ABSTRACT

Objective To investigate the anti-inflammatory effects and mechanism of durian peel extracts (DPE). Methods The in vivo anti-inflammation effects of DPE were examined by carrageenin-induced mice paw edema test and allergic contact dermatitis test induced by 2, 4-DNFB. And the in vitro anti-inflammation effects of DPE were evaluated with methyl thiazolyl tetrazolium ( MTT) assay in RAW 264.7 cell model of inflammation induced by lipopolysaccharide (LPS). Results The results of animal experiments showed that DPE groups could markedly relieve mice paw edema induced by carrageenin ( P<0.01 or P<0.001 compared with blank group). DPE could effectively inhibit the allergic contact dermatitis induced by 2, 4-DNFB in mice, showing good dose-effect relationship. The results of in-vitro test showed that DPE at the given concentrations had no influence on RAW 264.7 cell proliferation. Tumor necrosis factor alpha ( TNF-α) , interleukin 6 ( IL-6) , interleukin 1 beta (IL-1β), nitric oxide (NO) and nuclear factor kappaB (NF-кВ ) were observably inhibited, and anti-inflammatory cytokine IL-10 was enhanced by 25 and 50 mg/L of DPE. Conclusion DPE exert potential anti-inflammation effect, and the mechanism might be related to its inhibition of NF-кВsignal pathway.

ABSTRACT

【Objective】 To investigate the in-vitro anti-inflammatory mechanism of luteolin by observing the effect of luteolin on nuclear factor-kappa B(NF?B) and COX-2 expression as well as DNA-binding activity of NF-?B in RAW264.7 cells activated by lipopolysaccharides(LPS).【Methods】RAW264.7 cells at logarithmic growth phase were allocated to blank control group,model group(treated with LPS) and luteolin groups(treated with 5,15,and 45 ?mol/L luteolin respectively).After being treated with luteolin for 30 min and then incubated with 1?g/mL LPS for 12 hours,the change of prostaglandin E2(PGE2) level was observed by enzyme immunoassay(EIA),DNA-binding activity of NF-?B was detected by electrophoretic mobility shift assay(EMSA),mRNA expression of COX-2 was examined by reverse transcription polymerase chain reaction(RT-PCR),and NF-?B and COX-2 protein expression in RAW264.7 cells was analysed by Western blotting.【Results】Luteolin obviously inhibited the formation of PGE2,decreased DNA-binding activity of NF-?B and down-regulated mRNA expression of COX-2 as well as NF-?B and COX-2 protein expression in RAW264.7 cells activated by LPS.【Conclusion】The antiinflammatory mechanism of luteolin may be related with the down-regulation of COX-2 expression by inhibiting the expression of NF-?B and DNA-binding activity.

ABSTRACT

[Objective] To explore the anti - inflammatory mechanism of sinomenine, a monomer alkaloid from Caulis Sinomenii. [ Methods] In - vitro enzyme was applied to observe the effect of sinomenine on cyclooxygenase (COX) 1 and COX 2. [Results] Sinomenine at the concentration of 1 - 250 ?mol/L could inhibit the formation of PGE2 induced by COX 2 in a good dose - effect manner. [Conclusion] Sinomenine selectively inhibiting the activity of COX 2 may be the anti - inflammatory and anagesic mechanism of sinomenine with less gastrointestinal adverse effect.

ABSTRACT

【Objective】The effect of sinomenine on human cyclo-oxygenase(COX) and interleukin-1?(IL-1?) was observed to explore the anti-inflammatory mechanism of sinomenine.【Methods】Peripheral blood monocytes(PBMC) were isolated and incubated with sinomenine.After incubation,total RNA from PBMC was extracted and the effect of sinomenine on COX and IL-1? expression with or without the stimulation of lipopolysaccharide(LPS) was observed by the method of semi-quantitative reverse transcription polymerase chain reaction(RT-PCR).【Results】Sinomenine had no effect on COX-1 gene expression,had a slight inhibitory effect on COX-2 gene expression and had a strong inhibitory effect on IL-1? gene expression.【Conclusion】 The anti-inflammatory mechanism may be related to the inhibition of inflammatory cytokines.

ABSTRACT

【Objective】To observe the effect of different active ingredients from Chinese herbal medicine on interleukin 8(IL-8) secretion and Toll-like receptor 4(TLR4) mRNA expression in human intestinal tumor cell(HT-29)line.【Methods】Reverse transcription polymerase chain reaction(RT-PCR) was used to detect TLR4 mRNA expression,and enzyme-linked immunosorbent assay(ELISA) was used to examine IL-8 secretion.【Results】Astragaloside and emodin inhibited TLR4 mRNA expression stimulated by interferon ?(IFN-?),and emodin decreased IL-8 secretion stimulated by IFN-? and lipopolysaccharide(LPS).However,curcumin,naringin and jasminoidin had no effect on TLR4 mRNA expression.【Conclusion】The anti-inflammatory cellular mechanism of emodin may be related to the inhibition of TLR4 mRNA expression stimulated by IFN-?,and to the inhibition of IL-8 secretion stimulated by IFN? and LPS in HT-29 cell line.

ABSTRACT

Objective To explore the main pharmacological actions of Penyan Qing Capsule (PQC) for chronic pelvic inflammation. Methods The analgesic action of PQC was studied by mouse hot_plate method. Experiments of mouse auricular swell ing and rat agar_induced granuloma were carried out to investigate the anti_infl ammatory action of PQC. Effects of PQC on function of mouse mononuclear phagocyt es and rat mesenteric microcirculation were also studied. Results PQC increa sed the rat pain threshold, reduced mouse auricular swelling induced by xylene a nd the weight of rat agar_induced granuloma. PQC could also improved rat mesente ric microcirculation disorder induced by noradrenline and increase the function of mouse mononuclear phagocytes. Conclusion PQC exerts good anti_inflammatory and analagisic action and can improve microcirculation. This will supply pharm acodynamic evidence for the application of PQC in clinic.

ABSTRACT

Objective To investigate the therapeutic effects of matrine eye drops (MED) on experimental ocular inflammation in rabbits. Methods The effects of 0. 5 % and 1.0% MED, dexamethasone eye drops, saline solution were e-valuated in the rabbits with ocular inflammation induced by paracentesis of anterior chamber and capsicum tincture respectively. Results MED could significantly inhibited the increase of protein concentration in aqueous humor induced by anterior chamber paracentesis, and the anti - inflammatory effect of MED on capsicum - tincture - induced extra ocular and intraocular inflammation was similar to dexamethasone eye drops. There were no significant difference between 1.0% MED and dexamethasone eye drops. Conclusion MED has a good effect on experimental ocular inflammation in rabbits.
