ABSTRACT

Humankind has always been fascinated by venomous animals, as their toxic substances have transformed them into symbols of power and mystery. Over the centuries, researchers have been trying to understand animal venoms, unveiling intricate mixtures of molecules and their biological effects. Among venomous animals, Latrodectus Walckenaer, 1805 (widow spiders) have become feared in many cultures worldwide due to their extremely neurotoxic venom. The Latrodectus genus encompasses 32 species broadly spread around the globe, 14 of which occur in the Americas. Despite the high number of species found in the New World, the knowledge on these spiders is still scarce. This review covers the general knowledge on Latrodectus spp. from the Americas. We address widow spiders' taxonomy; geographical distribution and epidemiology; symptoms and treatments of envenomation (latrodectism); venom collection, experimental studies, proteome and transcriptome; and biotechnological studies on these Latrodectus spp. Moreover, we discuss the main challenges and limitations faced by researchers when trying to comprehend this neglected group of medically important spiders. We expect this review to help overcome the lack of information regarding widow spiders in the New World.(AU)

ABSTRACT Introduction: Accidents involving spiders bites usually cause mild medical reactions that lead to local symptoms and, less commonly, systemic effects. The most medically significant spiders belong to the genera Latrodectus and Loxosceles. This paper presents a possible case of steatodism in a young woman and her pet. Case description: 26-year-old female patient, who reports a clinical history characterized by paresthesia, malaise, fever, diarrhea and a painful papule in the left cheek after being bitten by a spider. Immediately after being bit, the patient hit the spider with the back of her hand and it fell to the ground, where her dog swallowed it. The dog presented with vomiting and general discomfort after ingestion. Symptomatic therapy was given for comfort, and neither the patient nor the dog required antivenin therapy. Both evolved favorably. Discussion: The relevance of this case is the involvement of two mammals (a human and her dog) due to the accidental contact with a spider, possibly of the genus Latrodectus or Steatoda. Conclusion: Two possible cases of steatodism are described. Since spider bites are a relatively frequent reason for medical consultation in Colombia, it is important to diagnose and manage them properly.


RESUMEN Introducción. Los accidentes producidos por arañas suelen ser eventos médicos poco severos. La mordedura provoca, por lo general, síntomas locales y, menos frecuente, efectos sistémicos. Las arañas más importantes desde el punto de vista médico pertenecen a los géneros Latrodectus y Loxosceles. Se reporta un posible caso de steatodismo en una mujer joven y su mascota. Presentación del caso. Paciente femenino de 26 años quien presenta cuadro clínico de 5 días de evolución caracterizado por parestesia, malestar general, fiebre, diarrea y una pápula dolorosa en la mejilla izquierda después de ser mordida por una araña. La joven golpea la araña inmediatamente con el dorso de la mano y esta cae al suelo, donde su perra la ingiere; esta última presenta vómito y malestar general después de la ingesta. Como tratamiento se administró terapia sintomática y ninguna de ellas requirió antiveneno. Ambas evolucionaron favorablemente. Discusión. Se presentan dos posibles casos de steatodismo, siendo el aspecto relevante del presente caso que tanto la paciente como su mascota presentaron síntomas debido al contacto accidental con una araña, posiblemente del género Latrodectus o Steatoda. Conclusión. Dado que los accidentes por mordedura de araña son relativamente frecuentes para la consulta médica en Colombia, es importante diagnosticarlos y manejarlos de forma adecuada.

Latrodectus geometricus (Araneae: Theridiidae), es una araña llamada vulgarmente "viuda marrón". Originaria de África, es una especie sinantrópica y cosmopolita, muy frecuente en varias ciudades del centro y norte de Argentina. A diferencia de otras especies del género no suele causar envenenamientos graves, habiéndose registrado en el mundo muy pocos casos que requirieron tratamiento los cuales evolucionaron siempre de forma favorable. Todos los casos registrados hasta la fecha fueron atribuidos a hembras, asumiendo que los machos no causan casos clínicos e incluso serían incapaces de atravesar con sus quelíceros la piel humana debido a su pequeño tamaño. Este trabajo presenta el primer registro de un accidente causado por un macho de L. geometricus describiendo los síntomas. El accidente ocurrió a una mujer de 21 años en Villa Gobernador Gálvez (33°01'31" S, 60°38'01" O), Santa Fe, Argentina, en marzo 2016. El ejemplar murió en el momento siendo conservado y luego identificado según su genitalia como un macho adulto de L. geometricus. Los síntomas se relevaron desde el accidente hasta el cese de las manifestaciones sin intervención terapéutica. En el momento la joven advirtió una sensación punzante seguida inmediatamente de ardor y eritema. Luego de 10 minutos el ardor aumentó, disminuyendo a los 20 minutos. Luego de una hora el ardor y el eritema disminuyó considerablemente. Los resultados observados respaldan que L. geometricus no reviste peligrosidad en Argentina y evidencian que los quelíceros de los machos pueden atravesar la piel humana, al menos en zonas de piel delgada.


