ABSTRACT

Objective: To observe the effects of NHE -1 (Na+/H+ exchanger 1) gene on the proliferative and invasive abilities of esophageal cancer cell line KYSE-70. Methods: The expressions of NHE-1 mRNA and protein in esophageal cancer EC-9706, KYSE-450, EC-1 and KYSE-70 cells were detected by realtime fluorescence quantitative-PCR and Western blotting, respectively. Chemically synthesized siRNA (small interference RNA) targeting NHE -1 was transfected into KYSE-70 cells. The expression levels of NHE-1 mRNA and protein in esophageal cancer KYSE-70 cells 24, 48 and 72 h after transfection with NHE-1 siRNA were detected by real-time fluorescence quantitative-PCR and Western blotting, respectively. The proliferative and invasive abilities of KYSE-70 cells were detected by MTT method and Boyden chamber assay, respectively. Results: NHE-1 mRNA and protein were highly expressed in KYSE-70 cells. As compared with the negative control (transfected with nonsense siRNA) and blank control (KYSE-70 cells without transfection), the expression levels of NHE-1 mRNA and protein in KYSE-70 were significantly decreased after transfection with siRNA targeting NHE -1 (all P < 0.05). As compared with the negative control, blank control and the KYSE-70 cells transfected with NHE-1 siRNA for 24 h, the proliferative and invasive abilities of KYSE-70 were inhibited after transfection with NHE-1 siRNA for 48 and 72 h (all P < 0.05). Conclusion: Silencing the expression of NHE -1 gene can inhibit the proliferative and invasive abilities of esophageal cancer KYSE-70 cells in vitro . NHE -1 gene is expected to become an important target for gene therapy of esophageal cancer. Copyright © 2013 by TUMOR.

ABSTRACT

Objective To analyze the characteristics of gastric stromal tumors(GST)under endoseopic ultrasonography(EUS)according to its aggressive risks.Methods The clinical data of 36 patients with GST,who underwent surgery from July 1997 to July 2007,were analyzed retrospectively.All the patients underwent EUS before operation and were classified according to Fleether's 4-tier system to predict the aggressiveness of the tumors.The features of the tumor under EUS including its size,ulceration,border,echo charateristies and growth pattern were recorded and the difference between each tumor group were analyzed by ANOVO and rank sam test.Results The mean maximal diameter of GST Was 7.3 cm(range 1.0-20.0 cm),and the size of tumors with hiisher aggressiveness risk Was significantly larger than that with lower risk(P<0.01).The mucosal ulceration,obscure border,irregular shape and echo heterogeneity were more commonly seen in the groups with higher risk(P<0.05).There Was no difference in the internal echo and growth pattern of GST between different groups.Conclusion EUS features are useful in differentiation of the aggressiveness risk of the GST.and guide the management of the tumors.

ABSTRACT

PURPOSE To provide evidence for the relationship between degree of invasion and tumor metastasis. The tumor cells of transplantable mouse histiocytic sarcoma (L1) were inoculated at the right hind footpads of inbred 615-strain mice. METHODS Once bearing the tumor, all the mice were sacrificed respectively on the 1st. 3th, 5th. 10th, 20th, 30th, and 40th day as to observe the degree of tumor invasion and the process of tumor metastasis. RESULTS When the degree of tumor invasion was grade Ⅲ or Ⅳ , the metastasis of tumor cells was found earliest in draining lymph nodes. However, the tumor metastasis in lung appeared later than in lymph nodes. CONCLUSION According to the time of tumor growth and degree of invasion and metastasis, the authors suggest a new classification for cancer staging, namely, latent, invasive and metastatic stages. During the stage of tumor invasion, it is redivided into early, middle and late invasive phases. During the stage of tumor metastasis, it is redivided into early, middle and late metastatic phases.
