ABSTRACT

Abstract Grape seed oil, which is usually extracted with highly toxic organic solvents that are harmful to human health, is produced from tons of grape pomace waste, generated during winemaking. Sometimes, this waste is used to make compost or is burnt, which causes environmental contamination. The functional qualities, antioxidant capacity (AC), α-tocopherol and total phenolic compounds content (TPC) of Black Borgoña (Vitis labrusca) grape seed oil, extracted by supercritical CO2, were evaluated. The high content of linoleic acid (ω-6) and monounsaturated fatty acids contributed to the beneficial effect on the functional quality indices, which were 0.20, 0.23, 11.80 for IA, IT and H:H, respectively. In addition, a POV of 6.23 ± 0.08 milliequivalents of peroxide/kg oil and an anisidine index of 2.70 ± 0.05 indicated a good quality oil. Also, a high concentration of a-tocopherol (9.82 ± 0.02 mg/100 g oil) and a high TPC ("4.14 ± 3.24 mg GAE/kg oil) were obtained. This study demonstrated that supercritical CO2 extraction is a suitable method for the delivery of a high-quality grape seed oil.


Resumen El aceite de semilla de uva que generalmente se extrae con disolventes orgánicos altamente tóxicos y perjudiciales para la salud humana, se produce a partir de toneladas de residuos de orujo de uva, generados durante la elaboración del vino. A veces, estos residuos se utilizan para hacer compost o se queman, lo que provoca la contaminación del medio ambiente. Se evaluaron las cualidades funcionales, la capacidad antioxidante (AC), el contenido de a-tocoferol y los compuestos fenólicos totales (TPC) del aceite de semilla de uva Borgoña Negra (Vitis labrusca), extraído mediante CO2 supercrítico. El alto contenido de ácido linoleico (ω-6) y de ácidos grasos monoinsaturados contribuyó al efecto beneficioso sobre los índices de calidad funcional que fueron de 0.20, 0.23, ''.80 para IA, IT y H:H, respectivamente. Además, un POV de 6.23 ± 0.08 miliequivalentes de peróxido/ kg de aceite y un índice de anisidina de 2.70 ± 0.05 indicaban una buena calidad del aceite. También se obtuvo una alta concentración de α-tocoferol (9.82 ± 0.02 mg/100 g de aceite) y un alto TPC ("4.14 ± 3.24 mg de GAE/ kg de aceite). Este estudio demostró que la extracción con CO2 supercrítico es un método adecuado para obtener un aceite de semilla de uva de alta calidad.


Resumo O óleo de semente de uva é geralmente extraído com solventes orgânicos altamente tóxicos que são prejudiciais à saúde humana, é produzido a partir de toneladas de resíduos de bagaço de uva, gerados durante a vinificação. Às vezes, esses resíduos são usados para fazer adubo ou são queimados, o que causa contaminação ambiental. Foram avaliadas as qualidades funcionais, capacidade antioxidante (AC), a-tocoferol e o teor total de compostos fenólicos (TPC) do óleo de semente de uva Borgoña Negra (Vitis labrusca), extraído por CO2 supercrítico. O alto teor de ácido linoleico (ω-6) e ácidos graxos monoinsaturados contribuiu para o efeito benéfico sobre os índices de qualidade funcional que foram 0.20, 0.23, 11.80 para IA, IT e H:H, respectivamente. Além disso, um POV de 6.23 ± 0.08 miliequivalentes de peróxido/ kg de óleo e um índice de anisidina de 2.70 ± 0.05 indicava uma boa qualidade de óleo. Também foi obtida uma alta concentração de α-tocoferol (9.82 ± 0.02 mg/100 g de óleo) e um alto TPC ("4.14 ± 3.24 mg de óleo GAE/ kg). Este estudo mostrou que a extração de CO2 supercrítico é um método adequado para a entrega de um óleo de semente de uva de alta qualidade.

