect on diabetic rats by inhibition of disaccharidases, the inhibitory rate of sucrase, maltase and lactase was 68.0%, 47.1%, 27.8% respectively, the difference of blood glucose concentration between portal and peripheral veins was also reduced. Conclusion The hypoglycemic effect of MTF on diabetic rats is probably via the inhibitory effect on small intestine disaccharidases in rats.
