ABSTRACT

ABSTRACT The CONVINCE study, recently published in the New England Journal of Medicine, reveals a groundbreaking 23% reduction in the relative risk of all-cause mortality among end-stage kidney patients undergoing high convective volume hemodiafiltration. This significant finding challenges the conventional use of high-flux hemodialysis and offers hope for improving outcomes in chronic kidney disease patients. While some controversies surround the study's findings, including concerns about generalizability and the causes of death, it is essential to acknowledge the study's design and its main outcomes. The CONVINCE study, part of the HORIZON 2020 project, enrolled 1360 patients and demonstrated the superiority of hemodiafiltration in reducing all-cause mortality overall, as well as in specific patient subgroups (elderly, short vintage, non-diabetic, and those without cardiac issues). Interestingly, it was shown that hemodiafiltration had a protective effect against infection, including COVID-19. Future research will address sustainability, dose scaling effects, identification of subgroups especially likely to benefit and cost-effectiveness. However, for now, the findings strongly support a broader adoption of hemodiafiltration in renal replacement therapy, marking a significant advancement in the field.


RESUMO O estudo CONVINCE, publicado recentemente no New England Journal of Medicine, revela uma redução inovadora de 23% no risco relativo de mortalidade por todas as causas entre pacientes renais em estágio terminal submetidos à hemodiafiltração de alto volume de convecção. Esse achado significativo desafia o uso convencional da hemodiálise de alto fluxo e oferece esperança de melhoria dos desfechos em pacientes com doença renal crônica. Embora algumas controvérsias cerquem os achados do estudo, incluindo preocupações sobre a generalização e as causas de óbito, é essencial reconhecer o desenho do estudo e seus principais desfechos. O estudo CONVINCE, parte do projeto HORIZON 2020, inscreveu 1.360 pacientes e demonstrou a superioridade da hemodiafiltração na redução da mortalidade por todas as causas em geral, bem como em subgrupos específicos de pacientes (idosos, HD de curta duração, não diabéticos e aqueles sem problemas cardíacos). Curiosamente, demonstrou-se que a hemodiafiltração teve um efeito protetor contra infecções, incluindo a COVID-19. Pesquisas futuras abordarão sustentabilidade, efeitos de escalonamento da dose, identificação de subgrupos especialmente propensos a se beneficiar e a relação custo-benefício. No entanto, por ora, os achados apoiam fortemente uma adoção mais ampla da hemodiafiltração na terapia renal substitutiva, marcando um avanço significativo na área.

ABSTRACT

Background: Moringa peregrina Forssk is a well-known plant in ethnomedicine due to its widespread uses in various diseases like cough, wound healing, rhinitis, fever, and detoxification. The plant seeds contain compounds that are cytotoxic to many cancer cells. During the therapeutic use of plants via the oral route, some compounds present in the plants may be cytotoxic to normal cell lines and red blood cells. Objective: This study was the first report of investigation of the cytotoxic profile on oral cancer, CAL 27, cell line, and hemolytic activities on human erythrocytes of Moringa peregrina seeds ethanolic extract (MPSE). Methods: MPSE was screened for its cytotoxic effect against oral cancer, CAL 27, cell line using 3-(4, 5-dimethylthiazol-2-yl)-2, 5,-diphenyltetrazolium bromide (MTT) assay. The toxicity of MPSE on human erythrocytes was determined by in vitro hemolytic assay. Results: MPSE showed significant anti-proliferative activity against oral cancer, CAL 27 cell line at lower concentrations with half maximal inhibitory concentration (IC50) value of 21.03 µg/mL. At 1,000 µg/ml of MPSE, the maximum hemolysis was found to be 14.3% which is within safer limit. Conclusions: This study revealed a potential anti-oral cancer of MPSE and provided a baseline for its potential use in oral cancer treatment with minimum hemolytic effect on human RBCs.


La Moringa peregrina Forssk es una planta muy conocida en etnomedicina debido a sus usos generalizados en diversas enfermedades como la tos, la cicatrización de heridas, la rinitis, la fiebre y la desintoxicación. Las semillas de la planta contienen compuestos citotóxicos para muchas células cancerosas. Durante el uso terapéutico de las plantas por vía oral, algunos compuestos presentes en ellas pueden ser citotóxicos para las líneas celulares normales y los glóbulos rojos. Objetivo: Este estudio fue el primer informe de investigación del perfil citotóxico sobre el cáncer oral, CAL 27, línea celular, y las actividades hemolíticas en eritrocitos humanos del extracto etanólico de semillas de Moringa peregrina (MPSE). Métodos: Se examinó el efecto citotóxico del MPSE contra la línea celular de cáncer oral CAL 27 mediante el ensayo con bromuro de 3-(4, 5-dimetiltiazol-2-il)-2, 5,-difeniltetrazolio (MTT). La toxicidad del MPSE sobre los eritrocitos humanos se determinó mediante un ensayo hemolítico in vitro. Resultados: MPSE mostró una actividad antiproliferativa significativa contra el cáncer oral, línea celular CAL 27 a concentraciones más bajas con un valor de concentración inhibitoria media máxima (IC50) de 21,03 µg/mL. A 1.000 µg/ml de MPSE, la hemólisis máxima fue del 14,3%, lo que está dentro del límite de seguridad. Conclusiones: Este estudio reveló un potencial anticancerígeno oral de MPSE y proporcionó una base para su uso potencial en el tratamiento del cáncer oral con un efecto hemolítico mínimo en los glóbulos rojos humanos.

