ABSTRACT

Abstract Background: In artificial insemination, chicken egg yolk is added to bovine semen to protect it during the cryopreservation process, although it contains substances that can affect the microbiological quality and metabolism of sperm. Objective: To evaluate post-thaw quality of bovine cryopreserved semen added with centrifuged and non-centrifuged egg yolk, low-density lipoproteins (LDL), and trehalose (T). Methods: Ten ejaculates from five bulls were cryopreserved under the treatments T1: pure egg yolk (PEY) at 20% v/v, T2: centrifuged egg yolk (CEY) at 20% v/v, T3: LDL at 8% v/v, T4: T at 100 mM, and T5: T at 100 mM plus LDL at 8% v/v (TLDL). Spermatic motility and kinetics, functional membrane integrity (FMI), structural membrane integrity (SMI), sperm vitality (SV) and abnormal morphology (AM) were assessed using the Sperm Class Analyzer (SCA®) system, hypoosmotic test (HOST), SYBR/PI probes, and eosin-nigrosin staining, respectively. A completely randomized design was used. Normal distribution of the variables was validated through the Kolmogórov- Smirnov test. A generalized linear model was used to determine sources of variation. Means were compared using the Tukey test. Results: Inclusion of CEY or LDL had a similar effect on sperm protection, and were superior for motility, kinetics and membrane integrity compared to the other treatments (p<0.05). CEY was superior for progressive motility (p<0.05). The cryoprotective action of LDL was similar to TLDL for motility and kinetics, SMI, SV, and AM (p<0.05). Inclusion of PEY and T resulted in the lowest semen quality (p<0.05). The use of T resulted in a reduction in FMI and SMI (p<0.05). No differences in AM between treatments were found (p>0.05). Conclusions: Egg yolk can be replaced by centrifuged egg yolk or low-density lipoproteins in the freezing extender used for bovine semen used in artificial insemination.


Resumen Antecedentes: la yema de huevo de gallina se agrega al semen bovino usado en inseminación artificial para protegerlo durante el proceso de criopreservación, aunque ésta tiene sustancias que pueden afectar el metabolismo espermatico y la calidad microbiológica del semen. Objetivo: evaluar la calidad post-descongelación del semen bovino criopreservado agregado con yema de huevo centrifugada y no centrifugada, lipoproteínas de baja densidad (LDL) y trehalosa (T). Métodos: diez eyaculados de cinco toros se criopreservaron bajo los tratamientos, T1: yema de huevo pura (PEY) al 20% v/v, T2: yema de huevo centrifugada (CEY) al 20% v/v, T3: LDL al 8% v/v, T4: T a 100 mM, y T5: T a 100 mM más LDL al 8% v/v (TLDL). La movilidad y la cinética espermática, la integridad funcional de la membrana (FMI), la integridad estructural de la membrana (SMI), la vitalidad espermática (SV) y la morfología anormal (AM), se determinaron mediante el sistema Sperm Class Analyzer (SCA®), la prueba hipoosmótica (HOST), las sondas SYBR/PI y la tinción con eosina-nigrosina, respectivamente. Se utilizó un diseño completamente al azar. La normalidad de las variables se validó mediante la prueba de Kolmogórov-Smirnov. Se utilizó un modelo lineal generalizado para determinar las fuentes de variación. Las medias de los tratamientos se compararon mediante la prueba de Tukey. Resultados: CEY y LDL tuvieron un efecto similar en la protección de los espermatozoides, siendo superiores a los demás tratamientos respecto a movilidad, cinética e integridad de la membrana (p<0,05). CEY fue superior para la movilidad progresiva (p<0,05). La acción crioprotectora de LDL fue similar a TLDL para movilidad y cinética, SMI, AM y SV (p<0,05). PEY y T resultaron en la más baja calidad seminal (p<0,05). El uso de T redujo la FMI y la SMI (p<0,05). No se encontraron diferencias en AM entre los tratamientos (p>0,05). Conclusiones: la yema de huevo puede reemplazarse por yema de huevo centrifugada o por lipoproteinas de baja densidad en el diluyente de congelación usado para semen bovino destinado a inseminacion artificial.


