ABSTRACT

Objective To investigate the association of serum levels of soluble intercellular cell adhesion molecule-1 (sICAM-1),soluble vascular cell adhesion molecule-1 (sVCAM-1) and high sensitivity C-reactive protein (hs-CRP) with peripheral vascular disease of lower limbs in patients with type 2 diabetes mellitus (T2DM).Methods One hundred and thirty T2DM patients admitted from October 2011 to October 2012,and 30 age/sex-matched healthy subjects were enrolled in the study.The serum levels of sICAM-1,sVCAM-1,hs-CRP and other clinical parameters were measured; the peripheral blood vessels of lower limbs were examined with color Doppler ultrasonography.Based on the extent of angiopathy of lower limbs T2DM patients were classified as normal vascular group (n =26),mild angiopathy group (n =45),moderate/severe angiopathy group (n =59).Results The serum levels of sICAM-1 and sVCAM-1 in moderate/ severe angiopathy group of T2DM patients were higher than those in mild angiopathy group,normal vascular group and healthy controls (t:4.15-8.93,all P <0.05) ; the serum levels of hs-CRP in moderate/severe angiopathy group were higher than those in mild angiopathy group,normal vascular group and healthy controls (t:2.18-4.27,all P < 0.05).The serum sICAM-1 level was positively correlated with total cholesterol (TC),low density lipoprotein cholesterol (LDL-C) and sVCAM-1.The serum sVCAM-1 level was positively correlated with course of disease,systolic blood pressure and CRP.Conclusions Serum levels of sICAM-1,sVCAM-1 and hs-CRP are correlated with the extent of angiopathy of lower limbs in T2DM patients,and the elevated sICAM-1 ; sVCAM-1 and hs-CRP levels are also associated with hyper blood pressure,dislipidemia and chronic inflammation.

ABSTRACT

ObjectiveTo investigate the effects of alpha-lipoic acid on serum soluble intercellular adhesion molecule-1 (sICAM-1) and high sensitive-C reactive protein (hs-CRP) levels in patients with early diabetic nephropathy (DN).MethodsA total of 61 patients with early DN were randomized into treatment group (n =31 ) and control group ( n =30 ).The two groups were both treated with dietary control and oral hypoglycemic drugs or insulin.Meanwhile,the treatment group was additionally given alpha-lipoic acid 300 mg/d by intravenous infusion for 20 days.The levels of sICAM-1 and hs-CRP in each group were detected.Results The levels of sICAM-1 and hs-CRP were decreased significantly in alpha-lipoic acid treatment group after 20 days of therapy ( [ 198.03 ±23.67] μg/L vs [271.17 ±34.66] μg/L,[5.16 ±0.43] mg/Lvs [7.95 ±0.88]mg/L,P <0.01 ).Conclusion alpha-lipoic acid may decrease the expression of slCAM-1 and hs-CRP,and that may be one of the mechanisms to postpone the progress of early DN.

ABSTRACT

Objective To explore the therapeutic effects and the possible defense mechanisms of Shenmai injection on acute lung injury.Methods SD rats were randomly divided into three groups:the control group(NS),the oleic acid group(OA)and the Shenmai injection group(SM).The modal of acute lung injury in rat(OA group and SM group)were induced by receiveing 0.1ml/kg oleic acid injection into tail vein,and NS group were injected 0.9% sodium chloride 0.1ml/kg.After in jecting loeic acid SM group were injected with Shenmai injection by 2ml each one.The expression of ICAM-1 was examined meanwhile examining breath frequency,wet/dry weight ratio,artery blood oxygen each group of rats,the histopathologieal change of lung tissue was observed.Results The W/D ratio,polymorphonuclear(PMN)infiltration,the lung interstitial edema,the extent of pulmonary alvedus wall destroys and the ICAM-1 expression in lung tissue of OA group increasingly increased compared with NS group,and SM group significantly decreased compared with OA group.Also along with time extending,the ICAM-1 expression of OA group devates gradually,while SM group does not change significantly.The arterial partial pressure of oxygen in group OA significantly decreased compared with NS group,while SM group increased compared with OA group.The microscope demonstrated that the extent of lung injury of OA group was more serious than that of NS group,while OA group was milder than OA group.Conclusion Shenmai injection can lighten oleic acid induced acute lung iniury,and cut down the expression of ICAM-1 in lung tissue.So it has a certain function to the early the acute lung injury.

