ABSTRACT

Foram avaliados o efeito do ácido salicílico (AS) no controle do Cowpea aphid-borne mosaic virus (CABMV), vírus que induz o endurecimento dos frutos do maracujazeiro (EFM), e a sua influência na expressão dos sintomas e na ativação das enzimas peroxidase e polifenoloxidase. O experimento foi delineado e conduzido de forma inteiramente casualizada. Os tratamentos consistiram em AS (2,5 mM) e controle (etanol 10%), aplicados 12 horas antes da inoculação mecânica do CABMV, agente causal do EFM no Brasil. Um experimento similar foi conduzido sob as mesmas condições, porém, quatro aplicações do AS foram realizadas semanalmente após a inoculação mecânica do CABMV. Em ambos os experimentos a avaliação da severidade da doença foi realizada empregando escala de notas que variaram de 0 a 3. Para avaliar a atividade das enzimas peroxidase e polifenoloxidase, as plantas foram tratadas (AS e controle) e, após 12 horas, inoculadas com o isolado de CABMV. Foram realizadas amostragens foliares 0, 12, 24 e 48 horas após os tratamentos (HAT), que foram processadas e analisadas em espectrofotômetro para a constatação da ativação da peroxidase e polifenoloxidase. Aos 30 dias após a inoculação (DAI), o AS aplicado uma única vez promoveu redução da severidade de 57,1%, quando comparado com o controle. Nas plantas submetidas às aplicações semanais de AS foi constatada a redução significativa da expressão dos sintomas aos 45 DAI. Nos ensaios bioquímicos foi observado aumento significativo de peroxidase nos intervalos de 12 horas (DAI)/24 horas (HAT). Para polifenoloxidase foi observado um aumento significativo de sua atividade nos intervalos de 24 horas (DAI)/48 horas (HAT). Sugere-se que o AS pode representar uma ferramenta adicional no manejo do EFM.(AU)


The effect of salicylic acid (SA) was evaluated in control of Cowpea aphid-borne mosaic virus (CABMV), which induces hardening of the fruits of passion fruit (EFM). Also, its influence on the expression of symptoms and the activation of peroxidase and polyphenol oxidases were evaluated. The experiment was designed and conducted in a completely random way. The treatments consisted of SA (2.5 mM) and control (10% ethanol), applied 12 hours before mechanical inoculation of CABMV, the causal agent of EFM in Brazil. A similar experiment was conducted under the same conditions, but four applications were performed weekly after CABMV mechanical inoculation. In both experiments a rating scale ranging from 0 to 3 was used to assess the severity of the disease. To evaluate the activity of peroxidase and polyphenol oxidase, the plants were treated (SA and control) and, after 12 hours, inoculated with isolated CABMV. Leaf samplings were performed at 0, 12, 24 and 48 hours after the treatments (HAT), processed and analyzed in a spectrophotometer to verify the activation of peroxidase and polyphenol oxidase. At 30 days after inoculation (DAI), the SA applied once promoted reduction of 57.1% of the severity, when compared with the control. In plants subjected to SA weekly applications, it was found a significantly reduction in the expression of symptoms at 45 DAI. In the biochemical assays, a significant increase in peroxidase in 12-hour intervals (DAI)/24 hours (HAT) was observed. For polyphenol oxidase, a significant increase of its activity was observed at 24 hour intervals (DAI)/48 hours (HAT). It is suggested that SA may represent an additional tool in the management of EFM.(AU)

Poucos são os estudos envolvendo o efeito do silício sobre a fisiologia e a resistência do cacaueiro a insetos-praga. Com o objetivo de avaliar as alterações fisiológicas, a incidência e o nível de dano provocado por insetospraga após aplicação de silicato de potássio em genótipos de cacau, foi conduzido em campo, um experimento no delineamento de blocos casualizados, em esquema fatorial 3 x 3 com 4 repetições, sendo 3 genótipos de cacau (TSH 1188, CCN 51 e Catongo) e 3 doses de silicato de potássio (0; 3 e 6 mL L-1). Avaliou-se o crescimento, a cinética da emissão da fluorescência da clorofila, o teor de Si, o índice de clorofila, além da incidência e nível de dano provocado por insetospraga. A aplicação de silicato de potássio na dose 6 mL L-1 reduziu a maioria das variáveis de crescimento. O TSH 1188 foi o único genótipo que apresentou incremento no índice de clorofila e no desempenho das variáveis de fluorescência com a aplicação de 3 mL L-1. A aplicação de silicato de potássio não influenciou o teor foliar de Si nos genótipos estudados, mas reduziu a incidência e o nível de dano provocado por insetos-praga.


