ABSTRACT

Aims: The paper aimed to clarify the effect of cucumber target leaf spot (TLS) under the Jingdusha (JDS) treatment. Study Design: We applied the method of artificial inoculation in the pot, and analyzed the changes in growth indexes and physiological characteristics. Place and Duration of Study: In 2018, these experiments were conducted in College of Bioscience and Biotechnology of Shenyang Agricultural University (Lab 240). Methodology: The seedlings in the two-leaf period were induced by the best application scheme of JDS, then inoculated Corynespora cassiicola for 24 h. Cucumber seedlings of each treatment group were randomly selected for photographing and growth index determination after inoculation for 5 d. The leaves of cucumber seedlings in each treatment group were randomly collected at 1 d, 3 d, 5 d, 7 d, and 9 d after inoculation for the determination of physiological and biochemical indicators. Results: When C. cassiicola infects cucumber, JDS can effectively improve the growth and photosynthetic pigment content of cucumber, reduce the degradation of chlorophyll (Chl) under the stress of C. cassiicola, strengthen the variety of metabolic responses in the plant, repair the enzyme protection system of cucumber leaves, reduce the accumulation of reactive oxygen species, shorten the process of membrane lipid peroxidation in blades. Conclusion: Taken together, these results suggest that JDS can improve the resistance of cucumber seedlings to C. cassiicola by regulating growth indexes and physiological characteristics. This work will provide a theoretical basis for further elucidating the molecular mechanism of JDS in cucumber defense against C. cassiicola.

ABSTRACT

BACKGROUND: Rice sheath blight (caused by Rhizoctonia solani) and tobacco mosaic virus are very important plant diseases, causing a huge loss in global crop production. Paenibacillus kribbensis PS04 is a broad-spectrum biocontrol agent, used for controlling these diseases. Previously, extracellular polysaccharides (EPS) from P. kribbensis PS04 had been purified and their structure was inferred to be fructosan. This study aimed to evaluate the effects of exogenous EPS treatment on plantpathogen interactions. RESULTS: Plant defense genes such as phenylalanine ammonia-lyase, catalase, chitinase, allene oxide synthase, and PR1a proteins were significantly induced by exogenous EPS treatment. Moreover, subsequent challenge of EPSpretreated plants with the pathogens (R. solani or tobacco mosaic virus) resulted in higher expression of defenseassociated genes. Increased activities of defense-associated enzymes, total phenols, and flavonoids were also observed in EPS pretreated plants. The contents of malondialdehyde in plants, which act as indicator of lipid peroxidation, were reduced by EPS treatment. CONCLUSIONS: This study comprehensively showed that EPS produced from P. kribbensis PS04 enhances disease resistance in plants by the activation of defense-associated genes as well as through the enhancement of activities of defense-related enzymes.

ABSTRACT

In Brazil, there are few records of insects associated with the cultivation of lima beans; among them, there is the black aphid Aphis craccivora Koch, 1854. The objective of this study was to evaluate the effects of silicon application on the resistance induction of lima bean plants, Phaseolus lunatus, to the black aphid A. craccivora. The experiment was conducted in the Entomology Laboratory of the Phytosanitary Sector of Centro de Ciências Agrárias, Universidade Federal do Piauí (UFPI), Brazil. The effects of the following treatments on biological aspects of the insect were evaluated: silicon applied to soil; silicon applied to soil + leaf; silicon applied to leaf; and control, without silicon application. The following biological variables were evaluated: generation period, reproductive period, and the fertility and daily average of produced nymphs per female. Plant silicon and lignin content were also evaluated. A 1% solution of silicic acid (2.0 g of product diluted in 200 mL of water) was applied around the plant stem (on soil), 15 days after emergence. Leaf application was performed with a 1-L spray, 5 days after the soil application. The non-preference of A. craccivora on lima beans was also evaluated. The evaluations were performed after 48 and 72 hours of infestation by counting nymphs and adults at each leaf section. Silicon application reduces nymph production, thereby interfering in the biological aspects of A. craccivora. Therefore, it can be used in cowpea pest management programs.(AU)

