ABSTRACT

Objective:To explore the effect of music relaxation training combined with Huangqi Jianzhong Decoction oral administration and foot bath on fatigue status and quality of life in patients with cancer related fatigue.Methods:A total of 90 patients with cancer related fatigue in our hospital from January 2019 to October 2020 were selected. The patients were divided into 2 groups by random number table method, with 45 in each group. Both groups were given health and diet education. The control group was given music relaxation training, and the observation group took Huangqi Jianzhong Decoction oral administration and foot bathing on the basis of the control group. Both groups have a course of treatment of 4 weeks, a total of 3 courses. The TCM syndrome scores were observed before and after treatment, and the Piper Fatigue Score (PFS) was used to evaluate fatigue status, and the Karnofsky functional state score was used to evaluate the quality of life of patients, evaluation of clinical efficacy.Results:After treatment, 43 patients in the observation group and 42 patients in the control group were analyzed. The total effective rate of the observation group was 83.72% (36/43), which was significantly higher than 64.29% (27/42) of the control group ( χ2=4.18, P=0.041). After treatment, the TCM syndrome scores in the observation group was significantly lower than that of the control group ( t=2.48, P=0.015). After treatment, the Karnofsky functional state score in the observation group was significantly higher than that of the control group ( t=2.83, P=0.006). After treatment, the emotion score (4.35±1.02 vs. 5.03±1.01, t=3.09), feeling score (4.31±1.06 vs. 5.00±1.02, t=3.06) and total score (4.23±1.07 vs. 4.87±1.10, t=2.72) in the observation group were significantly lower than those in the control group ( P<0.01). Conclusion:Music relaxation training combined with Huangqi Jianzhong Decoction oral administration and foot bathing can alleviate the fatigue state of CRF patients and improve the quality of life.

ABSTRACT

OBJECTIVE: To study the difference of the contents of copper, lead, cadmium, mercury and arsenic in the single decoction and combined decoction of Huangqi jianzhong decoction (containing Astragali radix, Cinnamomi ramulus, Paeoniae radix alba, prepared Glycyrrhiza uralensis, Zingiberis rhizoma recens, Jujubae fructus), and to provide reference for safe use of TCM in clinic. METHODS: Samples of medicinal materials, single decoction and combined decoction were prepared from different medicinal materials; the contents of copper, lead, cadmium, mercury and arsenic in various samples were determined by atomic absorption spectrophotometry, which were compared with the limits of copper (<20 mg/kg), lead (<5 mg/kg), cadmium (<0.3 mg/kg), mercury (<0.2 mg/kg) and arsenic (<2 mg/kg) content stated in 2015 edition of Chinese Pharmacopeia and Green Trade Standards of Importing Exporting Medicinal Plants Preparations. The difference of 5 elements contents were compared between single decoction and combined decoction. RESULTS: Among the 6 medicinal materials, the cadmium content of Cinnamomi ramulus and Zingiberis rhizoma recens exceeded the standard, 0.32, 0.74 mg/kg, respectively (copper, lead, mercury and arsenic all met the requirements), and the 5 elements content of other medicinal materials all met the requirements; the contents of copper, lead, cadmium, mercury and arsenic in the single and combined decoction meet the requirements; compared with the medicinal materials, the contents of copper, lead, cadmium, mercury and arsenic in the single decoction were lower (P<0.01); compared with mixture of single decoction, the contents of copper and mercury in combined decoction were lower (P<0.01), while the contents of lead, cadmium and arsenic in the decoction were higher (P<0.01). CONCLUSIONS: For Huangqi jianzhong decoction, combined decoction can reduce the dissolution of copper and mercury, and increase the dissolution of lead, cadmium and arsenic. The possible reason is that the interaction of various components in the co-decoction process changes the solubility of copper, lead, cadmium, mercury and arsenic.

ABSTRACT

Huangqi Jianzhong Decoction is recorded in Synopsis of the Golden Chamber by Zhong-jing Zhang in the Eastern Han Dynasty. It consists of seven herbs including Astragali Radix, Cinnamomi Ramulus, Paeoniae Radix Alba, Jujubae Fructus, Zingiberis Rhizoma Recens, Glycyrrhizae Radix et Rhizoma and Saccharum granorum. Clinically, it is mainly used to treat peptic ulcer, duodenal ulcer, gastroesophageal reflux and other diseases. By reviewing the research literatures of Huangqi Jianzhong Decoction and its components in recent years, the main chemical constituents, metabolic pathways, metabolic reactions and metabolites of Huangqi Jianzhong Decoction and its components were summarized provide scientific reference for the material basis research of Huangqi Jianzhong Decoction.

ABSTRACT

Objective To address the protective role Huangqi Jianzhong Decoction (HQJZ) against chronic atrophic gastritis (CAG) in rats with metabolites in serum, and illuminate its regulative approaches to the targets. Methods CAG rat model was constructed by alternant administrations of ammonia solution and sodium deoxycholate, combined with the hunger disorder method. Rats were randomly separated into five groups: control group, model group, positive group (teprenone), low-dose group, medium-dose group, and high-dose group of HQJZ for continuous ig administration for four weeks. Body weight, biochemical indexes, and histopathological exam were used to evaluate the efficacy of HQJZ after the model replicated successfully. 1H NMR-based metabonomics was employed to analyze the plasma metabolic features of HQJZ deviated from CAG rats. Partial least square regression analysis (PLS-RA) and MetPA analysis were utilized to explore the relevant pathways and the underlying mechanism involved in HQJZ against CAG. Results The pharmacodynamic results demonstrated that HQJZ possessed beneficial activities in treating CAG, which partially ascribed to the improvement of gastric PA and antioxidant system in vivo. A total of 18 plasma metabolites were selected as the potential biomarkers related to the development of CAG, 10 out of them including 3-hydroxybutyrate, lactate, acetate, succinate, etc. were significantly regulated by HQJZ. Three regulation pathways: arginine and proline metabolism, glycerolipid metabolism, and glycine, serine and threonine metabolism, were recognized to be the most relevant efficacy of HQJZ against CAG based on PLS-RA and MetPA analysis. Conclusion The efficacy of HQJZ against CAG were ascribed to the improvement of the pathological changes due to its regulation to the energy imbalance, excessive oxidative stress, immune disorders, as well as inflammation.