Resumo Antecedentes: a gema de ovo de galinha tem sido utilizada com a finalidade de proteger o sêmen bovino durante o processo de criopreservação, embora tenha substâncias que possam afetar o metabolismo dos espermatozóides e a qualidade microbiológica do sêmen utilizado para a inseminação artificial. Objetivo: avaliar a qualidade pós-descongelamento do sêmen bovino criopreservado com gema de ovo centrifugada e não centrifugada, lipoproteínas de baixa densidade (LDL) e trealose (T). Métodos: dez ejaculados de cinco touros foram criopreservados sob os tratamentos, T1: gema de ovo pura (PEY) 20% v/v, T2: gema de ovo centrifugada (CEY) 20% v/v, T3: LDL 8% v/v, T4: T 100 mM e T5: T 100 mM mais LDL 8% v/v (TLDL). Mobilidade e cinética espermática, integridade funcional da membrana (FMI), integridade estrutural da membrana (SMI), vitalidade espermática (SV) e morfologia anormal (AM) foram determinadas por o sistema Sperm Class Analyzer (SCA®), teste hiposmótico (HOST), coloração com SYBR/PI e eosina-nigrosina, respectivamente. Um design completamente aleatoriedade foi usado. A normalidade das variáveis foi validada pelo teste de Kolmogorov-Smirnov. Um modelo linear generalizado foi utilizado para determinar as fontes de variação. As médias dos tratamentos foram comparadas pelo teste de Tukey. Resultados: T2 (CEY) e T3 (LDL) tiveram efeito similar na proteção espermática, sendo superior aos demais tratamentos para mobilidade, cinética e integridade da membrana (p<0,05). T2 (CEY) foi superior para mobilidade progressiva (p<0,05). A ação crioprotetora de T3 (LDL) foi semelhante à T5 (TLDL) para motilidade e cinética, SMI, SV e AM (p<0,05). T1 (PEY) e T4 (T) tiveram a menor qualidade seminal (p<0,05). O uso de T4 (T) produziu uma redução na SMI e FMI (p<0,05). Não foram encontradas diferenças na AM entre os tratamentos (p>0,05). Conclusões: a gema de ovo pode ser substituída por gema de ovo centrifugada ou lipoproteínas de baixa densidade no diluente de congelamento de sêmen bovino.

ABSTRACT

The significant reason for morbidity and mortality in industrialised countries is coronary artery disease. Globally, it is considered as one of the major public health problems. There is emerging evidence of premature coronary artery disease occurring in Asian Indians at least 10 years earlier as compared to other ethnic groups. The most important atherosclerotic risk factor is dyslipidaemia. Recent studies have reported that Total Cholesterol (TC), Low Density Lipoproteins (LDL), Triglycerides (TG) levels have increased among young urban population over a 20-year period. Hence, this study was carried out to find out the prevalence of dyslipidaemia among different age groups in urban South Indian population. METHODSThis study is a descriptive cross-sectional study conducted among patients visiting the OPD of Department of General Medicine. The study subjects were divided into 3 groups as per age as 18 - 40 years, 41 - 60 years and > 60 years by involving 1989 subjects of whom 532 were in 18 - 40 years age group, 522 were in 41 - 60 years age group and 935 were over 60 years of age. In this study, fasting serum lipid profile was estimated. RESULTSThe study revealed higher prevalence of dyslipidaemia in the group < 60 years. Statistically significant difference in mean was found with cholesterol, LDL, and triglycerides in the age group of 18 - 40 years and more than 60 years. Statistically significant difference was not found between 18 – 40 years and 40 - 60 years for HDL. Mean serum cholesterol was high in the age group 18 – 40 years. The highest mean triglycerides were found in the age group of 41 - 60 years. The highest mean HDL levels were found in the above 60 years age group. CONCLUSIONHigher percentage of dyslipidaemia was found in the age groups less than 60 years. It is also the need of the hour to devise comprehensive strategy for awareness programme for periodically screening of all age groups.

ABSTRACT

Background: Stroke is one of the major global health problems. Stroke is the most common clinical manifestation of cerebrovascular disease of which more than 99% are due to arterial involvement and less than 1% due to venous involvement in the form of Cerebral Venous Thrombosis (CVT). Among arterial causes 85% are due to infarction and 15% due to haemorrhage.1,2 There is  difference in serum lipid levels in subtypes of strokes to guide lipid-lowering therapy which can reduce incidence of stroke and stroke related mortality by adapting primary and secondary preventive measures.3,4  Authors have endeavoured to correlate severity of lipid derangement and stroke.Methods: In this study 64 consecutive eligible ischaemic stroke cases and 64 eligible hemorrhagic stroke cases would be included. Cases of strokes will be divided into ischaemic and hemorrhagic as per clinical features and with help of brain imaging by CT scan and MRI at the time of admission and 8 hour fasting lipid profile was collected from all cases. All this information will be filled in preformed format.Results: Serum lipid profile of two categories of stroke showed raised serum total cholesterol in 39.1% patients of ischaemic stroke in contrast to 18.8% patients with haemorrhagic stroke (p=0.019).Stroke patients showed raised in LDL cholesterol in 29.7% patients of ischaemic stroke in contrast to 9.4% patients with haemorrhagic stroke, (p=0.007).Conclusions: Based on the finding of our study we conclude that ischemic stroke patient had higher lipid derangement as compare to haemorrhagic stroke in terms of raise total cholesterol, LDL cholesterol and decrease HDL cholesterol.