ABSTRACT

Abstract Plants can be attacked by a wide variety of herbivores. Thus, developing protective mechanisms for resistance against these agents is an advantage for survival and reproduction. Over the course of evolution, many resistance mechanisms against herbivory have been developed by the plants. Induced direct and indirect resistance mechanisms can manifest in plants after herbivore attack. The two-spotted spider mite Tetranychus urticae is not a pest of maize crops (Zea mays), despite being reported infesting plants that may have resistances against this herbivore. We tested the hypothesis that maize plants would be able to induce direct resistance against T. urticae after, evaluating the effect of T. urticae infestation in maize plants on the development and reproduction of conspecifics. We tested induced direct resistance performing infestation and measuring biological parameters upon a second infestation. Maize plants, 40 days after sowing, were divided into two groups: 30 not infested by T. urticae (clean plants clean) and, 30 infested by the spider mite. Infestation of maize plants by T. urticae reduced the conspecific female adult survival. However, no change in the survival of immature or reproduction was observed. These results suggest the induction of induced direct resistances in maize by T. urticae. This is first report of direct resistance induction in Z. mays by the two-spotted spider mite T. urticae.

ABSTRACT

Certain beneficial microorganisms isolated from rhizosphere soil promote plant growth and induce resistance to a wide variety of plant pathogens. We obtained 49 fungal isolates from the rhizosphere soil of paprika plants, and selected 18 of these isolates that did not inhibit tomato seed germination for further investigation. Based on a seed germination assay, we selected four isolates for further plant tests. Treatment of seeds with isolate JF27 promoted plant growth in pot tests, and suppressed bacterial speck disease caused by Pseudomonas syringae pathovar (pv.) tomato DC3000. Furthermore, expression of the pathogenesis-related 1 (PR1) gene was higher in the leaves of tomato plants grown from seeds treated with JF27; expression remained at a consistently higher level than in the control plants for 12 h after pathogen infection. The phylogenetic analysis of a partial internal transcribed spacer sequence and the β-tubulin gene identified isolate JF27 as Aspergillus terreus. Taken together, these results suggest that A. terreus JF27 has potential as a growth promoter and could be used to control bacterial speck disease by inducing resistance in tomato plants.

ABSTRACT

The aim of this study was to evaluate different alternative products, foliar sprayed either alone or in combination with fungicide fluazinam, to control white mold in soybean at three locations: Arapoti, Mauá-da-Serra, and Pinhão, in Paraná state, southern Brazil. The following chemical products were used: sodium dichloroisocyanurate, benzalkonium chloride, pyroligneous extract, sodium hypochlorite, cobalt + molybdenum, sucrose, sodium bicarbonate and acibenzolar-S-methyl. Incidence, severity, yield, and number of sclerotia produced were analyzed. The majority of the variables did not follow the normal distribution of the data according to the Kolmogorov-Smirnov test; therefore, the use of non-parametric analysis was necessary. The analysis of all the treatments individually revealed no significant effects so it was decided to separate the analysis into the following groups of treatments: 'control', treatments using alternative products ('alternatives'), treatment only with fungicide ('fungicide') and treatments with alternative products and fungicide ('alternatives + fungicide'). In Arapoti, no significant differences for any variables were observed. In Mauá-da-Serra, the 'fungicide' and 'alternatives + fungicide' had the lowest incidence and number of sclerotia, with higher yield when compared to 'control' and 'alternatives'. No differences were observed for severity. In Pinhão, the 'fungicide' and 'alternatives + fungicide' were superior for incidence, number of sclerotia produced, and yield when compared to the 'control' and 'alternatives'. The 'alternatives + fungicide' showed significantly lower severity when compared to the control, 'alternatives' and 'fungicide' in Pinhão experiment. In conclusion, the alternative products applied in combination with fluazinam resulted in lower severity of white mold in soybean in the experiment conducted in Pinhão, and the application of fluazinam alone or in combination with alternative products, was efficient to control white mold in soybean in locations with high disease incidence.

ABSTRACT

The excessive application of pesticides for agricultural production has raised quite some concern about environmental safety and sustainability. To reduce environmental impact of pesticide overuse, there is an increasing interest in using different elicitors including Jasmonic acid (JA) to induce resistance against pathogen and insect in crop. Chilli (Capsicum annuum L.), which is an important vegeTable cum spice crop around the world. The aims of this study were to compare the effectiveness of Jasmonic acid on growth, Phyto-physiological responses, yield and viruses control in chilli plant. It was evaluated the effectiveness of single spray of JA (JA1), double spray of JA (JA2), conventional pesticide (Malathion 50%) and without any of those or control. The experimental results showed that pesticide-treated plants perform much better at early growth stages and become less competitive to JA2 treatment at maturity. Double spray of jasmonic acid showed less stress symptoms in different antioxidant enzymes activities (GPX, APX and CAT), reduce percentage of disease incidence and severity as well as improve growth and yielding characters of chilli plant. Therefore application of Jasmonic acid in chilli could be a possible alternative of pesticide application and its two times exogenous spray (0.5mM) is most effective.