Latrodectus geometricus (Araneae: Theridiidae) is a spider commonly known as "Brown widow". Originally from Africa, is a synanthropic and cosmopolitan spider, very common in several cities in central and northern Argentina. Unlike other species of the genus, this spider does not cause serious poisoning, very few have been registered that required treatment and always evolved favorably. All cases listed to date were attributed to females, assuming that males do not cause clinical cases and would even be unable to pass through human skin due to the small size of their chelicerae. This work presents the first record of an accident by a male of L. geometricus and describes the symptoms. The accident occurred to a 21 year old woman from Villa Gobernador Gálvez (33°01'31" S, 60°38'01" W), Santa Fe, Argentina, in March 2016. The specimen died at the time, being preserved and then identified as an adult male of L. geometricus according to its genitalia. The symptoms were relieved from the accident to its end without applying treatments. At the time, the woman felt a sting followed immediately by burning sensation and erythema in the area. After 10 minutes the burning increased, decreasing at 20 minutes. One hour later, burning sensation disappeared and the erythema diminished considerably. The observed results support L. geometricus as not dangerous in Argentina and show that males actually are able to penetrate the human skin, at least in areas of thin skin.

ResumenSe reporta un accidente causado por la araña Latrodectus geometricus, incluida en el grupo conocido como “viudas negras”, en Costa Rica. El accidente le ocurrió a una mujer de 27 años vecina de la ciudad de Cartago, en su casa de habitación. Los síntomas mostrados cursaron la siguiente secuencia: dolor intenso en el área del piquete, enrojecimiento y aumento en la temperatura al tacto alrededor de ese punto, ampliación del área del dolor, sudoración, dolor de espalda, arritmia, taquicardia y aumento de la presión arterial. Los síntomas mostrados en este caso son muy similares a los descritos por personas picadas por la misma u otras especies de viudas negras en Suramérica y Centroamérica.


AbstractWe here report a case of human lactrodectism in Costa Rica, caused by the spider Latrodectus geometricus, included in the group known as “black widows”. A 27 year-old woman was bitten by a Brown Widow spider in her home in Cartago city. She showed a moderately severe case with the following sequence of symptoms: an acute, intense local pain begins just after being bitten, and this irradiates to the entire affected limb, redness, increase in temperature around the bitten area, sweating and backache. At the cardiovascular level, she showed tachycardia and hypertension. The symptoms showed in this case are quite similar to those cases described for the Brown Widow spider and for other Black Widow spiders, in Central and South America.

By at least 1841, the species had successfully established populations in South America and has more recently expanded its range to the southern states of North America. This highly adaptable spider has been far more successful in finding its niche around the world than its famous cousins, the black widow, Latrodectus mactans, found in the south-eastern states of North America, and the red-back, Latrodectus hasselti, found mostly in Australia, New Zealand and Japan.Methods:We performed an extensive web search of brown widow sightings and mapped the location of each sighting using ArcGIS. Specimens reputedly of the species L. geometricus were collected at three localities in Peninsular Malaysia. The spiders were identified and documented based on an examination of morphological characteristics and DNA barcoding.Results:The spiders found in Peninsular Malaysia were confirmed to be Latrodectus geometricus based on their morphological characteristics and DNA barcodes. We recorded 354 sightings of the brown widow in 58 countries, including Peninsular Malaysia.Conclusion:Reports from the Americas and the Far East suggest a global-wide invasion of the brown widow spider. Herein we report the arrival of the brown widow spider in Peninsular Malaysia and provide notes on the identification of the species and its recently expanded range.(AU)

