ABSTRACT

ObjectiveTo investigate the material basis of homologous and heterogeneous effect of Aurantii Fructus Immaturus(AFI) and Aurantii Fructus(AF) based on the total statistical moment analysis and molecular connectivity index(MCI). MethodRelevant literature at home and abroad and Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP) were consulted to establish the chemical composition database of AFI and AF, and set up their fingerprints by ultra-high performance liquid chromatography(UPLC), and the total statistical moments and similarity parameters of the fingerprint were calculated. According to MCI, all components of AFI and AF were divided into different component groups, the average values of 0-8th order(0χ-8χ) MCI of the common component groups of AFI and AF were calculated. ResultThe values of total zero-order moment(AUCT) of AFI and AF were (10.57±2.45)×106, (5.09±0.89)×106 μV·s, the values of total first-order moment(MCRTT) were (11.57±1.58), (12.10±1.29) min, the values of total second-order moments(VCRTT) were(24.49±2.30), (26.49±2.54) min2, respectively. It showed that qualitative and quantitative parameters of AFI and AF were significantly different. The components with high similarity such as neohesperidin, hesperidin and narirutin were screened as the common potential pharmacodynamic components of AFI and AF. The non-common components of AFI, such as alysifolinone and imperatorin, and the non-common components of AF, such as neoeriocitrin and isosakuranin, with high similarity were screened out as potential heterogeneous components of AFI and AF. The composition groups of AFI and AF were classified into six categories, and the similarities between the composition groups of AFI and AF and the total constituents were 0.872-0.979 and 0.918-0.997, the average values of 0χ-8χ MCI of alkaloids in AFI and AF were 3.65 and 3.14, the average values of 0χ-8χ MCI of flavonoids were 8.47 and 8.47, the average values of 0χ-8χ MCI of volatile oils were 2.71 and 3.48, respectively. It showed that there were some differences in MCI of chemical constituents(groups) between AFI and AF. ConclusionThe chemical constituents(groups) of AFI and AF not only differ in content and species, but also in structural characteristics and structure-activity relationship, which can provide a basis for further explaining the scientific connotation of homologous and heterogeneous effect of AFI and AF.

ABSTRACT

Objective:To evaluate the relationship between Karyopherin β2 (Kapβ2)-mediated nuclear translocation of nuclear inhomogeneous ribonucleoprotein A2/B1 (hnRNPA2/B1) and sevoflurane-induced brain neurotoxicity in a cellular experiment.Methods:The mouse hippocampal neuronal cell line HT22 cells were inoculated in confocal culture dishes and 6-well culture plates at a density of 2×10 5 cells/well and 1×10 6 cells/well and divided into 4 groups( n=12 each) by a random number table method: control group (GFP-C group) carrying green fluorescent protein (GFP) with empty adenovirus transfection, sevoflurane group (GFP-Sev group) carrying GFP with empty adenovirus transfection, control group (GFP-Sev group) transfected with Kapβ2 gene-overexpressing adenovirus, and sevoflurane group (Kapβ2-Sev group) transfected with Kapβ2 gene-overexpressing adenovirus. After 48 h of conventional incubation, empty adenovirus-carrying GFP (GFP-C and GFP-Sev groups) and Kapβ2 gene-overexpressing adenovirus (Kapβ2-C and Kapβ2-Sev groups) were transfected. After 48 h of transfection, the cells were conventionally incubated continuously in GFP-C and Kapβ2-C groups, and the cells were incubated for 3 h with 3% sevoflurane and then were conventionally incubated for 48 h in GFP-Sev and Kapβ2-Sev groups. The expression of Kapβ2, synaptophysin (SYP), postsynaptic density protein 95 (PSD95) and hnRNPA2/B1 nucleoplasmic ratio were measured in cells by Western blot. Immunofluorescence assay was used for hnRNPA2/B1 subcellular localization. Results:Compared with GFP-C group, the expression of SYP and PSD95 was significantly down-regulated, hnRNPA2/B1 nucleoplasmic ratio was decreased, and cytoplasmic hnRNPA2/B1 expression was up-regulated in GFP-Sev group, and Kapβ2 expression was significantly up-regulated in Kapβ2-C group ( P<0.05). Compared with Kapβ2-C group, the expression of SYP and PSD95 was significantly down-regulated, hnRNPA2/B1 nucleoplasmic ratio was decreased, and cytoplasmic hnRNPA2/B1 expression was up-regulated in Kapβ2-Sev group ( P<0.05). Compared with GFP-Sev group, the expression of Kapβ2, SYP and PSD95 was significantly up-regulated, hnRNPA2/B1 nucleoplasmic ratio was increased, and cytoplasmic hnRNPA2/B1 expression was down-regulated in Kapβ2-Sev group ( P<0.05). Conclusions:Kapβ2-mediated hnRNPA2/B1 nuclear translocation may be the endogenous protective mechanism against sevoflurane-induced brain neurotoxicity.

