ABSTRACT

Objective To study the effect of Fungi of Huai Er on human umbilicus vein endothelial cell((HUVEC)) and its inhibiting effect on liver cancer formation by liver cancer cells implanted under nude mice(subcutaneous) tissue, and to study the possible mechanism. Methods HUVEC was cultured with different concentrations of fungi of Huai Er and/or mitomycin(MMC) to observe the effect of Fungi of Huai Er on(HUVEC)′s hyperplasia , motility and adherence ability, and on the generation of blood vessels.In addition,the effect the Fungi of Huai Er on liver(cancer) formation by liver cancer cells implanted in nude mice(subcutaneous) tissue was also observed. Results Fungi ≥2mg/ml, can apparently reduce the hyperplasia(ability),inhibit motility and adherence ability of HUVEC,and the formation of blood vessels. The strongest effect on inhibiting the subcutaneous tumor formation was Fungi(3g/kg)plus MMC(500?g/kg),followed by MMC,and Fungi(3g/kg). Conclusions Possible mechanism of the inhibiting effect of Fungi on liver(cancer) is that the Fungi inhibits the(hyperplatic) ability, motility and adherence ability of HUVEC, and this(inhibits) the growth of blood vessels. Therefore, the growth of blood vessels is reduced and the development of liver cancer is suppressed.

ABSTRACT

Objective To investigate the inhibitiory effect of tacrolimus(FK506) and adriamycin(ADM) on hepatocellular carcinoma.Methods HepG-2 cells were cultured,and divided into 4 groups,namely(control)、FK506、ADM、FK506+ADM groups,the cells were trated by the drugs for 24 to 48 hours(respectively).The inhibitory rate of the cells was measured by MTT assay,and cell cycle and cell apoptotic rate were detected by flow cytometry(FCM).Results The ability of tumor cell growth were inhibited by FK506 after 48h;the apoptosis ratio was increased when FK506 was below 100?g/L,and when it(exceeded) that value,the apoptosis ratio was decreased.FK506 and ADM significantly prolonged cell G_2 phase;combined tacrolimus and adriamycin had synergistic role effect on arresting the cell in G_2 phase,FK506 combined with adriamycin demonstrated synergistic effect on apoptosis.Conclusions FK506 could inhibit the growth of hepatocellular carcinoma,arrest the cell in G_2 phase,and increase apoptosis.FK506 combined with adriamycin demonstrated synergistic inhibitory effect on hepatocellular carcinoma.

ABSTRACT

ObjectiveTo evaluate the effect of suture ligation around the tumor combined with intra-tumoral drug injection to treat hepatic cavernous hemangioma. MethodsTwenty-two cases of hepatic cavernous (hemangioma) underwent laparatomy and suture ligation around the tumor plus intra-tumoral drug injection under direct vision. ResultsThere was no postoperative mortality or morbidity. 22 patients were followed up for 3 months to 4 years. After this treatment for hepatic cavernous hemangioma, the success rate was 100%, and the significantly success rate was 76.19%. ConclusionsThe combined therapy for hepatic cavernous (hemangioma) is safe, reliable, simple and easy to perform.

ABSTRACT

Objective To investigate potential mechanism of arsenic trioxide (As2O3) and/or adriamycin (ADM) induces apoptosis in hepatocellular carcinoma (HCC) cells. Methods Human hepatocellular (carcinoma) cells, HepG2, were treated with As2O3 combined with/without ADM in various concentrations for various time, and then the expression of survivin was detected by immunocytochemistry and RT-PCR (respectively). Results (1)Before the cells were interfered with drugs, survivin was highly expressed in HepG2 cells. (2)Expression of survivin was reduced in cells treated with various concentrations of As2O3 or ADM , and the reduction was dependent on durg concentration and time, these two factors were interrelated. (3)Expression of survivin was reduced more markedly in cells treated with ADM than in those treated with As2O3 and the reduction was additionally increased in cells treated with a combination of the two compounds. Conclusions (1)Either As2O3 or ADM can downregulate the expression of survivin in HepG2 cells in a concentration-or time-dependent manner; (2)At the same concentration, ADM is more effective than As2O3 on the downregulation of survivin expression in HepG2 cells, but the effect can be enhanced by combination of these two drugs. (3)As2O3 and ADM may induce apoptosis in HepG2 cells through the downregulation of survivin expression and thus exert their anti-cancer effect. Their combination may enhance their anti-tumor effect, so that in clinical use, the dosage of each durg may be reduced.

ABSTRACT

Objective To explore the therapeutic effect of dual perfusion embolization and chemotherapy via hepatic artery and portal vein(combmation treatment) in the treatment of unresectable PHC.Methods Eighty-one cases of unresectable PHC were randomly divided into two gronps: (1) Combination treatment group.Forty-one cases,These cases received embolization and chemotherapy via hepatic artery and portal vein through a drug delivery system intraoperatively,and then embolization and chemotherapy via the drug pump were given periodically. (2) TACE group.Forty cases.These cases were treated with Seldinger's technique, the dosage of drugs were the same as used in the former group during laparotomy. After 3 times of treatment, AFP, the size of tumor, liver function, body weight, abdominal perimeter, survival time of the two groups were compared.Results The weight, AFP, decrease of tumour size in combination group were much better than those in TACE group( P 0.05). The median survival time in the two groups were 18.0 months and 11.1 months ( P =0.0001). The accumulating survival rate of 6, 9, 12, 24 months were 87.8%, 78.0% , 68.2%,31.7% in combination group, and 70.0%, 52.5%, 30.0%, 5.0% in TACE group, respectively . The factors affecting survival were therapeutic method, liver function, size of tumour.Conclusions Combination treatment is simple, convenient with less complications, and the effect is better than TACE. So it is an effective method for the unresectable hepatic carcinoma.

ABSTRACT

Objective To study the efficiency enhancement and toxicity reduction effects of scutellaria barbata polysaccharides(SBPS) on mice treated with cytoxan(CTX).Methods Transplanted H22 hepatoma mice were randomly divided into negative control group,CTX group and combined SBPS and CTX group.After 10 d of consecutive drug administration,the tumor inhibitory rate,blood white cell count,phagocytic index and activity of IL-2 were measured.Results SBPS increased the tumor inhibitory rate and reduced the toxicity of cytoxan,and enhanced the function of mononuclear phagocytes and the activity of IL-2 and TNF-?.Conclusions SBPS has efficiency enhancement and toxicity reduction effects on cytoxan,and the mechanism may be by increasing the immunologic function of the organism.