ABSTRACT

Background: Hypothyroidism is usually associated with the involvement of the cardiovascular system in the form of the cardiac abnormalities. The Research data on this has been found to be limited. The dysfunction of the cardiac caused by hypothyroidism is usually reversible. Objective was to study the incidence of cardiac abnormalities in patients with hypothyroidism.Methods: Total 40 confirmed cases of hypothyroid disease were included in the present study. General investigations, ECG, ECHO was done in all the patients to assess for cardiovascular involvement. The data was entered in the Microsoft Excel worksheet and analysed using proportions.Results: One fourth of the patients were found to have the bradycardia. The Delayed ankle jerk was seen in 67.5% of the cases and the next common CNS manifestation was the hoarseness of the voice. 30% of the case had normal ECG findings. 40% of the cases had the bradycardia. 35% of the cases had the low voltage complexes. 32.5% of the cases had the normal ECHO reports. 27.5% of the cases had the pericardial effusion. Same proportion of the cases were found to have the diastolic dysfunction. But most of them had the dysfunction and the severe cases were found to be very few. Only two patients had IVS thickness.Conclusions: Majority of the cases of the hypothyroidism in the present study were found to have the cardiovascular involvement. This points towards the importance of the early diagnosis of hypothyroidism and the prevention of complications like the cardiovascular system and the central nervous system involvement.

ABSTRACT

Background: Acute Myocardial Infarction (AMI) is one of the major causes of mortality in the world. Atherosclerosis leading to AMI is the most common and severe clinical manifestation observed. Dyslipidemia is one of main traditional risk factor for MI, but in more than 50% of CHD events dyslipidemia was absent.  Atherosclerosis is considered as both a chronic inflammatory condition and a disorder of lipid metabolism. The present study was aimed to estimate the levels of serum lipids (total cholesterol, LDL cholesterol, HDL cholesterol) and hs-CRP an inflammatory marker in newly diagnosed AMI cases and to find out any correlation between these two.Methods: The study was conducted on 50 newly diagnosed myocardial infarction cases, admitted in Cardiology Department Narayana Medical College, Nellore. Both the sexes were included. Fifty age and sex matched healthy individuals were selected as controls. Lipid levels are estimated by end point colorimetric assay (HUMANSTAR kit) and hs-CRP was estimated by immunofluorescence technique (Boditechkit).Results: Significant raised levels of hs-CRP (p value <0.0001) and low HDL cholesterol (p value =0.0085) levels among the cases was noticed. A significant positive correlation was observed between LDL cholesterol and hsCRP (r=0.109, p=0.040). A non-significant positive correlation between hsCRP and HDL cholesterol  (r=0.291, p=0.453) was observed.Conclusions: The results of the study implicate the role of inflammatory component in causing atherosclerosis or coronary artery disease rather than dyslipidaemias alone to be attributed for atherogenesis. Concomitantly HDL cholesterol levels were found to be low in cases that results from the inflammatory component in atherogenesis.

ABSTRACT

The dietary fats are composed primarily of triacylglycerols and some amount of phospholipids and cholesterol. Being hydrophobic in nature, these are insoluble in water, and hence cannot be transported in the blood plasma per se; to enable these lipids to be transported by the blood stream to various peripheral tissues, nature has devised the technique of making these soluble by binding them to proteins. These proteins involved in lipid transport are known as apolipoproteins, and the protein-lipid particle is known as lipoprotein. Thus, lipoproteins can be considered to be the primary transportmechanism to carry lipids from the alimentary tract to various parts of the body. Lipoproteins have gained prominence in medical field over the past few decades because of their role in the aetio-pathogenesis of cardiovascular diseases, principally atherosclerosis which is the cause of coronary artery disease and myocardial infarction. The various types and sub-types of lipoproteins have been found to have differing and even opposing roles in the development of arterial diseases. An understanding of the differing populations of lipoproteins, the associated proteins and other enzymes, and the myriad variety of inter-actions among themselves and with body cells is vital to our understanding the pathways involved in the development of cardio-vascular disordersand in determining the precise steps where pharmacological interventions can be introduced

Aim:This study investigated the relationship between small dense low density lipoprotein (sdLDL), tumour necrosis factor-alpha (TNF-α), aspartate aminotransferase (AST), alanine aminotransferase(ALT) and alkaline phosphatase (ALP) in chronic hepatitis B patients.Duration of Study: June2018-March 2019.Subjects and Methods:Sixty (60) participants were recruited for this cross sectional study. They comprised thirty (30) clinically diagnosed chronic hepatitis B virus (HBV) infected patients attending clinic at a tertiary hospital in Osogbo, Osun state, Nigeria. Thirty (30) apparently healthy volunteers were recruited as control subjects after fulfilling the inclusion criteria. Anthropometric measurements were performed using standard method. About 6mL of venous blood was collected from each study participant,serum was extracted and kept at -80oC until time of analysis. Small dense LDL, TNF-α, AST, ALT and ALP were determined using enzymelinked immunosorbent assay and colorimetric method as appropriate. Data analysis was doneusing Student’s t-test for Original ResearchArticle comparison of variables and Pearson’s correlation was used to determine the relationship between variables. P–value less than 0.05 was considered significant. Results:SdLDL, TNF-α, AST and ALT were significantly elevated in HBV patients when compared with the control subjects (P<0.05). SdLDL had a significant positive correlation with TNF-α (P=0.03), AST (P=0.01), ALT (P=0.00). TNF-α had a significant positive correlation with AST (P=0.02) and ALT (P=0.00).Conclusion:This study revealed a noteworthy positive relationship between sdLDL, TNF-α and hepatic aminotransferases in chronic hepatitis B patients