ABSTRACT

Aims: The purpose of this study was to investigate the action of Citrus limonum essential oil to control free radical-induced lipid peroxidation and preventing tissue damage in skin. Place and Duration of Study: Department of Internal Medicine (University of Roma “Tor Vergata) and A.R.P.A (Aging Research, Prevention and Therapy Association, www.antiaging. it), between January 2010 and June 2011. Methodology: The essential oil was subjected to GC-MS analysis. The superoxide anion scavenging activity of essential oil was evaluated by the enzymatic hypoxanthine/xanthine oxidase system. The same oil diluted in DMSO or grape-seed oil was spread on the face of human volunteers after UV exposition. A sample of skin lipids was collected and the presence of peroxyl radicals was detected based on the measurement of light emitted (chemiluminescence) when the excited carbonyl and singlet oxygen decay to ground state. Results: Our data demonstrate that the lemon essential oil is more active than - tocopherol against *O2 - and peroxide free radical inhibition at 1:100 dilution. A protocol for controlling free radical-induced lipid peroxidation in human skin was thus proposed. Conclusion: The scavenging action of lemon essential oil could have a practical application for treating human skin against oxidative damage.

ABSTRACT

The study analyzed the lipid patterns and fatty acid compositions of serum and liver tissues in groups of Sparague-Dawley rats. Some of the groups were fed with an basal diet, which contained corn oil (C), grape seed oil (GSO), or perilla oil (P), and the others were fed with a high fat diet, which had cholesterol (1%) and lard (10%) mixed with corn oil (CHF), grape seed oil (GSHF), or perilla oil (PHF). The amount of dietary intake was higher for the basal diet groups than the high fat diet groups. And diet efficiency was significantly low in the group of rats fed with the basal diet mixed with perilla oil. From the analysis of the serum lipid patterns, a significant decrease in total lipid concentration was observed in the group of rats fed on the basal diet mixed with perilla oil and the high fat diet group. The levels of triglyceride and phospholipid were significantly low in the basal diet group when perilla oil or grape seed oil was involved. The ordinary diet groups showed significantly higher in HDL-C than the high fat diet groups. There was no significant difference among the basal diet groups, whether the diet was mixed with grape seed oil, perilla oil, or corn oil. However, a significant increase in HDL-C was observed in the group of rats fed with the high fat diet containing perilla oil. For LDL-C, there was a significant difference between the high fat diet groups and the basal diet groups. LDL-C was especially low in the group of rats fed with the high fat diet to which perilla oil was added, and the grape seed-added high fat diet group showed a decreasing tendency in LDL-C. The content of total fat, total cholesterol, and triglyceride was the lowest in the group of rats fed with the perilla oil-containing basal diet, and this group was followed in order by the grape seed oil-containing diet group and corn oil-containing diet group. In the analysis of the fatty-acid composition in liver tissue, the high fat diet groups showed an increase in saturated fatty acids and polyunsaturated fatty acids, but a decrease in mono unsaturated fatty acids when compared to the basal diet groups. The composition ratio of fatty acids varied according to which type of oil the diet contains. Our finding suggest that grape seed oil was an apparent diet effect on the fatty-acid composition.

ABSTRACT

The study was performed to investigate the effects of isoflavone and/or grape seed oil supplementation on serum and liver lipid profiles and bone strength in ovariectomized female rats. Female Spraque-Dawley rats were assigned into 4 groups of ovariectomized (O), isoflavone (0.085 mg/100 g b.w/day) in ovariectomized rats, grape seed oil in ovariectomized rats and isoflavone and/or grape seed oil in ovariectomized rats. After 8 weeks, biochemical profiles of serum, liver and bone were analyzed. Total food intakes, body weight gains and FER (food efficiency ratio) were not statistically significantly different among groups. Total cholesterol, triglyceride and LDL-cholesterol levels in serum were decreased by fed of isoflavone and/or grape seed oils. However, crude lipid and total cholesterol contents in liver were not affect of isoflavone and/or grape seed oil. The hepatic glutathione contents were increased by isoflavone and/or grape seed oil fed. The hepatic glutathione-S-transferase activity in isoflavone and/or grape seed oil supplemented groups were higher than that O group. Bone (scapular and femur bone) dry weight, femur of max weight and bending strength were no significant difference among groups. Our finding suggest that isoflavone/grape seed oils might have potential role for serum lipid profiles improvement and bone strength in vivo.

ABSTRACT

The effects of grape seed oil on serum total cholesterol (TC) and high density lipoprotein cholesterol (HDL-C) levels were investigated in experimental hypercholesterolemic rats. The grape seed oil containing about 74% linoleic acid, multivitamins and trace elements. Each animal was given 4 ml/kg BW grape seed oil mixed in high fat ration for 40 days. The results in two experiments showed that TC concentration in the serum of rats fed grape seed oil was markedly lowered as compared to control, but serum HDL-C was less lowered. Furthermore, the ratio of HDL-C to TC in the experimental group was also significantly higher than that of control.