ABSTRACT

Since the 1970s, There have been studies of the venom of Latrodectus sp. spiders, in particular the latrotoxin (LTX) of Latrodectus mactans. Many of the studies were aimed at understanding the action of the venom on the muscular system. Now accepted that LTX is able to generate a calcium-permeable membrane pore and modulate the release of synaptic vesicles that activate a receptor and induce cellular changes. Interestingly, when work began with venom obtained from the Latrodectus sp present in Chile, it generated clinical indications similar to the bite of this spider in another country, with some differences in intensity. The purpose of the first studies was to understand the systemic mechanisms of this venom, and other active compounds were studied for biological interest. It was found that these molecules are capable of causing systemic effects such as changes in muscle contraction; of generating vascular relaxation and synaptic and cellular modulation; and of altering potassium conductance channels. Based on this evidence, we suggested biotechnological applications to characterize low molecular-weight compounds obtained from the Chilean Latrodectus venom and exploring the effects on the electrophysiology in oocytes and neurons, and the contraceptive effect on spermatozoa.


Desde los años 70, se han realizado estudios con el veneno de arañas Latrodectus sp, en particular la latrotoxina (LTX) de Latrodectus mactans. Muchos de estos estudios estuvieron enfocados a entender la acción del veneno sobre el sistema muscular. Hoy en día es aceptado que la LTX es capaz de generar un poro de membrana permeable a calcio y modular la liberación de vesículas sinápticas que activan un receptor e inducen cambios celulares. Interesantemente, cuando comenzamos a trabajar con el veneno obtenido de Latrodectus sp. presente en Chile, ésto generó indicaciones clínicas similares a la picadura de esta araña en otros países, con algunas diferencias en su intensidad. El propósito de estos primeros estudios fue entender los mecanismos sistémicos de este veneno y además otros compuestos activos fueron estudiados para interés biológico. Se ha encontrado que estas moléculas son capaces de causar efectos sistémicos así como cambios en la contracción muscular; generar relajación vascular y modulación sináptica y celular; y de alterar los canales de conductancia de potasio. Basados en estas evidencias, nosotros sugerimos usar aplicaciones biotecnológicas para caracterizar los compuestos de bajo peso molecular obtenidos del veneno de Latrodectus Chilena y explorar los efectos sobre la electrofisiología en ovocitos y neuronas, y el efecto anticonceptivo sobre los espermatozoides.

ABSTRACT

Brown widow spider (Latrodectus geometricus) venom (BrWSV) produces few local lesions and intense systemic reactions such as cramps, harsh muscle pains, nausea, vomiting and hypertension. Approximately 16 protein bands under reducing conditions and ~ 14 bands under non-reducing conditions on a 12.5% sodium dodecyl sulfate-polyacrylamide gel electrophoresis were observed. Neurotoxic clinical manifestations were confirmed in vivo, while proteolytic activity was demonstrated on gelatine film. Severe ultrastructural damages in mice skeletal muscles were observed at 3, 6, 12 and 24 h postinjection with at total of 45 µg of venom protein. Infiltration of eosinophils and ruptures of the cellular membranes were observed in the muscles along with swelling of the nuclear cover and interruption of the collagen periodicity. Altered mitochondrias and autophage vacuoles, nuclear indentation and mitochondria without cristae, slight increment of intermyofibrillar and subsarcolemic spaces and myelinic figures formation were also observed. In the capillary, endothelial membrane unfolding into the lumen was noticed; along with myelinic figures compatible with a toxic myopathy. Swollen sarcotubular systems with lysis of membrane, intense mitochondria autophagia and areas without pinocytic vesicles were observed. Swollen mitochondria surrounded by necrotic areas, myofibrillar disorganization and big vacuolas of the sarcotubular system, degenerated mitochondrium with formation of myelinic figure was seen. Glycogenosomes with small particulate, muscle type glycogen was noticed. Autophagic vacuole (autophagolysosomes) and necrotic areas were also noticed. These damages may be due to interactive effects of the multifactorial action of venom components. However, Latrodectus geometricus venom molecules may also be utilized as neuro therapeutic tools, as they affect neuronal activities with high affinity and selectivity. El veneno de la araña viuda marrón (Latrodectus geometricus) produce pocas lesiones locales pero intensas reacciones sistémicas, tales como calambres, dolores musculares severos, nauseas, vómitos e hipertensión arterial. Se observaron ~ 16 bandas de proteina bajo condiciones reducidas y ~14 bandas bajo condiciones no reducidas en electroforesis en geles de poliacrilamida al 12.5%. Las manifestaciones neurotóxicas clínicas fueron confirmadas in vivo, mientras que la actividad proteolítica fue demostrada en una placa de gelatina. Los músculos de ratón se estudiaron durante las 3, 6, 12 y 24 horas después de ser inyectados con 45 µg de proteina de veneno. Los músculos fueron seriamente dañados por este veneno. Se demostró una infiltracción de células eosinofílicas y rupturas de membranas celulares en tejido muscular, al mismo tiempo un fuerte incremento de la membrana nuclear y una interrupción de la periodicidad del colágeno. Se observaron daños en la mitocondria y sin cristaes, vacuolas autofágicas e indentación nuclear. Se notó un aumento de la luz de los espacios intermiofibrilares y subsarcolemicos. En los capilares fue visible un desdoblamiento de la membrana endotelial hacia el lúmen vascular. Del mismo modo, fue visto un hinchamiento del sistema sarcotubular con lisis de las membranas; intensa autofagia de mitocondrias y áreas sin vesículas pinocíticas. Fue además observado, glucogenosomas con glucogeno particulado. Se observaron vacuolas autofágicas (autofagolisosomas) y áreas de necrosis. Estos daños podrían ser atribuídos a los efectos interactivos de una acción multifactorial de los componentes del veneno.

