ABSTRACT

Abstract Background: Salmonella enterica serovar Enteritidis (SE) is one of the major causes of food-borne disease worldwide, mainly associated with the consumption of poultry products, such as eggs. Several control methods have been implemented in the egg production process, but they have not effectively reduced the outbreaks. Therefore, the use of bacteriophages for the biocontrol of food-borne pathogens is gaining increasing acceptance. Objective: To evaluate a bacteriophage cocktail's effectiveness in reducing SE counts in an experimentally contaminated mayonnaise-like matrix. Methods: Homemade mayonnaise was contaminated with SE (103 CFU/ml) with equal volume to a matrix (1:1) treated with a bacteriophage cocktail (five phages, MOI 105), and stored at 21 °C for 24 and 72 h. Bacterial counts were performed to evaluate the bio-controlling activity of the cocktail and compared with a contaminated but not treated group. Results: Significant reductions (up to 3.75 log10 CFU/ml) were observed in the bacteriophage-treated groups (p<0.0001). Conclusions: These results demonstrate the effectiveness of bacteriophages as biocontrol agents for Salmonella Enteritidis in a raw-egg-derivative foodstuff. Further studies are needed to prove the reduction in an undiluted homemade mayonnaise.


Resumen Antecedentes: La Salmonella enterica, serovar Enteritidis (SE), es una de las principales causas de enfermedades transmitidas por alimentos en todo el mundo, asociadas principalmente al consumo de productos avícolas tales como los huevos. Diferentes métodos de control se han ensayado en el proceso de producción de huevos, pero no han sido capaces de reducir eficazmente los brotes de salmonelosis en las personas. Por esta razón, el uso de bacteriófagos para el control biológico de patógenos transmitidos por los alimentos está ganando cada vez más aceptación. Objetivo: Evaluar la eficacia de un cóctel de bacteriófagos para reducir los recuentos de SE en una matriz similar a la de mayonesa contaminada experimentalmente. Método: La mayonesa casera fue contaminada con SE (103 UFC/ml) en igual volumen que la matriz (1:1), tratada con una mezcla de bacteriófagos (cinco fagos, MOI 105), y almacenada a 21 °C por 24 y 72 h. Se realizaron recuentos bacterianos para evaluar la actividad biocontroladora de la mezcla y compararlos con un grupo contaminado, pero no tratado. Resultados: Se observaron reducciones significativas (hasta 3,75 log10 CFU/ml) en los grupos tratados con bacteriófagos (p<0,0001). Conclusiones: Estos resultados demuestran la efectividad del uso de bacteriófagos como agentes de biocontrol de Salmonella Enteritidis en un alimento crudo derivado del huevo. Sin embargo, se necesita realizar más estudios para comprobar la reducción en mayonesa casera no diluida.

Diferentes métodos de controle foram implementados no processo de produção de ovos, mas não foram capazes de reduzir efetivamente os surtos nas pessoas. Por esse motivo, o uso de bacteriófagos para o controle biológico de patógenos de origem alimentar está ganhando crescente aceitação. Objetivo: Avaliar a eficácia de um coquetel de bacteriófagos na redução da contagem de SE em uma matriz experimentalmente semelhante a maionese contaminada. Método: A maionese caseira foi contaminada com SE (103 UFC/ml) no mesmo volume da matriz (1:1), tratada com uma mistura de bacteriófagos (cinco fagos, MOI 105) e armazenada a 21 °C por 24 e 72 h. As contagens bacterianas foram realizadas para avaliar a atividade de biocontrole da mistura e comparadas com um grupo contaminado, mas não tratado. Resultados: Reduções significativas (até 3,75 log10 UFC/ ml) foram observadas nos grupos tratados com bacteriófagos (p<0,0001). Conclusões: Esses resultados demonstram a eficácia do uso de bacteriófagos como agentes de biocontrole de Salmonella Enteritidis em alimentos crus derivados de ovos, mas são necessários mais estudos para verificar a redução da maionese caseira não diluída.

