ABSTRACT

Abstract Excess of protein in the diet can lead to both productive and reproductive problems in ruminants. Measuring the urea in the milk allows determining the ruminant availability of energy and protein and, therefore, to do some adjustments to the diet whenever necessary. This study aims to quantify the urea concentration in the milk in a farm that takes care of high-density multi-layer silvopastoral systems. A system with dual purpose was evaluated in El Cerrito, Valle del Cauca (Colombia). The animals were divided into three feeding groups (prairies with guinea-leucaena-algarrobo, estrella-leucaena-algarrobo and estrella-algarrobo, respectively). Samples of milk were gathered during the morning and afternoon milking from the animal selected in each group, and then the urea concentration was determined (Urease/ Berthelot). The urea concentrations in the morning and afternoon milk were correlated positively (r = 0.86), the average concentration was 22.7+9.4 mg/dl. However, the values varied between the feeding groups (p < 0.001), it was higher in the first sample (p < 0.001). Likewise, a broad variation was observed between the samples inside the same foraging basis. Multi-layer silvopastoral systems show an appropriate balance between the fermentable organic matter and the degradable protein in the ruminants.

ABSTRACT

All) A.C Smith.

ABSTRACT This work has assessed the effects of different dilutions of treated domestic wastewater, such as reuse water, on the development of Mimosa caesalpiniifolia Benth and Amburana cearensis (Fr. All) A.C Smith. seedlings. The experiment made use of sandy soil as substrate and irrigation with treatment consisted of five dilutions of reuse water and public supply water (PSW), with five repetitions each, as follows: T1 (100% reuse water + 0% PSW), T2 (75% reuse water + 25% public supply water), T3 (50% reuse water + 50% public supply water), T4 (25% reuse water + 75% public supply water), and T5 (0% reuse water + 100% public supply water). Effects of the different dilutions of treated wastewater were evaluated according to the following functional characteristics: number of leaves, size of primary root, dry matter content of leaves, dry matter content of primary root, and dry matter content of lateral roots. In addition, plant height and collar diameter were also measured over the period of 30, 60, and 90 days. Results obtained for A. cearensis show that dry matter content of leaves and dry matter content of primary root had the highest values with T2, T3, and T4. As far as diameter over time is concerned, there was a significant difference for the T3 treatment in 90 days. Treatments did not produce any meaningful differences in the case of M. Caesalpiniifolia. It was possible to conclude, therefore, that treated domestic wastewater had neutral or positive effects on development and could be a potential water source to irrigate seedlings of these species, in both diluted and concentrated forms.

ABSTRACT

Abstract Bauhinia monandra Kurz (Fabaceae) is an ornamental leguminous tree from Africa. Its leaf has been proven to be hypoglycaemic, and are a source of lecithin BmoLL, providing this species with great medicinal potential. Curiously, there are no reports of the anatomical description of the leaf of this species. The present work aimed at describing the leaf anatomy of B. monandra, and make histochemical inferences. To this end, anatomical procedures in light and scanning electron microscopy were performed from different parts of the leaves of plants growing in the state of Bahia, Brazil. The leaf is amphistomatous. It possesses papillate epidermis, nonglandular and navicular trichomes, and the mesophyll is dorsiventral. The central vein is distinct from the others of the first degree. The first, second, and third-degree veins possess thick fibre bundles with associated monocrystals. The other veins are a little fibrous and excessively reduced. The petiole vascularisation consists of circularly arranged collateral bundles, plus two accessory bundles. The pulvinus have a wide parenchymatic cortex with abundant druses and vascularisation restricted to the central region, surrounded by a crystalline sheath and by a starch sheath. The motile cushion has the vascular bundles arranged in series. Associated with the phloem from the pulvinus vascular bundles and the motile apparatus, occur non-lignified septate fibres that confer flexibility to these organs. The absence of lateral projections and the arrangement of vascular bundles in the petiole was the most distinctive anatomical features and of the greatest taxonomic potential observed in the leaf of B. monandra.

ABSTRACT

The genus Pseudomonas is an important source of proteases; however, in the food industry the use of this bacterium is restricted. Cloning allows for the use of the proteolytic activity of Pseudomonas proteases through their production in innocuous microorganisms. Leguminous are protein-rich sources from which value-added compounds can be obtained through enzymatic hydrolysis. In this study, an alkaline and thermostable recombinant protease (PT4) from Pseudomonas aeruginosa M211 was cloned and characterized in order to obtain protein hydrolysates from leguminous. Therefore, protease gene was cloned into the pJET1.2 / blunt vector using E. coli DHalpha as a host. Analysis of protease partial nucleotide sequence showed 99% homology with Peptidases M4 Family from Pseudomonas aeruginosa. The molecular weight of the recombinant enzyme was 80 kDa, it was active and stable under alkaline and thermophilic conditions, presented an optimum pH and temperature of 8 and 60 °C, respectively, and was inhibited by EDTA. In addition, it hydrolysed Glycine max, Phaseolus lunatus, Lupinus mutabilis y Erythrina edulis proteins, obtaining peptide fractions less than 40 kDa. This recombinant protease could be used in the elaboration of functional hydrolysates using protein from different sources and agricultural waste.

