ABSTRACT

Lysophosphatidylcholine (LPC) modulates the dynamic and integral process of macrophage polarization in immune responses, tissue inflammation and remodeling. Patatin-like phospholipase domain containing protein 7 (PNPLA7) was identified as an LPC-preferring lysophospholipase recently. However, the expression and role of PNPLA7 in macrophage polarization remained unknown. In the present study, PNPLA7 was found to be upregulated in the process of macrophage polarization toward an alternatively activated (M2) phenotype stimulated with interleukin 4 (IL-4) (P<0.05). We found that knockdown and overexpression of PNPLA1 decreased and increased the expression of M2 marker genes, including arginase 1 (Argl) and chitinase-like 3 (Ym\ ), respectively (P<0.05). Further studies showed that PNPLA7 regulated the expression of peroxisome proliferator activated receptor-γ (P P A R γ) at the mRNA and protein levels during M2 polarization (P < 0.05). However, the phosphorylation of signal transducer and activator of transcription 6 (STAT6) was not influenced by PNPLA7. These findings suggest that PNPLA7 favors macrophage anti-inflammatory M2 polarization through a PPAR

ABSTRACT

El análisis acústico de la voz es un método de evaluación vocal objetivo y no invasivo, utilizado con fines diagnósticos, terapéuticos e investigativos. Utiliza registros acústicos obtenidos directamente de la voz del paciente mediante el uso de un micrófono profesional. Existe una gran variedad de programas y métodos de graficación disponibles, que favorecen la versatilidad y especificidad del proceso. Esta revisión muestra los métodos de graficación de la voz más utilizados.


The acoustic voice analysis is an objective and non invasive vocal assessment method, used for voice diagnosis, therapy and research. It uses acoustic records obtained from the patient's voice using professional microphones. There is a variety of softwares and graphing programs available that allows process versatility and specificity. This review presents the most utilized methods of voice graphing.

ABSTRACT

The chiropterans constitute 25% of the world's mammal fauna. Due to the destruction of their natural ecosystem, the vampire bats have moved from nature to artificial roosts closer to man and domestic animals. This phenomenon has happened particularly in rural areas. Rabies is a viral anthropozoonosis, 100% lethal, and vampire bats (Desmodus rotundus) represent an important role in its epidemiology. D. rotundus were captured at night with mesh nets in partnership with the Botucatu Defense Office and sent to the Zoonosis Diagnostic Service, at the School of Veterinary Medicine and Animal Husbandry, UNESP. Serum samples from 204 bats were analyzed by enzyme-linked immunosorbent assay (ELISA) and fluorescent antibody viral neutralization test (FAVN) for rabies antibody detection. The results showed 7.4% of sera with titers higher or equal to 0.5 U for rabies antibodies, which demonstrated viral flow circulation among the studied region. Data suggest a need for constant monitoring accomplished by epidemiological and sanitary measures.(AU)

ABSTRACT

Lysophosphatidylcholine (LPC), which accumulates in atherosclerotic arteries, has been reported to inhibit endothelium-dependent relaxation (EDR) in many different species. However, the underlying mechanism of LPC-induced inhibition of EDR is still uncertain. In the present study, we measured simultaneously both isometric tension and cytosolic free Ca2+ ([Ca2+]i) in rabbit carotid strips, and examined the effect of LPC on tension and [Ca2+]i. In carotid strips with intact-endothelium, high K+ (70 mM) increased both tension and [Ca2+]i, and cumulative addition of acetylcholine (ACh) from 0.1 to 10microM induced dose dependent increase of [Ca2+]i with concomitant relaxation. In the presence of L-NAME (0.1 mM), ACh increased [Ca2+]i without affecting the amplitude of high K+-induced tension. These ACh-induced change of [Ca2+]i and tension was abolished by removal of endothelium or 10 nM 4-DAMP (muscarinic receptor antagonist) pretreatment. Pretreatment of LPC (10microM) inhibited ACh (10microM)-induced change of tension and [Ca2+]i in endothelium-intact carotid artery. On the other hand, LPC had no effect on ACh-induced change of tension and [Ca2+]i in endothelium denuded artery. In Ca2+-free external solution, ACh transiently increased [Ca2+]i, and pretreatment of LPC significantly inhibited ACh-induced transient [Ca2+]i change. Based on the above results, it may be concluded that LPC inhibits the ACh-induced [Ca2+]i change through inhibition of Ca2+ mobilization in vascular endothelial cells, resulting in decreased production of NO and concomitant inhibition of endothelium-dependent vascular relaxation.

This paper introduces the principles of an Environmental Temperature Supervising Network System, explain its hardware and software structure and the function of network system. The whole system is proved stable and reliable by experiment. It may have some profit in the market and great future in the application.

ABSTRACT

Effects of oxidized low-density lipoprotein (ox-LDL), l-alpha-stearoyl-lysophosphatidylcholine (LPC), on intracellular Ca2+ concentration were examined in mouse endothelial cells by measuring intracellular Ca2+ concentration ([Ca2+]i) with fura 2-AM and reverse transcription-polymerase chain reaction (RT-PCR). LPC increased [Ca2+]i under the condition of 1.5 mM [Ca2+]o but did not show any effect under the nominally Ca2+-free condition. Even after the store depletion with 30microM 2,5-di-tert- butylhydroquinone (BHQ) or 30microM ATP, LPC could still increase the [Ca2+]i under the condition of 1.5 mM [Ca2+]o. The time required to increase [Ca2+]i (about 1 minute) was longer than that for ATP-induced [Ca2+]i increase (10-30 seconds). LPC-induced [Ca2+]i increase was completely blocked by 1microM La3+. Transient receptor potential channel(trpc) 4 mRNA was detected with RT-PCR. From these results, we suggest that LPC increased [Ca2+]i via the increase of Ca2+ influx through the Ca2+ routes which exist in the plasma membrane.
