ABSTRACT

OB JECTIVE To study the protective effects of saponins from Gleditsia sinensis on ischemic stroke with phlegm and blood stasis (ISPBS)model rats ,and to explore its mechanism. METHODS Totally 119 rats were randomly divided into normal group (normal saline ),sham operation group (normal saline ),model group (normal saline ),nimodipine group (positive control group ,5 mg/kg),G. sinensis saponin low-dose ,medium-dose and high-dose groups (3.21,6.42 and 12.84 mg/kg),with 17 rats in each group. Except for normal group ,other groups were all given high-fat diet+suture-occluded method to induce ISPBS model. The neurological function score ,water content of brain tissue ,pathological morphology of brain tissue ,the changes of hemorheology indexes (whole blood viscosity , erythrocyte aggregation index , Casson-viscosity), four items of blood lipid [triacylglycerol (TG),total cholesterol (TC),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol(HDL-C)] and inflammatory factors in serum and oxidative stress indexes [malondialdehyde (MDA),nitric oxide (NO),superoxide dismutase (SOD)] in brain tissue were determined or observed in rats. The protein expressions of B lymphocytoma 2(Bcl-2),Bcl-2 associated X protein (Bax)and caspase- 3 in cerebral tissue were also detected. RESULTS Compared with normal group ,the score of nerve function ,5 kinds of serum indexes (TC,TG,LDL-C,TNF-α,IL-1β), hemorheology indexes ,the contents of MDA and NO and protein expressions of Bax and caspase- 3 in cerebral tissue were all increased significantly in model group (P<0.01). The levels of HDL-C and IL- 10 in serum ,SOD activity and protein expression of Bcl- 2 in cerebral tissue were decreased significantly (P<0.01),and obvious lesions such as nuclear pyknosis and cell membrane fragmentation occurred in brain tissue. Compared with model group ,above indexes of administration groups were improved to different extents ,among which there was statistical significance in above indexes of G. sinensis saponin high-dose group (P< 0.01). CONCLUSIONS Saponin from G. sinensis has a good protective effect on ISPBS model rats. Its mechanism may beassociated with reducing oxidative damage , reducing the production of pro-inflammatory mediators and resisting neuronal apoptosis.

ABSTRACT

OBJECTIVE: To discriminate and quantify of Gleditsia japonica Miq. thorn (SZJ) and Gleditsia microphylla Gordon ex Y. T. Lee thorn (YZJ) in the Gleditsia sinensis Lam thorn (GST). METHODS: Fourier transform near-infrared spectroscopy (FT-NIR) combined with linear discriminate analysis (LDA), support vector machine (SVM), as while as back propagation neural network (BPNN) algorithms were applied to construct the identification models. The SZJ and YZJ content in adulterated GST were determined by partial least squares regression (PLSR). RESULTS: The SVM models performance best compared with LDA and BP-NN models for it could reach 100% accuracy in training and validation set for identifying authentic GST and GST adulterated with SZJ and YZJ based on the spectral region of 5 000-4 200 cm-1 combined with SG+VN processing. The rp, RMSEP (the root mean standard error of prediction) and bias for the prediction by PLS regression model were 0.993, 2.919% and -0.330 3 for SZJ, 0.995, 2.57% and 0.364 9 for YZJ, respectively. CONCLUSION: Our results suggest that the combination of NIR spectroscopy and chemometric methods offers a simple, fast and reliable method for classifification and quantifification of SZJ and YZJ adulterants in the GST.