ABSTRACT

Mammalian testis exhibits remarkably high transcriptome complexity, and spermatogenesis undergoes two periods of transcriptional cessation. These make the RNA-binding proteins (RBPs) the utmost importance during male germ cell development. Heterogeneous nuclear ribonucleoproteins (hnRNPs) are a large family of RBPs implicated in many steps of RNA processing; however, their roles in spermatogenesis are largely unknown. Here, we investigated the expression pattern of 12 hnRNP family members in mouse testes and found that most detected members are highly expressed in the testis. Furthermore, we found that most of the detected hnRNP proteins (hnRNPD, hnRNPK, hnRNPQ, hnRNPU, and hnRNPUL1) display the highest signals in the nuclei of pachytene spermatocytes, round spermatids, and Sertoli cells, whereas hnRNPE1 exclusively concentrates in the manchette of elongating spermatids. The expression of these hnRNP proteins showed both similarities and specificity, suggesting their diverse roles in spermatogenesis.

ABSTRACT

Under the organization of Renji Hospital, Shanghai Jiao Tong University School of Medicine, a specialized disease database of inflammatory bowel disease (IBD) cohort was deployed, and a brief introduction of the database was made in this article. The IBD data set was established by referring to domestic and foreign standards. Through data extraction, cleaning, normalization and other information processing technologies, data from multi‑source heterogeneous platform were arranged to form a specialized major disease database of IBD cohort and the efficiency and quality of data collection in clinical practice, teaching and scientific research were guaranteed. The display and personalized export capacities of the database can promote the researches on IBD and assist the clinical decision‑making. It provides not only efficient, comprehensive and reliable research‑level data support for scientific research, but also a precise guidance for diagnosis and treatment of the disease. Furthermore, it can excavate the potential clinical principles based on medical big data.

ABSTRACT

OBJECTIVES@#Tongue squamous cell carcinoma (TSCC) is a common cancer in the oral and maxillofacial region, which seriously endangers people's life and health.Heterogeneous nuclear ribonucleoprotein A2/B1(hnRNP A2/B1) is an RNA-binding protein that regulates the expression of a variety of genes and participates in the occurrence and development of a variety of cancers. This study aims to investigate the role of hnRNP A2/B1 in TSCC progression.@*METHODS@#The differential expression of hnRNP A2/B1 in oral squamous cell carcinoma (OSCC) and normal oral mucosa cells and tissues was analyzed based on the gene expression profiles of GSE146483 and GSE85195 in the Gene Expression Omnibus (GEO) database. The correlation between hnRNP A2/B1 expression and disease-free survival of TSCC patients was analyzed based on TSCC related chip of GSE4676. TSCC cancer and paracancerous tissue samples of 30 patients were collected in Hunan Cancer Hospital from July to December 2021. Real-time RT-PCR and Western blotting were used to verify the mRNA and protein expression of hnRNP A2/B1 in TSCC patients'samples, respectively. Human TSCC Tca-8113 cells were transfected with hnRNP A2/B1 empty vector (a sh-NC group), knockdown plasmid (a sh-hnRNP A2/B1 group), empty vector overexpression plasmid (an OE-NC group) and overexpression plasmid (an OE-hnRNP A2/B1 group), respectively. The knockdown or overexpression efficiency of hnRNP A2/B1 was detected by Western blotting. The proliferation activity of Tca-8113 cells was detected by cell counting kit-8 (CCK-8), and the apoptosis rate of Tca-8113 cells was detected by flow cytometry.@*RESULTS@#Based on the analysis of OSCC-related chips of GSE146483 and GSE85195 in the GEO database, it was found that hnRNP A2/B1 was differentially expressed in the OSCC and normal oral mucosa cells and tissues (all P<0.01). Meanwhile, the analysis of TSCC related chip GSE4676 confirmed that the expression of hnRNP A2/B1 was negatively correlated with the disease-free survival of TSCC patients (P=0.006). The results of real-time RT-PCR and Western blotting showed that the relative expression levels of hnRNP A2/B1 mRNA and protein in TSCC tissues were significantly up-regulated compared with those in adjacent tissues (all P<0.01). The results of Western blotting showed that the expression level of hnRNP A2/B1 in Tca-8113 cells was significantly inhibited or promoted after knockdown or overexpression of hnRNP A2/B1 (all P<0.01). The results of CCK-8 and flow cytometry showed that inhibition of hnRNP A2/B1 expression in Tca-8113 cells reduced cell proliferation activity (P<0.05) and increased cell apoptic rate (P<0.01). Overexpression of hnRNP A2/B1 in Tca-8113 cells significantly increased cell proliferation (P<0.05) and decreased cell apoptosis (P<0.01).@*CONCLUSIONS@#HnRNP A2/B1 is a key factor regulating the proliferation and apoptosis of TSCC cells. Inhibition of hnRNP A2/B1 expression can reduce the proliferation activity of TSCC cells and promote the apoptosis of TSCC cells.