ABSTRACT

Objective To investigate the phenotypic and genetic characteristics of the lysostaphin‐resistant Staphylococcus aureus variants induced by recombinant lysostaphin in vitro .Methods Three clinical isolates of S . aureus ,including two resistant to methicillin (MRSA ) and one susceptible to methicillin (MSSA ) were induced by treatment with sub‐MIC of recombinant lysostaphin via one‐step selection in vitro .Susceptibility of the variants to antibiotics were determined and compared with their parental strains .The full length of femABX genes was amplified by polymerase chain reaction and sequenced to identify the potential mutation sites in these genes .The growth‐curve in liquid medium and virulence in a mouse systemic infection model of both parental and variant strains were observed . Results The frequency of lysostaphin resistance in S . aureus was between 10-4 to 10-8 following induction by lysostaphin . Resistance to lysostaphin was associated with a significant decrease in growth rate in vitro and virulence in vivo ,as well as increased susceptibility toβ‐lactams evidenced by the M IC of β‐lactams against the variants as low as 1/4 000 to 1/2 of the M IC against their parental strains . Sequencing of f emA BX genes showed mutation in femA gene in both variants ,which resulted in a premature termination codon .Conclusions Resistance of S . aureus to lysostaphin may develop following induction by recombinant lysostaphin in vitro . The lysostaphin‐resistant S . aureus variants are characteristic of lower growth rate , decreased virulence ,and higher susceptibility to β‐lactams .

ABSTRACT

With the objective to assess whether repeated non-toxic doses of sodium monofluoroacetate (MFA) induce resistance to poisoning by this compound, 18 sheep were randomly divided into two experimental groups of nine animals each. Sheep from Group 1 ingested non-lethal increasing doses of MFA for six periods: 0.05mg/kg for 5 days; 0.08mg/kg for 4 days; 0.08mg/kg for 4 days; 0.1mg/kg for 3 days; 0.1mg/kg for 3 days and 0.25mg/kg for 3 days. Between the first and second period of administration and between the second and third period the animals did not receive MFA for 10 consecutive days, between the third and fourth period and during the remaining periods of administration the sheep were left 15 days without ingesting MFA. Group 2 was not adapted to the ingestion of MFA and received a single dose of 1mg/kg of MFA at the same time that Group 1 was challenged. After challenge, seven sheep of Group 1 showed clinical signs of poisoning and one sheep recovered. In Group 2, all animals showed clinical signs of poisoning by MFA, however two sheep recovered. The mortality rate was 66.6 percent in Group 1 and 77.7 percent for Group 2. These results suggest that repeated administration of non-toxic doses of MFA does not protect against acute poisoning by this compound; therefore other alternatives of prophylaxis for poisoning by plants containing MFA should be searched, mainly the use of intraruminal bacteria that hydrolyze MFA.

ABSTRACT

Pepper is an economically important crop in many countries around the world but it is susceptible to many diseases. In Mexico, diseases caused by bipartite begomoviruses have emerged as important problems in pepper. Several control strategies have been explored wiht little success; most of them are based on the avoidance of virus transmission and the breeding for resistance. Abiotic inducers can act at various points in the signaling pathways involved in disease resistance, providing long-lasting, wide-spectrum resistance. Benzothiadiazole (BTH) shares the property of activating the systemic acquired resistance pathway downstream from the SA signaling. In this work, resistance to PepGMV infection was induced in pepper plants by activating the SA pathway using BTH treatment. The resistance was characterized by evaluating symptom appearance, virus accumulation and viral movement. Our results showed that BTH could be an attractive alternative to induce geminivirus resistance in pepper plants without a significant damage of the fruit quality and productivity.

