ABSTRACT

Whiteflies (Hemiptera: Aleyrodidae) are a diverse group of sap-sucking insect species that can cause severe damage in crops with global economic importance. Hops (Humulus lupulus L.) are a crop with considerable value for the brewing and pharmaceutical industries worldwide. The aim of the current study is to register whitefly species associated with this plant in Brazil. Specimens were collected from H. lupulus leaves in four different counties in Rio de Janeiro state (Cachoeiras de Macacu, Cordeiro, Nova Friburgo and Seropédica). Whiteflies belonging to nine genera and distributed in two subfamilies (Aleurodicinae and Aleyrodinae) were identified. Nine hop varieties (i.e., Brazylinsk, Cascade, Chinook, Columbus, Hallertau, Nugget, Saaz, Spalt, and Victoria) are used as host plants by these insects in Brazil. Except for Trialeurodes vaporariorum (Westwood), the other identified species were recorded in hop crops for the first time.

ABSTRACT

Objective To explore the effects of Humulus lupulus L. extract (HLE) and xanthohumol (XN) on preventing glucocorticoid-induced osteoporosis (GIOP). Methods The GIOP model was established by intraperitoneal injection of dexamethasone (DEX). Bone microstructure, bone mineral density and serum biochemical indexes were evaluated by Micro-CT and ELISA kits. The levels of cells proliferation and ALP activity, and the expression of bone formation related proteins were assayed with primary osteoblasts injured by DEX. Results HLE and XN significantly alleviated the bone microstructure damage, enhanced the bone mineral density, and improved the trabecular parameters in GIOP mice. In vitro experiments showed that HLE and XN can prevent bone loss not only by improving cell proliferation and ALP activity, but also through increasing the expression of bone γ-glutamic acid-containing proteins (BGP), bone morphogenetic protein 2 (BMP-2) and runt-related transcription factor 2 (Runx-2). Conclusion This study confirmed that HLE and XN had anti-GIOP effects for the first time. It provides a new resource for the development of anti-osteoporosis medications.

ABSTRACT

OBJECTIVE@#To systematically evaluate the protective effects of Humulus lupulus L. extract (HLE) on osteoporosis mice.@*METHODS@#In vivo experiment, a total of 35 12-week-old female ICR mice were equally divided into 5 groups: the sham control group (sham); the ovariectomy with vehicle group (OVX); the OVX with estradiol valerate [EV, 0.2 mg/(kg•d)] the OVX with low- or high-dose HLE groups [HLE, 1 g/(kg•d) and 3 g/(kg•d)], 7 in each group. Treatment began 1 week after the ovariectomized surgery and lasted for 12 weeks. Bone mass and trabecular bone mircoarchitecture were evaluated by micro computed tomography, and bone turnover markers in serum were evaluated using enzyme-linked immunosorbent assay (ELISA) kits. In vitro experiment, osteoblasts and osteoclasts were treated with HLE at doses of 0, 4, 20 and 100 µg/mL. Biomarkers for bone formation in osteoblasts and bone resorption in osteoclasts were analyzed.@*RESULTS@#Compared with the OVX group, HLE exerted bone protective effects by the increase of estradiol (P<0.05), the improvement of cancellous bone structure, bone mineral density (P<0.01) and the reduction of serum alkaline phosphatase (ALP), tartrate resistant acid phosphatase (TRAP), bone gla-protein, c-terminal telopeptides of type I collagen (CTX-I) and deoxypyridinoline levels (P<0.01 for all). In vitro experiment, compared with the control group, HLE at 20 µg/mL promoted the cell proliferation (P<0.01), and increased the expression of bone morphogenetic protein-2 and osteopontin levels in osteoblasts (both P<0.05). HLE at 100 µg/mL increased the osteoblastic ALP activities, and HLE at all dose enhanced the extracellular matrix mineralization (both P<0.01). Furthermore, compared with the control group, HLE at 20 µg/mL and 100 µg/mL inhibited osteoclastic TRAP activity (P<0.01), and reduced the expression of matrix metalloproteinase-9 and cathepsin K (both P<0.05).@*CONCLUSION@#HLE may protect against bone loss, and have potentials in the treatment of osteoporosis.