ABSTRACT

ObjectiveTo elucidate the mechanism of Osteoking against fracture, femoral head necrosis, osteoarthritis, and lumbar disc herniation by integrating heterogeneous information network mining and experimental validation. MethodOn the basis of the disease-related database and transcriptome expression profiling dataset, as well as the ETCM database, the gene sets related to four target diseases and the candidate target spectrum of Osteoking were obtained through the integration and analysis of bioinformatics data, and a "disease-syndrome-formula-target-pathway-effect" heterogeneous information network was constructed. In addition, by functional enrichment analysis, the core targets of Osteoking in interfering with the imbalance network of four kinds of bone injury diseases, the biological pathways involved, and the corresponding clinical symptoms were screened, and they were verified in animal experiments. ResultHeterogeneous information network mining indicates that Osteoking may commonly reverse the imbalance networks of fracture, femoral head necrosis, osteoarthritis, and lumbar disc herniation via regulating cell function and activity, inhibiting inflammatory response, reducing bone destruction, and improving the immune function of the body by modulating relevant core candidate targets such as RAC-alpha serine/threonine-protein kinase (Akt1), catenin beta-1 (CTNNB1), epidermal growth factor receptor (EGFR), heat shock protein 90-alpha (HSP90AA1), and phosphatidylinositol 3-kinase catalytic subunit alpha isoform (PI3KCA), as well as related biological pathways such as phosphatidylinositide 3-kinases/protein kinase B (PI3K/Akt), janus kinase/signal transducer and activator of transcription (JAK/STAT), tumor necrosis factor (TNF), nuclear factor kappa-B (NF-κB), and Toll-like receptors. In particular, Osteoking may improve the blood supply of the fracture end by regulating blood circulation at the target site of the disease, and it may maintain the balance of bone metabolism by regulating hormone-related pathways to promote fracture healing. In addition, Osteoking may relieve lipid metabolism disorders by targeting and regulating lipid-related pathways, accelerate bone formation and bone repair, and delay the progression of femoral head necrosis. Osteoking may relieve the symptoms of pain by acting on neurological pathways to reduce local nociceptive stimulation in patients with osteoarthritis and lumbar disc herniation. Further experimental validation demonstrates that the PI3K/Akt signaling pathway is the most significantly enriched pathway for the key network targets of Osteoking for the four diseases. The candidate target of Osteoking may have the strongest association with the network of fracture-related genes. Therefore, this study chooses fracture as the target disease to verify the efficacy of Osteoking. The results show that Osteoking can accelerate bone formation and promote fracture healing by inhibiting the activation of the PI3K/Akt signaling axis. ConclusionThe study shows that the main mechanism of "treating different diseases with an identical treatment" of four bone injury diseases with Osteoking involves cell function regulation and immune inflammation-related signaling pathways. Further experimental validation identifies that the PI3K/Akt signaling axis may be one of the key pathways of Osteoking to promote bone regeneration, bone reconstruction, and bone metabolism homeostasis.

ABSTRACT

The global industrialization of chemical dyes used for household products has resulted in various dyes leaching into the soil. Azo dye textile contamination could lead to infertile soil. This study investigates the ecological effect of azo textile dye contamination on bacterial biomass, selective pressure, and associated physicochemical changes in soil samples contaminated with textile azo dye in Itoku, Abeokuta, Nigeria. Microbiological and physicochemical changes of the soil and effluent samples collected were determined using standard procedures. Results revealed that the average count of total viable bacteria in the contaminated sampling sites ranged from 0.82±021 x 106 cfu/g to 1.65±0.02 x 106 cfu/g which was significantly lower than the control bacterial counts. The dye contaminated and the control soil samples each contained a heterogenous population of bacteria which included Paenibacillus validus, Bacillus licheniformis, Bacillus niacin, Seratia liquefaciens, Staphylococcus gallinarum, Bacillus subtilis, and Bacillus coagulans. The physicochemical analysis of the soil revealed high levels of pH in the azo dye contaminated soil (10.4–11.1) and electrical conductivity was also highest in the effluent (1250 to 2943 units) compared to the control (430 to 480). Importantly, the control samples were higher in all of the other tested soil parameters including cation exchange, the mean value of the organic matter, water holding capacity, particle density, moisture content, and total porosity. Conclusion- Azo dye contamination caused decreases in bacterial density and many other soil parameters along with higher PH and electrical conductivity which suggests that azo textile dye contaminated soil is unfavorable for agricultural purposes and potentially toxic to the ecosystem.

