ABSTRACT

Introdução: Sendo a metformina um dos principais tratamentos para o Diabetes Mellitus, torna-se necessário verificar seus efeitos colaterais. A redução nos níveis de vitamina B12 está entre os causadores de consequências importantes na população. Este estudo objetivou determinar o nível de vitamina B12 em usuários e não usuários de metformina de um ambulatório de geriatria no sul de Santa Catarina e avaliar fatores que possam interferir no nível desta vitamina. Métodos: Caracteriza-se como pesquisa quantitativa, observacional, retrospectiva, com coleta de dados secundários. Foram incluídos na pesquisa pacientes idosos com níveis de vitamina B12 documentados em prontuário e excluídos prontuários com ausência de dados e pacientes que fazem suplementação da vitamina. Após a seleção, foram analisados 131 prontuários de pacientes atendidos no período de 2016 e 2017 no Ambulatório de Geriatria das Clínicas Integradas de Saúde da Universidade do Extremo Sul Catarinense. Resultados: Dos pacientes que possuem deficiência de vitamina B12, 26,4% fazem uso de metformina. Dos pacientes que usam metformina, 36,8% possuem deficiência de vitamina B12. No uso de IBP, 39,6% apresentaram deficiência, e naqueles em uso concomitante de metformina e IBP, 9,4% apresentaram deficiência da vitamina. Conclusão: Apesar dos testes estatísticos não apresentarem associação estatisticamente significativa entre o uso de metformina e a deficiência de vitamina B12, os dados do presente estudo mostraram prevalência de deficiência da vitamina na amostra (40,4%). Sugere-se que isso é uma consequência importante da droga e mostra a necessidade de novos estudos na deficiência da vitamina e as suas consequências.


Introduction: As metformin is one of the main treatment options for diabetes mellitus, it is necessary to analyze its side effects. A reduction in B12 levels is one of the causes of important consequences to the population. This study aimed to determine B12 levels in patients who used metformin and those who did not in a geriatric outpatient clinic in southern Santa Catarina, assessing factors that could interfere with the levels of this vitamin. Methods: This is a quantitative, observational, retrospective study, with secondary data collection. We included older patients with B12 levels registered in their medical records and excluded records with missing data and patients who did vitamin replacement. After patient selection, we analyzed 131 medical records of patients seen between 2017 and 2017 at the geriatric outpatient clinic of Clínicas Integradas de Saúde, at Universidade do Extremo Sul Catarinense. Results: Among patients with B12 deficiency, 26.4% used metformin. Out of those using metformin, 36.8% had vitamin B12 deficiency. Among those who used proton-pump inhibitors (PPI), 39.6% had a vitamin deficiency, and considering those who simultaneously used metformin and PPI, 9.4% had a vitamin deficiency. Conclusion: Although statistical tests did not identify a statistically significant association between metformin use and vitamin B12 deficiency, our data showed a prevalence of vitamin deficiency in our sample (40.4%). We suggest that this could be an important consequence of this medication and highlight the need for new studies on B12 deficiency and its consequences.

And its regulation is mainly achieved through the "neuro-endocrine-immune" network system. Acupuncture at Neiguan (PC 6) and Hegu (LI 4) to inhibit intestinal peristalsis may have good application value in colonoscopy.

@#[Abstract] Objective: To investigate the effect of miR-3195 on the proliferation of laryngeal carcinoma Hep-2 cells and its molecular mechanism. Methods: From January 2008 to August 2012, the laryngeal cancer tissues and their corresponding paracancerous tissues from 29 patients with laryngeal cancer who were admitted to the Department of Otorhinolaryngology, Chenzhou First People's Hospital Affiliated to teaching hospital of University of South China were selected for this study. qPCR was used to detect the expression of miR-3195 in laryngeal carcinoma and the paracancerous tissues; Hep-2 cell line with stable and high expression of miR-3195 was constructed. The proliferation of miR-3195 over-expressed Hep-2 cells and the control cells was observed by MTT method. A nude mouse xenograft model was established to observe the proliferation of miR-3195 overexpressed Hep-2 cells in nude mice. Bioinformatics tools were used to predict the target gene of miR-3195; the luciferase vector of TBX1 3'UTR was constructed, and its luciferase activity was examined with dual luciferase detection system; Western blotting was used to detect the TBX1 protein expression in miR-3195 over-expressed cells and control cells. Results: The expression of miR-3195 in laryngeal carcinoma tissues was significantly lower than that in paracancerous tissues (P<0.01); miR-3195 up-regulation could inhibit the proliferation of Hep-2 cells (P<0.01) and significantly inhibit the growth of transplanted tumors in nude mice (P<0.05); The results of the Dual luciferase reporter gene assay indicated that miR-3195 might targetedly bind to TBX1 (P<0.05), and Western blotting proved that miR-3195 could inhibit the expression of TBX1 protein (P<0.05). Conclusion: miR-3195 has a significant inhibitory effect on the proliferation of Hep-2 cells, and its molecular mechanism may be related to the negative regulation of TBX1 expression.