Introducción. Numerosos estudios han demostrado una fuerte asociación entre el aumento de la concentración sérica del colesterol de las lipoproteínas de baja densidad (LDLc) y la incidencia de la enfermedad coronaria (EC). Objetivos. Derivar y validar la performance de una nueva ecuación para estimar el LDLc. Diseño. Estudio observacional y descriptivo. Institución. Hospital Edgardo Rebagliati Martins, Lima, Perú. Material. Informes de perfil lipídico. Intervenciones. La muestra de 4 644 informes de perfil lipídico extraídos de los archivos del laboratorio central, correspondiente a sujetos de ambos sexos, que presentaban edades entre 18 a 75 años, fue repartida en dos grupos. Del primer grupo conformado por 2 136 informes se derivó la ecuación para estimar el LDLc, mediante análisis de regresión múltiple. El segundo grupo conformado por 2 508 informes sirvieron para validar la performance de la nueva ecuación mediante la comparación con la medida del LDLc por el método directo homogéneo y estimado por la fórmula de Friedewald. Principal medida de resultados. Concentración sérica del LDLc. Resultados. La fórmula de Friedewald y la nueva ecuación infraestimaron el LDLc, obteniéndose sesgos de 14,01 mg/dL y 5,52 mg/dL, respectivamente, con respecto al método directo. Los errores sistemáticos en los tres niveles de decisión clínica para el LDLc resultaron ser mayores para la fórmula de Friedewald (12,6%, 12,0% y 11,7%) comparados a la nueva ecuación (4,2%, 5,7% y 6,7%). El factor de corrección de sesgo, que valora la exactitud, resultó ser más alto y más cerca de la línea de perfecta concordancia para la nueva ecuación, en toda la muestra y cuando esta se estratificó por niveles de triglicéridos. Conclusiones. La nueva ecuación ofrece una mayor exactitud y bajo error analítico comparada a la fórmula de Friedewald para estimar el LDLc, en los tres niveles de decisión clínica y en sueros con diferentes valores de triglicéridos.


Introduction: Various studies have shown a strong association between the increase in cholesterol low density lipoproteins (LDLc) serum concentration and the incidence of coronary heart disease. Objectives: To derive and validate the performance of a new equation to estimate the LDLc. Design: Observational and descriptive study. Institution: Hospital Edgardo Rebagliati Martins, Lima, Peru. Material: 4 644 lipid profile reports taken from the files of the hospital's central laboratory, belonging to subjects of bot. sexes, between 18 and 75 years old. Interventions: The sample consisted of 4 644 lipid profile reports obtained from the files of the hospital's central laboratory, and belonged to subjects of both sexes aged 18-75 years. It was divided into two groups: the first group was represented by 2 136 reports, and was used to derive the equation to estimate the LDLc by using multiple regression analysis; the second group comprised 2 508 reports that served to validate the performance of the new equation compared with the LDLc measurement by the homogeneous direct method and estimated by the Friedewald formula. Main outcome measure: LDLc serum concentration. Results: The Friedewald formula and the new equation underestimated the LDLc with a shortfall of 14.01 mg/dL and 5.52 mg/dL, respectively, with regard to the direct method. Systematic errors in the three clinical decision levels for LDL cholesterol were found to be higher for the Friedewald formula (12.6%, 12.0 %, and 11.7%) compared to the new equation (4.2%, 5.7% and 6.7%). The bias correction factor, which assessed accuracy, turned out to be higher and closer to the line of perfect match for the new equation, both in the whole sample and when the entire sample was stratified by levels of triglycerides. Conclusions: The new equation provided greater accuracy and lower analytical error compared to the Friedewald formula to estimate the LDL cholesterol in the three clinical decision levels and with different values of serum triglycerides.

