ABSTRACT

SUMMARY: Six Lonchura striata and six Copsychus saularis birds were selected in this study, morphological index of the small intestine was measured by quantitative biology and image analysis. The changes of goblet cells and Na+/K+ATPase were detected by AB-PAS staining and ELISA to inform the different mechanisms of the digestion and absorption of nutrients between the Lonchura striata and Copsychus saularis. The villus height, crypt depth and muscle thickness of each segment of small intestine of Lonchura striata were smaller than those of Copsychus saularis, and the difference of ileum muscle thickness was significant. In addition, the ileum villus height/crypt depth (VH/CD) value of Lonchura striata was significantly less than that of Copsychus saularis. The number of goblet cells in duodenum and jejunum of Lonchura striata and Copsychus saularis had no significant difference, but the number of goblet cells in ileum of Copsychus saularis was significantly larger than that of Lonchura striata. The vitality of Na+/K+-ATPase in different intestinal segments of the Lonchura striata and the Copsychus saularis was different. The vitality of Na+/K+-ATPase in the Lonchura striata was significantly higher than that of the Copsychus saularis. It can be concluded that the digestion and absorption capacity of Copsychus saularis and Lonchura striata are significantly different, and the reason may be due to their different diets and intestinal floras.

ABSTRACT

Objective To study the histomorphology structure of the femur in adult horses and adults, analyze the histological features and establish the method of identifying the species between humans and horses. Methods The 4 cm mid-diaphyseal segment of the right femur from adult human at autopsy was obtained. At the same time, the right femur of the horse was collected and the middle section was obtained about 4cm. After decalcification, a bone tissue section about 25 μm in thickness was obtained. Observe under an optical microscope, images under a microscope were input into a computer, and 25 indicators were selected for stepwise discriminant analysis. Results Significant differences between horses and human were observed on 13 indicators such as number of Haversian system and Haversian system diameter. Mathematical model for species identification was established based on these indicators. After a blind test, the discrimination reaches 99.6%. Conclusion Horse and human femur histological structure have obvious species characteristics and the established discriminant equation can effectively identify horses and human femur fragments.

ABSTRACT

Objective To explore effects of ephedrine on epinephrine (EPI) and estradiol( E2)concentration in blood, histological structures and activity of superoxide dismutase (SOD) and maleic dialdehyde (MDA) content in the ovary of female mice. Methods Pregnant female mice in the experimental group were treated with ephedrine (2g/L,4g/L, 6g/L), and the mice in the control group were treated with saline by intraperitoneal injection .Using ELISA method to detect concentration changes of EPI and E 2 in plasma, the changes in the enzymatic activity of SOD and MDA content were analyzed by colorimetry .The ovarian weight was measured , histological structures in ovary were observed by microscopy and the number of ovarian follicles were counted .Results EPI concentration in plasma of female mice increased , and the E2 concentration decreased dose-dependently in the experimental group .The activities of SOD were lower , and MDA content significantly increased compared to the control .The ovary weight of experimental groups was lower than the control group. The numbers of primary follicle , secondary follicle and atretic follicle increased and corpus luteum decreased in the experimental groups .There were pyknotic ovarian tissues , including oocyte ateophy , degradation granulose cells , and karyopyknotic granulose lutein cells in the experimental groups .Conclusion Ephedrine has a great effect on the ovarian histological structure and ovarian function of female mice , which may be correlated with the low activities of SOD .

ABSTRACT

Background Phlyctenular ophthalmia is a delayed hypersensitivity reaction to some microprotein and affected mainly by adolescent in high incidence. Objective This study was to investigate the microscopy findings of phlyctenular ophthalmia and evaluate the histological changes by laser scanning confocal microscope. Methods Twenty-nine eyes suffered from phlyctenular ophthalmia and twenty normal eyes were examined using laser scanning confocal microscope. The pictures were taken by a CCD camera. All the cases had initially chest X-ray,tuberculin test, bacterial and mycobacteria culture. Results Dendritic and inflammatory cells were increased and concentrated in conjunctiva, and epithelial cells were deformed and squamatizated. The capillaries engorged and the goblet cells were injured. The corneoscleral Vogt meshing of the phlyctenular keratitis was obscured and dendritic cells were intruded into the corneas. The corneal epithelium of phlyctenular keratitis was absent and the subepithelial nerve plexus were bended and fractured, and the dendritic and inflammatory cells were intruded.Scarring of corneal stroma was seen under the laser scanning confocal microscope. Conclusions Laser scanning confocal microscopy is valuable for basic research and clinical diagnosis of phlyctenular ophthalmia.

ABSTRACT

Objective Hermaphroditic planarians possess a very important position in the systematic evolutionary history of animal, as they are capacity of complete regeneration. Hence, the research on histological structure of autofluorescence has been carried out to provide a crucial insight into the developmental and regenerative biology. Methods Part of histological structure of planarian (Dugesia japonica) was revealed with HE method, Masson method and Van Gieson method. Their autofluorescence was observed with ultraviolet. There were six planarians in each stained group and the autofluorescence group. Results Epidermis, outer epidermis of pharynx, protonephridium, intestine, the photoreceptor cells and longitudinal nerve cords, all radiated blue autofluorescence. The epithelial dissociation side of copulatory bursa radiated yellow autofluorescence, its middle part radiated blue autofluorescence, its fundus side radiated weakly blue autofluorescence. Testis could hardly give off autofluorescence. Pigment cells of eyepot could not give off autofluorescence. Conclusion The research on configuration and autofluorescence of planarian eye may offer help for the study of origin and evolutionary law on eye of invertebrate.
