ABSTRACT

Objective To standardize the modeling and evaluation of type 2 diabetic gastric mucosal injury rat model of spleen deficiency. Methods Male SD rats were randomly divided into normal group and model group, type 2 diabetic rat model with gastric mucosal injury was established by the injection of small-dose streptozotocin (STZ) combined with feeding of high fat and sugar forage and gastric gavage of 30% ethanol solution. After the modeling, we examined the levels of fasting blood glucose ( FBG) , fasting insulin ( FINS) , superoxide dismutase (SOD), malondialdehyde (MDA) and nitric oxide (NO) in the rat serum, and gastric and pancreatic tissue pathology was evaluated after HE staining. Modern medical evaluation method was used for the statistics of information collected by four diagnostic methods of traditional Chinese medicine ( TCM) , and for the identification of properties and evolution of TCM syndromes. Results Type 2 diabetic gastric mucosal injury rat model had been established in 75% of the rats. And 10-16 weeks after the modeling, the model rats had the syndrome of spleen deficiency, showing as persistent emaciation, decreased grasping force, weak activities in open field test, pale tongue nature, decreased r value of the claw and tail, increased defecation frequency, softening of loose stool. Of all the model rats, 78% had the features of diabetes, one indicator of qi deficiency syndrome, and one or more specialized spleen-related indicators. Conclusion These results suggest that type 2 diabetic gastric mucosal injury rat model can be successfully induced by the injection of small -dose streptozotocin ( STZ) combined with feeding of high fat and sugar forage and gastric gavage of 30% ethanol solution, and the time window of spleen deficiency for type 2 diabetic gastric mucosal injury rat model is 10-16 weeks after the modeling.

ABSTRACT

Objective To observe the effect of Weipixiao, a compound recipe which has the actions of strengthening spleen, resolving stasis and removing toxins, on the histopathological changes of gastric mucosal tissue in rats with gastric precancerous lesions ( GPL) . Methods SD rats were randomly divided into normal group, model group, Vitacoenzyme group (0.2 g·kg-1·d-1), and high-, middle-, and low-dose Weipixiao groups ( in the dose of 15, 7.5, 3.75 g·kg-1·d-1, respectively) . Except for the normal control group, the rats in other groups received spontaneous intake of N-methyl-N’-nitro-nitrosoguanidine ( MNNG) solution combined with irregular diet and oral use of purgative herbs for 18 weeks to induce GPL. From the 9th week, the mediation groups were simultaneously given corresponding medicine for 10 weeks. At the end of the experiment, the histopathological changes of gastric mucosal tissue in all groups were observed. Results Pathological scores of intestinal metaplasia and epithelial dysplasia in rat gastric mucosa of the model group were significantly increased ( P<0.01 compared with those of the normal group) , but were decreased in three Weipixiao groups to various degrees, particularly in low-dose Weipixiao group ( P<0.05 or P<0.01) . Conclusion Weipixiao can block and reverse gastric intestinal metaplasia and dysplasia in GPL rats to certain degrees, and low-dose Weipixiao may have better long-term effect for the prevention and treatment of GPL.

ABSTRACT

【Objective】To observe the effect of Gastritis Prescription Ⅰ(GPⅠ) on the ultrastructure of gastric mucosa in spleen-deficiency atrophic gastritis(AG) patients.【Methods】Thirty chronic AG patients were treated with GPⅠ(mainly composed of Radix Codonopsis,Poria,Rhizoma Atractylodis Macrocephalae,Pericarpium Citri Reticulatae,Rhizoma Pinelliae,Fructus Amomi,Radix Angelicae Sinensis,Herba Taraxaci,Radix Paeoniae Alba,Radix Glycyrrhizae Preparata,etc.).The histological features of gastric mucosa sampled under gastroscope were observed before and after treatment under light microscope and electron microscope.【Results】After treatment,43.3% of moderate-degree and above atrophic area of gastric glands disappeared,and 16.7% became mild;36.7% of the disappeared gastric groove recurred and the thin gastric mucosa became normal.Microvilli of gastric gland became normal in 12 patients,accounting 54.5% in 22 patients with intestinal metaplasia.【Conclusion】GPⅠis effective in recovering the gastric mucosa of AG patients with deficiency of spleen and stomach Qi,and has an effect in counteracting intestinal metaplasia.

ABSTRACT

[Objective] To investigate the pathological features, secretion of interleukin-8 (IL - 8), tumor necrosis factor alpha (TNF-?) and secretory immunoglobulin A (sIgA) and helicobacter pylori (HP) infection in gastric mucosa (GM) of chronic superficial gastritis with spleen-stomach and damp-heat syndrome (SSDHS) and to explore their correlations. [Methods] Fifty-five cases of chronic superficial gastritis included 35 cases of SSDHS (Group A) and 20 cases of spleen-deficiency syndrome (Group B). Fifteen asymptomatic volunteers served as normal control group (Group C). Pathological changes, HP infection rate, levels of IL - 8, TNF -? and sIgA in GM were observed. [Results] HP infection rate was higher and inflammatory change was more obvious in Group A than those in Group B and Group C (P 0.05). [Conclusion] Gastric mucosa of chronic superficial gastritis with SSDHS presents inflammatory changes, which is related with the increase of IL - 8 and TNF -? secretion and the decrease of sIgA, and HP infection may be one of the factors inducing and enhancing the pathological changes.

ABSTRACT

One hundred forty cases were treated with Fuwei decoction, with a control group treated by Weimeisu and Sanjiuweitai. The effective rates were 93. 3% and 74. 3% respectively. Pathological exams showed that, in the treatment group the afective rates of atrophic gastric mucosa enterization and heterohyperlasia were 88. 6%, 86. 6% and control group were 72. 4% and 19. 4% respectively. with significant difference between all groups, demonstrating the definite efficacy of Fuweitang on these ailments.

ABSTRACT

Healthy male Wistar rats were prepared as model of precancerous lesion of gastric mucosa. The tongue, pH of tongue surface, dropped cells from tongue fur, histology of tongue. and corresponding gastric mucosa, pH of gastric juice, mucosal histological changes in both groups were observed. Results revealed that, in the rats with precancerous lesion, the tongues were bare or only with little fur, dark purplish tongue with petechia. pH of both tongue and gastric juice were high, which were both coincided with the syndrome type of asthenic gastric - qi and blockage of collaterals by stagnant blood.