ABSTRACT

ObjectiveTo investigate the expression of glial cell line-derived neurotrophic factor (GDNF) and androgen receptor (AR) in testicular peritubular cells (TPCs) of cryptorchidism mouse models and explore the theoretical significance of cryptorchidism-induced spermatogenesis dysfunction. MethodsA total of 30 five-week-old male ICR rats were divided randomly by using random number table method into 6 groups. Cryptorchidism was surgically induced in 3 randomly selected groups and the other 3 groups underwent sham surgery as the control groups. On days 4, 7 and 14 after surgery, we harvested the mice testes of the 3 groups and their corresponding control groups, then measured the testicular volumes, analyzed the testicular histopathology and detected the mRNA and protein expression levels of AR and GDNF in TPCs by immunofluorescence, real-time PCR and Western blot. ResultsIn normal control groups, on days 4, 7 and 14 after surgery, the testicular volumes were (125.58±19.22) mm3,(123.45±20.12) mm3, (140.09±13.62) mm3 , respectively. Clear layers of spermatogenic cells were well arranged and abundant sperm cells were found. Peritubular cells were morphologically homogeneous, with slim-spindle appearance and normal cell thickness. The mRNA expression levels of AR were 1.00±0.05, 1.06±0.07 and 1.19±0.13; GDNF mRNA 1.00±0.04, 1.09±0.05, and 1.10±0.07. The protein expression levels of AR were 1.01±0.01, 0.79±0.02 and 1.01±0.04; GDNF protein (18.68±0.43) pg/mL, (14.39±0.36) pg/mL and (16.88±0.37) pg/mL. In cryptorchidism groups, on days 4, 7 and 14 after surgery, the testicular volumes were (115.64±3.91) mm3, (69.51±14.97) mm3 and (44.86±5.56) mm3, respectively. Spermatogenic cells were disorganized, seminiferous tubules were disrupted, peritubular cells shrank, bent and fractured. The mRNA expression levels of AR were 0.76±0.06, 0.53±0.04, and 0.29±0.02; GDNF mRNA 0.72±0.05, 0.42±0.02 and 0.30±0.03. The protein expression levels of AR were 0.54±0.02, 0.98±0.04 and 0.31±0.01; GDNF protein (8.50±0.34) pg/mL, (17.44±0.32) pg/mL and (6.83±0.34) pg/mL. Statistically significant differences (P < 0.05) were found in 7-day and 14-day testicular volumes between control and cryptorchidism groups but not in the 4-day testicular volume (P > 0.05). Testicular volumes, AR and GDNF mRNA and protein expression in control groups had no statistically significant difference (P > 0.05), while those in cryptorchidism groups showed a trend of gradual decline in the amount and the differences between groups were statistically significant (P < 0.05). ConclusionsIn surgery-induced cryptorchidism mice, after the induction, the expression of AR and GDNF in TPCs showed a gradual decrease over time. AR and GDNF play a major role in mediating the TPCs damage in cryptorchidism. This study provides a theoretical basis for mechanism researches of cryptorchidism-induced spermatogenesis dysfunction.

@#A cemental tear is defined as an incomplete or complete detachment of the cementum along the dentino-cemental junction (CDJ) or the incremental line within the body of the cementum, which can also involve part of the root dentine adjacent to the cementum. The pathogenesis of cemental tears is not fully elucidated. From the literature review, possible predisposing factors were identified, including tooth type, sex, age, periodontitis, previous periodontal treatment or root canal treatment, history of dental trauma, and occlusal trauma or excessive occlusal force. The morphology of cemental tears can be either piece-shaped or U-shaped, which usually contributes to periodontal and periapical breakdown. Clinically, cemental tears have a unitary periodontal pocket and present with symptoms mimicking localized periodontitis, apical periodontitis, and vertical root fractures. Imaging examination is of great significance for the clinical diagnosis of cemental tears, which often manifest as thin ‘prickle-like’ radiopaque masses located longitudinally adjacent to the affected root surface. Exploratory surgery is needed in some cases. Although intraoperative cemental fragments and cemental lines on the root surface can assist in the diagnostic process, histopathology examination is the gold standard for the diagnosis of cemental tears. The treatment methods vary depending on the timing of the correct diagnosis and the clinical or radiological manifestations. With the development of regenerative biomaterials and the development of intentional replantation, an increasing number of affected teeth can survive for a long time. The aim of this review is to systematically describe the biological basis and predisposing factors, clinical features, radiographic and histological characteristics, diagnosis and clinical management of cemental tears, and treatment outcomes to help make a clear diagnosis and develop a personalized treatment plan.

italic>Mycobacterium tuberculosis, responsible for tuberculosis (TB), remains a major health problem worldwide and is one of the infectious diseases causing increased morbidity and mortality worldwide. Biotin, namely vitamin H, is an important cofactor necessary for fatty acid biosynthesis, gluconeogenesis and amino acid metabolism in organisms including Mycobacterium tuberculosis. Due to its inability to ingestion biotin from outside, Mycobacterium tuberculosis can only obtain biotin through biotin biosynthesis. Different from the classical BioC-BioH, BioI-BioW and non-classical BioZ pathways, Mycobacterium tuberculosis synthesized biotin by "BioC-BioH(2)" pathway in the early stage. This review focuses on the unique biotin synthesis pathway of Mycobacterium tuberculosis and its key genes, especially the response of this pathway and biotin-dependent carboxylase to tuberculosis first-and second-line drugs, as well as inhibitors and natural products targeting biotin synthesis.

ABSTRACT

Traditional Chinese medicines (TCMs) possess a rich historical background, unique theoretical framework, remarkable therapeutic efficacy, and abundant resources. However, the modernization and internationalization of TCMs have faced significant obstacles due to their diverse ingredients and unknown mechanisms. To gain deeper insights into the phytochemicals and ensure the quality control of TCMs, there is an urgent need to enhance analytical techniques. Currently, two-dimensional (2D) chromatography, which incorporates two independent separation mechanisms, demonstrates superior separation capabilities compared to the traditional one-dimensional (1D) separation system when analyzing TCMs samples. Over the past decade, new techniques have been continuously developed to gain actionable insights from complex samples. This review presents the recent advancements in the application of multidimensional chromatography for the quality evaluation of TCMs, encompassing 2D-gas chromatography (GC), 2D-liquid chromatography (LC), as well as emerging three-dimensional (3D)-GC, 3D-LC, and their associated data-processing approaches. These studies highlight the promising potential of multidimensional chromatographic separation for future phytochemical analysis. Nevertheless, the increased separation capability has resulted in higher-order data sets and greater demands for data-processing tools. Considering that multidimensional chromatography is still a relatively nascent research field, further hardware enhancements and the implementation of chemometric methods are necessary to foster its robust development.