ABSTRACT

Objective To explore the effects of Humulus lupulus L. extract (HLE) and its mechanism on improving bone formation of Aβ-injured osteoblasts. Methods Osteoblasts isolated from 24 h-old Wistar rats were injured by Aβ1-42 oligomer and intervened with HLE. The proliferation, differentiation and bone mineralization of osteoblasts were determined by MTT assay, alkaline phosphatase (ALP) activity assay and alizarin red staining, respectively. The apoptosis of osteoblasts was detected by flow cytometer. The expression levels of bone formation related proteins, and proteins of Nrf2 and FoxO1 pathways were measured by Western blotting analysis. The intranuclear expression of FoxO1 protein was detected by immunofluorescence. Results HLE significantly improved the cell proliferation, ALP activity and bone mineralization, and inhibited the apoptosis of Aβ-injured osteoblasts. HLE also significantly promoted the expressions of collagen type Ι (COL-I) and osteopontin (OPN) in Aβ-injured osteoblasts. HLE notably activated the Nrf2 and FoxO1 signaling pathways in Aβ-injured osteoblasts by promoting the expressions of related proteins and maintained bone metabolism through relieving oxidative stress. Conclusion This study confirms that HLE can alleviate Aβ-injury to osteoblasts, and preliminarily clarifies the mechanism being related to antioxidation, which provides a new reference for the mechanism research and drugs development for anti-osteoporosis.

ABSTRACT

OBJECTIVE:To establish HPLC fingerprint of Humulus lupulus ,and to investigate its correlation with the antioxidant activity. METHODS :The determination was performed on Diamonsil C 18 column with mobile phase consisted of 0.1% phosphoric acid solution-acetonitrile (gradient elution )at the flow rate of 1.0 mL/min. The column temperature was 30 ℃,and detection wavelength was set at 358 nm,with sample size of 2 μL. Using xanthohumol as reference,HPLC fingerprints of 11 batches of H. lupulus were determined. The similarity of 11 batches of samples was evaluated by TCM Chromatographic Fingerprint Similarity Evaluation System (2012 edition)to confirm common peaks. Using scavenging rate of DPPH and ABTS free radical as pharmacodynamic indicators of antioxidant effects ,SIMCA 14.1 analysis software was used for PLSR to establish the spectra-effect relationship ,and validation test of in vitro anti-oxidation was carried out. RESULTS :There were 19 common peaks in HPLC fingerprints of 11 batches of sample ,the similarity of which was higher than 0.830. Seven components were identified as xanthohumol,cohumulone,humulone,adhumulone,colupulone,lupulus and adlupulus. Eleven batches of H. lupulus had the ability to scavenge DPPH and ABTS free radicals. The spectrum-effect relationship showed that xanthohumol ,humulone and colupulone peaks were positively associated with its anti-oxidant ability ,and variable projection value was greater than 1. In vitro antioxidant results showed that scavenging effect of 0.1 mg/mL xanthohumol to DPPH free radicals was similar to that of 0.01 mg/mL vitamine C ,but scavenging effect of 10.0 mg/mL xanthohumol to ABTS free radical was less than that of 0.1 mg/mL vitamin C. CONCLUSIONS:Established HPLC fingerprint can be used for the quality evaluation of H. lupulus ;xanthohumol,humulone and colupulone are the main material basis for the antioxidant effect of H. lupulus

ABSTRACT

Humulus lupulus is a kind of special resource plant used both as medicine and food in Xinjiang. In addition to being widely used in beer brewing, Humulus lupulus has long been recognized for its medicinal value, especially for the treatment of postmenopausal osteoporosis. Modern pharmacological research shows that its active components have great potential in the development of anti-osteoporosis drugs. However, in recent years, the wild Humulus lupulus resources in China have been seriously degraded, and the contents of active components are quite different. Ensuring high-quality Humulus lupulus germplasm resources is a prerequisite for the development and utilization of Humulus lupulus. This paper reviews the major chemical components of Humulus lupulus and their effects and application in the prevention and treatment of osteoporosis, and discusses the existing problems, aiming to provide a reference for the development and utilization of Humulus lupulus against osteoporosis.

