ABSTRACT

OBJECTIVE: To study the improvement effect and mechanism of Chuanju zhitong capsules (CZC) on migraine model rats. METHODS: SD rats were randomly divided into normal group, model group, chemical drug positive control group (Zolmitriptan tablet, 0.004 05 g/kg), TCM positive control group (Compound yangjiao capsule, 4.32 g/kg), CZC high-dose, medium-dose and low-dose groups (1.6, 0.8, 0.4 g/kg), 10 rats in each group. All rats were relevant medicine intragastrically once a day, for successive 5 days. 30 min after last medication, except for normal group, rats in other groups were given glyceryl trinitrate subcutaneously (10 mg/kg) via head and neck to induce migraine model. The behavior changes of rats were evaluated using the times of scratching head as indexes within 2 h after modeling (30 min as a period of time). 4 h after modeling, hemorheological parameters as whole blood viscosity (low-shearing, medium-shearing, high-shearing), plasma viscosity, erythrocyte aggregation indexes and erythrocyte rigidity indexes were determined by automatic hemorheology instrument. The levels of NO, NOS, ET-1, CGRP in serum and the levels of  5-HT, 5-HIAA, DA and NE in cerebral tissue were determined by ELISA. RESULTS: Compared with normal group, the times of scratching head was increased significantly in model group at different periods (P<0.01). The serum levels of NO, NOS and CGRP were increased significantly (P<0.01), while ET-1 level was decreased significantly (P<0.01). The levels of 5-HT, DA and NE in cerebral tissue were decreased significantly (P<0.01), while 5-HIAA level was increased significantly (P<0.01). Whole blood viscosity (low-shearing), plasma viscosity, erythrocyte aggregation index and erythrocyte rigidity indexes were increased significantly (P<0.05),plasma viscosity were decreased significantly (P<0.05) . Compared with model group, the times of scratching head were decreased significantly in TCM positive control group (0-120 min), TCM positive control group (60-90 min), CZC high-dose (30-120 min) and medium-dose (60-90 min) groups (P<0.05 or P<0.01). The whole blood viscosity (low-shearing) and erythrocyte aggregation indexes of TCM positive control group, CZC high-dose and medium-dose groups were decreased significantly (P<0.05 or P<0.01); the serum levels of NO, NOS and CGRP in chemical drug positive control group and CZC high-dose and medium-dose groups were decreased significantly (P<0.05 or P<0.01), while the serum level of ET-1 was increased significantly (P<0.01); the levels of 5-HT, DA and NE were increased significantly in cerebral tissue (P<0.05 or P<0.01), while the level of 5-HIAA was decreased significantly (P<0.05). CONCLUSIONS: The improvement effect of CZC on migraine model rats is associated with decreasing the serum levels of NO, NOS and CGRP and increasing the levels of 5-HT, DA and NE in cerebral tissue.

ABSTRACT

OBJECTIVE: To study the improvement effects of Lanqian buccal tablets on scopolamine-induced learning and memory impairment of mice. METHODS: The mice were randomly divided into blank group (normal saline), model group (normal saline), positive group (Donepezil hydrochloride tablets, 1.52 mg/kg) and Lanqian buccal tablets high-dose, medium-dose and low-dose groups (800, 400, 200 mg/kg), with 14 mice in each group. Once a day, 30 days after continuous intragastric administration, except for blank group, other groups were intraperitioneal injected scopolamine hydrobromide 3 mg/kg to induce learning and memory impairment model. After modeling, the learning and memory ability of mice were evaluated with step through test (latency, mistake times of entering darkroom as indexes) and Morris water maze (the time of going up on the platform, the times of crossing the platform, swimming time in target quadrant as indexes). The levels of Ach, ChAT, AchE, SOD and MDA in cerebral tissue of mice were determined. RESULTS: Compared with blank group, latency, the times of crossing the platform, swimming time in target quadrant, the levels of ChAT, Ach and SOD were shortened or decreased significantly in model group (P<0.05 or P<0.01), while mistake times of entering darkroom, the time of going up on the platform, the levels of AchE and MDA were extended or rised significantly (P<0.01). Compared with model group, latency (except for Lanqian buccal tablet low-dose group), the times of crossing the platform, swimming time in target quadrant, the levels of ChAT, Ach and SOD were extended or rised significantly in positive group and Lanqian buccal tablet groups (P<0.05 or P<0.01). The mistake times of entering darkroom (except for Lanqian buccal tablet low-dose group), the time of going up on the platform (except for Lanqian buccal tablet low-dose group) and the levels of AchE and MDA (except for Lanqian buccal tablet medium-dose and low-dose group) were shortened or decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: Lanqian buccal tablet can improve scopolamine-induced learning and memory impairment of mice by increasing the levels of ChAT, Ach and SOD and decreasing the levels of AchE and MDA.