ABSTRACT

SATB1 plays a crucial role in the invasion and metastasis of breast cancer,and inhibition of SATB1 expression can effectively control breast cancer metastasis. In this study,homogeneous polysaccharides were isolated from Poria cocos and their sulfated derivatives were prepared to screen out the polysaccharide compositions with inhibitory effects on SATB1 expression. Smal-molecule components were removed from P. cocos by ethanol extraction,and P. cocos crude polysaccharide PPS was obtained by water extraction and ethanol precipitation. Then PPS was successively separated by DEAE Sepharose fast flow anion-exchange and Superdex-75 gel permeation chromatographic steps to give PPSW-1. The structure of PPSW-1 was identified and its sulfated derivatives were prepared. Then their inhibitory effects on human breast cancer MDA-MB-231 cells were investigated. A kind of polysaccharide,PPSW-1 with inhibitory effect on human breast cancer MDA-MB-231 cells,was obtained from P. cocos,with a relative molecular weight of 3. 06×104,and structure of 1,6-branched 1,3-α-D-galactan. PPSW-1 and its sulfated derivative Sul-W-1 showed good inhibitory effect on cells migration,and the water solubility of Sul-W-1 was better than that of PPSW-1. In addition,it was found that polysaccharide of P. cocos and its sulfated derivative can inhibit expression of SATB1. In this study,a kind of homogeneous polysaccharide with inhibitory effect on human breast cancer MDA-MB-231 cells was isolated from P. cocos,and its sulfated derivative with similar efficacy but better solubility was prepared,laying the foundation for the substance basis study of P. cocos.

ABSTRACT

As a glycopeptide antibiotic,vancomycin is the first choice for treatment of methicilin-resistant staphylococcus aureus (MRSA) infection.Because of the metabolic individual difference,if children were given the dose according to the instruction,few patients can achieve the valley concentration as recommended in the guideline.So it's necessary for optimizing individual vancomycin dosage regimen with the help of therapeutic drug monitoring (TDM) and population pharmacokinetics model-nonlinear mixed effect model (NONMEM),in order to realiz the combination of effect and safety.This article will introduce the population pharmacokinetics model of vancomycin in children and discuss about major parameters influencing vancomycin metabolism,including age,body mass index,renal function,health condition,and drug combination.With rational therapeutic drug monitoring and optimizing parameters of NONMEM,we hope to realize area under concentration-time curve/minimum inhibit concentration (AUC/MIC) ratio ≥400,and to provide the reference for safe and rational pediatric dosage regimen.

Biofilm is a bacterial lifestyle ubiquitously in natural environments. Bacterial biofilm leads to drug resistance, a main reason why many infectious diseases are difficult to control. Due to the prominent points of biofilms implicated in infectious disease and the spread of multi-drug resistance, it is urgent to discover new antibacterial agents that can regulate biofilm formation and development. This review introduces chemical agents that could modulate bacterial biofilm formation and development.

[Objective]Summarize the clinical experience of famous veteran doctor LIAN Jianwei in treating chronic diarrhea. [Methods] With the method of following Professor LIAN, analyse and induce his experience in treating chronic diarrhea by collecting cases. [Results] Professor LIAN is good at using classical prescriptions and current prescriptions to treat chronic diarrhea. According to the characteristics of diarrhea, to identify the viscera, cold and heat, asthenia and sthenia and flexible combination of prescriptions. Weakness of the spleen and the stomach using Shenling Baizhu Powder to nourish qi to invigorate spleen;Patients of liver stagnation and spleen deficiency are treated by Tongxieyao Formula to repress liver and supporting spleen;Sishen pill and decoction of four mild drugs treatment of spleen and kidney yang-deficiency by invigorating spleen and kidney;Patients of coldheat complex are treated by Wumei Pill to warm and nourish the stomach. [Conclusion] Professor's experience in treating chronic diarrhea is well worth learning for it always gets outstanding effect.