The Hop plant (Humulus lupulus L., Cannabinaceae) is a dioeciou twining perennial.The relevant papers at home and abroad in recent years were reviewed and a summary analysis and induction were conducted to research the progress of hops and provide a basis for further development and application of the medicinal materials.Pharmacological effects of hops were reviewed, which found that hops have a good research prospect and deserve further promotion and development.

ABSTRACT

OBJECTIVE@#To investigate the impact of Humulus Lupulus L. hydroalcoholic extract on the body weights, reproductive organs, sperm quality and hormone levels in male rats.@*METHODS@#By simple random sampling method, seventy male Sprague-Dawley rats were randomly assigned to 7 groups including control group [distilled water, 1 mL/(kg•d)], Tween 80 group [25% Tween 80 solution, 1 mL/(kg•d)], olive oil group [olive oil, 1 mL/(kg•d)], diethyl stilbestrol (DES) group [DES, 100 μg/(kg•body weight)], H50, H150 and H450 [50, 150 and 400 mg/(kg•d) of Humulus Lupulus L extract, respectively]. The administration was performed via gavage once daily for 7 weeks. Body and reproductive organs weights including testes, seminal vesicles, epididymis and prostate were weighted and epididymal sperm quality were determined by digital balance. Blood samples were collected and serum free testosterone (T), luteinizing hormone (LH), follicle-stimulating hormone (FSH), and estrogen (E2) levels were measured by rat specific enzyme-linked immunosorbent assay.@*RESULTS@#The percentage increase in mean body weights of rats in the DES and H50, H150 and H450 groups decreased significantly compared to olive oil and Tween 80 groups (all P0.05).@*CONCLUSION@#Humulus Lupulus L. extract significantly increased the seminal vesicle and testes weights and reduced the sperm motility.

ABSTRACT

Objective To determine the contents of total flavonoids and xanthohumol in hop from 29 different countries and regions .Methods Rutin colorimetric method was used to determine the content of total flavonoids .HPLC-UV method was established for the determination of xanthohumol in hops .HPLC method was performed by Dikma Technologies Diamonsil C18 (250 mm × 4 .6 mm ,5 μm ) column with mobile phase acetonitrile-1% glacial acetic acid solution at the flow rate of 1 .0 ml/min .The column temperature was 25 ℃ .The detection wavelength was 370 nm .Results The equation of linear re-gression of total flavonoids was A=30 .345C+0 .0168 ,r=0 .9999 .The equation of linear regression of xanthohumol was A=55446 C+9040 .5 ,r=0 .9999 .Their linear ranges were respectively 20 .2-404 .0 μg/ml ,2 .152-43 .040 μg/ml ,which indica-ted a good linear relationship .The RSDs of precision and repeatability were less than 2% .The average recoveries of flavonoids and xanthohumol were respectively 102 .71% and 100 .21% .Conclusion The contents of total flavonoids and xanthohumol in different hops varieties are significantly different and the import hops was better than the domestic hops in this study .

Objective: To investigate the effects of Humulus lupulus extract on the function of primary brown preadipocyte and its mechanisms, and try to provide new treatment for obesity and type 2 diabetes. Methods: Primary brown adipocytes in newborn rats were cultivated in vitro. After adding different doses of H. lupulus extract, MTT method was used to detect the cell proliferation, and oil red O staining method was used to analyze the lipid droplets consumption rate of H. lupulus extract on the brown adipocyte. The proteins expression of brown adipocytes (such as UCP1, p38αMAPK, and PGC1α) in different groups was detected by Western blotting. Results: The cell proliferation of primary brown preadipocytes was not effected by H. lupulus extract. Compared with control group, with the increasing dose, the lipid droplets consumption rate of H. lupulus extract on the brown adipocyte was increased. The expression of UCP1, p38αMAPK, and PGC1α was upregulated compared with the control group. Conclusion: H. lupulus extract may improve the heat production of brown adipocytes, and strengthen the energy metabolism. The possible mechanism may via enhancing p38MAPK-PGC1α-UCP1 cascade reaction, sequentially increasing the expression of brown fat specificity protein UCP1.