ABSTRACT

Pearl millet [Pennisetum glaucum (L.) R. Br.] downy mildew (DM) is caused by the fungus Sclerospora graminicola (SACC.) SCHRŐT. is the most widespread and destructive disease of pearl millet affecting yield and quality in all the millet cultivating tracts of India. Since pearl millet is a crop of low economic value grown by resource-poor farmers, conventional technological interventions are not cost feasible. Integration of indigenous knowledge with biocontrol agents appeared as a logical strategy in the present case. Studies were, therefore, undertaken to manage DM in rainfed crop of pearl millet using raw cow milk together with Gliocladium virens. Seed and soil treatments resulted in the lowest disease incidence. Biochemical constituents (metabolites and oxidative enzymes) were analysed to determine possible mode of action of Raw Cow Milk (RCM) and Gliocladium virens. A considerable increase in sugars, phenols and ortho-dihydroxy phenols (OD) in healthy and DM infected leaves of treated pearl millet plants was recorded when compared to untreated controls. A marked increase in all the photosynthetic pigments in both healthy and diseased treated plants was observed. The induction of resistance was accompanied by increased activities of defense related enzymes. It is assumed that the combination of RCM and G. virens is capable of stimulating different systemic responses in host plant.

ABSTRACT

The present work aimed to evaluate the persistence of the induced resistance, the efficiency of the number and the interval of ulvan applications and the influence of different inoculum concentrations in the control of bean anthracnose. Three independent experiments were carried out. Bean plants (P. vulgaris, cv. Uirapuru) were cultivated under green house conditions and, in the first experiment, they were treated with ulvan once at nine days before inoculation (dbi) V3(i); six dbi V3(m) or three dbi V3(f) or they were treated twice at nine and six dbi V3(i)/(m); nine and three V3(i)/(f) or six and three (dbi) V3(m)/(f), the they were inoculated at the same time. In the second and third experiments, bean plants were treated twice (six and three dbi) then inoculated in different moments or with different inoculums concentrations, respectively. The application of ulvan reduced the anthracnose severity by 50%. Two ulvan applications were more efficient in inducing defense responses in bean plants than a single application. The highest disease reduction (96%) was achieved by spraying ulvan twice at V3(m)/(f). The effect of two applications persists until nine days after the treatment. The reduction on the anthracnose severity was higher when intermediate (105 e 106 conidia per mL) inoculum concentrations were used.

ABSTRACT

The biotype B of Bemisia tabaci stands out among the main pests of the bean plant. The objective of this work was to evaluate the resistance induction to the whitefly by silicon application in bean plant. The experimental design was completely ranlomized with 4 treatments, in a 2 (cultivar Carioca and Brave) x 2 (with and without silicon application) factorial outline and 6 repiicates. Ten days after the emergence of the seedlings, 500 ml of 1 percent silicic acid solution was applied in the soil, at a dosage equivalent to 2 t SiO2/ha. Ten days after the silicon application, the plants were infested with non-sexed whitefly adults in an acclimatized room, wiht 100 whiteflys/vase. For the non-choice test, the same procedure was used, with each vase covered by cages of organza cloth. Previously to the infestation, a plant of each vase was removed, oven-dried to constant weight for the evaluation of phenol levels. After 48 hours of infestation, the liberated adults were removed from the plants for evaluation of the number of eggs. For that, a leaf of a plant/vase was chosen, selecting the third completely developed apical leaf. After fifteen days, the number of nymphs of 3rd and/or 4th instar were evaluated. There was no response among cultivars of common bean to silicon application. A lower number of eggs and lower number of nymphs were observed in the plants treated with silicon in the test with chance choice. In the no-choice test the silicon application did not affect the oviposition of the whitefly and the nymph development, as well as the phenol levels.

ABSTRACT

The silvering thrips, Enneothrips flavens Moulton, 1941 is considered the major pest on peanuts in Brazil, due to its widespread occurrence, high population levels, and high damage to the crop. The objective of this study was to evaluate the effect of silicon on E. flavens populations and on peanuts yield. Ten fully-opened or semi-opened leaflets per plant were sampled weekly from the peanut cultivar IAC 886. The following treatments were tested: leaf spraying of silicon at 20 days after plant emergence, two leaf spraying of silicon at 20 and 55 days, and the control check. A single silicon spraying provided protection to peanut by reducing the number of E. flavens adults and nymphs and by increasing the yield of peanut pods by 31.30 percent and peanut grains by 28.85 percent.