ABSTRACT

Heterogeneous nuclear rib nucleoproteins belong to a class of RNA-binding proteins and possess highly conserved structures. Among them, the heterogeneous nuclear rib nucleoprotein A2/B1 (heterogeneous nuclear rib nucleoprotein A2, hnRNPA2/B1) is involved in many processes such as transcription and translation. In recent years, in the field of neurodegenerative diseases, it has been found that hnRNPA2/B1 promotes the accumulation of fibrin in the cytoplasm and participates in the formation of stress granules in neurons, which involves a variety of processes related to cognitive function, and is closely related to neurodegenerative diseases such as amyotrophic lateral sclerosis, frontotemporal dementia, Alzheimer′s disease and so on. This article reviews the related research, summarizes the important role and mechanism of hnRNPA2/B1 in neurodegenerative diseases, and provides help for the diagnosis and treatment of neurodegenerative diseases in the future.

Objective@#To construct symptom-formula-herb heterogeneous graphs structured Treatise on Febrile Diseases (Shang Han Lun,《伤寒论》) dataset and explore an optimal learning method represented with node attributes based on graph convolutional network (GCN).@*Methods@#Clauses that contain symptoms, formulas, and herbs were abstracted from Treatise on Febrile Diseases to construct symptom-formula-herb heterogeneous graphs, which were used to propose a node representation learning method based on GCN − the Traditional Chinese Medicine Graph Convolution Network (TCM-GCN). The symptom-formula, symptom-herb, and formula-herb heterogeneous graphs were processed with the TCM-GCN to realize high-order propagating message passing and neighbor aggregation to obtain new node representation attributes, and thus acquiring the nodes’ sum-aggregations of symptoms, formulas, and herbs to lay a foundation for the downstream tasks of the prediction models.@*Results@#Comparisons among the node representations with multi-hot encoding, non-fusion encoding, and fusion encoding showed that the Precision@10, Recall@10, and F1-score@10 of the fusion encoding were 9.77%, 6.65%, and 8.30%, respectively, higher than those of the non-fusion encoding in the prediction studies of the model.@*Conclusion@#Node representations by fusion encoding achieved comparatively ideal results, indicating the TCM-GCN is effective in realizing node-level representations of heterogeneous graph structured Treatise on Febrile Diseases dataset and is able to elevate the performance of the downstream tasks of the diagnosis model.

ABSTRACT: Spontaneous polydactyly has been described in several species, but only one report about it in Swiss mice. The aim of the current study was to report the spontaneous occurrence of pre-axial polydactyly in Swiss mice. Clinical examination showed one extra toe laterally to the first digit, in the plantar region, alopecia in the back, altered face growth anatomy and changed perineal region anatomy. Pre-axial polydactyly in the tibial side, fused metatarsals and Y-shaped free phalanges were evidenced in the radiographic images. Pre-axial polydactyly observed in the plantar region differed from that in reports on albino Swiss mice with post-axial polydactyly (Po/Po+) phenotype featured by one extra toe in the ulnar side of one, or both, front limbs, which is the dominant feature. The observed findings highlight the importance of both clinical examinations and close attention by professionals involved in rodents' breeding on physical changes resulting from different causes, including the genetic ones, since they reveal mutations and, sometimes, new biomodels.


RESUMO: A polidactilia espontânea foi descrita em várias espécies, mas há apenas um relato em camundongos Swiss. O objetivo do presente estudo foi relatar uma ocorrência de polidactilia pré-axial em camundongos Swiss. O exame clínico revelou um dedo extra lateralmente ao primeiro dedo na região plantar, alopecia dorsal, anatomias facial e da região perineal alteradas. Nas imagens radiográficas foram evidenciados polidactilia pré-axial no lado tibial, metatarsos fundidos e falanges livres em forma de Y. A polidactilia pré-axial observada diferiu do relato existente de polidactilia pós-axial (Po/Po+) caracterizado por um dedo extra no lado ulnar de um ou ambos os membros anteriores e de característica dominante. Os achados observados destacam a importância dos exames clínicos e da atenção cuidadosa dos profissionais envolvidos na criação de roedores sobre as alterações físicas decorrentes de diferentes causas, inclusive as genéticas, em revelar mutações e, às vezes, novos biomodelos.

