ABSTRACT

OBJECTIVE: To investigate the effect of Wuling capsules on the activities of IL-2 and IL -6 in healthy mice and mice with immunological liver damage .METHODSBALB/C mice were treated with LPS(experimental group)or were not treated (control group) .12 days after giving Wuling capsule ig, the suspension of spleen cells of mice was cultured for 48h and 72h and the supernant fluid was collected for test.The IL - 2 was detected with MTT and IL - 6 with radio - immunoas-say. RESULTS: Wuling capsules could promote Th cells to secret IL- 2 and markedly elevate the liver - damage - induced low level of IL- 2.Wuling capsules did not effect the levels of IL-6 significantly in both healthy mice and mice with liver damage. CONCLUSION: Wuling capsules can promote the secretion of IL-2 and have immunoregulatory actions.

ABSTRACT

The results of the current studies demonstrated that the combined use of IL - 2 and IL - 7 could augment the in vitro proliferative responses of tumor - specific T cell lines and clones to antigen stimulation, increasing stimulation induces 6 to 8 times greater than using either IL - 2 or IL - 7 alone. Antigen - driven T cells maintained in culture using this combined cytokine regimen can be induced to grow and maintained functional in large numbers and survive long-term in cultrue with each antigen restimulation cycle prolonged to six weeks. The IL-2 doses used in this combined cytokine regimen can be reduced 10 to 100 times that of cultures using IL-2 alone. Thus, the combined use of IL-2 plus IL-7 is effective for procuring large numbers of antigen - specific functional T cells in vitro.

ABSTRACT

The Renca-bearing mice were treated by combined fibroblast-mediated IL-2 and G-CSF gene therapy. The survival periods of Renca-bearing mice were prolonged markedly and 75% of Renca-bearing mice were cured. The spleens of Renca-bearing mice treated by fibroblast-mediated G-CSF gene therapy were larger than the spleens of other mice. Histologic analysis of tumor showed that there were a number of infiltrating eosinophils. The NK, LAK and CTL activity induced from the splenocytes of the tumor-bearing mice increased siginificantly when the mice were treated by fibroblast-mediated IL-2 gene therapy and the cytotoxicity of macrophages also increased. The results demonstrated that the and - tumor responses might be induced more significantly by combined fibroblast-mediated IL-2 and G-CSF gene therapy and better antitumor effect may be achieved.

ABSTRACT

Enhancing effect of gin-senosides (GS) on immune function indirectly via hippocampus (HC was studied in this paper. The main results indicated that after the injection of GS into bilateral HC of rat directly by implantation at the dose of 5?10~8 kg per day for 4 days, splenocyte proliferative response to ConA was enhanced significantly as compared with control group. But after injection of GS into abdominal cavity with the same dose, no significant change of splenocyte proliferation to ConA was found. Be-sides , our data also indicated that GS, injected into HC, could promote NK cell activity, increase proliferative response of spleen B lymphocytes to SAC bacteria, enhance IL -2 production and IL -2R expression of spleen T lymphocytes. The study showed that GS could promote indirectly the immunce function of spleno-cytes via HC.