ABSTRACT

Objective To observe the role of Porphyromonas P.gingivalis(P.gingivalis)invasion on monocyte adhesion to endothelium and its effect on the intercellular adhesion molecule-1(ICAM-1)produc-tion in endothelial cells.Methods A cell culture model of human umbilical vein endothelial cells (HUVEC) Was used in vitro.The adhesion of monocytes to HUVEC was detected by Rose Bengal staining with or without P.gingivalis invasion.The ICAM-1 mRNA expression in HUVEC Was determined by RT-PCR or quantikine mRNA colorimetric quantification kits.Cell surface protein of ICAM-1 in HUVEC was de-termined by Western blot with or without P.gingivalis invasion.Results The adhesion of monocyte to endo-thelium and expression of ICAM-1(both at mRNA and protein level)were found to be enhanced by P.gingi-valis invasion.Pretreatment of HUVEC with anti-ICAM-l antibody partly abolished the increase in monocyte adhesion to endothelium.Furthermore.the effect of virulent strain W83 was stronger than that of avirulent strain ATCC33277.Condusion P.gingivalis plays an important role in monocyte-endothelium adhesion partly through up regulation of ICAM-1,which may be one of the pathologic mechallisms in atherosclerosis.

ABSTRACT

Objective To investigate the effects of hyperbaric oxygenation(HBO)therapy on serum con- centrations of soluble intercellular adhesion molecule-1(slCAM-1),soluble vascular cell adhesion molecule-1(sV- CAM-1),soluble E-selectin(sE-selectin)and matrix metalloproteinase-9(MMP-9)in patients with aeute cerebral infarction and their clinical implications.Methods One hundred and twelve cases of cerehral infarction in the ca- rotid artery system were assigned into two groups.Patients in the routine treatment group(RT group,n=62)were treated with routine clinical treatment regime,whereas those in the HBO group(n=50)were treated with H BO ther- apy in additioo to routine clinical treatment.Thirty age- and sex-matched normal subjects were recruited and served as controls.The serum concentrations of sICAM-1,sVCAM-1,sE-selectin and MMP-9 were measured hy using ELISA method before and 10 days after treatment.The assessment of neurological deficits using National Institutes of Health Stroke Scale(NIHSS)score was conducted before treatment,and at 10 and 30 days after treatment,and the therapeutic efficacy was evaluated.Results The concentrations of sICAM-1,sVCAM-1,sE-selectin and MMP-9 of the patients were significantly higher than those of the control subjects.These parameters were all decreased signifi- cantly after treatment in the two patient groups.Moreover,these parameters were lower in the HBO group than those in the RT group after treatment.The NIHSS scores of HBO group were significantly lower than that of the RT group at the 30th clay post-treatment.The effective rate of HBO group was higher than that of RT group.Conclusion HBO therapy can decrease the serum levels of sICAM-1,sVCAM-1,sE-seleetin and MMP-9,which might be one of the mechanisms of HBO in the treatment of cerebral infarction.

ABSTRACT

Background: TSH binding inhibiting imunoglobulins(TBII) are autoimmune antibody causing autoimmune thyroid diseases such as Graves disease or Hashimoto's thyroiditis, while intercellular adhesion molecule-1(ICAM-1) is known as a substance expressed at the site of autoimmune reaction in relation with lymphocyte infiltration. The serum TBII activity is used as an index of the disease course and prognosis of Graves disease treated with antithyroid drugs, propylthiouracil or methimazole. The aim of this study is to understand the change of serum ICAM-1 level according to the change of the degree of autoimmunity and clinical course of Graves disease. Methods: In order to study the change of soluble ICAM-1 and relationship to the immune mechanism of Graves' disease, we measured serum levels of TBII and ICAM-1 in patients(n 35) with Graves disease before and after treatment with antithyroid drugs and in relapsed patients using a highly sensitive ELISA method. Results: The serum levels of TBII and ICAM-1 were markedly elevated in patients with Graves disease before treatment than normal controls and there were good correlation between TBII and ICAM-1 level. In patients with normalized TBII levels after 22 months antithyroid drug treatment, the ICAM-1 levels became normal but in the patients with high serum TBII level showed high serum level of ICAM-1 even with clinical remission with same treatment. The serum levels of TBII and ICAM-1 in relapsed patients were elevated as those of patients before treatment. Conclusion: With the above results, we can conclude that not only the TBII level but seru ICAM-1 level also reflect the degree of autoimmune activity of Graves disease and may be used as an index of the disease course and prognosis of Graves disease treated with antithyroid drugs.