ABSTRACT

Mung bean (Vigna radiata L.) is one of the important leguminous plants of India, with shorter growing season. In this work, the intention was to correlate the effects of chromium on physical and biochemical responses of V. radiata L. Mung bean seeds were germinated and grown under controlled treatment, with different concentrations of waste soil mixed with garden soil. Physical parameters such as shoot length, root length, and fresh weight were found highest in control soil and 25% of contaminated soil, whereas 50% and pure contaminated soil showed poor growth stage and poor quality physical parameters. Biochemical parameters such as total chlorophyll content, protein content, and starch content were found highest in control soil and 25% contaminated soil, but these parameters were found to be less in 50% and pure contaminated soil. It was observed that physical and chemical parameters were declining with increasing chromium contamination.

ABSTRACT

Aims: This study was carried out to evaluate the antagonistic effect of ten isolated characterized Rhizobium sp. and three referenced strains against aggressive phytopathogenic fungi Fusarium spp., Aspergillus spp., Penicillium spp., Alternaria spp., Humicola spp. and Cladosporium spp. isolated from infected and wilted plants in vitro and in vivo. Study Design:  First, we have ten strains of rhizobia isolated from leguminous and characterized, after that,  some strains of fungi were isolated from infected  and wilt plants,  such as Fusarium, Aspergillus, Penicillium,  Alternaria, Humicola and Cladosporium. Finally, The investigation of the potential of the isolated rhizobia and three referenced strains was evaluated in dual culture, in pots experiments and on seeds. Place and Duration of Study: The study was carried out at the Department of Microbiology, Faculty of Nature and Life Sciences, Laboratory of Applied Microbiology, between April and November 2016. Methodology: Rhizobia isolates were identified upon their phenotypic traits as: the morphology of the colony, of the physiological characters (growth temperature, salt tolerance, resistance to antibiotics, metabolism of carbon source, generation time…) and also the production of extracellular metabolites as siderophores and proteases. Fungal isolates were identified by their macroscopic and microscopic characters. The antagonistic effect of rhizobia against phytopathogenic fungi was examined in vitro by: confrontation in Petri dishes and inoculation of seeds; and in vivo by inoculation of plants. Results: The isolated rhizobia were resistant to penicillin and amoxicillin with generation time ranged from 1.9 to 6.4 h, they were able to grow at temperatures from 4°C to 37°C, tolerate salt concentration (0.5 –2%) and produce siderophores and proteases. The results demonstrated the effectiveness of the rhizobia isolates and the referenced strains against the fungi isolates in vitro and in vivo. In dual culture, all rhizobia isolates inhibited the mycelial growth of the pathogens. The best disease control was obtained with isolate F3 from faba bean which inhibited the fungal growth with more than 70%. The highest inhibition growth was obtained against Alternaria spp.2, Penicillium spp.2, Cladosporium spp. and Humicola spp. with an inhibition rate ranging from 90 to 96%. In pot experiments, Rhizobia isolates from chickpea, lentil and faba bean reduced significantly all disease incidence  with more than 75%; where the best fight was observed in lentil plants, while in faba bean no wilted plants were observed. Seeds inoculated with rhizobia and fungi in Petri dishes showed that isolates from faba bean and lentil were the most effective in reducing fungi disease incidence. Conclusion: Rhizobia have a high potentiality to inhibit the growth of tested pathogens and could be fielded within an integrated disease management package.

ABSTRACT

This particular work deals with the Pucciniales fungi (Basidiomycetes) of Morocco. Surveys in northern Atlantic Morocco have allowed collecting dozen species of plants (Oxalis corniculata, Vicia sativa and Lotus edulis) infected by rust. The different symptoms that have been observed are described in the laboratory and a microscopic study of spores, allowed to identify the responsible pathogens for these symptoms which are like: Puccinia oxalidis, Uromyces ervi and Uromyces genistae-tincortoriae. The obtained results help to know the diversification of host Pucciniales and the description of other new species for that cause the fungal diversity of Morocco is to be known.