ABSTRACT

OBJECTIVE@#To identify phytochemical constituents present in the extract of flowers of Xanthoceras sorbifolia and evaluate their anti-oxidant and anti-hyperglycemic capacities.@*METHODS@#The AlCl3 colorimetric method and Prussian Blue assay were used to determine the contents of total flavonoids and total phenolic acids in extraction layers, and the bioactive layers was screened through anti - oxidative activity in vitro. The Waters ACQUITY UPLC system and a Waters ACQUITY UPLC BEH C18 column (2.0 mm × 150 mm, 5 μm) were used to identify the ingredients. And anti-oxidative ingredients were screened by off-line UPLC-QTOF-MS/MS-free radical scavenging. The ameliorative role of it was further evaluated in a high-fat, streptozotocin-induced type 2 diabetic rat model and the study was carried out on NADPH oxidase (PDB ID: 2CDU) by molecular docking.@*RESULTS@#Combined with the results of activity screening in vitro, the anti - oxidative part was identified as the ethyl acetate layer. A total of 24 chemical constituents were identified by liquid chromatography-mass spectrometry in the ethyl acetate layer and 13 main anti-oxidative active constituents were preliminarily screened out through off-line UPLC-QTOF-MS/MS-free radical scavenging. In vivo experiments showed that flowers of X. sorbifolia could significantly reduce the blood glucose level of diabetic mice and alleviate liver cell damage. Based on the results of docking analysis related to the identified phytocompounds and oxidase which involved in type 2 diabetes, quercetin 3-O-rutinoside, kaempferol-3-O-rhamnoside, isorhamnetin-3-O-glucoside, and isoquercitrin showed a better inhibitory profile.@*CONCLUSION@#The ethyl acetate layer was rich in flavonoids and phenolic acids and had significant anti-oxidant activity, which could prevent hyperglycemia. This observed activity profile suggested X. sorbifolia flowers as a promising new source of tea to develop alternative natural anti-diabetic products with a high safety margin.

OBJECTIVE@#To explore the mechanism of electroacupuncture (EA) in promoting recovery of the facial function with the involvement of autophagy, glial cell line-derived neurotrophic factor (GDNF), and phosphatidylinositol-3-kinase (PI3K)/mammalian target of rapamycin (mTOR) signaling pathway.@*METHODS@#Seventy-two male Sprague-Dawley rats were randomly allocated into the control, sham-operated, facial nerve injury (FNI), EA, EA+3-methyladenine (3-MA), and EA+GDNF antagonist groups using a random number table, with 12 rats in each group. An FNI rat model was established with facial nerve crushing method. EA intervention was conducted at Dicang (ST 4), Jiache (ST 6), Yifeng (SJ 17), and Hegu (LI 4) acupoints for 2 weeks. The Simone's 10-Point Scale was utilized to monitor the recovery of facial function. The histopathological evaluation of facial nerves was performed using hematoxylin-eosin (HE) staining. The levels of Beclin-1, light chain 3 (LC3), and P62 were detected by immunohistochemistry (IHC), immunofluorescence, and reverse transcription-polymerase chain reaction, respectively. Additionally, IHC was also used to detect the levels of GDNF, Rai, PI3K, and mTOR.@*RESULTS@#The facial functional scores were significantly increased in the EA group than the FNI group (P<0.05 or P<0.01). HE staining showed nerve axons and myelin sheaths, which were destroyed immediately after the injury, were recovered with EA treatment. The expressions of Beclin-1 and LC3 were significantly elevated and the expression of P62 was markedly reduced in FNI rats (P<0.01); however, EA treatment reversed these abnormal changes (P<0.01). Meanwhile, EA stimulation significantly increased the levels of GDNF, Rai, PI3K, and mTOR (P<0.01). After exogenous administration with autophagy inhibitor 3-MA or GDNF antagonist, the repair effect of EA on facial function was attenuated (P<0.05 or P<0.01).@*CONCLUSIONS@#EA could promote the recovery of facial function and repair the facial nerve damages in a rat model of FNI. EA may exert this neuroreparative effect through mediating the release of GDNF, activating the PI3K/mTOR signaling pathway, and further regulating the autophagy of facial nerves.

ABSTRACT

The development and application of processing technology is closely related to the quality of Chinese medicine. Currently, Chinese medicine processing is still in the mechanization stage with limited processing equipment, low levels of automation and intelligence. As a result, the imprecise control of parameters during processing leads to unstable quality of Chinese herbal pieces. However, with the arrival of the big data era and the continuous development of "Internet+", Chinese medicine processing technology and equipment have been continuously improved and updated, and gradually shifted to the development direction of automation and intelligence. The linkage production technology of Chinese herbal pieces optimizes the separate processing equipment coupling into the production line for continuous manufacture of Chinese herbal pieces, intending to improve production efficiency. The large-scale industrialized production of Chinese herbal pieces tends towards digital technology of processing experience and online inspection technology based on machine vision, electronic nose, and electronic tongue. These technologies are crucial prerequisites for standardizing the parameters of Chinese medicine processing. And further by docking the processing process and equipment with the internet, realizing the intelligent control of the production process is an important process for the transformation and upgrading of Chinese herbal piece industry in the future. In this paper, we summarized the development characteristics of different stages of Chinese medicine processing technology, combed application and development of processing theory, the evolution of processing equipment, and problems in the current industrial development stage of Chinese medicine processing, in order to provide ideas and methods for achieving digital and intelligent innovation of processing technology as well as high-efficient and high-quality production of Chinese herbal pieces.