Widow spiders (Latrodectus spp.), also known as "black widows", have a worldwide distribution and can cause latrodectism. To the best of our knowledge, in Brazil, only one case of Latrodectus geometricus (Koch, 1841) envenomation in a human has been reported. The aim of the present report is to describe a spider bite caused by Latrodectus geometricus in a patient who lives in Paranapanema, São Paulo state, Brazil.(AU)

ABSTRACT

The first case of envenoming by Latrodectus geometricus in Venezuela is described. The accident occurred at the victim’s home, in Aragua de Barcelona, Anzoátegui State. The 31-year-old female victim was bitten twice on the left scapular region, in quick succession (within seconds). She developed a hyperactive state of the central, autonomic and peripheral nervous systems with minor local symptoms.


Descreve-se o primeiro caso de envenenamento por Latrodectus geometricus na Venezuela. O acidente ocorreu na residência, em Aragua de Barcelona, Estado Anzoátegui. A paciente de 31 anos de idade foi picada, consecutivamente duas vezes, em segundos, na região escapular esquerda. Ela desenvolveu um quadro de hiperatividade do sistema nervoso central, autonômico e periférico com escassa sintomatologia local.

Introducción: El envenenamiento por mordedura de araña viuda negra es poco frecuente en edades pediátricas sin embargo no está exento de complicaciones y si no se trata a tiempo puede causar la muerte. En México se desconoce el número exacto de estos accidentes. Objetivo: Identificar los signos y síntomas característicos, evolución y tratamiento de los niños atendidos por morderua de araña en el Hospital Infantil del Estado de Sonora, con el propósito de establecer el tratamiento oportuno. Material y Métodos: Se revisaron 43 expedientes de niños hospitalizados por mordedura de araña viuda negra entre 1978-2004, las variables consideradas fueron: edad, género, lugar de origen, sitio del accidente, área corporal afectada, tratamiento previo; grado de envenenamiento por signos y síntomas, tratamiento, evolución clínica. Resultados: Predominó el género masculino 69.7%; sin diferencias entre el número de menores de seis años, escolares y adolescentes. El 44 y 37% de los casos se presentaron en el verano y en horas de la tarde, respectivamente. El 74% de los pacientes tuvieron el contacto con el arácnido dentro del domicilio; las áreas anatómicas más afectadas fueron, miembros superiores y tronco; los signos y síntomas más notables en lactantes y preescolares fueron: irritabilidad, llanto constante, sialorrea, agitación, eritema en área de lesión convulsiones. En escolares y adolescentes fueron los signos y sínotmas dolor en sitio de lesión, dolor abdominal, dolor torácico, ansiedad, espasmos musculares y temblor fino. Los pacientes que recibieron tratamiento con faboterapia específica tuvieron mejor evolución y menor tiempo de hospitalización, cabe señalar que no se presentó mortalidad. Conclusiones: La adecuada identificación de los signos y síntomas ocasionados por la morderura de araña en lactantes y preescolares permite el diagnóstico oportuno y ofrece la mejor alternativa de tratamiento.


