ABSTRACT

Resumen La candidemia es la micosis invasora más frecuente en los pacientes internados en hospitales de alta complejidad en el mundo. La infección fúngica en el sistema nervioso central constituye una complicación potencialmente mortal que agrava el pronóstico de los pacientes. El presente artículo aborda aspectos relevantes sobre las características clínicas de esta enfermedad, los mecanismos de invasión del hongo, la respuesta inmunitaria local frente a Candida albicans y el impacto de los defectos genéticos en receptores de la inmunidad innata, que aumentan la susceptibilidad a la neurocandidiasis.

ABSTRACT

This study aimed to evaluate the performance, apparent digestibility of diet, ingestive behavior which occurred in two moments, carcass traits, being evaluated constituent and non-carcass components, and also the effect the yeast culture could promote in the peripheral temperature of rumen, hull and body temperature. The diets consisted of a constant ratio of 50% forage (maize silage) and 50% concentrate. Thirty-six steers, ½ Angus Nelore, with average age of 11 months and average initial body weight of 339.5±10kg were used in the experiment. The inclusion of yeast culture promoted a higher daily dry matter intake (8.83 vs 9.35kg day-1) and, consequently, a better daily weight gain (1,143 vs. 1,325kg day-1) in the initial feedlot phase, with no difference in other periods. The apparent digestibility of the diet containing yeast culture was higher than the control diet (69.69 versus 68.32%, respectively), and its use did not interfere with the feeding behavior of the animals. Based on our findings, supplementation with yeast culture may bring positive results in the initial feedlot phase.(AU)


O objetivo deste estudo foi avaliar o desempenho; a digestibilidade aparente da dieta; o comportamento oral ingestivo, o qual ocorreu em dois momentos, as características de carcaça, sendo avaliados componentes integrantes e não integrantes da carcaça; bem como o efeito que a cultura de leveduras pudesse promover perante a temperatura periférica de rúmen, casco e temperatura corpórea, sendo aferida por meio da temperatura retal. As dietas foram constituídas em uma constante relação de 50% de volumoso (silagem de milho) e 50% de concentrado. Utilizaram-se no experimento 36 novilhos, ½ sangue Angus Nelore, com idade média de 11 meses e peso vivo médio inicial de 339,5 ± 10kg. O uso de cultura de leveduras promoveu maior consumo diário de matéria seca (8,83 contra 9,35 kg dia-1) e consequentemente melhor ganho de peso diário (1,143 contra 1,325kg dia-1) na fase inicial do confinamento, não havendo diferença nos demais períodos. A digestibilidade aparente da dieta que continha cultura de leveduras foi superior à da dieta controle (69,69 contra 68,32%, respectivamente), e seu uso não interferiu no comportamento ingestivo dos animais. Com base nos resultados do presente estudo, a suplementação com cultura de leveduras pode trazer resultados positivos na fase inicial de confinamento.(AU)

ABSTRACT

ABSTRACT Bioactive compounds are substances present in low doses in foods mostly from the plant kingdom. Their consumption can influence physiological or cellular processes and has a beneficial effect on health. Fucoidans and beta-glucans are bioactive compounds that share the characteristic of being carbohydrates of the polysaccharide type, specifically of the soluble fiber kind. These compounds can be found in foods available in Chile and part of the Chilean diet; foods include, brown algae and some cereals. The concentration of these compounds in foods is variable, and depends on factors like season, cultivation, germination, and method of preparation and conservation. The current literature shows the potential beneficial effects of fucoidan and beta-glucans compounds in human health, which include: anticoagulant, immuno-modulator and antidiabetic and immunomodulating. The effects depend, among other factors, on their bioavailability and molecular weight. The objective of this review was to describe the potential beneficial effects of these bioactive compounds, analyze their characteristics and properties, and provide consumption recommendations that may lead to achieving the expected beneficial effects. To that end, relevant and recent scientific literature was analyzed.


