ABSTRACT

La disponibilidad de cepas bacteriana para el estudio de la resistencia bacteriana es clave para los avances en la investigación básica y clínica respecto del tema. Existen pocos biorrepositorios o bancos de bacterias con mecanismos de resistencia conocidos, aisladas de infecciones clínicamente significativas. Una revisión de la literatura revela que sólo en los Estados Unidos de América existe un biobanco de aislados resistentes disponibles para estudios. En esta publicación se cuenta cómo se creó el primer biorrepositorio de bacterias resistentes en Chile asociados a la Red de Laboratorios MICROB-R, con la participación de 11 centros distribuidos a lo largo del país, que a la fecha cuenta con más de 3.500 aislados bacterianos estudiados fenotípica y genotípicamente, disponibles para la comunidad científica chilena.


The availability of bacterial strains for the study of bacterial resistance is key to advances in basic and clinical research. There are few biobanks of bacteria with known resistance mechanisms, isolated from clinically significant infections. A review of the literature reveals that only in the United States of America is there a biobank of resistant isolates. This publication shows the creation of the first biorepository of resistant bacteria Chile associated with the MICROB-R Laboratory Network, with the participation of 11 centers distributed throughout the country, which to date has more than 3,500 bacterial isolates studied phenotypically and genotypically, available to the Chilean scientific community.

ABSTRACT

Objective@#To evaluate the running status of Qinghai provincial measles laboratory network during 2012—2017.@*Methods@#To analyze serological and virological surveillance database developed in Qinghai provincial measles laboratory network in 2012—2017, and evaluate the indicators for the running status of measles laboratory network in Qinghai Province.@*Results@#It was shown that 5 763 suspected measles cases were reported in Qinghai Province during 2012—2017, and 4 167 serum samples were collected from suspected measles cases, the collection rate is 72.31%; among them, 3 697 were IgM positive for measles and the positive rate was 88.72%; 68 were IgM positive for rubella and the positive rate was 1.63%. And 515 throat swab specimens were collected from suspected measles cases, 82 measles virus isolates were obtained and the positive rate was 15.92%. The result of sequencing and analysis showed that all the measles viruses belonged to genotype H1 and subgenotype H1a, which were predominant genotype circulated in China in recent years. In addition, Qinghai provincial measles network labs passed all the serological confirmatory test and proficiency test, and on-site review held by national or provincial measles laboratory and WHO during 2012—2017, respectively.@*Conclusions@#Qinghai provincial measles laboratory network has been established and running well. It provided important scientific basis for measles elimination in Qinghai province.

ABSTRACT

Objective To investigate the effect of measuring value transfer for human serum samples assigned by the reference laboratory network on improving the trueness of seven enzyme activities in clinical laboratories,such as ALT,AST,GGT,LDH,CK,AMY and ALP.Methods Depending on the medical imtitutions at all levels contacted by 5 reference laboratories in North China,South China,East China and Southwest China,the corresponding clinical laboratory measuring value transfer/traceability network was established.The frozen human serum samples with good interehangeability and standard material characteristics,including calibrator,sample 1 and sample 2,were provided by Beijing Aerospace General Hospital,and were assigned by 5 reference labotatories in four regiom.These samples were sent to 48 clinical laboratories.These clinical laboratories measured sample 1 and sample 2 according to their standard operating procedures,and then measured.the two samples again after adjusting their measurement system by using the supplied calibrator.The changes of trueness of detection results in these laboratories were evaluated according to the WS/T 403-2012 standard,and the changes of consistency for ALT and AST before and after measuring value tramfer were investigated.Results The results of AMY,ALP,GGT,CK and LDH calibrator,sample 1 and sample 2 assigned by the established network were 138.7 U/L,278.5 U/L and 68.3 U/L,265.3 U/L,94.5 U/L and 134.4 U/L,195.8 U/L,89.0 U/L and 158.9 U/L,393.7 U/L,260.0 U/L and 645.3 U/L,and 302.0 U/L,250.0 U/L and 452.7 U/L,respectively.The percentages of sample 1 and sample 2 which met the bias requirements of the WS/T 403-2012 standard before measuring value transfer for AMY,ALP and GGT were 65.9% and 61.0%,76.6% and 78.7%,and 66.7% and 70.8%,respectively,while after measuring value transfer,they were 89.2% and 83.8%,86.7% and 80.0%,and 85.4% and 91.7%,respectively.The percentages of sample 2 which met the bias requirements of the WS/T 403-2012 standard before measuring value transfer for CK and LDH were 64.6% and 58.3%,respectively,while after measuring value trander,they were 93.5% and 84.8%,respectively.The coefficients of variation (consistency) of sample 1 and sample 2 for ALT and AST before measuring value tramfer were 12.9% and 11.3%,and 10.2% and 8.9%,respectively,while after measuring value transfer,they were 9.3% and 8.2%,and 5.6% and 5.9%,respectively.Conclusion The calibration of routine measurement systems based on the measuring value transfer for human serum samples assigned by the reference laboratory network may improve the comparability of 7 enzyme actvities measurement results in chnical laboratories at all levels obviously,which deserves to be further spread.