ABSTRACT

PURPOSE: Humulus japonicus pollen (Hop J) is a major cause of inhalant allergy in autumn of the Far East countries, and its allergenic potency has been increasing with climate changes. Allergen immunotherapy has been considered in Hop J-sensitized allergic patients; however, Hop J allergen extracts for immunotherapy are not commercially available. We speculate that Humulus lupulus pollen (Hop L) belonged to the same genus may share cross-reacting allergens with Hop J and evaluated allergenic relationships between these 2 pollens. METHODS: Thirteen patients with allergic rhinitis and/or asthma sensitive to Hop J pollens were enrolled in Ajou University Hospital, Suwon, Korea. Hop J pollens were collected locally and lyophilized extracts were prepared, while lyophilized Hop L extracts were provided by Lofarma S.p.A. IgE-ELISA/enzyme-linked immunosorbent assay (ELISA) inhibition tests, sodium dodecyl sulphate-polyacrylamide gel electrophoresis and IgE-immunoblot/immunoblot inhibition analysis using sera from the enrolled subjects were performed. RESULTS: All patients had high serum specific IgE to both Hop J and Hop L extracts by ELISA, but no significant correlation was found between these 2 extracts. ELISA inhibition tests showed significant dose-dependent inhibitions on IgE-bindings to Hop L with serial additions of Hop J extracts in a dose-dependent manner, while minimal inhibitions of IgE binding to Hop J were noted with additions of Hop L. IgE-immunoblot analysis demonstrated that the major allergenic component of Hop J at 12 kDa was inhibited by Hop J, while no inhibitions were noted by Hop L extracts on IgE-immunoblot inhibition analysis. CONCLUSION: These findings suggest that there may not be a significant cross-allergenicity between Hop J and Hop L.

Hops, the female inflorescences of the hop plant (Humulus lupulus), are widely used in the brewing industry to add bitterness and aroma to beer. Combining with the relevant literature, the chemical composition(resinae, volatile oil, polyphenol and polysaccharide) in hops and their pharmacological effects are reviewed in this paper so as to present some sights for further application research and development.

ResumenObjetivo:Analizar la relación del consumo de cerveza sin alcohol (fuente de antioxidantes) con los valores del perfil de lípidos de un grupo de adultos del cantón San Mateo de Alajuela durante 35 días.Métodos:La condición nutricional y el perfil lipídico de los adultos, que participaron en el presente estudio se obtuvo mediante una anamnesis nutricional; Esto incluyó, en primer lugar, una evaluación antropométrica. En segundo lugar, el análisis sanguíneo de los perfiles lipídicos tanto antes como después del consumo de una cierta cantidad de cerveza no alcohólica (0,5 l por día) durante un período de 35 días; Finalmente, una dieta basada en los hábitos de alimentación y su valor nutricional.Resultados:Los sujetos estudiados presentaron varios factores de riesgo asociados con el desarrollo de enfermedades cardiovasculares, sobresalieron: el sobrepeso, la obesidad, el consumo excesivo de grasas y carbohidratos simples, deficiencia de frutas, vegetales, alimentos integrales, y el sedentarismo. El consumo moderado de cerveza demostró mejorar los valores de Colesterol HDL en la mayoría de los participantes; por el contrario, el Colesterol Total, Colesterol LDL y Triglicéridos presentaron variaciones.Conclusión:La investigación evidenció mejorías en algunos indicadores del perfil de lípidos; sin embargo, se necesita investigar y ahondar más en el efecto de un consumo moderado de cerveza sin alcohol en la salud de grupos más disciplinados y controlados.


AbstractObjetive:To analyze the relation between the consumption of nonalcoholic beer and nourishment as well as the lipid profile values of a group of adults from the canton of San Mateo, Alajuela in a 35-day period.Methods:The nutritional condition and the lipid profile of the adults, that took part in the current study gotten by means of a nutritional anamnesis; this included, first, an anthropometric evaluation. Second, the blood analysis of lipid profiles both before and after the consumption of a certain amount of nonalcoholic beer (0.5 l per day) during a 35 day- period; finally, a diet based on feeding habits and their nutritional value.Results:The studied people presented several risk factors associated with the development of cardiovascular illnesses; among others, overweight, obesity, nourishment eating habits that promote an excessive consumption of fat and simple carbohydrates, a deficient consumption of fruits, vegetables, whole-grain sources, and sedentary lifestyle. A moderate consumption of nonalcoholic beer showed a development in HDL cholesterol in most patients; as opposed, the global cholesterol, the LDL cholesterol and triglycerides showed given variations related to the cardiovascular risk factors of patients under study.Conclusion:This research showed evidenced of gan improvement in the lipid profile indicators; however, a further investigation is required for testing the effects of moderated consumption of nonalcoholic beer in a more disciplinary and medicine-controlled population of adults. Besides, a relationship between good eating habits and physical activities to normal lipid profiles was demonstrated

