ABSTRACT

Aim To investigate the potential protective effect of quercetin on chronic ethanol exposure induced autophagy dysfunction .Methods The male C57 BL/6 J mice were divided randomly into four groups: Nor-mal control group ( Con);Ethanol group ( Eth);Etha-nol plus Quercetin group ( Eth+Qu); Quercetin con-trol group ( Qu ) .The mice were pair-fed with either regular or ethanol-containing Lieber De Carli liquids diets for 15 weeks and the related parameters were as-sayed .Result Compared to Con group , chronic etha-nol feeding led to serious aminotransferases leakage , hepatic lipid accumulation , and liver oxidative dam-age.Additionally, long-term ethanol exposure caused lysosome damage , cathepsin B leakage to cytoplasm and decreased the expression of LAMP 2A which was the key protein for the autophagosome-lysosome fusion . As expect , quercetin supplementation evidently allevia-ted lysosome damage and decreased the abnormal LC 3-II and p62 accumulation , accelerated the autophago-some-lysosome fusion and increased the expression of LAMP2A.Conclusion Quercetin may alleviate the liver injury induced by chronic alcohol exposure by a-meliorating the lysosomal damage and up-regulating LAMP2 A expression .

ABSTRACT

Objective To observe the effects of Xianfu Wenyang Tongluo Drink (XFWYTLD) on ANCA associated antigen in rats with thromboangiitis obliterans;To discuss its mechanism. Methods Totally 72 SPF male Wistar rats were randomized into sham-operation group, model group, Mailuoning Granule group and XFWYTLD low, medium, and high dose groups. Method of femoral artery injecting sodium laurate was used to duplicate models. From the second day after modeling, the rats in sham-operation group and model group were fed with distilled water, while other groups received gavage with relevant medicine. 15 days later, the activity of MPO in serum was detected through the method of ultraviolet spectrophotometry;the protein expressions of PR3 and LAMP-2 in femoral artery and the surrounding tissues were detected through the method of immunohistochemisty. Results Compared with sham-operation group, the activity of MPO in serum and the protein expressions of PR3 and LAMP-2 in rats of model group were significantly higher;Compared with model group, the activity of MPO in serum and the protein expressions of PR3 and LAMP-2 in rats of all medication administration groups decreased, among which the XFWYTLD medium dose group showed the most obvious decrease. Conclusion XFWYTLD may lower levels of ANCA associated antigen, and further inhibit humoral immune function.

ABSTRACT

Danon disease is characterized clinically by the triad of cardiomyopathy, myopathy and mental retardation. It was originally reported as a lysosomal glycogen storage disease with normal acid maltase by Danon. Danon disease results from mutations in lysosome associated membrane protein-2 (LAMP-2) gene. The LAMP-2 gene is located on Xq24-25. We report a case of suspected Danon disease in patient who had hypertrophic cardiomyopathy and mental retardation along with abnormal findings in electromyography.