ABSTRACT

Objective To summarize and review the comparative results of the isotope dilution mass spectrometry in Cholesterol Reference Method Laboratory Network (CRMLN)project of the Centers for Disease Control and Prevention (CDC)of the United States in order to provide quality controls for determination of blood lipid.Methods The isotope dilution liquid chromatography tandem mass spectrometry (ID-LC/MS/MS)methods for determination of serum total cholesterol (TC)and triglycerides (TG)were developed to participate in the comparison of CRMLN.The survey was conducted every three months before 2016 and every half year from 2016.Four kinds of reference materials with two parallel tubes for each material and each tube in duplicate were determined in every survey.At least two certified reference materials used as quality control samples were analyzed simultaneously in each determination.Results In the 15 comparisons the CV of the TC determination method in our laboratory was 0.43% while the CV of all the participated laboratories in CRMLN was 0.42%.The bias from the overall mean value in our laboratory was 0.22% while the bias from the CDC target values was 0.58%.The CV of the TG determination method in 15 tests of our laboratory was 0.62% and the bias was-0.98% from the overall mean value and-0.80% from the target values of CDC.Among the 60 results for comparison,98% (59/60)of CV in the TC determination met with CDC requirement for precision (CV≤ 1%),and 70% of bias (42/60) of the results met with CDC requirement for accuracy (bias ≤ 1%).For the 60 results in comparison of TG determination,92% (55/60)of bias of the results met with the accuracy requirement of CDC (bias ≤2.55%).Conclusion In CRMLN comparison the results of TC and TG determined by ID-LC/MS/MS method were consistent with the values which were certified by CDC and determined by other network laboratories.The comparative surveys may play an important role in the standardization of lipid determination,and should be expected to provide experiences and technical supports in the comparative plan for reference measurement in our country.

ABSTRACT

Se describe la participación del Laboratorio Nacional de Referencia e Investigaciones en Tuberculosis y Micobacterias del Instituto de Medicina Tropical Pedro Kourí, en la organización del recurso bacteriológico para el diagnóstico y la búsqueda de casos, fundamentalmente en la atención primaria de salud, en apoyo al Programa Nacional de Control de la Tuberculosis. Se hace referencia a las funciones y principales actividades científicas de este Laboratorio Nacional, Centro Colaborador OPS/OMS, como cabecera de los laboratorios de la red de diagnóstico de la tuberculosis en el país. La introducción y aplicación de nuevas tecnologías de avanzada permite contar con valiosas herramientas para dar respuesta a los diferentes retos que plantean la tuberculosis y las micobacteriosis en la actualidad. Con la ejecución de un proyecto del fondo mundial de lucha contra el sida, la tuberculosis y la malaria, se ha fortalecido la red de diagnóstico, lo que debe contribuir, en un futuro cercano, a la eliminación de la tuberculosis como problema de salud en Cuba


The participation of the National Laboratory of Reference and Research on Tuberculosis and Mycobacteria of Pedro Kouri Institute of Tropical Medicine in the organization of the bacteriological resources for the diagnosis and screening of cases in the primary health care, in support of the national program for control of tuberculosis, was described in this paper. Reference was made to the functions and main scientific activities of this laboratory, which is a PAHO/WHO collaborating center at the forefront of the tuberculosis diagnosis laboratory network in the country. The introduction and the implementation of state-of-the-art technologies have allowed valuable tools to be used to meet several challenges posed by tuberculosis and mycobacteria nowadays. Thanks to the implementation of a World Fund against AIDS, tuberculosis and malaria project, the diagnosis network has strengthened, which must contribute to the elimination of tuberculosis as a health problem in Cuba in the near future
