ABSTRACT

To explore the mechanism of Hedyotis Diffusae Herba-Smilacis Glabrae Rhizoma(HDH-SGR) in treating lung adenocarcinoma based on big data bioinformatics combined with network pharmacology analysis and molecular docking technology. The chemical components and potential therapeutic targets of HDH-SGR were obtained from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP). Lung adenocarcinoma-related genes were obtained from The Cancer Genome Atlas(TCGA), Therapeutic Target Database(TTD), Pharmacogenetics and Pharmacogenomics Knowledge Base(PharmGKB), Online Mendelian Inheritance in Man(OMIM), DrugBank, and GeneCards. "Drug component-target" network was constructed using Cytoscape to screen out key compounds. STRING was used to build protein-protein interaction(PPI) network and core targets were screened out by Cytoscape-CytoNCA topology analysis. Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) analyses of target genes were performed by R-clusterProfiler. Finally, key compounds were docked to core target genes using AutoDock. The results showed that 22 active compounds and 499 potential therapeutic targets were obtained from HDH-SGR. A total of 14 332 lung adenocarcinoma-related targets were screened out through six data platforms, including 182 common targets. Fifteen core targets were screened out from the PPI network. GO and KEGG analyses revealed significant enrichment of relevant target genes in various biological processes, cellular functions(e.g., response to lipopolysaccharide, nuclear receptor activity, and ligand-activated transcription factor activity) and close relationship between target genes and non-small cell lung cancer signaling pathways. Based on the results of molecular docking validation, diosgenin, quercetin, naringenin, taxifolin, 2-methoxy-3-methyl-9,10-anthraquinone, stigmasterol, and β-sitosterol were able to bind tightly to the core targets. HDH-SGR can intervene in lung adenocarcinoma through multiple targets and signaling pathways, such as non-small cell lung cancer signaling pathways. The binding of active components in Chinese medicine to key targets is presumedly one of the mechanisms that produce therapeutic effects.

ABSTRACT

Objective: To evaluate the effects of ethanol extract of Hedyotis Diffusae Herba (EEHDH) on chronic ulcerative colitis (UC) and the inhibition on intestinal tissue abnormalities hyperplasia. Methods: The C57 mice were randomly divided into five groups: normal, DSS, low-, mid-, and high-dose (2.5, 10.0, and 20.0 g/kg) EEHDH groups. The DSS-induced UC model was employed. The body weight, hematochezia, and stool property were recorded to evaluate the change of physiology daily. The pathology changes of colonic mucosa tissue of mice were evaluated by HE staining, and the expression changes of inflammatory factors IL-6 and TNF-α in serum and tissues were detected by ELISA and real-time PCR, as well as immunohistochemistry was used to analyze the positive rate of Ki67 of the nuclear antigen in epithelial cells of colonic tissues. Results: Compared to the model group, the treatment with mid- and high-dose EEHDH has a relatively slower weight loss and a faster recovery when the stimulation is gone; Meanwhile the symptoms of diarrhea and hematochezia are significantly improved, the pathological damage and intestinal tissue dysplasia are alleviated; The inflammatory factors IL-6, TNF-α, and the positive rate of Ki67 are significantly decreased. Conclusion: EEHDH could attenuate DSS-induced UC and decrease pathological injury, inflammation invasion, and abnormalities hyperplasia, by inhibiting the expression of inflammatory cytokines, such as IL-6 and TNF-α, which indicates that EEHDH is of great value in the treatment of UC and colitis-associated cancer.

ABSTRACT

Objective: To study the difference of substantial base of Baihuasheshecao (Hedyotis Diffusae Herba) Injection (BSCI) within the nationwide. Methods: To determine 16 batches of samples by UPLC-QTOF-MS and analyze the data according to a variety of retention time and m/z by principal component analysis (PCA). Results: The results showed that the difference itself was smaller but between each other was bigger among the different enterprises. Seven markers greatly impacted on the components through loading plot analysis were found. Conclusion: BSCIs from different enterprises within nationwide are not only obviously different in color but also different in substantial base, which especially indicates the content of flavonoids and organic acids.