RESUMEN Los compuestos bioactivos corresponden a aquellos elementos presentes en los alimentos, que influyen en procesos fisiológicos o celulares, cuyo consumo produce un efecto beneficioso para la salud, encontrándose en alimentos principalmente del reino vegetal y cuyas concentraciones en los alimentos son en mínimas dosis. Los fucoidanos y los beta-glucanos, son compuestos bioactivos que comparten la característica de ser un carbohidrato, de tipo polisacárido, específicamente fibra de tipo soluble, encontrándose presente en alimentos disponibles en nuestro país y que son parte de nuestra dieta, como las algas pardas y algunos cereales. La concentración de estos compuestos en los alimentos, es fluctuante y depende de factores como la estacionalidad, cultivo, germinación, método de preparación y conservación. En la bibliografía actual, se evidencian los potenciales efectos beneficiosos de fucanos y beta-glucanos para la salud humana como anticoagulante, antidiabético e inmunomodulador, la intensidad de estos efectos dependen de su biodisponibilidad y del peso molecular. El objetivo de esta revisión fué profundizar respecto a los potenciales efectos beneficiosos de estos compuestos bioactivos, analizar sus características, propiedades, además de relacionar una recomendación de consumo que permita alcanzar los efectos beneficiosos esperados. Para ello se analizó la bibliografía científica atingente, relevante y reciente.

Resumen Introducción. Se calcula que más de 300 millones de personas alrededor del mundo padecen asma y se estima que para el año 2025 esta cifra se incremente a 400 millones debido a los contaminantes criterio. Sin embargo, dadas sus limitaciones, los estudios epidemiológicos son controversiales sobre la contaminación y el desarrollo de asma. Objetivos. Describir las diferencias y similitudes de la respuesta inmunológica de pacientes asmáticos y los modelos animales de asma alérgica después de la exposición a contaminantes criterio y elementos biológicos, para así identificar los factores inmunológicos relacionados con el desarrollo de asma. Materiales y método. Se realizó una búsqueda sistemática en las bases de datos sobre asma y los diferentes contaminantes criterio. Resultados. La respuesta Th2 es activada por la inhalación de ozono, dióxido de nitrógeno, azufre y la exposición aguda a material particulado, mientras que el contacto con ciertos tipos de pólenes y glucanos y la exposición crónica de partículas incrementa la respuesta Th1, la cual inhibe a la respuesta Th2 produciendo un "efecto protector". Conclusiones. La respuesta Th1 podría causar baja o nula asociación entre la exposición a contaminación y el desarrollo de asma en las diferentes ciudades, adicionando de esta manera otra limitación a los estudios epidemiológicos.


Abstract Introduction: More than 300 million people around the world suffer from asthma, and estimations indicate that this figure will increase to 400 million by 2025 due to criteria pollutants. However, given their limitations, epidemiological studies on pollution and its role in the development of asthma are controversial. Objectives: To describe the differences and similarities of the immunological response of asthmatic patients and animal models to allergic asthma after exposure to criteria pollutants and biological elements, in order to identify the immunological factors related to the development of asthma. Materials and methods: A systematic search was conducted in asthma databases and criteria pollutants. Results: The Th2 response is activated by the inhalation of ozone, nitrogen dioxide, sulfur and acute exposure to particulate matter. On the other hand, contact with certain types of pollens and glucans and the chronic exposure of particles increases the Th1 response, which inhibits Th2 response producing a "protective effect". Conclusions: Th1 response could cause low or no association between exposure to pollution and the development of asthma in different cities, which constitutes another limitation in epidemiological studies.

ABSTRACT

ß-glucans are glucose polymers (polysaccharides) of high molecular weight naturally present in the cell wall of various living organisms such as bacteria, yeasts, fungus and plants (mainly cereals like barley and oat). ß-glucans are GRAS and are currently used as texturing agents by the food industry. Furthermore, scientific evidence suggest that, depending on their physicochemical structure and source, their intake would be associated with health promoting effect such as the decrease of plasma cholesterol and the improvement of the glycemic index of the ß-glucan-containing foodstuffs. Accordingly, a health claim related with the intake of cereal ß-glucans and plasma cholesterol improvement has been accepted by the FDA in U.S.A. and by the EFSA in the E.U. For this reason, ß-glucans are increasingly used in the world for the development of functional foods. However, the effect of yeast ß-glucans on immune system stimulation is still discussed.