Introducción: las enfermedades transmitidas por alimentos son una problemática mundial a la que la Organización Mundial de la Salud y los países brindan una atención especial. Dentro de los grupos de alto riesgo se encuentran los productos de origen animal, como la leche y sus derivados. Staphylococcus aureus es un microorganismo causante de brotes de las enfermedades transmitidas por alimentos, lo que genera la necesidad de estudiar la carga microbiana y establecer los controles adecuados. Objetivo: evaluar la calidad microbiológica en el proceso manufacturero en el área de producción de la Cooperativa de Producción Agropecuaria Chone LTDA, con el fin de asegurar la ausencia del Staphylococcus aureus en el queso fresco 100% chonero. Materiales y métodos: Se realizó un estudio de evaluación de la calidad en cuatro fases. Se realizaron exámenes microbiológicos iniciales, se identificaron las causas esenciales de contaminación identificadas por observación directa, revisión de documentos y entrevistas, como base para la elaboración e implementación de POE (Procedimientos Operativos Estandarizados) y POES (Procedimientos Operativos Estandarizados de Saneamiento). Se cumplieron los requerimientos éticos y se realizaron los análisis estadísticos utilizando porcentaje e índices como medida resumen y con el uso de IBM SPSS, versión libre, realizándose regresión logística binaria para determinar la probabilidad de éxito al aplicar dichos procedimientos. Resultados: los análisis microbiológicos iniciales (antes de implementación de POE y POES), dieron como resultado: 98, 116, 79, 120 y 13 UFC/g para las muestras: DiQ1, DiQ2, DiQ3, DiQ4 y DiQ5, respectivamente. Las causas esenciales de contaminación fueron falta de control microbiológico, falta de control del aspecto del personal y falta de control del agua) Una vez implementados los POE y POES, el análisis mostró ausencia en todas las muestras analizadas. Conclusiones: se concluye que la aplicación de POE y POES tiene un efecto significativo sobre la ausencia del Staphylococcus aureus, con probabilidad de éxito del 100% (AU).


Introduction: Diseases transmitted by foodstuffs (DTF) are a worldwide problem that receives special attention by WHO and different countries. High risk groups include foodstuff of animal origin, like milk and its by-products. Staphylococcus aureus is a microorganism responsible for DTF outbreaks which generates the necessity of studying its microbial burden and establishing the adequate controls. Objective: to evaluate the microbiological quality of the manufacturing process in the production area of the Farming and Stockbreeding Cooperative Chone LTDA, to assure the absence of Staphylococcus aureus in the manufacturing process of fresh cheese. Materials and methods: A study to assess the quality in four stages was carried out: initial microbiological tests, identification of the essential contamination causes by direct observation, documental review and inquires. They were used to elaborate and implement the Standardized Operational Procedures (SOP) and the Standardized Operational Procedures of Sanitation (SOPS). The ethical requirements were fulfilled and the statistical tests were performed using percentages and indexes as summary measures through IBM SPSS, free version; binary logistic regression was made to determine the success when applying those procedures. Results: initial microbiological controls (before implementing SOP and SOPS) gave these results 98, 116, 79, 120, and 13 UFC/g for samples DiQ1, DiQ2, DiQ3, DiQ4 and DiQ5 respectively. The main contamination causes were lack of microbiological control, lack of control on the staff's cleanliness and lack of control on the water. After implementing SOP and SOPS microbiological control found particularly clean samples. Conclusions: SOP and SOPS have a significant effect on the amount of Staphylococcus aureus, with 100% success probability (AU).

ABSTRACT

In recent years, an increase in the occurrence of antimicrobial resistance among Salmonella enterica has been observed in several countries, which is worrisome because S. enterica is one of the most common causes of human gastroenteritis worldwide. The aim of this study was to characterize class 1 integrons and antibiotic resistance genotypes in Salmonella enterica isolates recovered from foodstuff and related sources. Nineteen multidrug-resistant (MDR) Salmonella enterica isolates were recovered. Higher resistance rates to tetracycline (90 percent), streptomycin (80 percent), sulfamethoxazole-trimethoprim (80 percent), ampicillin (60 percent) and nalidixic acid (70 percent) were related to the presence of the tetA, aadA, sul1/sul2, blaTEM-1 genes, and a codon mutation at position 83 of the gyrA gene, respectively. Class 1 integrons harboring aadA, blaTEM-1, sul1 or dhfr1 genes were detected in nine (45 percent) Salmonella enterica strains belonging to serotypes Brandenburg, Panama, Agona, Mbandaka and Alachua. Finally, clonal dissemination of S. Panama, S. Derby and S. Mbandaka was confirmed by PFGE. Detection of clonally related MDR Salmonella enterica suggests that endemic serotypes can be supported by class 1 integron-borne gene cassettes and/or mutations in drug targets. Emergence and dissemination of multidrug-resistant Salmonella enterica can have a major public health impact in an environment where large-scale suppliers ship their products.

