ABSTRACT

Lamiophlomis Herba, one of the medicinal herbs commonly used by the Tibetan ethnic minority, has the effect of activating blood, stopping bleeding, dispelling wind, and relieving pain and is frequently used to treat trauma, traumatic bleeding, rheumatic arthralgia, and dampness and dampness-heat. By a review of ancient Tibetan medicine classics, materia medica classics, and modern literature, this paper systematically analyzed the name, nature, taste, original plant, medicinal part, harvest, processing, efficacy, and indications of Lamiophlomis Herba. The textual research showed that Lamiophlomis Herba was first recorded in the Somaratsa (《月王药诊》) in the middle of the eighth century. This medicinal herb was mainly recorded with a plain and warm nature and a sweet, bitter, and astringent taste. The herb was recorded as non-toxic in other books except the Tibetan Medicinal Plants in Gannan of Qinghai-Tibet Plateau (《青藏高原甘南藏药植物志》). In the books of the past dynasties, the aboveground part of Lomiophlomis rotatd was used as medicine. In addition, there were also records of using the whole herbs of Ajuga ovalifolia, A. ovalifolia var. calantha, and Oreosolen wattii as the medicine. In ancient times, the herb was mainly harvested before Frost's Descent in the 8th-9th months of the lunar calendar, while it was mainly harvested during the flowering-fruiting stage in autumn in modern times. Due to the decreased reserve of Lamiophlomis Herba, the medicinal part evolved from whole herb or fresh leaves to the aboveground part. According to the ancient and modern records, this herb mainly has the functions of nourishing bone and marrow, eliminating dampness and dampness-heat, stopping bleeding, and relieving pain. In ancient times, it was used for treating parasitic diseases, menostaxis, spermatorrhea, diarrhea, and nourishing the body. This paper aims to provide a basis for further development and study of Lamiophlomis Herba through the textual research.

ABSTRACT

OBJECTIVE To evaluate the quality of total flavonoids of Lamiophlomis rotata from different producing areas. METHODS Total flavonoids of S1-S15 batches of L. rotata from different producing areas were extracted by percolation and purified by polyamide column. The content of total flavonoids was determined by UV spectrophotometry, and its purity was calculated. HPLC fingerprint chromatograms and control fingerprint of total flavonoids from 15 batches of L. rotata were established with Similarity Evaluation System of Chromatographic Fingerprint of TCM (2012 version). Their similarities were analyzed. Cluster analysis, principal component analysis and orthogonal partial least squares-discriminant analysis were used to evaluate the quality of total flavonoids from 15 batches of L. rotata, and the main components which affected the quality of total flavonoids were analyzed. RESULTS The purities of total flavonoids from 15 batches of L. rotata were 55.82%-94.12%, with an average value of 77.72%; a total of five common peaks were identified in the fingerprint, and No. 3 peak was luteolin; the similarities between the fingerprint of each batch of samples and the control fingerprint were 0.925-1.000. By cluster analysis, S1 and S3-S9 were clustered into the first class, which were samples from Qinghai Province and Tibet Autonomous Region. S14 and S15 were clustered into the second class, which were samples from Yunnan Province. S10-S13 were clustered into the third class, which were all samples from Sichuan Province. S2 was clustered into the fourth class. The principal component analysis showed that the qualities of samples from the first and fourth classes were better; peaks 2, 3 and 5 were identified as the main components that caused the differences among different batches of samples by orthogonal partial least squares-discriminant analysis. CONCLUSIONS The qualities of total flavonoids of L. rotata from Qinghai Province and Tibet Autonomous Region are better.

ABSTRACT

Objective: The macroporous adsorptive resin chromatographic column combining with polyamide chromatographic column was used to separate the flavonoides, phenylethanoidglycosides, iridoid glycosides, and high polarity ingredients (such as alkaloid salts and sugars) in industrialization. Methods: Water extract from Lamiophlomis rotata (WELR) was separated and enriched by series and parallel method of polyamide and macroporous adsorptive resins and CCPP serial laminar analysis, and was ultrafiltrated by LM-125 ultrafiltration apparatus, then was dried under the reduced pressure by JYT-50 LN multipurpose kinesis back flow extract concentration subassembly. WELR, 70% ethanol solution eluate I (70% ethanol solution eluted polyamide column), 70% ethanol solution eluate II (70% ethanol solution eluting macroporous resin column), and ultrafiltration matter. The total flavonoids and total phenylethanoid glycosides were determined by aluminum nitrate-sodium nitrite UV spectrophotometry. The total iridoid glycosides were determined by the first derivative spectrophotometry. Shanzhiside methylester and 8-O-acetyl shanzhiside methylester were determined and the changes of total iridoid glycosides ingredients were investigated in the different elution requirement by HPLC. Results: Ethanol (70%) was used as dissolvent. Using the "introduction from superior surface" sample introduction mode and "introduction from superior surface" eluting mode in polyamide chromatographic column, the "introduction from inferior surface" sample introduction mode and "introduction from superior surface" eluting mode in the macroporous adsorptive resin chromatographic column, yields of 70% ethanol solution eluate I, 70% ethanol solution eluate II, and ultrafiltration matter were 2.43%, 20.80%, and 82.60%, respectively. Conclusion: This production technology may transform the experimental achievement to industrial production successfully.

ABSTRACT

Objective To study the effect of Tibet medicine of Herba Lamiophlomis rotata on promoting hemostasis and blood coagulation and to explore its mechanism. Methods High dose(2 g/kg),middle dose(1 g/kg) and low dose (0.5 g/kg) of the aqueous extract from Herba Lamiophlomis rotata(HLRE)were given to mice by gastric gavage for 3 days, and then bleeding time (BT),clotting time(CT) and platelet count(PLC)were determined. Different doses of HLRE(3,1.5,0.75 g/kg)were administered by gastric gavage to rats for 14 days, and then blood samples were collected from common carotid artery for the determination of prothrombin time (PT),activated partial thromboplastin time (APTT),thrombin time(TT),fibrinogen(FIB),hepatic function and blood lipid indexes. Results Compared to the blank control group, the bleeding time and clotting time were obviously shortened in 2g/kg HLRE and 1g/kg HLRE groups and positive control group, but the difference of PLC was insignificant. PT and APTT values in all of the treatment groups did not differ from those in the blank control group. However, TT values were obviously shortened in 3 g/kg HLRE group,FIB and Albumen(Alb)values increased, the aminotransferase and blood lipid values tended to decrease. Conclusion The aqueous extract from Herba Lamiophlomis rotata has an effect on promoting hemostasis and blood coagulation ,and its mechanisms may be related to the increase of FIB and Alb contents and the shortening of TT value.