SUMMARY: Anatomical studies describe the vastus medialis (VM) as being subdivided into two morphologically distinct components, the vastus medialis obliquus (VMO) and the vastus medialis longus (VML). However, there are discrepancies regarding the functional differentiation of these components. The aim of this study was to compare the levels of activation of the VMO and the VML by high density surface electromyography. Twelve healthy young women (age: 21.4 ± 2.0 years; weight: 58.1 ± 7.5 kg; height: 1.6 ± 0.1 m), performed an open kinetic chain knee exercise during which the EMG activity of the VMO and the VML was recorded with two- dimensional matrices of 32 surface electrodes. The exercises were performed with three levels of resistance (5, 10 and 15 % of the body weight (BW)), considering three phases: concentric, isometric and excentric. In the isometric phase the VMO had greater activation than the VML with the three levels of resistance (p<0.05). In the excentric phase, the VMO also showed greater activation than the VML with the 10 and 15 % BW resistance levels, while in the concentric phase, the VMO showed greater activity than the VML with only the 15 % BW resistance. The results indicated significant differences in the activation level of the two components of the VM. This bears importance in the development of exercises intended to achieve a greater or more selective activation of the VMO. In the sample subjected to evaluation, the EMG recordings describe a greater activation of the VMO in comparison to the VML, which is more important in the isometric and excentric phases of the flexion/extension of the knee in an open kinetic chain. These findings suggest a functional compartmentalization of the VM.


RESUMEN: Los estudios anatómicos describen que el músculo vasto medial (VM) se subdivide en dos componentes morfológicamente distintos, el vasto medial obliquus (VMO) y el vasto medial largo (VML). Sin embargo, existen discrepancias con respecto a la diferenciación funcional de estos componentes. El objetivo de este estudio fue comparar los niveles de activación del VMO y el VML mediante electromiografía de superficie de alta densidad. Doce mujeres jóvenes sanas (edad: 21,4 ± 2,0 años; peso: 58,1 ± 7,5 kg; altura: 1,6 ± 0,1 m), realizaron un ejercicio de rodilla de cadena cinética abierta durante el cual se registró la actividad EMG de la VMO y la VML con dos matrices dimensionales de 32 electrodos de superficie. Los ejercicios se realizaron con tres niveles de resistencia (5, 10 y 15% del peso corporal (PC)), considerando tres fases: concéntrica, isométrica y excéntrica. En la fase isométrica el VMO tuvo mayor activación que el VML con los tres niveles de resistencia (p <0,05). En la fase excéntrica, el VMO también mostró mayor activación que el VML con los niveles de resistencia de 10 y 15% BW, mientras que en la fase concéntrica, el VMO mostró mayor actividad que el VML con solo el 15 % de resistencia al BW. Los resultados indicaron diferencias significativas en el nivel de activación de los dos componentes de la VM. Esto tiene importancia en el desarrollo de ejercicios destinados a lograr una activación mayor o más selectiva del VMO. En la muestra sometida a evaluación, los registros EMG describen una mayor activación del VMO en comparación con el VML, que es más importante en las fases isométrica y excéntrica de la flexión / extensión de la rodilla en cadena cinética abierta. Estos hallazgos sugieren una compartimentación funcional de la VM.

ABSTRACT

Splicing factor Heterogeneous Nuclear Ribonucleoprotein A2/B1 (HNRNPA2B1 ) is associated with mouse lifespan and human longevity.It also plays a causal role in cancer development.However, whether it participates in cellular senescence, a biological process that contributes to individual aging and inhibits cancer, remains unknown.Here, we report that HNRNPA2B1 showed significantly increased expression in various cancer types while consistently decreased expression in multiple cellular senescence models.Knocking down HNRNPA2B1 in cancer cells leads to a series of senescence- associated phenotvpes.In line with its function as a splicing factor, HNRNPA2B1 downregulation causes alternative splicing changes in over one thousand genes, including those known to have a causal role in senescence.Our results also suggests that the E2F transcription factor 1 (E2F1 ) could regulate the expression of HNRNPA2BI, and E2F1-HNRNPA2B1 may be a new regulatory axis functioning in both cancer and cellular senescence, which might also have potential medical implications for cancer therapies.