ABSTRACT

The purpose of this study was to investigate the sanitation management of school foodstuffs and the sanitation knowledge of delivery company workers. A questionnaire that identified employee's food safety sanitation management, knowledge, and practices was developed based on a review of the literature. The subjects consisted of 201 delivery company workers from 38 delivery companies in the Daegu and Gyeongbuk area. The workers were 30-39 years of age (41.3%) with 1-3 years of working experience (30.8%). Approximately 62% of the respondents were educated and had 1-2 years of food safety experience, 52.7% of the respondents delivered foodstuffs to two schools in the morning. The total score for delivery company worker sanitation knowledge was 3.75/7.00, which was low. The total mean sanitary performance score for factory workers was 4.58/5.00. They indicated that the most difficult part of the operation is delivering on time. Temperature management was another difficulty. In order to secure the best quality of school food to be supplied safely, the thorough sanitation education must be conducted to the school dieticians and delivery company managers.

ABSTRACT

Objective To study the effect of applying food composition table and foodstuff substitution method on nutritional status in patients with diabetic nephropathy and undergoing peritoneal dialysis.Methods Forty patients with diabetic nephropathy,treated with peritoneal dialysis for over three months was included in this study.Under the guidance of the food composition table and foodstuff substitution method for six months,nutritional status was compared before and after treatment. Results There was a significant difference in HGS,triceps skin foId thickness(TSF),mid-arm circumference(MAC),midarm muscle circumference(MAMC),Hb,albumin(Alb),serum crcatinine after 6 mortths of intervention compared with those before intervention. Conclusions Food composition table and foodstuff substitution method can obviously impwve the nutritional status for patients with diabetic nephwpathy undergoing peritoneal dialysis and increase the patients'diet compliance as well.

ABSTRACT

Background: Foodstuff safety hygiene plays a vital role in the health of humans. Therefore, studying foodstuff production and finding suitable methods to control foodstuff contamination is an urgent issue. Objectives: (1) To evaluate various factors of foodstuff contamination in food services and households in Hoan Kiem and Tay Ho districts of Ha Noi. (2) To discover the knowledge, attitude and practices of foodstuff hygiene of the people who process food. Subject and method: A descriptive epidemiology study that included 34 restaurants, 31 kindergartens and 72 households in the 2 districts of Hanoi. Results: The environmental hygiene in foodstuffs service spots and in the families does not fully satisfy the requirements of foodstuffs safety hygiene. Knowledge, attitude and practice of foodstuffs hygiene of the people who process food in Hoan Kiem and Tay Ho districts are not good. Conclusion: It is very necessary to strengthen the knowledge for co-operating practice guidance of basic foodstuffs safety hygiene for people. Food services, kindergartens, etc\u2026 must have training certificate of foodstuff safety hygiene.

ABSTRACT

A simple and rapid method was described for determining nitrogen content of a Kjeldahl digest by pulse polarography. The amount of sample needed was as small as 200mg. The digest was diluted and pipetted into a solution of formaldehyde in an acetate buffer. The hexamethylenetetramine produced by reaction between ammonia and formaldehyde was determined electrochemically at -0.9v.This method proved itself a better reproducibility. The recovery rate of nitrogen was 94.5% to 103.1%. Calibration curve of nitrogen in the range of 20 to 120 ?g/ml showed linear relation.Human milk and eleven different food-stuffs were analysed. There was no significant difference between the results by this method and Kjeldahl method.