There are few studies of the effect of silicon on the physiology and resistance to insect pests in cocoa. With aim to evaluate the physiological changes, the incidence and level of damage caused by insect pests after application of potassium silicate in cacao genotypes was conducted in the field, an experiment in randomized blocks in factorial scheme 3 x 3 with four replications, with three genotypes of cocoa (TSH 1188, CCN 51 and Catongo) and three doses of potassium silicate (0, 3 and 6 mL L-1). We evaluated the growth kinetics of chlorophyll fluorescence emission, the Si content, chlorophyll content, and the incidence and level of damage by insect pests. The application of potassium silicate at a dose 6 mL L-1 reduced the majority of growth variables. The TSH 1188 was the only genotype that showed increase in chlorophyll content and in performance of variable fluorescence with the application of 3 mL L-1. The application of potassium silicate did not influence the content of Si in leaf genotypes, but reduced the incidence and level of damage by insect pests.

Este trabalho objetivou a purificação parcial, por precipitação com sulfato de amônio (SA) e cromatografia de filtração em gel (CFG), de compostos presentes no decocto de Adiantum capillus-veneris (avenca) eficientes na indução de fitoalexinas em mesocótilos de Sorghum bicolor sorgo. Decocto de A. capillus-veneris a 1% (peso seco/volume) foi precipitado com concentrações de SA variando de 0 a 100% (em intervalos de 20%), e essas frações foram submetidas à CFG. Para o decocto não precipitado foram obtidos nove picos proteicos e um pico glicídico com massas moleculares variando de 0,61 à 0,01 KDa. Para a precipitação fracionada obteve-se: na fração 0-20% dois picos proteicos (menores que 0,01 KDa) e dois glicídicos com concentração de açúcares variando de 4,1 a 17,5 µg mL-1; na fração 20-40% três picos proteicos (111,5 à 0,98 KDa) e cinco glicídicos (11,3 a 73,7 µg de açúcares mL-1); na fração 40-60% dois picos proteicos (111,5 à 0,09 KDa) e dois glicídicos (5,6 a 7, 5 µg de açúcares mL-1); na fração 60-80% seis picos proteicos (menores que 0,02 KDa) e dois glicídicos (16,5 a 51,3 µg de açúcares mL-1); e na fração 80-100% três picos proteicos (menores que 0,09 KDa). Mesocótilos de sorgo foram tratados com as frações provenientes da CFG, além do decocto a 1%, acibenzolar-S-metil (ASM) (125 mg L-1 do i.a. como eliciador de referência) e tampão fosfato de sódio 10 mM pH 6,0. O pico proteico II (0,09 KDa) do decocto não precipitado induziu fitoalexinas, 6,68% superior a ASM. Entre os precipitados, a fração 60-80% de SA induziu 76% mais que ASM. Dessa forma, pôde-se obter frações proteicas e/ou glicídicas indutoras de fitoalexinas em sorgo de maneira superior ao extrato (decocto) do qual é originária, indicando o potencial dessas moléculas para trabalhos futuros sobre indução de resistência.


This study aimed to partially purify the compounds present in decoction of Adiantum capillus-veneris, which are efficient in the induction of phytoalexins in sorghum mesocotyl, by ammonium sulphate (AS) fractionation and gel filtration chromatography (GFC). The decoction of A. capillus-veneris at 1% (weight/volume) was precipitated with AS at the concentration of 0-20%, 20-40%, 40-60%, 60-80% and 80-100%, and these fractions were subjected to GFC. For the decoction not precipitated with AS, nine protein peaks and one carbohydrate peak were obtained with molecular weights ranging from 0.61 to 0.01 KDa. For the AS precipitation, we obtained: for the fraction 0-20%, two protein peaks (0.01 KDa) and two carbohydrate peaks with concentration of sugars ranging from 4.1 to 17.5 µg of sugar mL-1; for the 20-40%, three protein peaks (0.98 to 111.5 KDa) and five carbohydrate peaks (11.3 to 73.7 µg sugar mL-1); for the 40-60%, two protein peaks (0.09 to 111.5 KDa) and two carbohydrate peaks (5.6 to 7.5 µg of sugar mL-1); for the 60-80%, six protein peaks (lower than 0.02 KDa) and two carbohydrate peaks (16.5 to 51.3 µg of sugar mL-1); and for the 80-100%, three protein peaks with molecular weight equivalent to 0.09 KDa. The sorghum mesocotyls were treated with GFC fractions, decoction (1%), acibenzolar-S-methyl (ASM) (125 mg L-1 a.i. as elicitor reference) and sodium phosphate buffer (10 mM, pH 6.0). The protein peak II (0.09 KDa) from the decoction not precipitated was effective in inducing phytoalexin, exceeding in 6.68% the ASM. Among the fractions, the one with 60-80% of AS increased in 76% the induction of phytoalexin compared to ASM. According to the results, we could obtain protein and/or carbohydrate fractions capable of inducing phytoalexins in sorghum better than the decoction from which they are derived from, showing the potential of these molecules for future research studies on the induction of resistance.