ABSTRACT

Rho iso-alpha acids-rich extract (RIAA) from Humulus lupulus (hops) and proanthocyanidins-rich extracts (PAC) from Acacia nilotica exert anti-inflammatory and anti-diabetic activity in vitro and in vivo. We hypothesized that a combination of these two extracts would exert enhanced effects in vitro on inflammatory markers and insulin signaling, and on nonfasting glucose and insulin in db/db mice. Over 49 tested combinations, RIAA:PAC at 5:1 (6.25 microg/mL) exhibited the greatest reductions in TNFalpha-stimulated lipolysis and IL-6 release in 3T3-L1 adipocytes, comparable to 5 microg/mL troglitazone. Pretreatment of 3T3-L1 adipocytes with this combination (5 microg/mL) also led to a 3-fold increase in insulin-stimulated glucose uptake that was comparable to 5 microg/mL pioglitazone or 901 microg/mL aspirin. Finally, db/db mice fed with RIAA:PAC at 5:1 (100 mg/kg) for 7 days resulted in 22% decrease in nonfasting glucose and 19% decrease in insulin that was comparable to 0.5 mg/kg rosiglitazone and better than 100 mg/kg metformin. RIAA:PAC mixture may have the potential to be an alternative when conventional therapy is undesirable or ineffective, and future research exploring its long-term clinical application is warranted.

ABSTRACT

Humulus lupulus L., Cannabaceae, é usada como sedativo e ansiolítico na medicina popular. O método de HPLC-DAD-ESI-MSn representa uma ferramenta poderosa para a análise de produtos naturais, desde que ela fornece o espectro de UV e informações estruturais sobre os constituintes da mistura. O objetivo deste trabalho foi o de caracterizar os constituintes encontrados no extrato hidroalcoólico. Os constituintes 1-9 foram tentativamente caracterizados através do UV/DAD e ionização por electrospray (MS/MS) depois da separação usando fase reversa, tempo de retenção e dados da literatura. Os principais compostos fenólicos (baseados na área dos picos) foram caracterizados como ácido hulupínico (9), coulupona (8), dois alfa-ácidos amargos oxidados (principais constituintes), um deles sendo um derivado da coumulinona oxidada (5) e o outro um derivado da humulinona oxidada (7), junto com uma procianidina B (3) e os flavonoides rutina (4) e o canferol-7-O-rutinosídeo (6). Esta planta conhecida devido às suas propriedades ansiolíticas e por ser um componente da cerveja, mostrou derivados oxidados de alfa-ácidos, como principais constituintes do extrato hidroalcoólico.


Humulus lupulus L., Cannabaceae, is commonly used as light sedative and anxiolytics in folk medicine. HPLC-DAD-ESI-MSn represents a powerful tool for the analysis of natural products, since it can simultaneously provide a UV chromatogram and significant structural information about compounds in complex mixture. The aim of this work was characterize the constituents present in hydroethanolic extract. Compounds 1-9 were tentatively characterized on the basis of UV, MS/MS, after reversed phase separation, retention time and literature data. The main phenolic compounds (based on peak area) were characterized as hulupinic acid (9), cohulupone (8), two oxidized hop alfa-bitter acids (principal constituents), one being a oxidized cohumulinone (5) and the other an oxidized humulinone (7) derivatives, together with a procyanidin dimer B (3), flavonoids rutin (4) and kaempferol-7-O-rutinoside (6). This plant known, due to anxiolytic property and beer flavoring, showed oxidized hop bitter acids, as principal constituents, in its hydroethanolic extract.