Este artigo tem como objetivo abordar as transformações ocorridas no trabalho institucional do Lugar de Vida -Centro de Educação Terapêutica durante o período de distanciamento social acarretado pela pandemia do COVID-19. Retoma-se a história da instituição, ressaltando o caráter inventivo inerente à clínica psicanalítica com crianças com entraves estruturais na constituição psíquica. This article introduces the transformations that took place in the institutional work of Lugar de Vida -Centro de Educação Terapêutica during the period of social distance caused by the COVID-19 pandemic. The institution's history is presented, emphasizing the inventive character in psychoanalytic clinic with children with structural barriers in their psychic constitution. Clinical situations ilustrate the questioning and reformulation of therapeutic projects in this scenario and the way the institution continues with its work with children, family and school. For some children, it is considered that remote sessions function as a treatment of the Other. It is a new way of blocking and protecting the invasive or chaotic Other. Screens can be subjectivating devices if on one side there is an interlocutor-subject and, on the other side, a supposed-subject


Cet article traite des transformations qui ont eu lieu dans la pratique clinique du Lugar de Vida -Centro de EducaçãoTerapêutica, pendant la période de distanciation sociale à cause de la pandémie COVID-19. L'histoire de l'institution est citée, soulignant le caractère inventif de la clinique psychanalytique des enfants avec des barrières structurelles dans leurconstitution psychique. Situations cliniques déclenchent la reformulation des projets thérapeutiques et la manière dont l'institution poursuit son travail. Pour certains enfants, on considère que la séance à distance peut être une manière de traiter l'Autre, puisque ça possibilite des nouvelles façons de bloquer l'Autre invasif ou chaotique de l'enfant autiste ou psychotique. ABSTRACT

OBJECTIVE@#To explore the integration method and technical realization of artificial intelligence bone age assessment system with the hospital RIS-PACS network and workflow.@*METHODS@#Two sets of artificial intelligence based on bone age assessment systems (CHBoneAI 1.0/2.0) were developed. The intelligent system was further integrated with RIS-PACS based on the http protocol in Python flask web framework.@*RESULTS@#The two sets of systems were successfully integrated into the local network and RIS-PACS in hospital. The deployment has been smoothly running for nearly 3 years. Within the current network setting, it takes less than 3 s to complete bone age assessment for a single patient.@*CONCLUSIONS@#The artificial intelligence based bone age assessment system has been deployed in clinical RIS-PACS platform and the "running in parallel", which is marking a success of Stage-I and paving the way to Stage-II where the intelligent systems can evolve to become more powerful in particular of the system self-evolution and the "running alternatively".

ABSTRACT

Objective: To investigate the expressions of heterogeneous nuclear ribonucleoprotein I) (hnRNPD) and epithelial cell adhesion molecule (EpCAM) in the oral squamous cell carcinoma (OSCC) tissue and normal oral mucosa tissue, and to discuss the associations between their expressions and the clinicopathological parameters of the OSCC patients, and to elucidate their effects on the occurrence and development of OSCC. Methods: The tumor tissue of 38 OSCC patients without preoperative radiotherapy and chemotherapy (OSCC group) and the normal oral mucosa tissue of 11 patients with tooth extraction (control group) were selected. Immunohistochemistry staining was used to detect the expression levels of hnRNPI) and EpCAM in the tumor tissue and normal oral mucosa tissue. the expression levels of hnRNPD and EpCAM in tumor tissue of the OSCC patients with different clinicopathological parameters were analyzed∗ and the correlations between the expression levels of hnRNPD and EpCAM in tumor tissue of the OSCC patients were detected by Spearman correlation test. Results: Compared with control group, the expression levels of hnRNPD and EpCAM in the tumor tissue in OSCC group were increased (Z=-4. 936, P= 0.000; Z= 2.780, P=0. 005); there was statistically significant difference in the expression level of EpCAM in tumor tissue of the OSCC patients with different genders and ages (Z=-2.471, P= 0.013; Z=-1.967; P= 0.049), there was no significant difference in the expression level of hnRNPD in tumor tissue of the OSCC patients with different clinicopathological parameters ( P>0. 05). The correlation between the expression levels of hnRNPD and EpCAM was weak (p=0. 227, P=0. 17). Conclusion: The expression levels of hnRNPD and EpCAM in tumor tissue of the OSCC patients are up-regulated, and they promote the occurrence and development of OSCC.