ABSTRACT

Objective To study the expression of plasma oxidized low-density lipoprotein (oxLDL) in children with acute phase Kawasaki disease (KD), and investigate its value for early prediction of coronary artery lesions in KD. Methods Totally 80 children with KD were collected. Children were divided into four groups by the results of echocardiogram of coronary artery in different periods: CAL1 group (children with coronary artery lesions (CAL+) both in acute and sub-acute phase, 8 cases), CAL2 group (children with CAL+in acute phase but recovery normal (CAL-) in sub-acute phase, 10 cases), NCAL1 group (children with CAL-in acute phase but occur CAL+ in sub-acute phase, 10 cases) and NCAL2 group (children with CAL- both in acute and sub-acute phase, 52 cases). The serum samples (before the use of intravenous immunoglobulin) were collected in acute phase. Twenty healthy controls and twenty fever controls were enrolled into the study, and their serum samples were collected. OxLDL was measured by enzyme linked immunosorbent assay (ELISA). They were compared using ANOVA, pairwise comparison LSD-t test. And ROC curve analysis was used to determine the threshold. Results Compared with the control groups,plasma oxLDL levels were higher in children with KD, both CA+and CAL-[(15.0±3.3) mU/L, (12.3±3.5) mU/L vs (9.2±2.2) mU/L, (8.0±2.3) mU/L, F=20.435, P<0.05]. Plasma oxLDL levels were increased more significantly in children with CAL+ than children with CAL- in KD [(15.0 ±3.3) mU/L vs (12.3 ±3.5) mU/L, t=2.28, P=0.002]. There was significant difference in the concentration of oxLDL between the groups of Kawasaki disease (F=5.068, P=0.003). Plasma oxLDL levels were significantly higher in the NCAL1 group than those in the NCAL2 group [(14.5 ±3.8) mU/L vs (11.9±3.3) mU/L, t=2.29, P=0.02], but there were no statistically significant difference between the NCAL1 group and CAL1 or CAL2 group [(14.5±3.8) mU/L vs (15.9±3.9) mU/L, (14.5±3.8) mU/L vs (14.2±2.7) mU/L, t=0.73, 0.20;P=0.41, 0.84]. ROCs analysis indicated that oxLDL≥13.83 mU/L, could be the threshold for the prediction of coronary artery lesions with the sensitivity of 0.607 and a specificity of 0.75. Conclusion OxLDL plays an important role in coronary artery lesions in KD. The coronary endothelial dysfunction is earlier than coronary dilatation, and oxLDL is expected to become a reliable early predictor of coronary artery lesions in KD.

ABSTRACT

La terapia biológica basada en anticuerpos monoclonales, constituyen una nueva herramienta terapéutica, que de la mano del desarrollo de la biología molecular permiten intervenir puntos claves en la etiopatogenia de la enfermedad, permitiendo una terapia personalizada. Se han desarrollado nuevos anticuerpos monoclonales que intervienen en las enfermedades cardiovasculares, por lo que el entendimiento de los principios terapéuticos, los tipos y el mecanismo de acción de estos fármacos es esencial para un uso adecuado. En este artículo se discutirá el perfil de los anticuerpos monoclonales en la enfermedad cardiovascular, como los inhibidores de la PCSK-9 y el antídoto del Dabigatran, pasando por las moléculas ya conocidas como el abciximab, el antídoto de la digoxina y la terapia inmunomoduladora en el trasplante cardiaco. Del mismo modo, se evalúa una posible aplicación de los anticuerpos monoclonales usados en reumatología que podrían tener un papel importante en el manejo de las enfermedades cardiovasculares.


The monoclonal antibodies (mAb) constitute a new therapeutic tool in cardiovascular medicine, based in the development of the molecular biology in this area. The understanding of therapeutic principles, types and mechanism of action of these drugs is essential for proper use and management of adverse reactions. We discuss the profile of the PCSK-9 inhibitors, the Dabigatran antidote, the abciximab, the digoxin antidote, the therapy in heart transplantation, and a possible cardiovascular use of monoclonal antibodies used in rheumatology.

ABSTRACT

O presente estudo teve como objetivo avaliar a capacidade crioprotetora das lipoproteínas de baixa densidade (LDL) presentes no plasma de gema de ovo, adicionado ao trihidroxiaminometano (TRIS) para congelar sêmen ovino. Trinta e seis ejaculados foram coletados para formar 12 "pool". Cada alíquota de sêmen foi diluída em TRIS-gema de ovo (TRISG) ou TRIS- plasma de gema de ovo (TRISP) antes de congelar o sêmen. Para a obtenção do plasma da gema de ovo, foi utilizado o método de ultracentrifugação. Após o descongelamento, não houve diferença entre os dois extensores em relação aos parâmetros seminais (motilidade, viabilidade, membrana acrossômica e plasma). No entanto, no Teste de Termo Resistência Lenta (TTRL - 4h/38°C), o sêmen congelado com TRISP resultou no aumento do número de espermatozoides com acrossoma intacto (P<0,032). Conclui-se que o diluente TRISP é uma alternativa para criopreservação do sêmen de carneiros, pois permite uma melhora da característica seminal em relação ao diluente TRIS gema, mas sem a necessidade do processo de purificação das LDL.


The present study aimed to evaluate the cryoprotectant low-density lipoprotein (LDL) present in the plasma of egg yolk added to the extender trihidroxiaminometano (TRIS) for freezing ram semen. Thirty-six ejaculates were collected to form 12 pool. Each aliquot of semen was diluted in TRIS-egg yolk (TRISG) or TRIS-egg yolk plasma (TRISP) before freezing the semen. The plasma of egg yolk was obtained by ultracentrifugation. After thawing, no difference was detected between the two extenders in relation to seminal parameters (motility, viability, plasma membrane and acrosome). However, in the thermal resistance slow test (4h in a water bath at 38°C), the semen frozen with TRISP resulted in higher number of sperm with intact acrosome than those with TRISG (P<0.032). It was concluded that the TRISP extender is an alternative for cryopreservation of ram semen, once it improves the semen characteristics in comparison to TRISG, avoiding the purification process of LDL.