ABSTRACT

This study assessed the toxicity of seed water extracts of 15 leguminous species upon Aedes aegypti larvae. A partial chemical and biochemical characterization of water extracts, as well as the assessment of their acute toxicity in mice, were performed. The extracts of Amburana cearensis, Anadenanthera macrocarpa, Dioclea megacarpa, Enterolobium contortisiliquum and Piptadenia moniliformis caused 100 percent of mortalit y after 1 to 3 h of exposure. They showed LC50 and LC90 values ranging from 0.43 ± 0.01 to 9.06 ± 0.12 mg/mL and from 0.71 ± 0.02 to 13.03 ± 0.15 mg/mL, respectively. Among the secondary metabolite constituents, the seed water extracts showed tannins, phenols, flavones, favonols, xanthones, saponins and alkaloids. The extracts also showed high soluble proteins content (0.98 to 7.71 mg/mL), lectin (32 to 256 HU/mL) and trypsin inhibitory activity (3.64 = 0.43 to 26.19 = 0.05 gIT/kg of flour) The electrophoretic profiles showed a great diversity of protein bands, many of which already described as insecticide proteins. The extracts showed low toxicity to mice (LD50 > 0.15 = 0.01 g/kg body weight), but despite these promising results, further studies are necessary to understand the toxicity of these extracts and their constituentsfrom primary and secondary metabolism upon Ae. aegypti.

ABSTRACT

In a greenhouse experiment, morpho-anatomical and micromorphometrical analyses of two soybean cultivars, MG/BR46 (Conquista) and BR16-tolerant and sensitive to drought, respectively-were used to study their water-deficit-tolerance strategies. Drought treatments were applied at reproductive stages from R2 to R7, where evaluations were conducted at 30 days and 45 days after stress started, respectively. The total length of Conquista plants (shoot + root) was greater than of BR16 plants. Pod dry weight was adversely affected due to the lack of moisture, decreasing productivity even of Conquista plants. Both the cultivars had normal development of root hairs; however, it was observed a decrease in the cortex:central cylinder ratio in BR16 stressed for 30 days, and they also showed similar leaflet thickness and stomata distribution. Differences in drought tolerance observed between the two cultivars seemed to be related to factors other than morphological traits since this species has a short lifecycle.

ABSTRACT

The quantitative analysis with different types of indicators is crucial in determining the sustainability of crop succession. This study aimed to appraise that in tilled plain areas. The research was carried out in Pelotas, RS, Brazil at Embrapa Clima Temperado research area. The treatments consisted of successions of winter and spring-summer crops species for five years, followed by two years of fallow and three years of rice cropping. The spring-summer crops were corn and soybean, besides rice; the winter crops were cereals, leguminous pastures, mixtures species, turnip and native grass species. Seeding was performed either by no-till (NT) or conventional (CT). The experiment was planned in a split-plot randomized blocks design with three replications, where the summer crops occupied the plots, while the winter's species were allocated to the subplots. The parameters measured were transformed into index to standardize the variables, each one in relation to the better measurement behavior, and arranged in four categories of analysis: (a) Agronomics indices (dry weight and grain yield); (b) Environmental indicators (soil fertility); (c) Energy Indicators (gain and energy efficiency); d) Economic Indicators (profitability and gross value of production). Through harmonic average rates calculated for each indicator, comparisons to the crop succession were then performed, and its sustainability inferred. Analyses of contrasts between the crop successions were accomplished too. The results provided the following conclusions: (a) All crop succession which had maize participation had a higher index sustainability; (b) the crops successions S1 [Cereals x soybean x rice(NT)] and S4 [Turnip x soybean x rice(NT)] had, respectively, the highest and the lowest sustainability among the ones with the soybean crop participation; (c) S5 [Native grass x soybean x rice(CT)] showed the lowest sustainability level, (d) S8 [Species mixtures x corn x rice(NT)] ...

ABSTRACT

Soluble (a) and potentially degradable (b) fractions and degradation rate of b fraction (c) of dry matter (DM) and crude protein (CP) of Parkia platycephala pods in particle sizes 2 and 5mm were estimated by in situ nylon bag method in sheep. The times of incubation were 3, 6, 12, 24, 48, 72 and 96 hours, and the effective degradability (ED) was determined considering passage rates of 2, 5 and 8 percent/h. The a fractions for DM and CP was 69.6 and 49.9 percent, respectively, revealing elevated DM solubility: The b fractions for DM and CP was 24.7 and 43.9 percent, denoting reduced DM in situ degradation. The stabilization of the DM and CP degradation occurred at 72h and 48h after incubation, respectively. In situ degradability of constituents of P. platycephala pods, in special CP, were not depressed for their tannin contents