ABSTRACT

Objective Non-contrast enhanced magnetic resonance angiography ( MRA) has become the focus of research at home and abroad .The aim of the article was to optimize the scan parameters of renal artery with inflow-sensitive inversion recovery ( IF-IR) MRA. Methods 30 cases of renal artery patients into two groups were collected from December 2013 to May 2014 in our hospi-tal.They were divided into stenosis group(n=12) and non-stenosis group(n=48).IFIR-MRA sequences were carried out on each patient by using various BSP-TI with 1200,1300 and 1400 ms, all other parameters were identical .The image signal to noise ratio, a renal artery branch and venous artifact were evaluated by two experienced radiologists , and the best BSP-TI was obtained from different groups. Results BSP-TI=1400 ms was optimal for renal artery stenosis group while 1200 ms was the most suitable for non-stenosis group. Conclusion IFIR-MRA of renal artery can achieve better image quality of renal artery and its branches and it is feasible in clinical routine practice , especially for those patients with renal insufficiency .

ABSTRACT

Metformin has become the first-line drug for the treatment of type 2 diabetes, which is stably efficacious, safe, and cheap. Previous studies have shown that the application of metformin can decrease the rate of tumor incidence, recurrence, and metasta-sis. It can also improve the prognosis of cancer patients and their lifetime after long-term use. Considering its multiple advantages, met-formin has become an emerging choice for the treatment and prevention of cancer. Therefore, the mechanisms through which metfor-min inhibits tumor metastasis in cancer therapy are worth investigating.

ABSTRACT

Breast cancer is the most common malignancy in women, both in the developed and developing countries. Anthocyanins are natural coloring of a multitude of foods, such as berries, grapes or cherries. Glycosides of the aglycons delphinidin represent the most abundant anthocyanins in fruits. Delphinidin has recently been reported to inhibit the growth of human tumor cell line. Also, delphinidin is a powerful antioxidant that reportedly exerts beneficial effects in patients with advanced cancer by reducing the level of reactive oxygen species and increasing glutathion peroxidase activity. This study investigates the effects of delphinidin on protein ErbB2, ErbB3 and Akt expressions associated with cell proliferation and Bcl-2, Bax protein associated with cell apoptosis in MDA-MB-231 human breast cancer cell line. MDA-MB-231 cells were cultured with various concentrations (0, 5, 10, and 20 micromol/L) of delphinidin. Delphinidin inhibited breast cancer cell growth in a dose dependent manner (p < 0.05). ErbB2 and ErbB3 expressions were markdly lower 5 micromol/L delphinidin (p < 0.05). In addition, total Akt and phosphorylated Akt levels were decreased dose-dependently in cells treated with delphinidin (p < 0.05). Futher, Bcl-2 levels were dose-dependently decreased and Bax expression was significantly increased in cells treated with delphinidin (p < 0.05). In conclusion, I have shown that delphinidin inhibits cell growth, proliferation and induces apoptosis in MDA-MB-231 human breast cancer cell lines.

Objective Study of the effects of hepatocyte growth factor on inhibit Intimal hyperplasia of the anastomotic stoma after carotid artery bypass grafting.Methods Thirty-two New Zealand white rabbits were randomly divided into control group and the experimental group.The veins were pretreated with saline solution(control group)only or pretreated with HGF(experimental group ;100ng/ml).The vein grafts were harvested at 14 days,28days after operation,HE Stain and Elastic fibrin Stain,The thickness of Intima and media in the vein grafts,intima-media ratio(I/M) was calculated by computer image analysis system.PCNA Immunohistochemistry was performed.Results The thickness of Intima and media in the vein grafts of control group surpassed experimental group significantly(P <0.01).At 14d I/M in the vein grafts of control group (0.81 ± 0.05) surpassed experimental group (0.47 ± 0.05) (P < 0.01),At 28d I/M in the vein grafts of control group(0.73 ± 0.01)surpassed experimental group (0.65 ± 0.01) (P < 0.01).The vascular smooth muscle cell proliferation in experimental group was significantly lower than that in control group (P < 0.01).Conclusion Treatment of veins grafts with HGF can significantly inhibit intimal hyperplasia in a rabbit carotid artery bypass grafting model.