Resumo Introdução: Atualmente o Brasil mostra um expressivo aumento da sobrevida e consequente elevação do número de idosos na sua população. Do ponto de vista de propostas para a melhoria da qualidade de vida deles, a sexualidade deve ser compreendida a partir do princípio holístico, não somente do fator biológico. Nesse cenário, novas reflexões de profissionais da saúde tornam-se indispensáveis para o planejamento de ações específicas, objetivando a atenção integral à saúde do idoso. Relato de experiência: O presente relato de experiência traz o início de um diálogo com acadêmicos de Medicina de uma universidade pública sobre a complexidade da sexualidade e infecções sexualmente transmissíveis (IST) em idosos, buscando identificar o conhecimento desses discentes sobre o assunto e idealizar propostas de ação ao público-alvo. Para isso, estruturou-se uma estratégia de ensino-aprendizagem cuja abordagem consistiu na adaptação e aplicação da metodologia do Arco de Maguerez para o debate sobre IST no contexto da saúde pública brasileira, o qual foi realizado com base nas diretrizes do ensino remoto emergencial. Nossos resultados vão de encontro à literatura que visa compreender as representações sociais acerca da sexualidade dos idosos. Neles, verificou-se que as representações sociais acerca da sexualidade na terceira idade assemelham-se à descrição científica, apresentado similitude entre o senso comum e o conhecimento erudito. Discussão: Diante da discrepância descrita na literatura quanto aos conceitos errôneos por parte dos idosos e dos familiares e pelos profissionais de saúde acerca da sexualidade na terceira idade, são imprescindíveis a realização de campanhas e o desenvolvimento de medidas e estratégias preventivas voltadas para idosos, a fim reduzir a incidência de IST nessa comunidade. Para isso, são fundamentais estratégias de educação em saúde no ensino acadêmico e de educação profissional continuada para sedimentar os conhecimentos necessários na idealização de propostas eficazes. Conclusão: A partir da formação médica adequada e por meio da discussão dos aspectos complexos sobre o assunto, serão possíveis a construção, a implementação e a avaliação de políticas públicas de saúde para o enfrentamento das IST, de modo a diminuir as barreiras relacionadas à prevenção de doenças e à promoção de saúde sexual na população idosa.


Abstract Introduction: Brazil is currently reporting a significant increase in survival and consequent rise in its elderly population. From the point of view of proposals to improve their quality of life, sexuality should be understood from a holistic, and not just a biological, perspective. In this scenario, new reflections by health professionals become indispensable for the planning of specific actions aimed at providing comprehensive health care for the elderly. Experience report: This experience report describes the beginning of a dialogue with medical students from a public university about the complexity of sexuality and sexually transmitted infections among the elderly, seeking to identify their knowledge of the subject and devise proposals for action with the target audience. To this end a teaching-learning strategy was structured based on an approach that adapted and applied the Arc of Maguerez methodology to the debate on sexually transmitted infections (STIs) in the context of Brazilian public health, which was conducted out within the guidelines of emergency remote learning. Our results are in line with the literature that seeks to understand the social representations about sexuality among the elderly. It was found that the social representations about sexuality in the elderly are similar to the scientific description, with similarities between common sense and scholarly knowledge. Discussion: Given the discrepancy described in the literature regarding the misconceptions on the part of the elderly, family members and health professionals about sexuality in old age, it is essential that campaigns be conducted, and preventive measures and strategies developed for the elderly to reduce the incidence of STIs in this community. Hence, health education strategies are needed in academic teaching and continuing professional education to consolidate the knowledge required to devise effective proposals. Conclusions: Based on fit-for-purpose medical education, the discussion of complex aspects about this subject will support the construction, implementation, and evaluation of public health policies to tackle STIs, reducing barriers related to the prevention and promotion of sexual health in the elderly population.

ABSTRACT

O objetivo deste trabalho foi discutir acerca da utilização de um dispositivo de pesquisa: o caderno digital itinerante, a partir do qual foi possível promover a construção conjunta de uma pesquisa, partindo do método cartográfico. A pesquisa de campo que deu origem a esta reflexão metodológica teve por objetivo principal cartografar experiências sapatão na cidade, por meio da construção conjunta de um caderno digital itinerante no qual as participantes, juntamente com a pesquisadora, escreviam sobre suas experiências como mulheres lésbicas e/ou pessoas que se reconheciam enquanto sapatão. Tendo como um dos instrumentos a análise do diário de campo, produzido durante a pesquisa, foi possível discutir em profundidade o enfoque metodológico adotado e questionar a dicotomia pesquisadora-objeto, provocando algumas reflexões acerca da temática da participação e do desenvolvimento de pesquisas on-line, sobretudo no período pandêmico. Por fim, apostar na cartografia enquanto método de pesquisa com e não sobre permitiu a produção de uma pesquisadora sapatão que pudesse se ocupar da escuta, lendo sobre diferentes processos de subjetivação e, com isso, produzindo outros sentidos para seu objeto de pesquisa. The aim of this study was to discuss the use of a research tool: the itinerant digital notebook, which facilitated the collaborative construction of research based on the cartographic method. The field research that gave rise to this methodological reflection aimed to map dyke experiences in the city through the collaborative construction of an itinerant digital notebook. In this note-book, participants, alongside the researcher, wrote about their experiences as lesbian women and/or individuals who identified as dykes. Using the analysis of the field diary produced during the research as one of the instruments, it was possible to deeply discuss the adopted methodological approach and question the researcher-object dichotomy, prompting reflections on the theme of participation and the development of online research, especially during the pandemic period. Ultimately, embracing cartography as a research method with, not on, participants allowed the production of a dyke researcher who could engage in listening, reading about different processes of subjectivation, and thereby generating alternative meanings for her object of study. El objetivo de este trabajo fue discutir el uso de un dispositivo de investigación: el cuaderno digital itinerante, a partir del cual fue posible promover la cons-trucción conjunta de una investigación, a partir del método cartográfico. La investigación de campo que dio origen a esta reflexión metodológica tuvo como objetivo principal mapear las experiencias lesbianas en la ciudad, a través de la construcción conjunta de un cuaderno digital itinerante en el que las parti-cipantes, junto con la investigadora, escribieron sobre sus experiencias como lesbianas y/o personas que se reconocían como bolleras/camioneras. Teniendo como uno de los instrumentos el análisis del diario de campo producido durante la investigación, fue posible discutir en profundidad el abordaje metodológico adoptado y cuestionar la dicotomía investigadora-objeto, provocando algunas reflexiones sobre el tema de la participación y el desarrollo de investigación, especialmente en el período de pandemia. Finalmente, apostar por la carto-grafía como método de investigación con participantes y no sobre las partici-pantes permitió producir una investigadora lesbiana que podía ocuparse de escuchar, leer sobre diferentes procesos de subjetivación y, con eso, producir otros significados para su objeto de investigación. ABSTRACT

