ABSTRACT

Objective: To observe the effect of modified Erchentang on the expression of interleukin-19 (IL-19), interleukin-20 (IL-20)and their receptor IL-20R1, IL-20R2 in bronchioles of rats with chronic obstructive pulmonary disease (COPD), and to explore the molecular mechanism of modified Erchentang on anti-inflammatory of COPD. Method: The model of rat with COPD was established by cigarette smoke combined with lipopolysaccharide (LPS). The experimental rats were randomly divided into 6 groups:normal group, model group, modified Erchentang high, medium and low dose group, and Jizhitangjiang group. Normal group and model group fed with normal saline 4 mL · d-1, modified Erchentang high, middle, low dosage group(20,10,5 g · kg-1 · d-1).The dosage of Jizhitangjiang group was 12 g · kg-1 · d-1, all groups were given intragastric administration for 14 days, twice a day. To observe the general situation of rats.To evaluate the pulmonary function of rats. To detect the contents of IL-10, IL-19 and IL-20 in serum by enzyme-linked immunosorbent assay (ELISA).To observe the pathological changes of bronchiole tissue by light microscopy. To detect the expression of IL-20R1 and IL-20R2 in bronchiole tissue by immunohistochemistry. Result: Compared with normal group, peak expiratory flow(PEF), forced expiratory volume in one second(FEV1), forced vital capacity (FVC), and FEV1/FVC in model group were significantly increased (PPP1 and in bronchioles tissue significantly increased (P1, FVC, FEV1/FVC of Jizhitangjiang group, modified Erchentang high, medium and low dosage group were significantly increased(PPP1 and IL-20R2 in bronchioles tissue was significantly decreased (PConclusion: Modified Erchentang can improve the lung function and protect the tissue structure of bronchioles in COPD rats, which may be related to the inhibition of the expression of IL-19, IL-20 and their receptor IL-20R1, IL-20R2 in bronchioles of rats with modified Erchentang.

ABSTRACT

AIM:To explore the differential expression of interleukin-19 (IL-19) and its receptors (IL-20R1/IL-20R2) in the samples of chronic rhinosinusitis (CRS) with or without nasal polyps (CRSwNP and CRSsNP), and to investigate the correlation of IL-19 and its receptors with tissue remodeling factors, including matrix metalloproteinase (MMP)-2, MMP-9 and tissue inhibitor of metalloproteinase (TIMP)-1 in CRS.METHODS:The polyps from CRSwNP patients (n=30), the sinus mucosa from CRSsNP patients (n=15) and the inferior turbinal mucosa of nasal septum de-viation patients (n=15) were collected. The expression of IL-19 and its receptors (IL-20R1/IL-20R2) in each group was detected by real-time RT-PCR and immunohistochemistry. The expression of tissue remodeling factors MMP-2, MMP-9 and TIMP-1 in different groups was detected by real-time RT-PCR and ELISA.RESULTS:Increased mRNA and protein expression levels of IL-19, IL-20R1, IL-20R2 and MMP-9 were observed in CRSwNP group compared with CRSsNP and control group (P<0.05), while elevated expression level of TIMP-1 was observed in CRSsNP group compared with CRSwNP and control group (P<0.05).The relative expression of MMP-2 among the 3 groups showed no obvious difference. The expression of IL-19 and its receptors showed significantly positive correlations with MMP-9 in CRSwNP (P<0.05).No significant correlation between IL-19 and/or its receptors with TIMP-1 in CRSwNP group was observed.CONCLUSION:The high expression levels of IL-19, IL-20R1 and IL-20R2 and their positive correlations with MMP-9 in CRSwNP indica-te that IL-19 and its receptors may be involved in the tissue remodeling of chronic rhinosinusitis.

ABSTRACT

[ ABSTRACT] AIM:To evaluate the effect of intravenous injecting plasmid encoding interleukin-19-IgG on exper-imental autoimmune myocarditis ( EAM) in rats.METHODS:Cardiac myosin was emulsified with equal volume of com-plete Freund’ s adjuvant. The animal model of EAM was established by injecting with the preparation in both footpads of the Lewis rats. The rats were intravenously injected with the plasmid encoding IL-19-IgG on day 6.Echocardiography was per-formed before the rats were sacrificed on day 17.The effect of IL-19-IgG plasmid injection was evaluated by measuring the heart weight/body weight, myocarditis area, relative expression levels of atrial natriuretic peptide ( ANP) and brain natri-uretic peptide (BNP) in the hearts. The mRNA expression levels of related cytokines including IL-18, IL-1β, IL-12p35 and IFN-γwere detected.RESULTS:The rats in model group showed significant myocardial damage and a decrease in the left ventricular functions. The rats in the treatment group injected with IL-19-IgG plasmid showed an improvement of the cardiac functions. The ratio of heart weight/body weight, the area of myocarditis and the mRNA levels of ANP and BNP were significantly lower in IL-19-IgG treatment group than those in model group. The mRNA levels of IL-18, IL-1β, IL-12p35 and IFN-γwere also significantly decreased in IL-19-IgG treatment group.CONCLUSION:Intravenous injection of plasmid encoding IL-19-IgG effectively prevents the development of the left ventricular remodeling and myocardial damage in EAM rats.

ABSTRACT

Objective To study the level of interleukin(IL)-19 in peripheral blood of patients with asthma and investigate its importance. Methods Serum samples were obtained from 45 asthmatic patients (acute attack group and catabasis group) and 45 subjects (control group). The level of IL-19 and IgE in peripheral blood was respectively determined with ELJSA.The accurate number of eosinophile granulocyte (EOS) in peripheral blood was counted with cytoanalyze. Results The levels of IL-19,IgE and the quantity of EOS were (327.92 ± 245.79) ng/L,(260.48 ± 108.77) kU/L,(498.29 ± 96.46) x 107L in acute attack group,(165.54 ± 114.76) ng/L,(164.33 ± 59.08) kU/L,(224.67 ± 86.28) x 106/L in catabasis group, and (49.67 ±40.48) ng/L,(68.12 ± 25.70) kU/L,(31.84 ± 28.27) x 106/L in control group.Compared with control group, the levels of IL-19,IgE and the quantity of EOS in acute attack group and catabasis group were obvious higher than those in control group (P < 0.05).The level of IL-19 and the quantity of EOS in acute attack group were higher than those in catabasis group (P <0.05),but there was no significant difference in the level of IgE between acute attack group and catabasis group (P > 0.05).Furthermore,the results of correlation analysis showed that the level of IL-19 positively correlated with the level of IgE and the quantity of EOS respectively (r = 0.826 >0.738,P< 0.05).Conclusions The ascending degree of the IL-19 level is different during various periods of asthma. It may be inferred that IL-19 can play an important role in the pathogenesis of asthma, and observing the change of the IL-19 level dynamically is useful in diagnosing asthma and monitoring patient's condition.
