ABSTRACT

To investigate the changes of immunological function of LAK,NK cell activity and T lymphoctyes subsets before and after operation in patients with hepatic cancer.The activity of Lymphokine-activated Killer Cell (LAK|,Natural Killer Cell(NK) and T lymphocytes subsets were detected in 31 patients with hepatic cancer of preoperation and postoperation,so did the 30 normal volunteers(as control group).The results showed that the activities of LAK,NK cell and T lymphocytes subsets of CD3,CD4 and the ratio of CD4/CD8 in patients with hepatic cancer were lower than that in normal control group(P

ABSTRACT

Purpose:To study effect of peripheral blood lymphocyte subsets in autologous LAK cell treatment of advanced lung adenocarcinoma and to discuss the prognosis.Methods:21 surgically resected cases, were confirmed as primary lung adenocarcinoma stage Ⅲ or Ⅳ. ABSTRACT

This essay investigated the in vitro tumoricidal activity of Con A primed murine LAK cells (Con A-LAK), discussed the role of mTNF-? in the killing effect. The results showed that Con A-LAK cells could kill NK sensitive YAC-1, TNF sensitive L929 and H-2 low/non expression HCa-F target cells; The cytotoxicitity of Con A-LAK cells was performed by direct contact, the non-secretary pathway mediated by interaction of surface ligands of effector cells and receptors of target cells play an important role; Con A-LAK cells expressed mTNF-?, which participated in the killing of TNF sensitive tumor cells and might be one of the important effector molecules in MHC non-restricted, antigen non-specific cvtotoxicitv.

ABSTRACT

Using a syngeneic transplantable leukemia model of 615 inbred mice, named L615, we (1) compared the adoptive immunoprophylactic(AIP) effects of spleen cells from various normal dornor mice with different genetic background and from different immune-deficient mice;(2) studied the adoptive immunotherapeutic (AIT) and adoptive chemo-immunotherapeutic(ACIT) effects of immune spleen cells from those survival mice in the AIP groups;(3) analysed the antitumor activities of various murine spleen cells either nonspecifically activated in vitro or specifically induced in vivo and resensitized in vitro by MMC-treated L615 tumor cell vaccine (TCV).Our results might provide some new experimental evidences for clinical application of cancer ACI.

ABSTRACT

Objective:To study the effects of WAK on immune function in mice.Methods: After different doses of WAK were given to mice ig, its effects on delayed type hypersensitivity (DTH), hemolysin production, NK and LAK cell killing activity were observed.Results: WAK could enhance DTH and hemolysin production in immunosuppressive mice and improve NK and LAK cell killing activity in mice.Conclusions: WAK could exhibit its antitumor effect by increasing immune function.

ABSTRACT

Two hour after LAK—like cells were co—cultured with gastric cancer cells,the mixed sus-pension was spread on slids and dyed with histochemical method.Through analysing the dyedcells,we directly confirmed what subset the LAK—like cells belonged to.The featrue of LAK—like cells adhering to cancer cell was observed under electron micrscope in our experiment.A-mong the lymphoctes which were adherent to cancer cells,the number of CD8~+ cells was in-creased.Most of adherent to cancer cells,the number of CD8~+ cells was increased.Most of ad-herent cells were TAC~+,suggesting that the expression of TAC antigen plays an important rolein cell mediated cytotoxity.

ABSTRACT

The super severe combined immunodeficient mice(superscid)is a newly established mouse strain which was constructed by introducing the bg gene into C.B-17 scid mice.To identify its cellular immunologic haracteristics,we examined the T、natural killer(NK)and lymphokine-activated killer (LAK)cell activity in this mouse strain.The NK cell activity was studied by measuring the cytotoxicity to NK sensitive cell(YAC-1)and the influence of poly Ⅰ-C on it.The LAK cell activity was examined by measuring the cytotoxicity aganist NK resistant cell(p815).The spleen cells of B.C.B-17 scid-beige mice lacked responsiveness to a T cell mitogen (concanavalin A 5?g/ml,600.004?207.84cpm,266.67?29.84cpm for scid mice,p0.05);showed a low LAK cell activity(6.40?6.9 for B.C.B-17 Scid-beige mice,vs 72.3?14.1% for C.B- 17 scid mice at IL-2 250u/ml;E/T 00:1,3 days,P

ABSTRACT

In the present experiment, we investigated the im munoprophylactic and immunotherapeutic effects of transferring spleen LAK cells or " LAK-88/DC " cells from 615 inbred mice against mouse leukemia. The results showed that transferring LAK cells or "LAK-88/DC" cells and injecting conditional medium for a week 21 days before L615 inoculation induced significant prophylactic effects, which cured 80% of the L615-sufferring mice. Injecting conditional medium alone was also effective, which cured 60% of the mice, While transferring LAK cells or "LAK-88/DC" cells at the same time with L615 inoculation or 1 day after L615 inoculation presented no apparent therapeutic effect. However, when combined with cyclophosphamide (CY) chemotherapy, the transfer of LAK cells or "LAK-88/DC " cells 5 days after L615 inoculation could prolong the mean survival time and even cured 25%-28.6% of the advanced L615 leukemic mice.