Los ß-glucanos son polímeros de glucosa (polisacáridos) de elevado peso molecular que se encuentran en forma natural en la pared celular de diversos organismos vivos como bacterias, levaduras, hongos y plantas (cereales como avena y cebada). Los ß-glucanos son inocuos y actualmente son utilizados por las empresas de alimentos como agentes texturizantes. Además, antecedentes científicos sugieren que, dependiendo de su estructura fisicoquímica y de su origen, su consumo se asociaría a efectos beneficiosos para la salud del ser humano como la disminución de la concentración plasmática de colesterol total y la reducción del índice glicémico de los alimentos que lo incluyen. Un mensaje de salud sobre consumo de ß-glucanos de cereales y disminución del colesterol ha sido aceptado tanto por la FDA de EE.UU. como por la EFSA de la U.E. Por esta razón los ß-glucanos están siendo utilizados cada vez más para el desarrollo de alimentos funcionales en varios países del mundo. Sin embargo, el efecto de los ß-glucanos de levadura sobre la estimulación de la respuesta inmune aún está en discusión.

ABSTRACT

Objetive. This study was conducted to evaluate, by means of lectinhistochemistry (LHC), the expression of carbohydrates in granulomas induced by the bacillus Calmette-Guerin (BCG) in muscle tissue of Piaractus mesopotamicus after 33 days. Material and methods. Histological sections with 3 μm thick were incubated with the following lectins :WGA (Wheat germ agglutinin), DBA (Dolichos biflorus agglutinin) and HPA (Helix pomatia agglutinin), and the results were evaluated by light microscopy. Results. Acid fast bacilli were stained by Ziehl Neelsen (ZN) and strong labeled by WGA in the cytoplasm of macrophages. Labeling with DBA was intense in fibroblasts and weak in macrophages. On the other hand, HPA binding was stronger in macrophages, especially in those that were in close contact with epithelioid cells, without evidence of binding to fibroblasts. The epithelioid cells were not labeled by the used lectins, but they were identified by Hematoxilin-Eosin (HE). The lectins labeled specific type saccharides in glycoproteins, as N-acetylglucosamine present in bacilli and macrophages, as well as N-acetyl-galactosamine in macrophages. The control group showed no inflammation or lectin binding. Conclusions. This technique may be useful in identifying receptors for WGA, DBA and the HPA lectins in epithelioid granuloma induced by BCG in P. mesopotamicus.


El presente estudio fue realizado para evaluar por medio de lectinhistoquímica (LHC), la expresión de carbohidratos en granulomas inducidos por el bacilo de Calmette-Guérin (BCG) en músculo de Piaractus mesopotamicus después de 33 días. Materiales y métodos. Cortes histológicos de 3 µm de grosor fueron incubados con las siguientes lectinas: WGA (Wheat germ aglutinin), DBA (Dolichos biflorus agglutin) y HPA (Helix pomatia agglutinin), y los resultados evaluados por medio de microscopia de luz. Resultados. Bacilos ácido resistentes fueron identificados por la tinción de Ziehl Neelsen(ZN). Se observó un marcaje intenso con WGA en el citoplasma de macrófagos. El marcaje con DBA fue intenso en fibroblastos y débil en macrófagos. Con la lectina HPA el marcaje fue intenso en macrófagos, principalmente en los que estaban en estrecho contacto con las células epitelióides, externamente se observó marcaje débil en fibroblastos. Las células epitelióides no fueron marcadas por las lectinas, pero fueron identificadas con la tinción de Hematoxilina-Eosina (HE). Las lectinas tuvieron un tipo de marcaje específico en algunos monosacáridos, como N-acetilglucosamina presente en los bacilos y en macrófagos, y N-acetilgalactosamina en macrófagos. En el grupo control no fue observada inflamación así como tampoco marcaje con las lectinas. Conclusiones. Esta técnica resultó eficiente en la identificación de receptores para las lectinas WGA, DBA y HPA en el granuloma epitelióide inducido por BCG en P. mesopotamicus.

