ABSTRACT

Aims: Mentha spicata medicinal properties are well known. In this study, Total phenolic and flavonoid contents and the antioxidant activity of the methanol extract of Mentha spicata were determined as well as evaluation of the cytotoxic activity of it. Also, the identification of some bioactive compounds in the plant was analysed. Place and Duration of Study: Extraction and antioxidant at Medicinal Chemistry Department, Theodor Bilharz Research Institute, cytotoxic evaluation and GC-Ms analysis at Al-Azhar University, Egypt. Methodology: The antioxidant activity was determined by two methods, DPPH (2, 2-diphenyl-1-picrylhydrazyl) and ABTS (2-2՝azinobis (3-ethylbenzthiazoline-6-sulphonic acid) while, the cytotoxic assay was determined by MTT assay on HepG-2 (Human hepatocellular carcinoma) and MTC-116 (Human colon carcinoma). Regarding to the chemical identification of methanol extract was carried out by GC-MS analysis. Results: The study proved that Mentha spicata has high Total phenolic and flavonoid contents (388.20±2.38mg GAE/gm of extract &204.01±17.93mg RE/gm of extract) respectively. Also, exhibited promising antioxidant activity by DPPH & ABTS (IC50=65.13±1.29 µg/ml & 52.31±0.81 µg/ml) respectively. The methanol extract of the plant showed a good cytotoxic effect on HepG2 and HTC-116 (IC50=25.2±3.6µg/ml & 62.1±4.9µg/ml) respectively. GC-MS analysis of the methanol extract of Mentha spicata showed 43 oxygenated hydrocarbon compounds. The major ones are Hexadecanoic acid, methylEster (palmitic acid ester) (31.51%) followed by 9,12,15-Octadecatrienoic acid,methyl ester (CAS)(methyl linolenate) (22.10%), 2-Pentadecanone,6,10,14-trimethyl- (CAS)(6.82%), Phytol (6.20%), 9,12-Octadecadienoic acid(Z,Z)-, methyl ester (6.18%), Hexadecanoic acid (palmitic acid ) (5.95%) and Methyl stearate (4.49%). Conclusion: Mentha spicata is a potential antioxidant and anticancer agents.

ABSTRACT

Objective To evaluate the endogenous anticoagulation potential of bone fracture patients and its relationship with deep venous thrombosis formation (DVT).Methods A total of 95 DVT patients after bone fracture and 100 healthy subjects as control from Beijing Jishuitan Hospital were included in this study.On the third day after orthopedic surgery,the citrated anticoagulant plasma samples were collected and fibrin degradation products (FDP) and D-dimer (DD) were measured.Protac-induced coagulation inhibition percentage (PICI) was measured with ThromboPath (ThP) chromogenic assay as the marker of endogenous anticoagulation potential.The serum samples from patients were collected to measure the concentration of high-density lipoprotein cholesterol (HDL-C).Results Both FDP and DD values of DVT patient group were significantly higher than those of healthy control group (all P < 0.01).The PICI (%) of DVT patients group was significantly lower than that of healthy control group (82.8 ± 7.2 vs 87.8 ± 3.6,P < 0.01).The PICI of patients ≥65 years old was 4.8 percent lower than that of the patients <65 years old (P <0.05) in DVT group.When the cut-off value for PICI was set as 84.2%,a significant difference was showed between DVT and control group by Chi-Square test (P <0.0 1).PICI was negatively correlated with FDP and DD (r =-0.318,-0.336,both P < 0.01),but positively correlated with HDLC (r =0.284,P < 0.01).Logistic regression analysis suggested that PICI was a risk factor of DVT with odds ratio 1.243 (P < 0.01).Conclusion The endogenous anticoagulation potential may be severely impaired in DVT patient after bone fracture.

ABSTRACT

Hojas de las plantas Hura crepitans y Codiaeum variegatum se recolectaron en la ciudad de Medellín (Colombia), para determinar la actividad biológica (Actividad citotóxica [AC] y antiviral [AAV]) de cuatro extractos de éstas (hexánico, en acetato de etilo, metanólico y acuoso) sobre los virus de Estomatitis Vesicular (VSV) serotipos Indiana y New Jersey y Herpes Virus Bovino tipo-1 cepa Bogotá (BHV-1B). El tamizaje de actividad biológica de los extractos se realizó en monocapas de células BHK-21. Luego los extractos promisorios, pasaron a la fase cuantitativa de AAV por el ensayo de MTT [3-(4,5-dimethylthiazol–2-yl)-2,5-diphenyltetrazolium bromide] para determinar la concentración citotóxica 50 (CC50), la concentración inhibitoria 50 (IC50) y el índice de selectividad (IS). Ninguno de los extractos de las cuatro plantas presentó AAV contra VSV, mientras que los extractos hexánico, en acetato de etilo y metanólico de H. crepitans confirieron resistencia a la infección por BHV-1B con IC50 de 17.41, 7.87 y 2.75 µ g/ml, respectivamente. El extracto con mejor IS (relación entre IC50 y CC50) fue el hexánico de H. crepitans (IS > 17.4). Estos resultados sugieren que pueden existir compuestos en el extracto hexánico de H. crepitans con una actividad promisoria anti BHV-1B. Este estudio es pionero en demostrar actividad antiviral de extractos de plantas de la especie Euphorbiaceae contra BHV-1B.


Leaves from Hura crepitans and Codiaeum variegatum plants were collected in Medellín, Colombia to determine its cytotoxic [CA] and antiviral activity [AVA]. Four extracts were prepared using hexane, ethyl acetate, methanol and water; the antiviral activity was tested against Vesicular Stomatitis Virus (VSV), serotypes Indiana and New Jersey and Bovine Herpes Virus (type 1 Bogotá strain). The preliminary biological activity of each of the eight extracts was determined on BHK-21 cell monolayers using 96 well-plates. Then, promissory extracts were further tested using the MTT [3-(4,5-dimethylthiazol–2-yl)-2,5-diphenyltetrazolium bromide] assay to determine the cytotoxic concentration 50 (CC50), the inhibitory concentration 50 (IC50 ), and the selectivity index (SI). None of the evaluated plants exhibited AVA against VSV, whereas the extracts in hexane, ethyl acetate and methanol from Hura crepitans protected cell monolayer from infection against BHV-Bogotá with IC50 of 17.41, 7.87 and 2.75 µ g/ml respectively. The best IS was hexane of H. crepitans (SI > 17.4). These results suggest that this extract contains antiviral compounds. This research is pioneer demonstrating AVA against BHV-1B in extracts from plants of the Euphorbiaceae family.
