ABSTRACT

Cassia fistula Linn, a paramount medicinal plant consumed by huge population all over the world. Its bioactive metabolites include flavonoids, phenolics, and polyphenols. Major objective of current study is to find right media combination and age of callus for maximum accumulation of free flavonoids in Cassia fistula cultures. In this study, the calli were induced in MS + 1.0mg/L 2,4-D + 0.05 mg/L BAP with 50 mg/L ascorbic acid. These calli were then transferred to basic MS medium with 1.0 mg/L2,4,5-T and varied BAP concentrations. Four and 8 weeks old calli were used to find out the right concentration of BAP and age for upscaling of free flavonoids. The samples were extracted and HPTLC analyzed for free flavonoids at 260 nm along with the leaf explants. The study revealed that the amount and number of free flavonoids in 4 weeks old calli increased as the amount of cytokinin (0.5 to 1.0 mg/L BAP) increased but decline with further addition of BAP (2.0 mg/L BAP). In aged calli number and amount of free flavonoids exhibit a tremendous increase when compared to the calli in the same medium at 4 weeks. This is inferred that accruing age of callus, reduced quantities of plantgrowth regulators (optimized for callus growth) accumulate free flavonoids. The best medium for free flavonoid production is MS+ 1.0 mg/L 2,4,5-T with 1.0 mg/L BAP may be recommended for producing a high amount of free flavonoids in C. fistula in vitro.The results were justified with the results of quantification.

ABSTRACT

Peltophorum dubium (Spreng.) Taub. is a species belonging to the family Fabaceae, and is known popularly as golden shower tree. Its wood has multiple uses in reforestation programs and as an ornamental tree, and is also considered an endangered species, requiring studies that assist in its preservation. The present work was conducted with the objective of determine the ideal substrate type and temperature to perform germination and vigor tests with P. dubium seeds. The experiment was carried out at the Seed Analysis Laboratory (Centro de Ciências Agrárias, Universidade Federal Paraíba, Areia, Paraíba, Brazil), in a completely randomized design. The treatments were distributed in a 4 x 6 factorial scheme; temperatures (constant temperatures of 25, 30 and 35°C; and alternate temperatures of 20-30°C) and substrate (paper towel, over blotting paper, in sand, in vermiculite, Bioplant® and Plantmax®) on four replications of 25 seeds. The following parameters were analyzed: germination percentage, germination speed index, first germination count, length and dry mass of seedlings. The constant temperature of 30°C and 20-30°C alternate, and the substrates into sand and paper towel can be recommended for germination and vigor tests of P. dubium seeds. The temperature of 25°C should not be used in germination and vigor tests of P. dubium seeds in any of the tested substrates.


Peltophorum dubium (Spreng) Taub. é uma espécie pertencente à família Fabaceae, conhecida popularmente como canafístula. A espécie pode ser utilizada como ornamental e sua madeira tem múltiplos usos em programas de reflorestamento, é também considerada uma espécie em extinção, fazendo-se necessários estudos que auxiliem em sua preservação. Este trabalho teve como objetivo recomendar o substrato e a temperatura ideais para condução de testes de germinação e vigor para sementes desta espécie. O experimento foi realizado no Laboratório de Análise de Sementes (Centro de Ciências Agrárias, Universidade Federal Paraíba, Areia, Paraíba, Brasil), em delineamento inteiramente ao acaso, com os tratamentos distribuídos em esquema fatorial 4 x 6 (temperaturas constantes de 25, 30 e, 35°C e alternada de 20-30°C e os substratos: papel toalha, sobre papel mata-borrão, entre areia, entre vermiculita, Bioplant® e Plantmax®), distribuídas em quatro repetições de 25 sementes. Foram analisadas as seguintes características: germinação, índice de velocidade de germinação, primeira contagem de germinação, comprimento e massa seca de plântulas. Recomendam-se para condução dos testes de germinação e vigor das sementes de P. dubium os substratos entre areia ou papel toalha, na temperatura constante de 30°C e alternada 20-30°C. A temperatura de 25°C não deve ser utilizada para testes de germinação e vigor de sementes de P. dubium, em quaisquer dos substratos utilizados.

ABSTRACT

A eficiência da baixa temperatura no prolongamento da vida de vaso de várias flores de corte tem sido bastante estudada. No entanto, o efeito da temperatura depende da duração do armazenamento e da espécie floral. O experimento foi realizado objetivando avaliar a longevidade e as alterações metabólicas ocorridas durante o armazenamento refrigerado de inflorescências cortadas de Oncidium varicosum 'Samurai', armazenadas em diferentes temperaturas (5, 10 e 20°C). Foram avaliados os seguintes parâmetros: conteúdo relativo de água, carboidratos solúveis e açúcares redutores, conteúdo de carotenóides, coloração e longevidade. Os resultados indicaram diferenças nos parâmetros avaliados nas três temperaturas de armazenamento. As flores de Oncidium varicosum 'Samurai', armazenadas a 5°C, apresentaram melhor manutenção da qualidade do que nas outras temperaturas, o que foi evidenciado pela menor redução do conteúdo relativo de água, melhor manutenção do conteúdo de carotenóides, dos carboidratos solúveis e açúcares redutores, coloração e maior longevidade.


The effectiveness of low temperatures on extending vase life of several cut flowers has been largely investigated. However, the temperature effect depends on the length of the storage period and of the floral species. The experiment was accomplished to evaluate the longevity and the metabolic changes which occurred during the cold storage of cut inflorescences of Oncidium varicosum 'Samurai' kept under different temperatures (5, 10 and 20°C). The following parameters were analyzed: relative water content (RWC), soluble carbohydrates, reducing sugars, carotenoids contents, color and longevity. The results indicated differences among the measured parameters at the three storage temperatures. The inflorescences of Oncidium varicosum 'Samurai' stored at 5ºC presented better maintenance of flowers quality than at the other temperatures. It was evidenced by the lower reduction of RWC, better maintenance of carotenoids, soluble carbohydrates and reducing sugars. The flowers kept at 5°C also presented maintenance of coloration and greater longevity.