ABSTRACT

Objective: To investigate the active ingredients and molecular mechanism of Astragalus membranaceus var. mongholicus, Angelica sinensis, Angelicae dahurica, and Gleditsia sinensis in Tuoli Xiaodu Powder in promoting of diabetic wound healing. Methods: UPLC-Q-TOF/MS in positive and negative ion modes was applied to analyze the components in the ethanol extract from Tuoli Xiaodu Powder. Molecular docking technology was used to predict the targets proteins of these components. The function and pathway annotations of target proteins were performed through relevant databases such as Uniprot and KEGG. The drug components-target-function diagram was constructed using Cytoscape software. Results: Twenty-eight compounds containing flavonoids, saponins, coumarins, alkaloids, and triterpenoids were identified in positive and negative ion modes. Among these compounds, 17 compounds could interact with 17 target proteins, and there were 210 pairs of component-target relationships by analyzing the results of molecular docking. Among them, five targets were related to immune regulation, six targets were related to antibacterial and anti-inflammatory effects, six targets were related to cell differentiation, 10 targets were related to cell migration, six targets were related to angiogenesis, two targets were related to stimulation of epithelial growth factor, six targets were related to vasodilation, and two targets were related to estrogen. Conclusion: The flavonoids, saponins, coumarins, steroids, and triterpenoids contained in the simplified formula possess many biological effects such as antibacterial, anti-inflammatory, immune regulation, and angiogenesis. These functions may be related to its modulation of NF-κB, PI3K/Akt/eNOS, and MAPK pathway through regulating NF-κB, MAPK, PI3K, and ERK2 targets.

ABSTRACT

Objective To investigate the chemical constituents from the ethyl acetate extract of Gleditsiae Spina. Methods The chemical constituents were isolated and identified by chromatography on silica gel, Toyopearl HW-40C, Sephadex LH-20, MCI Gel CHP-20, ODS, and RP-HPLC. Their structures were elucidated on the basis of physicochemical properties and spectral analyses. Results Eighteen compounds were isolated from the ethyl acetate extract of Gleditsiae Spina, and identified as scopoletin (1), (-)-(7R,8S)-erythro-guaiacylglycerol (2), 5-methoxy-guaiacylglycerol (3), (-)-(7R,8R)-threo-guaiacylglycerol (4), 3,4’- dihydroxypropiophenone (5), dihydroferulic acid (6), p-hydroxybenzoic acid (7), indole-3-carbaldehyde (8), protocatechuic acid methyl (9), protocatechuic aldehyde (10), syringic acid (11), 2-guaiacylpropane-1,3-diol (12), (2R*,3R*,4S*)-2,3-diguaiacyl-4- hydroxyl tetrahydrofuran (13), 3-(2-oxopropyl)-3-hydroxy-indolin-2-one (14), juglanin D (15), C-veratroylglycol (16), 3-(4-hydroxyl-3-methoxyphenyl)-propan-1,2-diol (17), and (-)-syringaresinol (18). Conclusion Eighteen compounds are isolated from this plant for the first time.

ABSTRACT

To study the breeding system and pollination characteristics of Gleditsia sinensis, we observed the development of flower development and the processing of pollination, and determined the pollen viability and stigma acceptability by TTC and benzidine-hydrogen peroxide method and detected its breeding system using OCI value, P/O ratio and artificial pollination.The results showed that: ①G. sinensis are racemes, divided into bisexual inflorescences (only a small amount of inactive pollen) and male inflorescences (occasionally a few bisexual flowers), flowers hermaphrodite. ②Male flowers had the strongest pollen activity 4 h after flowering; the stigma receptivity of bisexual was the highest at 1 h after flowering, and pollination was the best in this time. ③The pollen tube had a few elongation when the bisexual flower is half-opened. The number of pollen tube and length significantly increased when blooming. The flower reaches the ovary and even enters the ovule to complete the fertilization. ④When the OCI=4 and P/O=11 684, it means that the breeding system was facultative, outcrossing, and requiring pollinators based on the results of the bagging experiment.There was parthenogenesis. ⑤The characteristics of saponin pollination were wind pollination and insect vector pollination, and pollinators were initially identified as Apis mellifera ligustica. All these results provides a theoretical and technical foundation for the new germplasm of G. sinensis.

