ABSTRACT

Objective To assay Icariine, Epimedin C, asperosaponin Ⅵ, psoralen and angelicin in Xianlinggubao capsules via multi-wavelength HPLC method.Methods Separation was carried out on Welch Ultimate○R XB-C18 column.The mobile phase was acetonitrile-water system and a linear gradient elution was used.The column temperature was 30 ℃.The detection wavelength for Icariine, Epimedin C,asperosaponin Ⅵ was set at 212 nm, psoralen and angelicin at 246 nm.Results Five components reached baseline separation, the linearity was good when sample size was in the range of 0.008 2-0.328 μg for Icariine(r=0.999 5), 0.055 6-2.224 μg for Epimedin C (r=0.999 6), 0.144 1-5.764 μg for asperosaponin Ⅵ(r=0.999 6), 0.005 4-0.215 2 μg for psoralen(r=0.998 0), 0.006 6-0.265 6 μg for angelicin(r=0.998 5).The average recoveries were 97.59%, 98.58%, 98.11%, 97.86%, 98.22% respectively.The RSDs of recovery were all less than 2.0%.Conclusion This method is simple, accurate, with good separation, high sensitivity for the assay of multiple components in Xianlinggubao capsule.

ABSTRACT

Objective: To develop a method for controlling the quality consistency of Qianlie Shule Capsule (QSC) by blending raw marerials. Methods: Blending raw materials were blended by medicine calculator V2. The content differences of epimedin C and icariine in QSC made from raw materials and their mixtures were compared. Results: The RSD of index component epimedin C from blended and raw materials were 11.2% and 82.5%, the RSD of icariine were 8.70% and 35.5%, the difference between index components decreased obviously, and it showed that the quality consistency in QSC was greatly increased. Conclusion: Using the method to control the quality coherence of Chinese patent medicine has the obvious results, it is easy to operate and suitable for actual production.

ABSTRACT

Objective To study the biological effect of Icariine on human umbilical cord derived mesenchymal stem cells(huCMSCs) and inducing them to differentiate into osteoblasts. Methods MSCs were isolated and expanded in vitro and their surface antigens of huCMSCs were detected by immunohistochemistry. HuCMSCs were assigned into two groups. In the blank control group, huMSCs were incubated in basic medium. In the experimental group huMSCs were incubated in Icariine medium, The proliferation and differentiation of huMSCs were examined by inverted microscope, Alkaline phosphatase (ALP) staining and the determination of ALP activities. Results HuCMSCs were strongly positive for CD44, and negative for CD34; strongly positive expression for ALP and calcium deposition was detected from the second day to the fourth day in Icariine group; blank control group and Icariine group had significant differences (P < 0.05).Conclusion HuCMSCs can be successfully cultured from the adherent tissue pieces, Icariine enhances proliferation and osteogenic differentiation of huCMSCs and be used as an osteoinductive factor.

ABSTRACT

OBJECTIVE:To establish an HPLC method for the determination of Icariine in Qianlietong tablets.METHODS:Hypersil ODS(200 mm?4.6 mm,5 ?m) column was used for the HPLC determination,and the mobile phase consisted of acetonitrile-water(30∶70) at a flow rate of 1.0 mL?min-1.The detection wavelength was 270 nm.RESULTS:Icariine has good linear relationship in the range of 0.050 6~0.506 0 ?g(r=0.999 9).The average recovery was 102.67%(RSD=0.99%,n=6).CONCLUSION:The established method is simple,reproducible,specific and suitable for the quality control of Qianlietong tablets.

ABSTRACT

AIM: To determine quantitatively active ingredients,icariine and osthole,and to identify Fructus Cnidium and Scorpio in Ruxinan Capsule ( Folium Epimedii,Fructus Cnidii,Scorpio). METHODS: Fructus Cnidium and Scorpio were identified by TLC and the contents of icariine and osthole were determined by HPLC. RESULTS: Fructus Cnidium and Scorpio were clearly separated in the TLC chromatogram. The results of HPLC analysis for icariine were: a linear range of 61. 7 ng-740. 4 ng,r = 0. 999 6,an average recovery of 99. 86% with a RSD of 0. 82% and the corresponding data for osthole were: a linear range of 57. 4 ng-688. 8 ng,r = 0. 999 8, an average recovery of 99. 55% with a RSD of 0. 71% . CONCLUSION: This method is simple and feasible and can be used as quality standards for Ruxinan Capsule.

ABSTRACT

AIM: To establish the quality standard of Gusongbao Dispersible Tablets(Herba Epimedii,Radix Dipsaci,Radix Pasoniae rubra,Rhizoma Chuanxiong,etc.) METHODS: Herba Epimedii,Radix Paeoniae rubra,Rhizoma Chuanxiong,Rhizoma Anemarrhenae were identified by TLC.Icariine content was determined by HPLC.C_18 column(ODS,4.6 mm?200 mm,5 ?m) as chromatographic column,acetonitrile-water(30∶70) as mobile phase,flow rate kept to 1.0 mL/min,detection wavelength at 270 nm,and under room temperature condition.The dissolubility test met with Chinese Pharmacopoeia for Icariine. RESULTS: The relevant spots in Herba Epimedii,Radix Paeoniae rubra,Rhizoma Chuanxiong,Rhizoma Anemarrhenae could be identified by TLC.The content of icariine can be determined by HPLC.The linearity of icariine was good in the range of 0.13-0.78 ?g(r=0.999 9).The average recovery of icariine was 98.90%,RSD=0.92%(n=6).Dissolubility test used for icariine coformed to homogeniety. CONCLUSION: The established quality standard is simple,feasible and repeatable,and can be used for quality control of Gusongbao Dispersible Tablets.
