ABSTRACT

Aim To explore the therapeutic effect of DZ2002, a reversible S-adenosyl-L-homocysteine hydrolase inhibitor, on psoriasis-like skin lesions of guinea pig and its mechanism. Methods The guinea pig model of psoriasis was established with 50 g • L-1 propranolol hydrochloride liniment. The pathological changes of the skin were determined by hematoxylin and eosin (HE). Then the Baker score and epidermal thickness were measured based on HE. The infiltration of neutrophils was marked by immunohistochemical staining. The expression of chemokines in TNF-a/IFN-7-treated HaCa T cells in the present of DZ2002 or not were determined by real-time polymerase chain reaction (RT-PCR), and the production of chemokines from HaCa T cells were quantified by ELISA and Luminex x-MAP technology. In the same condition, supernatants were used to test the Chemotaxis effect on Jurkat and THP1 cells via Chemotaxis assays. Results Pathological features such as acanthosis, inflammatory cell infiltration, Munro microabscess, hyperkeratosis and parakeratasis appeared in the psoriasis-like skin lesions of guinea pigs. The Baker score and epidermal thickness of psoriasis-like guinea pig ear both increased significantly. Compared with vehicle group, DZ2002 cream not only significantly improved the pathological manifestations of guinea pig ear skin, but also reduced the skin Baker score and epidermal thickness. DZ2002 significantly down-regulated the expression of chemokines including IL-8 and CXCL9 in TNF-0/IFN-7treated HaCaT cells, and inhibited the Chemotaxis of THP1 and Jurkat cells. Conclusions DZ2002 cream can significantly improve the psoriasis symptoms in guinea pig model of psoriasis via inhibiting the secretion of chemokines by keratinocytes and reducing the infiltration of inflammatory cells.

OBJECTIVE@#To investigate the electrophysiological changes of autonomic cells in left ventricular outflow tract in guinea pigs with iron deficiency anemia complicated with chronic heart failure.@*METHODS@#Guinea pigs model of iron deficiency anemia complicated with chronic heart failure in 10 guinea pigs of the experimental group was made by feeding a low iron diet, pure water and subcutaneous injection of isoproterenol. The control group consisting of 11 guinea pigs was given normal food, normal water and injected with normal saline. The left ventricular outflow tract model specimen was also prepared. The standard microelectrode technique was used to observe electrophysiological changes of autonomic cells in the outflow tract of left ventricular heart failure complicated with iron deficiency anemia in guinea pig model. The indicators of observation were maximal diastolic potential, action potential amplitude, 0 phase maximal depolarization velocity, 4 phase automatic depolarization velocity, repolarization 50% and 90%, and spontaneous discharge frequency.@*RESULTS@#Compared with the control group, 4 phase automatic depolarization velocity, spontaneous discharge frequency and 0 phase maximal depolarization velocity decreased significantly (P < 0.01) and action potential amplitude reduced (P < 0.01) in model group. Moreover, repolarization 50% and 90% increased (P < 0.01).@*CONCLUSIONS@#There are electrophysiological abnormalities of the left ventricular outflow tract in guinea pigs with iron deficiency anemia complicated with heart failure.

ABSTRACT

Objective To establish a suitable guinea pig model for evaluating the protective effects of new therapeutic TB vaccines in preclinical study .Methods The guinea pigs were subcutaneously injected with single-cell suspension of multi-drug resistant M.tuberculosis at a dose of 1000 CFU, 0.2 ml per animal.The study was divided into experimentⅠand experimentⅡ.In experimentⅠ, the guinea pigs were given immuno-therapy and/or chemical treatment on day 3 after infection .Four guinea pigs in each group were dissected at weeks 5, 7 and 9 after infection for evaluating lesion scores and histopathology changes of liver , spleen and lung, as well as bacterial load in spleen .In experimentⅡ, the guinea pigs were given immunotherapy and/or chemical treatment with different doses on day 14 after infection .All guinea pigs were dissected at week 5 after infection for evaluating lesion scores of liver , spleen and lung , as well as bacterial load in spleen .Results In experimentⅠ, all of the three treatments including immunotherapy combined with chemotherapy , immunotherapy alone and chemotherapy alone could effectively prevent organ lesion and reduce bacterial load in spleen , which were significantly different from negative control group .The immunotherapy combined with chemotherapy showed better treatment effects than other two treatments .Along with a prolonged period after drug withdrawal , the de-gree of organ lesion in immunotherapy group and chemotherapy group rebounded sharply , but only slightly in im-munotherapy combined with chemotherapy group .In experimentⅡ, animals in all treatment groups showed alle -viated organ lesion and reduced bacterial load in spleen .A relatively better treatment effect was observed in im-munotherapy combined with chemotherapy group .Conclusion The established guinea pig model of M.tubercu-losis infection showed an advantage of good repeatability .It might be used to evaluate the protective effects of new therapeutic vaccines against tuberculosis in preclinical study .

ABSTRACT

OBJECTIVE:To compare the therapeutic effects of8-methoxypsoralen(8-MOP)liposomal gel with other dosage forms of8-MOP in equal concentration on guinea-pig models of vitiligo.METHODS:The guinea-pigs were divided into normal control,model,8-MOP tincture,8-MOP gel and8-MOP liposome gel groups.The vitiligo model was established by chemical decoloration method in all groups except normal control group,then corresponding preparations were topically ap?plied.The therapeutic effects were compared and the melanogenesis of hair follicles,serum acetylcholinesterases and skin MDA contents were observed in all groups.RESULTS:8-MOP liposomal gel had better therapeutic effects on the animal models,and could improve the dermatic melanogenesis and acetylcholinesterase of the animal model,and also had effect on MDA of skin.The differences between8-MOP liposomal gel group and model group were highly significant(P
