ABSTRACT

Tropical diseases such as malaria, tuberculosis, trypanosomiasis, and leishmaniasis, account for a large number of deaths annually. Herbs are an excellent source of tropical medicines. Many advancements and discoveries have taken place in the field of drug discovery but still, a major population of tropical diseases relies on herbal traditional medicine. There are some challenges related to policy implementation, efficacy, resistance and toxicity of tropical medicines. There are many tropical diseases such as such as schistosomiasis, leishmaniasis, African sleeping sickness, filariasis and chagas disease which are neglected because very few pharmaceutical companies have shown their interest in developing therapeutics against these diseases of poor people. There are many benefits associated with herbal medicine such as the cost of production, patient tolerance, large scale availability, efficacy, safety, potency, recyclability, and environment friendly. A large number of natural extracts such as curcumin, artemisinin, morphine, reserpine, and hypericin, are in use for treatment of different tropical diseases for a long time. The current review is to discuss the overview of tropical medicinal herbs, its scope and limitations in the modern drug discovery process.

ABSTRACT

During the curing of complex disease, prospective therapeutic effect has been more and more difficult to achieve by using single-target inhibitors. Along with the development of systems biology and network pharmacology, the nosogenesis and the pathological process have been studied in depth, and people find that multi-target inhibitors can overcome many defects by using single-target inhibitors. Multi-target inhibitors include multicomponent and single component, but single component takes more advantages. The treatment of AIDS is a world problem, single-target inhibitors and multicomponent multi-target inhibitors (i.e. HAART) are used; However, viral height variability and severe drug resistance restrict the wide application of the two therapeutics. Therefore, the single component multi-target inhibitors with low toxicity and highperformance have been the most important choice to treat AIDS caused by HIV-1. The single component multi-target HIV-1 inhibitors mostly come from rational drug design and screening. Some HIV-1 inhibitors have been discovered from the natural products which have extensive sources and wide variety of kinds. That is an important way to develop lead compounds of HIV-1 inhibitors. This paper summarizes the advances in studies on HIV-1 multi-target drugs from natural products.

ABSTRACT

Natural products have been an important source of new drugs, which also played a dominant role in the discovery and research of new drugs for the treatment of hypertension. This review article reviews the recent progress in the research and development of natural lead compounds with antihypertensive activity, including alkaloids, diterpenes, coumarins, flavonoids, and peptides. We summarized their structures, sources, as well as the antihypertensive mechanisms. These information provides instructive reference for the following structural modifications and optimization.

ABSTRACT

Cancer is a serious threat to human life and health .Therefore, there is an emergent need to develop novel anti-cancer agents with new structural type , new mechanism of action and higher efficacy .Natural products had played a key role in the dis-covery of anticancer agents .The anti-cancer activity , mechanism of action , structure and activity relationship of several lead-com-pounds which were derived from natural products were summarized in this review .

ABSTRACT

As a summary of the research progress of chemical modification of lead compounds by saccharides and their derivatives, this article studies the linkage style of saccharides with lead compounds and bioactivity changes of the modified lead compounds. The result shows that the modified lead compounds can decrease the toxicity and side effect efficiently, and enhance the pharmacal effect. Therefore, it is a promising research project to modify lead compounds by saccharides.