Objective@#To investigate the role of human papillomavirus (HPV) 16 early genes E2 and E6 and heterogeneous nuclear ribonucleoprotein (hnRNP) E2 and their interaction effects in the progression of the cervical cancer.@*Methods@#Women with normal cervix (NC), low cervical intraepithelial neoplasia (CIN Ⅰ) and high cervical intraepithelial neoplasia (CIN Ⅱ/Ⅲ) from the cervical lesions cohort in Jiexiu County of Shanxi Province from June 2014 to September 2014, and patients with cervical squamous cell carcinoma (SCC) treated at the Second Hospital of Shanxi Medical University in the same period were enrolled in this study. There were 257 participants, about 67 NC cases (26.07%), 69 CIN Ⅰ cases (26.85%), 68 CIN Ⅱ/Ⅲ cases (26.46%), and 53 SCC cases (20.62%), respectively. The information of demographic characteristics, life health habits and cervical lesions were collected by using the structured questionnaire. Cervical exfoliated cells and cervical biopsy tissues were collected to detect the infection of HPV16 and the protein expression levels of hnRNP E2, HPV16 E2 and E6. According to the median-value of the protein expression levels of hnRNP E2, HPV16 E2 and E6 and E2/E6 ratio in the NC group, the study participants were divided into the high and low expression groups/ratio groups. The multivariate logistic regression model was used to analyze the correlation between HPV16 early gene E2 and E6, hnRNP E2 and cervical cancer. The interaction effect was analyzed by using the generalized multifactor dimensionality reduction (GMDR) model.@*Results@#The ages of NC, CIN Ⅰ, CIN Ⅱ/Ⅲ and SCC groups were (47.00±9.07), (47.64±7.35), (46.37±8.67) and (51.26±8.03) years old, respectively. The multivariate logistic regression model analysis showed that the HPV16 E2 low expression, E6 high expression and E2/E6 low ratio could increase the risk of CIN Ⅱ/Ⅲ, about OR (95%CI) values 11.11 (1.63-75.56), 8.00 (1.28-50.04), and 9.75 (1.22-77.72), respectively and SCC, about OR (95%CI) values 14.22 (2.11-95.88), 10.33 (1.67-64.00), and 12.38 (1.56-97.91), respectively. The hnRNP E2 low expression could increase the risk of CIN Ⅱ/Ⅲ and SCC, about OR (95%CI) values 3.35 (1.39-8.10) and 5.53 (1.54-19.88). The result of GMDR showed that there were interaction effects of the hnRNP E2 low expression, HPV16 E2 low expression and HPV16 E6 high expression in both CIN Ⅱ/Ⅲ and SCC groups.@*Conclusion@#The HPV16 E2 low expression, HPV16 E6 high expression and hnRNP E2 low expression could increase the risk of high-grade cervical intraepithelial neoplasia and cervical cancer, and they might have an important interaction effect in the progression of the cervical cancer.

ABSTRACT

Abstract The organic compound caffeine when detected in environmental matrices such as surface waters and groundwater is considered as an emerging contaminant, in which its effects are still unknown. Therefore, in the present research, zinc oxide-based catalysts impregnated with iron and silver were prepared for the reaction of caffeine degradation by heterogeneous photocatalysis. The wet impregnation method with excess solvent was applied to the preparation of the materials, later they were characterized by adsorption of N2, X-ray diffraction and photoacoustic spectroscopy. Then, the photodegradation, photolysis and adsorption tests were carried out, in which it was observed that only the presence of the radiation or photocatalysts could not sufficiently degrade the caffeine, however when combined radiation with all the catalysts studied here presented degradation above 70% at the end of 300 minutes of the reaction, and the best catalyst studied was that containing 8% Ag in non-calcined ZnO. Thus, these results point out that the methodology employed in this research, both for the preparation of the catalysts and in the process of the photocatalysis reaction, was efficient in the degradation of the emerging contaminant, caffeine, which could later be used for a mixture of other contaminants.

ABSTRACT

Abstract This work reports the study of the potential application of Zn/TiO2 catalysts, obtained by the sol-gel method, in processes of environmental decontamination through the reactions of photodegradation of textile dye, followed by electrospray mass spectrometry. The catalysts synthesis was performed according to a 2² factorial design with repetition at the central point. The characterization techniques used were: N2 adsorption measurements (BET method), scanning electron microscopy with energy dispersive X-ray (MEV/EDS), X-ray diffraction and point of zero charge (PZC). The photocatalytic tests were performed in batch in the presence of sunlight, and to evaluate the degradation kinetics study, a rapid direct injection electrospray mass spectrometry (DI-ESI-MS) method has been developed. By the photocatalytic tests, the calcination temperature of 400 °C has shown the best results of discoloration for the reactive Orange-122 dye (99.76%) in a reaction time of 2h. The discoloration kinetics were a pseudo-first order, and a statistical analysis was performed to investigate the effects of the variables and to optimize the conditions of discoloration to the dye. After the reactional time of 2 h, an ion of m/z 441.5 was detected by ESI-MS, indicating that the photocatalytic process was effective for the degradation of the dye to secondary compounds.

