ABSTRACT

OBJECTIVE: To establish an HPLC method for determination of ferulic acid in Ligusticum Wallichii,which is the active ingredient of Xinmaishu capsules. METHODS: The content was determined by HPLC. The analysis was carried out on C18(4.6 mm×250 mm,5 μm) column. The mobile phase consisted of acetonitrile-0.3% acetic acid(13∶87). The flow rate was 1.0 mL·min-1, and the UV detection wavelength was set at 322 nm. RESULTS: The linearity range of the calibration curve for ferulic acid was from 1.0 to 10.0 μg·mL-1 (r=0.999 8). The average recovery rate was 99.57% with RSD of 0.75%(n=9). CONCLUSION: This method is sensitive, accurate,reproducible and simple. It can be used for the quality control of ferulic acid in Xinmaishu capsules.

ABSTRACT

OBJECTIVE: To study preliminarily the cadmium absorption and accumulation in Ligusticum Wallichii to provide a scientific basis for solving the problem of cadmium excess. METHODS: The aboveground and underground parts of Ligusticum Wallichii in different growth periods and the rhizosphere soil were collected from Aoping Town, Pengzhou City and Yongshou Town, Meishan City. The cadmium content was determined, and the enrichment coefficient and transfer coefficient were calculated. RESULTS: The average cadmium content in the underground part was 77.21% higher than that in the aboveground part. The cadmium enrichment coefficients and cadmium transfer coefficients of different parts were both greater than 0.5, and the enrichment coefficient of the underground part was 64.75% higher than the aboveground part. CONCLUSION: Ligusticum Wallichii has a certain capacity to absorb and transfer cadmium, and most cadmium is enriched in the medicinal roots.

ABSTRACT

Objective: Kangxian Pills, containing Angelicae Sinensis, Chuanxiong Rhizoma, radix paeoniae rubra, and 12 other kinds of Chinese materia medica, have the functions of softening and dispersing blood stasis. It has been used for liver injury and liver fibrosis. The current study was designed to evaluate the anti-hepatic injury activity and the mechanism of Kangxian Pills on a CCl4-induced animal model. Methods: To induce chronic liver injury, mice were treated with CCl4 twice a week for four weeks. Kangxian Pills (6 or 12 g/kg) and Compound Biejia Ruangan Tablet (0.901 g/kg) were ig given to mice once daily for four weeks after CCl4 was withdrawal. The anti-hepatic injury activities and mechanisms of Kangxian Pills were assessed by hepatic histology and by measuring the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), albumin (ALB), and total protein (TP) of serum as well as superoxide dismutase (SOD) and glycogen (Gn) of the liver. Results: Kangxian Pills significantly decreased the levels of liver index, ALT, and AST in mice liver injury models in treatment group. Moreover, Kangxian Pills and Compound Biejia Ruangan Tablet inhibited the CCl4-induced reduction of SOD and Gn levels in the liver. The histological study showed that Kangxian Pills could reduce cellular swelling and infiltration of inflammatory cells in liver injury. Conclusion: Kangxian Pills possess the potent abilities to alleviate chronic liver injury, suggesting that Kangxian Pills exert this effect by enhancing the anti-oxidant ability and metabolism of the liver.

ABSTRACT

Objective To compare the difference in bioavailability of ligusticum wallichii microemulsion and decoction by detecting the concentration of Ferulic acid in rabbit plasma. Methods RP-HPLC was used to detect the concentration of Ferulic acid in rabbits plasma at different times, and software 3p87 was used to analyze the pharmacokinetics parameter. Results The pharmacokinetics parameters of ligusticum wallichii microemulsion and decoction were as follows: AUC0-9=2 123.6 μg/(ml?min), Cmax=5.086 μg/ml, Tpeak=56.26 min for microemulsion;Decoction:AUC0-9=878.35 μg/(ml?min), Cmax=2.427μg/ml, Tpeak=128.86 min for decoction, F(%)=241.77. Conclusion Microemulsion of ligusticum wallichii has a higher biological availability and shorter peak time than decoction.

ABSTRACT

[Objective]To observe the indexes of quality control of nature and quantity of Yanluotong Infusion.[Method]Make authentication to the 2 main components of Ligusticum wallichii and rhizoma sparganii on their nature with Silica gel G thin-layer chromatography;make content measurement to puerarin with RP-HPLC,take Kromasil 100-5 C18(4.6mm?250mm,5?m)as chromatogram column,methanol-water(30:70)as flow phase;the measured wavelength is 250nm.[Result]The puerarin sampling in the range 0.06~0.65?g is in good linear relationship with the summit area,the relative coefficient r=0.9999,the intraday and days nicety RSD are respectively 0.85% and 0.48%(n=5);the average sample reclaim rates of high,middle and low concentrations(n=3)are respectively 101.2%,101.1% and 101.4%,RSD is 1.1%,0.94% and 0.41% separately.[Conclusion]Silica gel G thin-layer chromatography and RP-HPLC are correct and reliable for nature and quantity control,with good repeatability,and can be taken as quality control standard of the preparation.

ABSTRACT

The activity of erythrocyte membrane Na+-K+ATPase was determined from 43 patients with essential hypertension and 10 normotensive controls. Erythrocyte membrane preparation was obtained by washing after hypotonic hemolysis and repeating high speed centrifugal sedimentation. Ouabain was used as a specific inhibitor in Na+-K+ATPase assay. A significantly decreased activity of erythrocyte membrane Na+-K+ATPase was found in these patients of hypertension. Although their clinical manifestations were improved, there were no differences in Na + -K+ATPase activity between treated with Ligusticum Wallichii Hort and untreated groups. The mechanisms involved in these changes were discussed.