La rueda de reconocimiento es una diligencia de investigación policial orientada a identificar al autor de un crimen. Se trata de una práctica relativamente frecuente que puede tener como resultado consolidar sospechas o también descartar líneas de investigación. La rueda de reconocimiento suele estar precedida de la exposición de un álbum de fotos y también es frecuente solicitar al mismo testigo que haga una descripción o un retrato robot del atacante. Existen fundadas sospechas de que la concatenación de estas diligencias policiales de identificación desvirtúa la eficacia de la declaración del testigo presencial para elegir al verdadero culpable, en caso de que esté presente en la rueda. También existen algunas controversias con respecto a la fiabilidad de la rueda de reconocimiento en función de la edad del testigo, del número de personas que la conforman, del procedimiento para llevarla a cabo, entre otras. Con el objetivo de acotar la evidencia empírica en torno a estas cuestiones, se presenta una revisión de estudios meta-analíticos y diversos estudios experimentales, desde los cuales se han podido identificar los procedimientos más adecuados para optimizar la eficacia en la precisión de la declaración de los testigos presenciales de un delito en el contexto de una rueda de reconocimiento.


The line-up is a police investigative procedure aimed at identifying the perpetrator of a crime. It is a relatively common practice that can result in consolidating suspicions or ruling out lines of investigation. The identification parade is usually preceded by the exhibition of a photo album and it is also common to ask the same witness to make a description or sketch of the perpetrator. There are strong suspicions that the concatenation of these police identification steps undermines the effectiveness of the eyewitness statement in identifying the real culprit, if present at the line-up. There is also some controversy regarding the reliability of the identification parade, depending on the age of the witness, the number of people involved, the procedure for carrying it out, among others. In order to narrow down the empirical evidence on these issues, a review of meta-analytical studies and various experimental studies is presented, based on which it has been possible to identify the most appropriate procedures to optimise the effectiveness in the accuracy of eyewitness testimony in the context of a line-up.


A roda de reconhecimento é um procedimento de investigação policial que visa identificar o autor de um crime. É uma prática relativamente comum que pode resultar na consolidação de suspeitas ou na exclusão de linhas de investigação. A roda de identificação é geralmente precedida pela exibição de um álbum de fotos e é comum pedir à mesma testemunha que faça uma descrição ou um esboço do autor do crime. Há fortes suspeitas de que a concatenação dessas etapas de identificação policial prejudica a eficácia do depoimento da testemunha ocular na identificação do verdadeiro culpado, se presente nessa fase. Há também alguma controvérsia quanto à confiabilidade da roda de reconhecimento, dependendo da idade da testemunha, do número de pessoas envolvidas, do procedimento para realizá-la, entre outros. A fim de restringir as evidências empíricas sobre essas questões, é apresentada uma revisão de estudos metanalíticos e experimentais, a partir dos quais foi possível identificar os procedimentos mais adequados para otimizar a eficácia na precisão do depoimento de testemunhas oculares no contexto de uma roda de reconhecimento.

Introducción. Los tumores neoplásicos se caracterizan por su invasividad y metástasis. Las células neoplásicas tienen heterogeneidad genética, por lo cual pueden desarrollar resistencia a los quimioterápicos. Por esta razón, las plantas continúan siendo una fuente importante de nuevos productos anticancerígenos. Objetivo. Evaluar la actividad citotóxica y antiproliferativa de un extracto rico en fucoidan de Lessonia trabeculata nativa (EFLt) sobre la línea celular de adenocarcinoma mamario murino, triple negativo 4T1. Métodos. La citotoxicidad y la IC50 se determinaron en monocapas de 4T1 empleando el reactivo MTT. Para demostrar la actividad antiproliferativa se aplicaron los métodos de cierre de herida y anticlonogénico utilizando las IC50 del EFLt y Dox (doxorubicina). El cierre de herida fue evaluado mediante barrido de tiempos discretos; el efecto anticlonogénico fue evaluado 7 días postratamiento mediante el conteo de colonias y se determinó la fracción de sobrevivencia. Adicionalmente, se evaluaron la citotoxicidad y actividad antiproliferativa combinando las IC50 de EFLt y Dox. El porcentaje de migración y conteo de colonias se realizó con el programa ImageJ. Resultados. La IC50 del EFLt (950 μg/mL) produjo 56% de citotoxicidad, 80,3% de inhibición de la migración celular, 68% de inhibición en la formación de colonias. La IC50 de Dox fue 0,5 μg/mL. Conclusiones. El EFLt ejerce citotoxicidad dependiente de la concentración y tiene efecto antiproliferativo sobre 4T1. Se requiere continuar los ensayos en modelos de mayor complejidad que permitan esclarecer el potencial antitumoral del EFLt.


Introduction. Neoplastic tumors are characterized by invasiveness and metastasis. Neoplastic cells are genetically heterogeneous and can develop resistance to chemotherapeutic agents. For this reason, plants continue to be an important source of new anticancer products. Objective. To determine the cytotoxic and antiproliferative activity of a fucoidan-rich extract of native Lessonia trabeculata (EFLt) on the tripe negative murine mammary adenocarcinoma cell line 4T1. Methods. Cytotoxicity and IC50 were determined in 4T1 monolayers using the MTT reagent. To demonstrate antiproliferative activity, "wound" closure and anticlonogenic methods were applied using the IC50 of EFLt and Doc (doxorubicin). "Wound" closure was evaluated by discrete times sweep to determine percentage inhibition of cell migration; the anticlonogenic effect was evaluated by colony counting 7 days after treatment and the survival fraction was determined. In addition, cytotoxicity and antiproliferative activity were evaluated by combining the IC50 of EFLt and Dox. Percent migration and colony counts were performed using ImageJ software. Results. The IC50 (950 μg/mL) of EFLt was 56% cytotoxicity, 80,3% inhibition of cell migration, 68% inhibition of colony formation.The ICof Dox was 0,5 μg/mL. Conclusions. EFLt exerts concentration-dependent cytotoxicity and antiproliferative effect on 4T1. Further studies in more complex models are needed to elucidate the anti-tumor potential of EFLt.

