ABSTRACT

Objective: To review the radiopathological correlation of lesions of uncertain malignant potential (B3) diagnosed in our institution, as well as to evaluate the therapeutic conduct and to establish the percentage of underestimation of the excised lesions. Material and method: This work consists of a retrospective study of the database which in- cludes the interventional procedures performed in the Imaging Department of the Alexander Fleming Institute between May 2016 and December 2019. Patients with a histological outcome in the percutaneous biopsy of one or more of the following diagnosis were included: flat epithelial atypia (FEA), radial scar (RS) /complex sclerosing lesion (CSL), atypical ductal hyperplasia (ADH), classic lobular neo- plasia (LN), papillary lesión (PL) or phyllodes tumor (PT). Results: Out of the 67 analyzed patients, 43.3% were perceived as nodules at imaging examinations and 37.3% as grouped microcalcifications. Papillary lesion was the most frequently diagnosed B3 lesion (44.8%), followed by flat epithelial atypia (16.4%). 98.5% of patients presented an accurate imaging-pathology concordance. 48 patients underwent surgery and the remaining 19 patients continued with clinical and radiological follow-up. The histopathology of the surgical specimen reflected that in 33.3% of the cases there was an underestimation in relation to the percutaneous biopsy; in over half of these cases they corresponded with DCIS. Conclusions: The percentage of underestimation with final diagnosis of DCIS or low grade IDC coincided with the parameters found in current literature. Even though we were able to reduce the surgical rate to 71% of patients with B3 lesions, a proper selection of cases which could be candidates for vacuum assisted excisional biopsy could further reduce the number of unnecessary surgeries and probably the un- derestimation rate as well.

ABSTRACT

HDA9, a member of the deacetylase family, plays a vital role in regulating plant flowering time through flowering integrator SOC1 and AGL24. However, it remains elusive how HDA9 interacts with SOC1 and AGL24 in flowering time control. Here, HDA9 was cloned in Brassica juncea and then its three active sites were separately replaced with Ala via overlap extension PCR. Thus, mutants of HDA9(D172A), HDA9(H174A) and HDA9(D261A) were constructed and fused into the pGADT7 vector. The yeast one-hybrid assays indicated that HDA9 mutants remained the interactions with the promoters of SOC1 and AGL24. Furthermore, the aforementioned results were confirmed in the dual luciferase assays. Interestingly, the DNA-protein interactions were weakened significantly due to the mutation in the three active sites of HDA9. It suggested that flowering signal integrator SOC1 and AGL24 were regulated by the key amino acid residues of 172th, 174th and 261th in HDA9. Our results provide valuable information for the in-depth study of the biological function and molecular regulation of HDA9 in Brassica juncea flowering time control.

ABSTRACT

Objective To explore the protective effects of 10-HDA combined with AA-2G on UVA-induced photodamage in fibroblasts. Methods The primary cultured human skin fibroblasts were divided into three groups: blank control group, UVA irradiation group and 10-HDA+AA-2G group. The cell viability and cellular senescent state were analyzed using CCK-8 and senescence associated-β-galactosidase (SA-β-gal) staining, respectively. Fluorometric assays were performed to detect the formation of reactive oxygen species (ROS) in the cells. Results Compared with those in control group, the cell viability was decreased (P < 0.05), and the SA-β-gal positive cells and ROS level were significantly increased (P<0.01 for both) in UVA-irradiated group. Compared with those in UVA-irradiated group, the cell viability was significantly increased (P < 0.01), and the SA-β-gal positive cells and ROS level were significantly reduced (P < 0.05, P < 0.01, respectively) in the 10-HDA combined with AA-2G group. Conclusion Fibroblasts treated with 10-HDA and AA-2G are significantly protected from UVA-induced cytotoxicity, cellular senescence and ROS, indicating that 10-HDA combined with AA-2G has photoprotective effects. Therefore, 10-HDA combined with AA-2G may be a potential combination for the prevention and treatment of skin photoaging.

ABSTRACT

Objective: To establish the quality standard of Jinwang Capsules. Methods: The technique of TLC was used to identify 10-Hydroxy-2-decylenic acid (10-HDA). Its content was determined by dual-wavelength UV spectrophotometry. Results: 10-HDA can be detected by TLC. The content of 10-HDA wasn't lower than 3.0mg per granule. Volatile alkalescent substance wasn't more 100mg per 100g.Conclusion: These methods are able to effectively control the quality of Jinwang Capsules.

ABSTRACT

AIM:To prepare the Lyophiled Royal Jelly Soft Capsule and study its stability and Influential factors.METHODS:The suspending agent and processing method were optimized using sedimentation volume rate as the index.Soft capsules were prepared and product stability under high temperature and high humidity environment was studied according to the determination of the content of 10-HAD by HPLC.RESULTS:The finished product yield in pilot test was more than 90%,the soft capsule products stored in cold were stable, while those stored under room temperature or high temperature and high humidity were unstable with a noticeable decrease in quality.Water content in capsule shell affects the 10-HDA content of the finished product.CONCLUSION:The preparative process is feasible and the products should be storaged in cold enviroment.