O estudo buscou avaliar as respostas fisiológicas e de desempenho à adição de diferentes níveis de β-glucanos na dieta de leitões recém-desmamados. Foram utilizados 30 machos de linhagem comercial, com 34 dias de idade e peso de 10,9±0,63kg, alojados em gaiolas individuais, durante 14 dias. Os tratamentos variaram somente nos níveis de β-glucanos que foram de 35, 70, 140 e 280g/T. No final desse período, em metade dos animais foram injetados 2,7mg de lipopolissacarídeo (LPS) em 1mL de solução (PBS)/leitão, e nos demais 1mL/leitão de PBS. Foram avaliadas temperatura retal (TR), frequência respiratória (FR), perfil bioquímico sanguíneo e sinais clínicos aos 30, 90, 120 e 390 minutos seguintes à injeção. As respostas de desempenho não foram afetadas pelos níveis de β-glucanos. Foi observado o aumento de TR, tanto em função do LPS como em função da hora em que a temperatura foi medida. No entanto, os animais LPS-injetados, que receberam 280g/T de β-glucanos, tiveram a mesma TR daqueles LPS-não injetados. Os sinais clínicos foram compatíveis com um quadro de inflamação aguda e foram observados, nos animais LPS-injetados, vômito, prostração e diarreia, sendo que a inclusão de β-glucanos não conseguiu reverter esses sintomas. O uso de β-glucanos por 14 dias na dieta de leitões recém-desmamados mostrou um efeito anti-inflamatório em situação de desafio agudo. Para reverter o desafio imunológico utilizado, o nível de 280g/T de β-glucanos mostrou-se o mais recomendado.

Thirty males from a commercial line with 34 days of age and 10.9±0,6kg initial weight were housed in individual metabolic cages during 14 days. The treatments only differed in β-glucans levels: 35; 70; 140 and 280g/T. On the 14th day, half of the pigs were inoculated with 2.7mg LPS (lipopolysaccharide)/mL of solution (PBS)/pig and the other half received 1mL PBS/pig. Rectal temperature (RT), respiratory frequency (RF), biochemical profile and animal behavior (120 and 390 minutes after LPS inoculation) were evaluated. As the experiment was conducted, performance was not affected by β-glucans. An RT increase was observed due to LPS and time of temperature measurement. LPS-injected animals receiving 280g/T of β-glucans had the same RT as the LPS- non injected group. Clinical signs were near the expectation for acute inflammation. LPS-injected presented prostration, diarrhea and vomit and the β-glucans did not reverse this situation. The use of β-glucans in weaning pigs for fourteen days showed an anti-inflammatory action. To revert the immunologic challenge, the level of 280g/T of β-glucans was the most recommended.

ABSTRACT

La caries dental es una de las enfermedades más frecuentes en el ser humano. En su etiología multifactorial, desempeñan un papel importante determinadas bacterias cariogénicas, que en interacción con la superficie del diente promueven su desmineralización. Dentro de los mecanismos mediadores de la adhesión bacteriana, se encuentra la producción de polisacáridos extracelulares bacterianos. En particular los glucanos sintetizados por las glucosiltransferasas, no solo permiten la adherencia, sino que también constituyen una fuente nutricional para las bacterias, por lo tanto, la actividad de dichas enzimas se considera un factor de virulencia bacteriana en la caries dental. Esta revisión bibliográfica tiene el objetivo de esclarecer los aspectos relacionados con la estructura, biosíntesis y función de los glucanos, y enfatizar en la aplicación de estos conocimientos en la prevención de la caries dental(AU)


Dental caries is one of the most common diseases in the human being. Certain cariogenic bacteria play an important role in its multifactorial etiology, since in their interaction with the dental surface they promote its demineralization. The production of extracellular bacterial polyssacharides is among the mechanisms mediating bacterial adhesion. The glucans synthesized by glycosyltransferases not only allow the adherence, but they also are a nutritional source for bacteria and that's why the activity of such enzymes is considered a factor of bacterial virulence in dental caries. This bibliographic review is aimed at making clear the aspects related to the structure, biosynthesis and function of glucans and at giving emphasis to the application of this knowledge in the prevention of dental caries(AU)