ABSTRACT

OBJECTIVE: To observe the improvement effects of angiotensin converting enzyme inhibitor (ACEI) fosinopril, perindopril and benazepril on ventricular remodeling in patients with acute myocardial infarction (AMI), and to evaluate its safety. METHODS: A total of 96 AMI patients selected from our hospital during Jan. 2014-Oct. 2016 were divided into group A, B, C according to random number table, with 32 cases in each group. All patients received symptomatic treatment, underwent percutaneous coronary intervention, and then given ACEI after blood vessels recanalization and keeping blood pressure stable. Group A was given Fosinopril sodium tablets 10 mg, qd; group B was given Perindopril tert-butylamine tablets 4 mg, qd; group C was given Benazepril hydrochloride tablets 10 mg, qd. All groups were treated for consecutive 6 months. Cardiac structure and function indexes (LVESD, LVEDD, IVSD, LVPWD, LVEF, CO), hemodynamic indexes (SBP, DBP, HR) and related lab indexes (FPG, TG, TC, HDL-C, LDL-C, AST, ALT, Scr, BUN) of 3 groups were observed before and after treatment. The occurrence of ADR was recorded. RESULTS: Before treatment, there was no statistical significance in cardiac structure and function indexes, hemodynamic indexes or related lab indexes among 3 groups (P>0. 05). After treatment, the levels of LVESD, LVEDD, LVPWD, CO, HR, FPG, TG, TC and LDL-C in 3 groups were decreased significantly, while the levels of LVEF and SBP were increased significantly, with statistical significance (尸<0. 05). There was no statistical significance in above indexes among 3 groups after treatment (P>0. 05). After treatment, the level of Scr in group B was significantly increased and higher than group A and C, with statistical significance (P<0. 05). There was no statistical significance in the levels of IVSD, DBP, HDL-C, AST, ALT or BUN among 3 groups before and after treatment as well as the level of Scr between group A and C (P> 0. 05). There was no statistical significance in the incidence of ADR among 3 groups(P>0. 05). CONCLUSIONS: Fosinopril, perindopril and benazepril can significantly improve ventricular remodeling in AMI patients, narrowing the heart cavity, increasing systolic pressure, lowering heart rate, reducing the oxygen consumption of the ventricle, with similar effects. Perindopril may increase the level of Scr, so fosinopril and benazepril are safe and suitable for AMI patients with renal function disorder.

ABSTRACT

OBJECTIVE:To investigate the improvement effect of Eucalyptol enteric soft capsule on mice with lipopolysaccha-ride(LPS)-induced acute lung injury(ALI). METHODS:60 mice were randomly divided into blank control group,model control group,Eucalyptol enteric soft capsule low-dose,medium-dose,high-dose groups(100,300,900 mg/kg),12 in each group. Mice in administration groups were intragastrically given relevant medicines,mice in blank control group and model control group were intragastrically given equal volume of normal saline(0.1 mL/10 g). After 2 h of administration,except for the blank control group, ALI was induced in other groups by atomized LPS. After 6 h of modeling,the mice were sacrificed,alveolar lavage fluid(BALF) and lung tissue were taken. Morphological changes of lung tissue were observed under microscope;number of total cells,neutro-phils in BALF were calculated by blood cell count plate and staining by wright-giemsa respectively. Total protein concentration in BALF supernatant was detected by BCA method;TNF-α,IL-6 contents in BALF supernatant were determined by enzyme-linked immunosorbent assay. RESULTS:Compared with blank control group,lung tissue of mice in model control group showed obvious pathological damage and severe pulmonary edema;number of total cells,neutrophils in BALF,total protein concentration and TNF-α,IL-6 contents in BALF supernatant were significantly increased(P0.05). CONCLUSIONS:High-dose Eucalyptol enteric soft capsule can obviously improve LPS-induced ALI of mice.

ABSTRACT

Objective] To explore a new method of synthesis of resveratrol oligmer induced by photosensitizers, further analyze its free radical scavenging activity and analgesic activity. [Method] Resveratrol was dispersed in pH 7.4 phosphate buffer solution, mixed with pheophorbide, then illuminated by laser at 630nm. The oligmers in the filtrate were analyzed by HPLC-MS and isolated by preparative liquid chromatography. Free radical scavenging activity was determined by DPPH and ABTS method, and the analgesic effect was measured by hot plate test and acetic acid writhing test in mice. [Result] A new peak occurred and reached stable in reactive solution after 20 min illumination. It was identified as the dimer of resveratrol with conversion of 35%. Its concentrations on inhibition of 50% DPPH and ABTS radical were respectively 230 μg·mL-1 and 273 μg·mL-1. It reached maximum pain threshold at 180 min, and the inhibition rate of writhing number was 62.6% with better effects than resveratrol. [Conclusion] Reseratrol can be polymerized to the dimer after pheophorbide excited by red light, the resveratrol and its dimer both have the ability of eliminating free radicals and analgesic effects, but the dimer has the stronger activity.

ABSTRACT

ObjectiveTo explore the influence of washable type subglottic attract on incidence of ventilator-associated pneumonia (VAP) in patients with mechanical ventilation. Methods356 cases in need of mechanical ventilation from July 2009 to July 2010 in our hospital were chosen as the research object.They were randomly divided into the observation group and the control group with 178 cases in each group.The control group was treated with conventional endotracheal tube for mechanical ventilation.The observation group used washable type subglottic endotracheal tube to attract for mechanical ventilation.The VAP rate for the two groups occurred during mechanical ventilation in ventilator was compared.Migration of the dominant bacteria of the subglottic secretion was detected,underwent pathogenic examination and drug sensitivity test. ResultsThe VAP rate was significantly lower in the observation group than that of the control group.186 bacteria strains were detected from subglottic secretion,and 169 were detected from lower respiratory tract. ConclusionsWashable type subglottic attract for mechanical ventilation can reduce the risk of ventilator-associated pneumonia for patients with mechanical ventilation significantly,and it is worthy of clinical application.