BACKGROUND: Newborn neurons have bean shown to induce long-term potentiation (LTP). Activation of N-methyl-D-aspartic acid (NMDA) receptor subunit NR2B plays an important role in mature neurons-induced LTP. But there have been no reports addressing on the effects of NR2B activation on newborn neuron-induced LTP.OBJECTIVE: To investigate the effects of NR2B receptor antagonist Ro25-6981 on LTP induced by newborn neurons in adult rat dentate gyrus.DESIGN, TIME AND SETTING: An electrophysiological recording trial was performed at the Department of Neuroblology,Shanxi Medical University from February to June 2007.MATERIALS: Twenty-six male Wistar rats, aged 3 months, were provided by Laboratory Animal Center, Shanxi Medical University.METHODS: Following sacrifice for brain harvesting under anesthesia, the hippocempus was taken to preparation of 400 μ mol/L brain slices. Using extracellular field potential recordings, low-frequency stimulation was performed in the medial molecular layer of dentate gyrus with insulated bipolar tungsten electrodes. After having stable recordings, LTP was induced under high-frequency tetanic stimulation. LTP was induced with a protocol developed previously (4 trains, 500 ms each, 100 Hz within the train, repeated every 20 s). Only those slices which produced the field excitatory postsynaptic potential of 1 mV or cerebrospinal fluid (ACSF)-induced LTP (ACSF-LTP): brain slices were divided into 2 groups: ACSF group, in which, slices were continuously perfused using ACSF bubbled with 95% O2 and 5% CO2; ACSF+ Ro25-6981 group: a 10-minute treatment with 3μ mol/L Ro25-6981 was performed prior to tetanic stimulation, and the remaining procedures were the same as ACSF divided into 2 groups: BIC group: a 10-minute treatment with 10 μmol/L BIC was performed prior to titanic stimulation; BIC+Ro25-6981 group: 3μ mol/L Ro25-6981 and 10μ mol/L BIC were simultaneously perfused 10 minutes prior to tetanic stimulation.MAIN OUTCOME MEASURES: LTP recording results.minutes of titanic stimulation, LTP was (164.67±2.40)% and (147.56±6.63)% in the BIC and BIC+ Ro25-6981 groups,respectively, and a significant difference existed between the two groups (P < 0.05).

ABSTRACT

A fused functional gene of human OPG and Mhsp65 was amplified by PCR,and cloned into the prokaryotic expression vector pET-28a.The BL21(DE3) strain of E.coli was transformed using the recombinant plasmid pET-28a-OPG-HSP65 and the expected protein was expressed by induction with IPTG.Result of SDS-PAGE indicated that the expected recombinant protein of 23 kDa was expressed with high yield as inclusion body.The fusion protein could be specifically recognized by both the anti-His antibody and anti-human OPG monoclonal antibody in Western blot analysis.The purified and refolded fusion protein could inhibit osteoclast proliferation and bone absorption in vitro.The results of mouse ear swelling assay and expressions of TNF-?,IFN-? and IL-17 mRNAs detected by real-time quantitative PCR demonstrated that the fusion protein had an anti-inflammation activity.The results suggest that the fusion protein of human OPG and Mhsp65 may act as a potential therapeutics for rheumatoid arthritis.

Interleukin-12 can induce the production of INF-?, up-regulate the inhibitive factor of angiogenesis such as IP-10, MIG, TIMP, TNF-?, down-regulate the induced factor of angiogenesis such as VEGF, bFGF, MMP, inhibit the angiogenesis of tumor together and accelerate the regression of tumor. Interleukin-12 can inhibit the angiogenesis of tumor, which is independent of the immune system.

ABSTRACT

It is known that inflammatory processes result from intraocular surgery or ocular trauma mediated by prostaglandin, which cause the protein amounts to increase in anterior chmaber, congestion of vessels, miosis and intraocular pressure to increase. In order to compare the anti-inflammatory effects of antiprostaglandin agents such as Aspegic(R), Profenal(R) and Ocufen(R) in 40 eyes of 20 rabbits, we measured the pupil diameter and amounts of PG E2 and total protein before and after paracentesis. The inhibitory effects of Aspegiz(R), Profenal(R) and Ocufen(R) to pupillary constnztion were significantly greater (p<0.05) than control. The lowering effect to PG E2 and total protein were greater in Aspegic(R), Profenal(R) and Ocufen(R) than control. Through over experiment we suggested that the topical non-steroidal anti-inflammatory agents such as Aspegic(R), Profenal(R), and Ocufen(R) can inhibit the PG activator in antivator chamber and interrupt the miosis and rising intraocular pressure during surgery.