INTRODUCTION: Poisoning caused by black widow spider bite, is not commonly observed during childhood, it is not exempt from complications, and must be treated in time or the patient can die. In Mexico we do not have accurate data on the exact number of accidents due to this condition. OBJECTIVE: Identify the signs and characteristic symptoms of children treated at the Children's Hospital from the state of Sonora with the aim of starting early treatment. MATERIAL AND METHODS: We reviewed 43 clinical records of hospitalized patients admitted for black widow spider bite between 1978 - 2004. Demographic variables were: age, gender, place of origin, place where accident took place, treatment given prior to hospital admittance, degree of envenomation measured by signs and symptoms, treatment and clinical evolution. RESULTS: 69.7% of cases were male, we did not find differences in the number of patients between infant, pre-school, elementary school children and teenagers. During summer and afternoon hours we registered 44% and 37% cases respectively; 74% patients were poisoned by the spider at home; the anatomical areas most affected were: arms, hands and thorax. Signs and symptoms in infants were: irritability, constanty cry, sialorrea, agitation erythema on wounded areas and seizures. Among elementary school children and teenagers signs included pain in wounded area, abdominal and thoracic, muscle spasms, fine tremor and residual signs. Patients who received fabotherapic treatment had better evolution and less time of hospitalization. No deaths were reported. CONCLUSIONS: The different signs and symptoms between infants, preschool, elementary school children and teenagers allowed us to clearly identify evenommation and differentiate it form confounding diseases. In our study, specific fabotherapy constituted the best treatment choice.

Em 02/07/1995, foi atendido no Instituto de Medicina Tropical do Amazonas, paciente masculino, 11 anos, acidentado em Manaus, por picada na região retroauricular direita, clínicamente compatível com aquele causado por Latrodectus. Observavam-se abalos musculares, febre, calafrios e sudorese intensa. Instituída terapêutica com neostigmine precedido de atropina, gluconato de cálcio, cimetidina, diazepam e hidrocortisona. No terceiro dia apresentava-se melhorado, consciente, orientado e com diminuição importante do edema palpebral. A despeito de uma melhora progressiva diária, no quinto dia surgiu eritema máculo-pápulo-vesiculoso. Em 14/07/1995 teve alta, assintomático. O caso relatado é o primeiro descrito na região Amazônica, ocorrido na periferia de Manaus e pode ter sido uma consequência da expansão urbana das duas últimas décadas.


In July 2, 1995 arrived at the Instituto de Medicina Tropical do Amazonas as eleven-year-old male with complaining of spider bite on his right retroauricular region, presenting typical findings of latrodectism. The accident was reported as having occurred in the suburbs of Manaus. The patient was given neostigmine preceded by atropine, calcium gluconate, cimetidine, diazepan and hydrocortisone. Within three days the patient showed improvement, and was aware, orientated and with significant palpebral oedema reduction. Muscle spasms are still present, as well as fever, shivering, and intense sweating. In spite of a daily progressive improvement, at the fifth day appeared a spotted papular erythema. The patient was discharged without symptoms after the 13th day. This is the first such reported case which took place in Amazonian region, it might have been happened a consequence of the urban sprawl which has characterized the growth of the city of Manaus, in the last twenty years. It is not possible at this point to evaluate the epidemiological resound of the event, but in any case, it seems plausible to recommend that black widow antivenom become available.

ABSTRACT

Se reportan 4 casos de picaduras por arañas compatibles con el tipo viuda negra, en pacientes adultos de la provincia de Ciego de Ávila. Los síntomas y signos sobresalientes fueron: parestesias, vómitos, sudación, cólicos abdominales, adinamia e hipotensión. Los pacientes fueron tratados fundamentalmente con hidrocortisona endovenosa y administración de volumen. La evolución fue satisfactoria y rápida, con recuperación al cuarto día.


4 cases of spider bites, which are compatile with the black widow type were reported in adult patients from the province of Ciego de Avila. The most important symptoms and signs were: paresthesia, vomits, sweating, abdominal colics, acratia and hypotension. The evolution was satisfactory and fast. Patients recovered on the fourth day.
