ABSTRACT

Objective To explore the effect of Heshouwuyin on the steroidogenic acute regulatory protein (StAR) and cytochrome P450 cholesterol side chain cleavage enzyme(P450scc) protein expression in Leydig cells of rats. Methods Juvenile male Wistar rat, 10-20 days old, were used to isolate the Leydig cells and cultured, and then collected them. The oxidative damage technique was used to establish Leydig cell aging model. This study had a Leydig aging model, Heshouwuyin and Shouwu pill treatments, and normal and Heshouwuyin control group. Results The intensities of immunohistichemical staining for StAR and P450scc were significantly lower in the Leydig aging model group than those in the normal group and Heshouwuyin control group (P < 0.01), and were significantly higher in the Heshouwuyin and Shouwu pill group than that in the aging model group (P < 0.05). Heshouwuyin group was better than Shouwu pill group. Conclusion Heshouwuyin can increase the protein expression of StAR and P450scc in Leydig cells, improve the synthesis of testosterone, thus delaying the senescence of Leydig cells.

ABSTRACT

ObjectiveTo find the mechanism of Heshouwuyin(HSWY) in anti-aging, so as to provide theoretical and experiment evidence for the application of HSWY. Methods50 eight-week female SD rats ( SPF degree)were used in this study. The rats were randomly divided into 5 groups:normal control group, 10 rats; model group,10 rats; HSWY anti-aging group, including low, middle and high dose group,10 rats in each group. The sub-acutely aging rats were made by ip injection of D-galactose for 60 days continually. The rats in HSWY groups were administered with various doses of intragastric HSWYduring D-galactose injection. The parameters of ovary levels of SOD,GSH-PX, TAOC were detected. ResultsThe MDA in ovaries of model rats was significantly higher than that of negative control group;while the SOD, GSH-PX, TAOC of model rats were significantly lower than negative control group.The MDA content in ovaries after taking HSWY was significantly lower than that of model rats; while the SOD, GSHPX,TAOC of the taking HSWY rats was significantly higher than that of model rats. ConclusionThe HSWY could increase antioxida tive activity of rat ovaries to stay D-galactose induced consenescence.

ABSTRACT

Objective To find the mechanism of traditional Chinese medicine Heshouwuyin(HSWY) in anti-aging in ovary of the aging model rats,so as to provide theoretical and experimental evidence to anti-aging in female sexual gland.Methods Thirty-six rats were divided into three groups at random: normal group,aging model group,prevent aging group.Rats the in prevent aging group were subdivided into four groups: HSWY low dosage group,HSWY medium dosage group,HSWY high dosage group and Heshouwan(HSWW) group.The sub-acutely aging model rats were induced by D-galactose,and rats in the prevent aging group were received intragastric administration of HSWY and HSWW with D-galactose intraperioneal injection.The gene expressions of p53,p19~(ARF),Rb and p16 were detected in the ovaries of each group with reverse transcription polymerase chain reaction,and the protein expressions of Rb and p16 in the ovaries with Western blotting.Results The model group rats were in the aging status with low spirits and withered and yellowed hair.The ovaries of the rats in the model group expressed aging change.The gene expressions of p53,p19~(ARF),Rb and p16 were up-regulated and the protein expressions of Rb and p16 were increased in the ovaries of aging rats.HSWY significantly suppressed the gene expressions of p53,p19~(ARF),Rb and p16(P