ABSTRACT

To study morphological characteristics change of the development process and quercetin and polyphenols of accumulation dynamic of the spines of Gleditsia sinensis, measure and compare morphological indexes using Vernier caliper, ruler and balance, calibrate and analysis quercetin and polyphenols content using HPLC and colorimetric method. The spines of G. sinensis development process is divided into formation period (the beginning of August to the beginning of November), dormant period (the beginning of November to the end of March in the following year), germination period (the end of March to the middle of April), fast growth period (the middle of April to the middle of August), browning period (the middle of August to the end of August) and mature period (the beginning of September to the end of December). Formation period the spines primordium divides and forms the scale bubs; dormant period the scale bubs are in a dormant state; germination period the bubs scales fall off, spines primordium began to development; fast growth period rapid growth to maximum; browning period browning from the tip and browns until the whole becomes brown; mature period The early stage of maturity is full of luster, gradually the color deepened and the luster faded. The accumulation of quercetin was gradually decreasing after increasing. The total polyphenol accumulation was significantly decreased and then gradually increased, decreased finally. The content of quercetin was increased from 0.000 4%-0.002 6%, and the polyphenol content decreased from 0.761 9%-0.049 1% and then slowly increased to 0.286 9% in the fast growth period.The quercetin continuous increase to 0.004 3% and total polyphenol increased to 0.421 6% in the browning period. In the mature period, the quercetin content significantly decreased after reaching 0.009 6% in September, and the polyphenols content decreased after reaching 0.723 5% in October. Using principal component analysis results: September first, October 2nd, November 3rd. The morphological characteristics change of the development process and quercetin and polyphenols accumulation were determined. The development process is divided into six periods, the best harvest time is the early stage of mature period. Provide theoretical support for the utilization of the spines of G. sinensis and cultivation techniques of high yield.

BACKGROUND: Metronidazole has been known as the most effective drug for treatment of Trichomonas vaginalis-related diseases. However, it has been reported that metronidazole has adverse effects and incidence of metronidazole-resistant T. vaginalis (CDC085) has increased. Development of new drug, which is effective against metronidazole-resistant T. vaginalis and showing no adverse effects, has been required. METHODS: The purpose of this study was to investigate effects of various extracts from herbs such as Quisqualis indica, Gleditsia sinensis, Prunus armeniaca, Morus alba, Platycodon grandiflorum, Ailanthus altissima, Stemona japonica, Biota orientalis, Dryobalanops aromatica, and Cimicifuga heracleifolia on metronidazole resistant strain of T. vaginalis in vitro (CDC085). RESULTS: Anti-Trichomonas activities were observed in T. vaginalis treated with G. sinensis, P. armeniaca, and P. grandiflorum on the growth and fine structure of metronidazole resistant strain of T. vaginalis. Of the three standard extracts that showed the most effective anti-trichomonas activity, G. sinensis was the most effective. The inhibitory effects of fraction extracts of this drug were shown on the growth of T. vaginalis. The fine structure of the cytoplasm was changed after application of G. sinensis extract. The number of polyribosome and hydrogenosome decreased whereas the number of food vacuole and vacuole in the cytoplasm increased, compared with that of untreated control group. CONCLUSION: The results of our study indicate that G. sinensis may induce the inhibition of cell multiplication as well as impairment of protein synthesis of metronidazole resistant strain of T. vaginalis in vitro.

Object To study on the structure of sapogenins from pod of Gleditsia sinensis Lam Methods Pod of G. sinensis was extracted with solvents and the constituents in the extraction isolated, after hydrolised completely, with the aid of flash chromatography to obtain 2 gledgenins Results The structures of the 2 gledgenins were proved to be 3 hydroxy 12 oleanen 28 oic acid and 3, 16 dihydroxy 12 oleanen 28 oic acid respectively as shown by IR, NMR, MS and crystallographic data The structures were further verified by acylation, methylation and followed by reaction with 3, 5 dinitro benzoyl chloride Conclusion New crystalline derivatives were obtained to provide further evidence of their structures