ABSTRACT

Objective: To predict the active ingredients and their potential targets of Huangqi Jianzhong Decoction (HQJZ) against chronic atrophic gastritis (CAG) based on the “single drug-active ingredient-action targets” network. Methods: Using pharmacokinetic parameters [oral bioavailability (OB) and drug likeness (DL)] of traditional Chinese medicine system pharmacology platform (TCMSP) to screen the active ingredient of HQJZ, and then integrating target prediction database (STPD), human gene database (Genecard), and Online Mendelian Inheritance in Man (OMIM) to predict and screen its target genes for the treatment of CAG, and using Cytoscape software to construct the network of “single drug-active ingredient-action targets”. The molecular docking of those main active ingredient and action targets were confirmed through Systems Dock Web Site. String database and Cytoscape software were used to draw the protein interaction network, and DisGeNET database was used to assign their target type. Finally the biological function and metabolic pathway of these targets were analyzed by DAVID database. Results: A total of 118 potential active ingredients were screened based on their OB and DL parameters, and 16 CAG-related genes were involved, 52 active ingredients were related to disease target, which mainly involved in the regulations of cancer pathway, focal adhesion, arginine and proline metabolism, vascular endothelial growth factor signal transduction and neurotrophic factor signaling pathway in the treatment of chronic atrophic gastritis. Conclusion: The therapeutic mechanism of HQJZ reflects the characteristics of multi-component, multi-target, and multi-pathway of traditional Chinese medicines, which provides a scientific basis for further explaining the action mechanism and the material basis of HQJZ against CAG.

ABSTRACT

Objective To investigate the effect of modified Huangqi-Jianzhong decoction on healing quality of gastric ulcer in rats and to explore its mechanism. Methods 40 Wistar rats were randomly divided into a normal group, a model group, a modified Huangqi-Jianzhong decoction group and a omeprazole group (n=10) according to the random number table method. These rats were used to model of chronic gastric ulcer induced by acetic acid 100%. On third day after surgery, the rats in the modified Huangqi-Jianzhong decoction group were treated with 10% of the modified Huangqi-Jianzhong decoction according to the dose of 20 ml per kilogram of body weight. The rats in the omeprazole group were treated with a 3% omeprazole suspension according to the of 20 ml per kilogram of body weight. The rats in the normal group and the model group were treated with the same volume of saline. After treated with corresponding treatment for 16 days, the gastric mucosa pathology, ulcer index, regenerative mucosal thickness, mucosal surface mucus thickness, muscularis mucosal layer width of the defect, cystically dilated glands number and level of serum prostaglandin (6-K-PGF1α), epidermal growth factor (EGF), nitric oxide (NO) were observed. Results Compared with the model group, the rat ulcer index (8.95 ± 2.78 mm2 vs. 20.82 ± 5.12 mm2), mucosal muscularis defect width (123.56 ± 32.89 μm vs. 229.32 ± 49.69 μm), cystic glandular expansion (1.65 ± 0.76 vs. 6.12 ± 1.23) were lower in the modified Huangqi-Jianzhong decoction group (P<0.01). Thickness of regenerated mucosa (329.55 ± 32.22 μm vs. 198.22 ± 5.83 μm), the surface thickness of mucus (44.3 ± 2.8 μm vs. 24.5 ± 7.8 μm), serum 6-K-PGF1α (93.7 ± 8.9 pg/ml vs. 58.5 ± 5.8 pg/ml), EGF (2.11 ± 0.29 ng/L vs. 0.82 ± 0.20 ng/L) , NO (0.51 ± 0.03 μmol/L vs. 0.22 ± 0.05 μmol/L) was higher (P<0.01). In addition to the ulcer index, other each target improvement flavored were better in the modified Huangqi-Jianzhong decoction group than in the omeprazole group (P<0.05). Conclusion Modified Huangqi-Jianzhong decoction can improve the quality of ulcer healing by promoting gastric mucosa ulcer repair, reducimg ulcer index, mucous muscularis layer defect width and cystic glandular expansion and improving the thickness of regenerative mucosa, mucosal surface mucus thickness and cyst, EGF, NO level and thus improve the experimental gastric ulcer healing quality.

[Objective]To explore the change of serum gastrin and G-cell in gastric antrum in rat model of spleen-qi deficiency and the curative effect of huangqi jianzhong decoction(HQJZD).[Methods]Spleen-qi deficiency syndrome was induced in rats by compound methods.After the models had been established,they were treated by different dosages of HQJZD.And at different time points,serum xylose,creatine phosphokinase,gastrin and G cells were observed.[Results]These indexes in groups of different dosages of HQJZD recovered gradually.Especially after 21 days' treatment,in groups of the middle and the high dosages,these indexes were increasing significantly when compared with those in the model control group(P