ABSTRACT

Objective This study aims to investigate the effects of high-fat diet rich in perilla oil on the expression of key genes that regulate hepatic VLDL synthesis in obese rats .Methods Sixty healthy male 5-week old SD rats were randomly divided into 2 groups.The rats in the normal control group (NC, n=12) were given normal diet, and the rats in the high fat group ( HF, n=48) were given a pure high fat diet in order to induce rat models of obesity .In the intervention period, the obesity model rats were randomly divided into 4 subgroups including consistent high fat group (CHF) and three intervention groups depending on perilla oil substitution rate of lard in CHF:20%PO, 50%PO and 100%PO.The serum triglyceride (TG) of the rats was measured after 4 weeks.Real-time PCR was applied to measure microsomal triglyceride transfer protein ( Mtp) and apolipoprotein B ( Apob) mRNA, and western blot assay was used for detecting the expression of MTP and APOB in the liver .Results Compared with the NC group , the CHF rats exhibited significantly high fat deposi-tion.The serum TG was markedly higher and the MTP and APOB were decreased at gene and protein levels in the CHF group compared with the NC group .After the intervention , PO remarkably reduced the level of serum TG and decreased he-patic fat deposition as it showed by pathological examination .At the gene and protein levels , MTP and APOB were upregu-lated by PO to different degrees .Conclusions All the three PO intervention can promote VLDL synthesis and secretion , and decrease the hepatic fat deposition in the obese rats .Furthermore , PO upregulates the expression of MTP at gene and protein levels in a dose-dependent manner .

INTRODUCCIÓN: actualmente hay consenso en que el proceso aterosclerótico se inicia en la infancia, y la dislipidemia es uno de los principales factores de riesgo aterogénicos que deben ser estudiados, así como la valoración nutricional para una adecuada prevención. OBJETIVO: identificar algunas señales de aterosclerosis tempranas como el sobrepeso y las dislipidemias en adolescentes. MÉTODOS: se realizó un estudio descriptivo transversal de 372 adolescentes de la Secundaria Básica "Protesta de Baraguá". Se hicieron mediciones de peso, talla, índice de masa corporal y circunferencia de la cintura. Se tomó muestra de sangre venosa con ayuno de 12 horas. Se midió el colesterol total, el colesterol unido a lipoproteína baja y alta densidad, y triglicéridos. En el análisis estadístico se realizaron prueba de comprobación de media entre variables de valoración nutricional y lipídicas entre sexos (prueba t), y se trabajó con una probabilidad de error menor de 0,05. RESULTADOS: la media del peso, talla y circunferencia de la cintura fueron mayores en el sexo masculino (p< 0,05). El exceso de peso fue 23,7 %, y a partir del percentil 90-97 hubo un 21,8 % de exceso de grasa abdominal. La media del colesterol total, lipoproteínas de alta y baja densidad y triglicéridos, fue similar en los diferentes sexos, sin relación significativa (p> 0,05). El 18,5 % presentó colesterol total limítrofe alto, el 26,6 % tenía triglicéridos limítrofe alto y 7,5 % alto, con predominio del sexo femenino. CONCLUSIONES: alrededor de la cuarta parte de los adolescentes tenían exceso de peso e incremento de la grasa abdominal, casi la cuarta parte tenía el colesterol total limítrofe y alto, y la alteración lipídica observada con mayor frecuencia fue la hipertrigliceridemia.


INTRODUCTION: there is a current consensus that the atherosclerotic process begins at childhood and that dyslipidemia is one of the atherogenic risk factors to be studied together with the nutritional assessment for the adequate prevention of atherosclerosis. OBJECTIVE: to identify some early signs of atherosclerosis such as overweight and dyslipidemias in teenagers. METHODS: a cross-sectional descriptive study of 372 teenagers from "Protesta de Baragua" junior high school. Weight, height, body mass index and waist circumference were measured. Blood samples were taken after 12 hour fasting. Total cholesterol, low and high density lipoprotein cholesterol and triglyceride were all measured. The statistical analysis included the t test among nutritional assessment and lipid variables between sexes and the error probability was lower than 0.05. RESULTS: weight, height and waist circumference means were higher in males (p< 0.05). Overweight was 23.7 % and from the 90-97th percentile, the excessive abdominal fat was 21.8 %. The mean of total cholesterol, high and low density lipoproteins and triglycerides was similar in both sexes, with no significant relation (p> 0.05). In this group, 18.5 % of adolescents presented with borderline high total cholesterol, 26.6 % showed borderline high triglyceride rate and 7.5 % had high cholesterol, being females predominant. CONCLUSIONS: one fourth of adolescents approximately exhibited overweight and increased abdominal fat; almost 25 % had reached borderline and high total cholesterol whereas the most observed lipid disorder was hypertrigliceridemia.