El presente estudio tuvo como objetivo el diseño y vali- dación de una escala para evaluar comportamiento con- traproducente en línea que incluye el análisis conjunto de conductas destructivas (antisociales) y constructivas (prosociales). Además, se propuso determinar qué ras- gos de personalidad (normales, patológicos, negativos o positivos) predicen dicho comportamiento. Participaron 351 sujetos de población general argentina (156 hombres, 191 mujeres, 2 se identificaron como no binarios), con una edad promedio de 39.35 años (de = 13.79). Para el cumplimiento de todos los objetivos, aparte de la prueba construida, se emplearon los siguientes instrumentos: Big Five Inventory, Personality Inven- tory for DSM-5 Brief-Form, Positive Traits Inventory Short-Form, Dark Triad Scale, y una encuesta diseñada ad hoc para evaluar el uso de internet y redes sociales. El análisis factorial exploratorio reveló una estructura de dos factores orientados a la evaluación de los dos tipos de conductas, que explicaban el 57.5 % de la varianza total. Como resultado, se dio lugar a una escala de 18 ítems con muy buenos valores de fiabilidad ­coeficiente omega­ (conductas destructivas ω = 0.88; conductas constructivas ω = 0.79). Por último, se realizaron análisis de regresión jerárquica que determinaron que los rasgos de la tríada oscura son los que mejor predicen el desarrollo de conductas destructivas, en tanto que los rasgos positivos predicen las conductas constructivas. The present study aimed to design and validate a scale to assess online counterproductive behavior that includes the joint analysis of destructive (antisocial) and constructive (prosocial) behaviors. In addition, it was studied which personality traits (normal, pathological, negative or positive) predict this counterproductive behavior. 351 subjects from the general Argentine population participated (156 men, 191 women, 2 identified themselves as non-binary), with an average age of 39.35 years (SD = 13.79). In order to achieve the objectives, in addition to the constructed test, the follow- ing instruments were used: Personality Inventory for dSM-5 Brief-Form, Positive Traits Inventory Short-Form, Dark Triad Scale, and a survey designed ad hoc to assess internet and social media sites use. The exploratory factor analysis revealed a two-factor structure ­oriented to the evaluation of the two types of behaviors­ that explained 57.5 % of the total variance. As a result, a scale of 18 items with very good reliability values ­omega coefficient­ (destructive behaviors ω = 0.88; constructive behaviors ω = 0.79) was developed. Finally, hierarchical regression analyzes were performed. The results showed that the Dark Triad traits best predict destructive behaviors, while positive traits predict constructive behaviors.


O objetivo deste estudo foi a concepção e validação de uma escala para avaliar o comportamento on-line contraproducente que inclui a análise conjunta de com- portamentos destrutivos (antissociais) e construtivos (pró-sociais). Além disso, foi proposto determinar quais traços de personalidade (normal, patológico, negativo ou positivo) predizem tal comportamento. Participaram 351 sujeitos da população geral argentina (156 homens, 191 mulheres, 2 identificados como não binários), com idade média de 39.35 anos (dP = 13.79). Para atender a todos os objetivos, além do teste cons- truído, foram utilizados os seguintes instrumentos: Big Five Inventory, Personality Inventory for DSM- 5 Brief-Form, Positive Traits Inventory Short-Form, Dark Triad Scale e um questionário elaborado ad hoc para avaliar o uso da internet e das redes sociais. A análise fatorial exploratória revelou uma estrutura de dois fatores orientada para a avaliação dos dois tipos de comportamentos, que explicaram 57.5 % da variância total. Como resultado, foi criada uma escala de 18 itens com valores de confiabilidade muito bons ­coeficiente ômega­ (comportamentos destrutivos ω = 0.88; comportamentos construtivos ω = 0.79). ABSTRACT

The purpose of this study was to test the effectiveness, and the sustained effect (follow-up) of a fourweek mindfulness intervention in reducing anxiety and stress in a group of Brazilian university students, the intervention was adapted to be made available through the application Neurosaúde, created as part of this work to serve as a platform for intervention. The research as a whole took place in a virtual environment, with forty-eight participants recruited who were randomly allocated to the intervention group or to the waiting list control. Measures were used to access the levels of mindfulness, anxiety and stress before, after the intervention and at the 4-week follow-up. Applying the intention-to-treat analysis, we found significant differences between the groups for the measurement of anxiety in the follow-up with a large feat size (F (2.92) = 10.275, p = 0.000, η2 = 0.183). Our results suggest that the 4-week mindfulness intervention through a mobile application was able to act as a protective factor against the anxiety developed by university students during the occurrence of the pandemic caused by Covid-19, whereas while the treatment group maintained levels reduced anxiety the control group demonstrated a continuous increase in anxiety in the post-test and follow-up that coincided with the first wave of Covid-19 in the region where the participants lived.


El propósito de este estudio fue probar la eficacia y el efecto sostenido (seguimiento) de una intervención de atención plena de cuatro semanas para reducir la ansiedad y el estrés en un grupo de estudiantes universitarios brasileños, la intervención se adaptó para estar disponible a través del app. Neurosaúde, creada como parte de este trabajo para servir como plataforma de intervención. La investigación en su conjunto se llevó a cabo en un entorno virtual, siendo reclutados cuarenta y ocho participantes que fueron asignados aleatoriamente al grupo de intervención o al control en lista de espera. Se utilizaron medidas para evaluar los niveles de atención plena, ansiedad y estrés antes, después de la intervención y en el seguimiento a las 4 semanas. Al aplicar el análisis por intención de tratar, encontramos diferencias significativas entre los grupos para la medida de ansiedad en el seguimiento con un tamaño de efecto grande (F (2.92) = 10.275, p = 0.000, η2 = 0.183). Nuestros resultados sugieren que la intervención de mindfulness de 4 semanas a través de una aplicación móvil fue capaz de actuar como factor protector contra la ansiedad desarrollada por estudiantes universitarios durante la ocurrencia de la pandemia de Covid-19, ya que mientras el grupo de tratamiento mantuvo niveles, el grupo control demostró un aumento continuo de la ansiedad en el postest y seguimiento que coincidió con la primera ola de Covid-19 en la región donde residían los participantes.