ABSTRACT

A demanda mundial por alimentos isentos de agrotóxicos tem impulsionado a pesquisa para a busca de métodos alternativos ao controle de patógenos em plantas. A ativação dos mecanismos de defesa com o uso de indutores vem demonstrando ser uma alternativa viável e promissora. O uso de extratos de plantas medicinais tem demonstrado capacidade para induzir a produção de fitoalexinas, como um mecanismo de defesa em plantas tratadas. O objetivo deste trabalho foi verificar o potencial da planta Eugenia uniflora L. (pitangueira) em induzir fitoalexinas em cotilédones de soja (Glycine max). Os derivados de extrato alcoólico, infusão, maceração e decocção, obtidos de folhas de pitangueira, foram usados nas concentrações de 0,1; 1; 10 e 40 por cento, além de óleo essencial. Água foi utilizada como controle e quitosana (1 por cento) como indutor de referência. Os preparados de pitangueira apresentaram capacidade de indução das fitoalexinas gliceolinas em cotilédones de soja, respondendo ao aumento das concentrações dos preparados. O óleo essencial apresentou destacável efeito na indução de fitoalexinas, sendo superior aos demais preparados. Quitosana induziu fitoalexinas em cotilédones de soja e pode ser utilizado em estudos similares como um indutor de referência.


The worldwide demand for food without pesticides has stimulated the research on alternative methods to control pathogens in plants. The activation of defense mechanisms by inductors seems a viable and promising alternative. The use of medicinal plants extracts has demonstrated capacity to induce the production of phytoalexins, as a mechanism of defense in treated plants. The objective of this research was to verify the potential of Eugenia uniflora L. (surinan cherry) to induce phytoalexins in cotyledons of soybean (Glycine max). The derivatives alcoholic extract, infusion, maceration and decoction, obtained of surinan cherry were used in the concentrations of 0.1; 1; 10 and 40 percent, beyond essential oil. Water was used for the control and chitosan (1 percent) as inductor reference. The preparations of surinan cherry presented capacity of induction of the phytoalexins glyceolin in cotyledons of soybean, with the increasing concentration of the preparations. The essential oil presents detachable effect in the induction of phytoalexins in relation to the other preparations. Chitosan induces phytoalexins in cotyledons soybean and can be used in similar studies as a reference inductor.

ABSTRACT

Objective Antibiotic resistance has increasingly been recognized as the major cause of treatment failure for Helicobacter pylori infection. The study was designed to compare the propensity of Helicobacter pylori to develop in vitro resistance to five commonly used antibiotics and to investigate the rates of resistance to these five antibiotics. Methods The serial passage tests were done in 7 sensitive Helicobacter pylori strains (2 type strains and 5 clinical isolates) to induce the resistance to amoxycillin, tetracycline, furazolidone, metronidazole or clarithromycin in vitro. The agar dilution tests detecting the minimal inhibitory concentration (MIC) were performed in 165 strains of Helicobacter pylori isolated from 2000 to 2001 to determine the resistance rates to the 5 antibiotics mentioned above. Results Serial passage tests showed that 5 of 7 strains of Helicobacter pylori were induced to be resistant to metronidazole and tetracycline; the inducible multiple of metronidazole was the highest. No strain was induced to be resistant to clarithromycin, but the inducible multiple of one strain was relatively high. No strain was induced to be resistant to amoxycillin or furazolidone, and the inducible multiple of furazolidone was the lowest. The resistance rates of Helicobacter pylori to metronidazole, clarithromycin, amoxycillin, tetracycline and furazolidone were 49.7% (82/165), 7.3%(12/165), 1.2% (2/165), 2.4%(4/165) and 1.2%(2/165), respectively. Conclusions Our study indicate that it is quite easy to induce the resistance to metronidazole,not difficult to clarithromycin, and relatively difficult to furazolidone or amoxycillin. The relative degree of difficulty in inducing the resistance to the drugs other than tetracycline is associated with the resistance rates detected, the fact that may help predict the changing trend of resistance rates in general population.