ABSTRACT

Human serum paraoxonase is physically associated with HDL and has been implicated in the detoxification of organophosphates and possibly in the prevention of LDL lipid peroxidation and therefore retards atherosclerosis. HDL levels are inversely related to the risk of developing atherosclerosis. We investigated the serum activity and concentration of paraoxonase and HDL levels in 104 subjects (42 diabetic patients without complications, 42 controls, 20 diabetic patients with complications.). Paraoxonase activity was found to be lower in diabetic patients than in controls. Similarly there was reduction in HDL levels in cases suggesting a positive correlation between HDL and paraoxonase levels.

ABSTRACT

The present study was conducted to find out the role of serum lipids in the development of diabetic retinopathy in type II Diabetes Mellitus. One hundred fifty subjects aged 30-70 years attending OPD at Old Civil hospital, Surat, participated in the study and were divided into three groups. Group I included 50 healthy non-diabetic subjects who served as control. Group II included 50 diabetic subjects with no signs of diabetic retinopathy and Group III included 50 diabetics with diabetic retinopathy. Funduscopy under homatropine was done in all the subjects. Serum triglycerides and total cholesterol were estimated by enzymatic methods and High Density Lipoprotein by precipitation method. Serum Low density lipoprotein was calculated using Friedewald’s formula. It was found that triglyceride levels were significantly raised (p<0.05) in subjects with diabetic retinopathy as compared to those without diabetic retinopathy showing a positive association of Triglycerides with the incidence of diabetic retinopathy. Whereas no such association was found between low density lipoprotein and total cholesterol levels with the prevalence of diabetic retinopathy.

Se realizó un estudio descriptivo y transversal de 400 habitantes (mayores de 15 años de edad) de las comunidades pehuenches de Alto Biobio en Chile, de mayo a octubre del 2011, a fin de determinar la prevalencia de dislipidemias en esta población. La información necesaria se recolectó sobre la base de la normativa y los criterios del Ministerio de Salud y como resultados generales de las concentraciones plasmáticas promedio y la prevalencia de dislipidemias figuraron: colesterol total de 169,20 ±26,36 mg/dL y 8,2 %; lipoproteínas de baja densidad de 89,93 ±23,31 mg/dL y 4,5 %; triglicéridos de 145,89 ±48,96 mg/dL y 53,0 %; y lipoproteínas de alta densidad de 50 ±8,87 mg/dL y 28,3 %. Las cifras fueron inferiores en el grupo etario de 15-24 años y en personas de ascendencia pehuenche, con una pobre asociación a sobrepeso u obesidad abdominal; en general, resultaron menores a las de los citadinos.


A descriptive and cross-sectional study was carried out in 400 people (over 15 years) from Pehuenche communities of the Chilean Alto Biobio, from May to October 2011, in order to determine the prevalence of dyslipidemias in this population. Necessary information was collected on the basis of regulations and criteria of the Ministry of Health, and as general results of average plasma levels and prevalence of dyslipìdemia were: total cholesterol 169.20 ± 26.36 mg/dL and 8.2%; low-density lipoproteins 89.93 ± 23.31 mg/dL and 4.5%; triglycerides 145.89 ± 48.96 mg/dL and 53.0%; and high-density lipoproteins 50 ±8.87 mg/dL and 28.3%. The values were lower in the age group of 15-24 years and in Pehuenche people with poor association with abdominal obesity or overweight; in general, they were lower than those of the city people.

ABSTRACT

Menopause is the permanent cessation of mensturation due to loss of ovarian follicular function, which results in decreased production of estradiol and other hormones. Decreased levels of estrogen leads to variations in lipid profile leading to atherosclerosis thus by increasing risk of coronary artery disease in post menopausal women. The present study was conducted to evaluate the relationship between serum estradiol and lipid profile status in postmenopausal women to assess risk for coronary artery disease. Study includes 70 subjects, 30 healthy premenopausal women (25-40yrs) and the healthy postmenopausal women in the age group of (55 – 65yrs).Serum estradiol and lipid profile status were estimated in both cases and controls. Reduced estradiol (P0.0001) in post menopausal women and p value is highly significant for total cholesterol, triglycerides, low density lipoproteins. Conclusion: Thus the study outlines that the low levels of estrogen exerts a significant effect on the plasma lipids and lipoprotein metabolism. The risk of coronary artery disease increases in women after menopause.