O propósito deste estudo foi testar a eficácia e o efeito sustentado (follow-up) de uma intervenção de mindfulness de quatro semanas na redução da ansiedade e estresse em um grupo de estudantes universitários brasileiros. A intervenção foi adaptada para ser disponibilizada pelo aplicativo Neurosaúde, criado como parte deste trabalho para servir de plataforma para a intervenção. A pesquisa como um todo ocorreu em ambiente virtual, sendo recrutados 48 participantes que foram alocados randomicamente para o grupo intervenção ou para o controle de lista de espera. Medidas foram utilizadas para acessar os níveis de mindfulness, ansiedade e estresse antes, após a intervenção e no follow-up de quatro semanas. Aplicando a análise por intenção de tratar, encontramos diferenças significativas entre os grupos para a medida de ansiedade no follow-up com um tamanho de efeito grande (F (2.92) = 10.275, p = 0.000, η2 = 0.183). Nossos resultados sugerem que a intervenção de mindfulness de quatro semanas por meio de um aplicativo móvel foi capaz de agir como um fator protetor contra a ansiedade desenvolvida pelos estudantes universitários durante a ocorrência da pandemia causada pelo Covid-19, visto que, enquanto o grupo tratamento manteve níveis reduzidos de ansiedade, o grupo controle demonstrou um contínuo aumento de ansiedade no pós-teste e follow-up que coincidiu com a primeira onda de Covid-19 na região onde os participantes residiam.

RESUMO As novas Tecnologias de Informação e Comunicação estão cada vez mais presentes no cotidiano de pessoas e instituições em nível global. As informações que elas veiculam podem estar desatualizadas, incompletas, incorretas ou deliberadamente mentirosas. Vive-se um contexto de 'desordem informacional'. Na saúde, uma informação como essa pode causar dano ao indivíduo e à sociedade. Algumas alternativas têm sido apresentadas para enfrentar esse problema. Este artigo discute como a avaliação da qualidade da informação sobre saúde on-line e a certificação de sites podem servir para o enfrentamento da 'desordem informacional'. Ele apresenta uma metodologia de avaliação da acurácia da informação on-line seguindo as informações atualizadas e cientificamente comprovadas pela medicina baseada em evidências. Analisa o caso da leishmaniose visceral: uma das doenças negligenciadas mais prevalentes no mundo. Seguindo esse método, foi construída uma ferramenta com indicadores e foram avaliados sites de organizações não governamentais internacionais e dos Ministérios da Saúde do Brasil e da Índia, países nos quais a leishmaniose visceral é endêmica e que concentram grande parte dos casos mundiais da doença. Os resultados revelam que nenhum dos sites avaliados obteve mais de 50% de conformidade com os indicadores utilizados.


ABSTRACT New Information and Communication Technologies are increasingly present in the daily lives of people and institutions globally. The information they convey may need to be updated, complete, correct, or deliberately untrue. We live in a context of information mayhem. Health-related information like this can harm individuals and society. Some alternatives have been put forward to address this issue. This article discusses how online health information quality assessment and website certification can serve in confronting information disorder. It presents a methodology for assessing the accuracy of online information following the updated and scientifically proven information from evidence-based medicine. It analyzes the case of visceral leishmaniasis: one of the most prevalent neglected diseases in the world. Following this method, a tool with indicators was built, and the websites of international non-governmental organizations and the Ministries of Health of Brazil and India were evaluated. The results show that all the evaluated websites obtained at most 50% compliance with the indicators used.

ABSTRACT

Health care–associated infections (HAI) directly influence the survival of children in pediatric intensive care units (PICU), the most common being central line–associated bloodstream infection (CLABSI) 25–30%, followed by ventilator-associated pneumonia (VAP) 20–25%, and others such as catheter-associated urinary tract infection (CAUTI) 15%, surgical site infection (SSI) 11%. HAIs complicate the course of the disease, especially the critical one, thereby increasing the mortality, morbidity, length of hospital stay, and cost. The incidence of HAI in Western countries is 6.1–15.1% and in India, it is 10.5 to 19.5%. The advances in healthcare practices have reduced the incidence of HAIs in the recent years which is possible due to strict asepsis, hand hygiene practices, surveillance of infections, antibiotic stewardship, and adherence to bundled care. The burden of drug resistance and emerging infections are increasing with limited antibiotics in hand, is still a dreadful threat. The most common manifestation of HAIs is fever in PICU, hence the appropriate targeted search to identify the cause of fever should be done. Proper isolation practices, judicious handling of devices, regular microbiologic audit, local spectrum of organisms, identification of barriers in compliance of hand hygiene practices, appropriate education and training, all put together in an efficient and sustained system improves patient outcome.

ABSTRACT

OBJETIVO: Os serviços tecnológicos tiveram um crescimento exponencialmente acelerado nos últimos anos, principalmente com a chegada da pandemia coronavírus. Entre esses serviços estão aqueles relacionados à saúde, como a psicoterapia on-line. OBJETIVO: Esse estudo buscou compreender as representações sociais associadas ao atendimento on-line pelos psicólogos clínicos. MÉTODO: Utilizou-se como instrumentos: 1) teste de associação livre de palavras para investigação das representações sociais da psicoterapia on-line; 2) questionário semiestruturado para investigação das práticas sociais e atitudes relativas ao objeto; 3) questionário contendo questões de caracterização dos participantes e itens sobre o posicionamento atitudinal frente ao objeto. RESULTADOS E DISCUSSÃO: Os participantes foram 151 psicólogos residentes no Brasil, acessados pela técnica snowball. Para análise utilizou-se os softwares SPSS, EVOC 2000 e IRaMuTeQ. Como resultados pode-se destacar a representação social da psicoterapia online muito relacionada a aspectos de facilidade, acesso e conexão, que parecem estar relacionadas a possibilidades de alcance de alguns grupos sociais, atrelados ao vínculo e acolhimento necessários neste serviço. Estas representações aparecem também como ancoradas nas de psicoterapia presencial. CONCLUSÃO: Conclui-se que no momento desta pesquisa os profissionais estavam voltados à adaptação da psicoterapia presencial para a psicoterapia on-line e destaca-se a importância da ampliação dos conhecimentos sobre esse novo setting psicoterapêutico.