Resumen En el presente trabajo se evaluó la degradación fotocatalítica del herbicida glifosato en solución acuosa con un catalizador comercial (TiO2 Degussa P25) y un catalizador sintetizado a partir de TiO2 dopado con manganeso (TiO2-Mn), soportados en anillos de borosilicato de diámetro interno y externo de 6,52 mm y 7,59 mm, respectivamente, y una longitud aproximada de 9,43 mm, mediante el uso de un reactor continuo de lecho empacado. El catalizador fue caracterizado por FTIR, SEM-EDS y AFM, con lo cual se determinaron algunas propiedades físicas y químicas del mismo. Las condiciones de operación del reactor fueron un caudal de alimentación de 4,25 mL min-1 de una solución de glifosato de pH natural de 4,45 y un tiempo de retención de 1 h y 25 min, en el cual se llevaron a cabo ensayos de fotocatálisis heterogénea, fotólisis y adsorción por un tiempo de 150 min. De lo anterior, se obtuvieron los porcentajes de remoción y el orden de la reacción fotocatalítica para el catalizador soportado en los anillos. A partir de los estudios de degradación realizados, con el TiO2-Mn soportado en los anillos, se logró un porcentaje máximo de degradación de 39.19%, mientras que, con el catalizador comercial TiO2 Degussa P25, se alcanzó un 28.6% de remoción. El modelo de reacción que sigue la degradación del glifosato es de difusión intrapartícula, debido a los procesos difusivos en los que la molécula de glifosato es adsorbida en los poros del catalizador para luego ser degradada.


Abstract In this work, the photocatalytic degradation of the herbicide glyphosate in aqueous solution was evaluated. Assays were performed on a commercial catalyst (TiO2 Degussa P25) and a catalyst synthesized from TiO2 doped with manganese (TiO2-Mn) supported on borosilicate rings, with an internal and external diameter of 6.52 mm and 7.59 mm respectively, and an approximate length of 9.43 mm, using a continuous packed bed reactor. The synthesized catalyst was characterized with techniques as FTIR, SEM-EDS, and AFM, which allowed to evaluating its chemical and physical properties. The reactor operating conditions were a feed flow rate of 4.25 mL min-1 of a pH 4.45 glyphosate solution and retention time of 1 h and 25 min. In such experiments, heterogeneous photocatalysis, photolysis, and adsorption test were carried out for 150 min, obtaining results of degradation percentages and the order of photocatalytic reaction for the catalyst supported in the rings and the powder in suspension. From the removal studies, a maximum degradation percentage of 39.19% was reached with TiO2-Mn supported in the rings. In contrast, the commercial catalyst TiO2 Degussa P25 had a 28.6% of removal. The glyphosate degradation follows an intraparticle diffusion model due to a diffusive process, where the glyphosate molecule is adsorbed in the catalyst pores and then degraded.


Resumo Neste trabalho, a degradação fotocatalítica de glifosato em solução aquosa foi avaliada com um catalisador comercial (TiO2 Degussa P25) e sintetizado a partir de TiO2 dopado com manganês (Mn-TiO2) com suporte em anéis de borosilicato de diâmetro interno e externo de catalisador 6,52 mm e 7,59 mm, respectivamente e um comprimento de aproximadamente 9,43 mm, usando um leito empacotado reator contínuo. O catalisador sintetizado foi caracterizado com FTIR, SEM-EDS and AFM, o que permitiu a sua morfologia e composição. As condições de operação do reactor foi alimentada à velocidade de 4,25 mL min-1 de uma solução de pH natural de 4,45 glifosato e um tempo de retenção de 1 hora e 25 minutos; em que foram realizados ensaios de fotocatálise heterogénea, fotólise, tempo de adsorção de 150 minutos, obtendo-se como percentagens resultados de remoção e com a reação fotocatalítica para o catalisador suportado em anéis. A partir dos estudos, foi obtido uma taxa máxima de degradação com TiO2-Mn suportado em anéis de 39,19% em comparação com o catalisador comercial de TiO2 Degussa P25 com os quais obtiveram porcentagens de degradação de 28,6%. A degradação do glifosato segue um modelo de difusão intrapartícula devido ao processo difusivo em que a molécula de glifosato é adsorvida no catalisador poros logo a ser degradada.