ABSTRACT

The ability of benzothiadiazole (BTH) and/or humic acid (HA) used as seed soaking to induce systemic resistance against a pathogenic strain of Fusarium oxysporum was examined in four soybean cultivars under greenhouse conditions. Alone and in combination the inducers were able to protect soybean plants against damping-off and wilt diseases compared with check treatment. These results were confirmed under field conditions in two different locations (Minia and New Valley governorates). The tested treatments significantly reduced damping-off and wilt diseases and increased growth parameters, except the number of branches per plant and also increased seed yield. Application of BTH (0.25 g/L) + HA (4 g/L) was the most potent in this respect. Soybean seed soaking in BTH + HA produced the highest activities of the testes of oxidative enzymes followed by BTH in the four soybean cultivars. HA treatment resulted in the lowest increases of these oxidative enzymes. Similar results were obtained with total phenol but HA increased total phenol more than did BTH in all tested cultivars.

ABSTRACT

The objective of the present study was to evaluate the protection of passion fruit plants against CABMV by using preparations from Agaricus brasiliensis and Lentinula edodes mushrooms. In experiments carried out in the greenhouse, the fruiting body extracts from some of the isolates of both mushrooms significantly reduced CABMV incidence in passion fruit plants. This protective effect occurred when the plant leaves, pre-treated with extracts, were later inoculated mechanically with the virus. However, the extracts did not protect the plants in experiments involving CABMV transmission by aphid vectors. An inhibitory effect of mushroom extracts on the virus particles was also demonstrated on Chenopodium quinoa, a CABMV local lesion host, by inoculating the plants with a mixture of extracts and virus suspension. Still in C. quinoa, the mushroom extracts from some isolates induced systemic resistance against the virus. These results showed that aqueous extracts from A. brasiliensis and L. edodes fruiting bodies had CABMV infectivity inhibitors, but that was not enough to control the viral disease on passion fruit plants at all, considering they were infected through a vector.

ABSTRACT

O pulgão-verde Schizaphis graminum (Rond.)

The greenbug Schizaphis graminum (Rond.) (Hemiptera: Aphididae) is one of the main pest insects on wheat crop, being capable of leading a plant to death by sap suction and toxin injection, in addition to transmitting viruses. A strategy recommended to the management of aphids on wheat is the use of selective insecticides, mainly in the seed treatment. This way, in this research, the objective was to monitor the feeding behavior of S. graminum on wheat plants treated with silicon and/or imidachloprid, in addition to evaluate some biological features of the insect. The experiment was carried out at the Entomology Department, UFLA, on wheat cultivar Embrapa 22, S. graminum, 1% silicic acid, and imidachloprid seed treatment. The biology assay was conduced, starting at 15, 30, and 45 days after the seed germination, and the honeydew excretion was evaluated. The treatments were distributed in factorial scheme 2 (with silicon and without silicon) x 2 (with imidachloprid and without imidachloprid), with 10 replicates. It was observed that the systemic insecticide imidachloprid acts as a deterrent on adults and it is lethal on nymphs of S. graminum up to 45 days after seed treatment. Furthermore, the plants treated with silicon showed resistance to S. graminum.

ABSTRACT

The aphid Myzus persicae (Sulzer) is an important pest of potato and causes direct harm, due to the quantity of sap extracted and for being vector of important phytovirus. This work was carried out to evaluate the action of silicon as a resistance inducer of potato to M. persicae. Four treatments were tried: foliar fertilization with silicon acid at 1 percent; soil fertilization with 250 ml silicic acid solution at 1 percent; foliar fertilization with silicon acid at 1 percent + soil fertilization with 250 ml silicic acid solution at 1 percent; and a control. The treatments were applied thirty days after the explants emergence. Fifteen days after the application of the treatments, feeding preference and some biological aspects of the aphids were evaluated. After, the content of tannins and lignin present in the leaves and the activity of the enzymes peroxidase and phenylalanine ammonia-lyase were also determined. The silicon fertilization did not affect the preference of the aphids; however it reduced fecundity and the rate of population growth of the insects. The lignin percentage increased in the leaves of plants fertilized with silicon via soil and/or foliar and the percentage of tannins increased only in the leaves fertilized via soil plus foliar. The silicon acted as a resistance inducer to M. persicae in potato.