OBJECTIVE: Technological services have grown exponentially in recent years, especially with the arrival of the coronavirus pandemic. Among these services are those related to health, such as online psychotherapy. OBJECTIVE: This study sought to understand the social representations associated with online care by clinical psychologists. METHOD: The following instruments were used: 1) free word association test to investigate social representations of online psychotherapy; 2) a semi-structured questionnaire to investigate social practices and attitudes related to the object; 3) a questionnaire containing questions about the characterization of the participants and items about the attitudinal position towards the object. RESULTS AND DISCUSSION: The participants were 151 psychologists residing in Brazil, accessed through the snowball technique. For analysis, SPSS, EVOC 2000 and IRaMuTeQ software were used. As a result, the social representation of online psychotherapy can be highlighted, which is closely related to aspects of ease, access and connection, which seem to be related to the possibilities of reaching some social groups, linked to the bond and welcome needed in this service. These representations also seem to be anchored in face-to-face psychotherapy. CONCLUSION: It is concluded that at the time of this research, professionals were focused on adapting face-to-face psychotherapy to online psychotherapy, and the importance of expanding knowledge about this new psychotherapeutic setting is highlighted.


OBJETIVO: Los servicios tecnológicos han crecido exponencialmente en los últimos años, especialmente con la llegada de la pandemia del coronavirus. Entre estos servicios se encuentran los relacionados con la salud, como la psicoterapia en línea. OBJETIVO: Este estudio buscó comprender las representaciones sociales asociadas a la atención en línea por parte de psicólogos clínicos. MÉTODO: Se utilizaron los siguientes instrumentos: 1) prueba de asociación de palabras libres para investigar las representaciones sociales de la psicoterapia en línea; 2) cuestionario semiestructurado para investigar prácticas y actitudes sociales relacionadas con el objeto; 3) cuestionario que contiene preguntas sobre la caracterización de los participantes e ítems sobre la posición actitudinal hacia el objeto. RESULTADOS Y DISCUSIÓN: Los participantes fueron 151 psicólogos residentes en Brasil, accedidos a través de la técnica de bola de nieve. Para el análisis se utilizaron los software SPSS, EVOC 2000 e IRaMuTeQ. Como resultado, se puede destacar la representación social de la psicoterapia en línea, que está íntimamente relacionada con aspectos de facilidad, acceso y conexión, que parecen estar relacionados con las posibilidades de llegar a algunos grupos sociales, ligados al vínculo y acogida que se necesita en este servicio. Estas representaciones también parecen estar ancladas en la psicoterapia cara a cara. CONCLUSIÓN: Se concluye que en el momento de esta investigación, los profesionales estaban enfocados en adaptar la psicoterapia presencial a la psicoterapia en línea, y se destaca la importancia de ampliar el conocimiento sobre este nuevo escenario psicoterapéutico.

Resumo Este artigo pretende analisar experiências de empreendedorismo sustentável na comunidade rural José Gomes, Nordeste do Brasil, e promover uma reflexão sobre a relevância das políticas de desenvolvimento local na construção de projetos coletivos regionais. Este estudo caracteriza-se como exploratório e descritivo na abordagem qualitativa. Para tanto, foram utilizados técnicas rapport, entrevistas semiestruturadas (82 moradores), observação direta e diário de campo, além de pesquisas bibliográficas. A pesquisa abordou as políticas de desenvolvimento local como mecanismos de avanço e melhoria na qualidade de vida, na organização econômica e na conservação do meio ambiente, que devem ser incorporadas ao planejamento municipal e às ações do poder público. Os resultados apontaram potencialidades socioprodutivas e limitações estruturais e infraestruturais que esgotam as possibilidades de difusão do empreendedorismo sustentável e a consolidação do desenvolvimento local.


Resumen Este artículo pretende analizar experiencias de emprendimiento sostenible en la comunidad rural José Gomes, Nordeste de Brasil, y promover una reflexión sobre la relevancia de las políticas de desarrollo local en la construcción de proyectos colectivos regionales. Se utilizaron técnicas de rapport, entrevistas semiestructuradas (82 residentes), observación directa y diario de campo, además de la investigación bibliográfica. Las políticas de desarrollo local se entendieron como mecanismos para el avance y mejoramiento de la calidad de vida, la organización económica y la conservación del medio ambiente que deben incorporarse a la planificación municipal y a las acciones del poder público. Los resultados apuntaron potencialidades socioproductivas y limitaciones estructurales e infraestructurales que agotan las posibilidades de difundir el emprendimiento sostenible y consolidar el desarrollo local.


Abstract This article analyzes experiences of sustainable entrepreneurship in the José Gomes rural community in Northeast Brazil, reflecting on the relevance of local development policies in the construction of local collective projects. This qualitative, exploratory, and descriptive study adopted rapport techniques, semi-structured interviews (82 dwellers), direct observation, field diary, and bibliographic research. Local development policies were understood as mechanisms for advances and improvements in the quality of life, economic organization, and environmental conservation, which must be incorporated into municipal planning and governmental actions. The results pointed to socio-productive potentials and structural and infrastructural limitations, which hamper the possibilities of spreading sustainable entrepreneurship and consolidating local development.

ABSTRACT

Background: The COVID-19 pandemic had a widespread impact over the health-care system all over the world. This had resulted in varying degrees of psychological issues on healthcare workers. Aim and Objectives: The aim of the study was to assess the prevalence of depression among healthcare workers of a COVID-19 first line treatment center (CFLTC). Materials and methods: In this cross-sectional study, 240 healthcare workers who volunteered for the study at COVID first line treatment centre-3 NIT mega boys’ hostel, Kozhikode, Kerala were recruited. They were interviewed, basic demographic data and patient health questionnaire-9 was administered. The data were analyzed with Microsoft Excel. The prevalence among various categories of healthcare workers were compared. Results: The prevalence of depression among healthcare workers was 52%. The prevalence of depression is each category of staff-doctors, staff nurses, cleaning staffs, and patient caretakers-separately. It was found that depression is inversely proportional to the knowledge, experience, medical qualification, and training. Conclusion: The prevalence of undetected depression is high among healthcare workers working in unconventional environment as in warfront situations such as COVID pandemic. The present study emphasizes the importance of proper screening of depression among healthcare workers in such circumstances in years to come.