La aterosclerosis es una enfermedad vascular crónica que afecta las bifurcaciones de las grandes arterias. Las enfermedades cardiovasculares asociadas a esta afección representan la primera causa de mortalidad en el mundo occidental. La fisiopatología de la aterosclerosis interrelaciona una serie de fenómenos moleculares y celulares complejos que aún no están del todo esclarecidos. Sin embargo, se han postulado algunas hipótesis para dar explicación a los procesos patogénicos que tienen lugar durante la aterogénesis. El objetivo del presente trabajo es ofrecer una actualización sobre los principales cambios moleculares que ocurren durante el inicio y progreso de esta enfermedad, así como las alternativas terapéuticas para su tratamiento y control. Para ello se realizó una revisión de publicaciones científicas en la base de datos MEDLINE durante los últimos 10 años. Se profundizó en la complejidad de dicha afección y se demostró que aún no existe una terapia totalmente eficaz para su tratamiento.


Atherosclerosis is a chronic vascular disease which affects bifurcations of major arteries. Atherosclerosis-associated cardiovascular diseases represent the first cause of mortality in the western world. The physiopathology of atherosclerosis associates a variety of molecular and cellular complex events, which are not completely understood. However, some hypothesis have been postulated to explain the pathogenic events during atherogenesis. The objective of the present article is to offer an updating on the principal molecular events during the atherosclerosis development and therapeutic alternatives for its treatment and control. To this end, a literature review was made in MEDLINE database, which covered the scientific publications of the last 10 years. The complexity of this illness was analyzed in depth and it was demonstrated that there was not any completely effective therapy to treat it.

Tomando en cuenta que aún no existe una metodología estándar de rutina para la determinación del colesterol de lipoproteínas de baja densidad (LDL-c) se decidió evaluar su determinación analítica utilizando tres técnicas: determinación enzimática homogénea, precipitación con sulfato de polivinilo y fórmula de Friedewald. Fueron procesadas 98 muestras de suero a las cuales se les determinó triglicéridos (TG), colesterol total (CT), colesterol de lipoproteínas de alta densidad (HDL-c) y colesterol de lipoproteínas de baja densidad (LDL-c). Los valores promedio de CT fueron 194,46 ± 43,54 mg/dL, HDL-c 51,12 ± 12,36 mg/dL y TG 132,88 ± 76,93 mg/dL. Aun cuando el análisis de regresión mostró una buena correlación entre los valores de LDL-c, los resultados indicaron una diferencia estadísticamente significativa en los mismos cuando los niveles de TG superaron los 200 mg/dL. La misma se observó principalmente entre el método de precipitación y la fórmula de Friedewald, siendo los valores significativamente más bajos en esta última (LDL-c por precipitación: 141,3 ± 26,2 mg/dL; LDL-c por fórmula de Friedewald: 110,1 ± 35,4 mg/dL). De la misma manera se vio afectada la proporción de individuos clasificados según su riesgo coronario. Es necesario comparar las técnicas aplicadas en este estudio con la cuantificación beta para evaluar cuál tiene un mayor nivel de exactitud.


Considering that there is still no standard methodology for routine determination of low density lipoprotein (LDL-c) it was decided to evaluate their analytical determination using three techniques: homogeneous enzymatic determination, polyvinyl sulphate precipitation and Friedewald formula. Ninety-eight serum samples were processed; triglycerides (TG), total cholesterol (TC), high-density lipoprotein (HDL-c) and LDL-c were determined. Mean total cholesterol was 194.46 ± 43.54 mg/dL, HDL-C was 51.12 ± 12.36 mg/dL and TG was 132.88 ± 76.93 mg/dL. Although regression analysis showed a good correlation between LDL-c, the results showed a statistically significative difference in them when TG levels exceeded 200 mg/dL. It was mainly observed in the precipitation method and the Friedewald formula, the latter values being significantly lower (LDL-C by precipitation: 141.3 ± 26.2 mg/dL, LDL-C by the Friedewald formula: 110, 1 ± 35.4 mg/dL). Moreover, this difference affected the proportion of individuals classified according to their coronary risk. It is necessary to compare the techniques applied in this study with beta quantification to assess which has a higher level of accuracy.


Levando em consideragao que ainda nao existe uma metodologia padrao de rotina para a determinagao do colesterol de lipoproteínas de baixa densidade (LDL-c) se decidiu avaliar sua determinagao analítica utilizando tres técnicas: determinagao enzimática homogénea, precipitagao com sulfato de polivinil e fórmula de Friedewald. Foram processadas 98 amostras de soro as quais lhes foi determinado triglicerídeos (TG), colesterol total (CT), colesterol de lipoproteínas de alta densidade (HDL-c) e colesterol de lipoproteínas de baixa densidade (LDL-c). Os valores médios de CT foram 194,46 ± 43,54 mg/dL, HDL-c 51,12 ± 12,36 mg/dL e TG 132,88 ± 76,93 mg/dL. Inclusive quando a análise de regressao mostrou uma boa correlagao entre os valores de LDL-c, os resultados indicaram uma diferenga estatisticamente significativa nos mesmos quando os niveis de TG superaram os 200 mg/dL. A mesma se observou principalmente entre o método de precipitagao e a fórmula de Friedewald, sendo os valores significativamente mais baixos nesta última (LDL-c por precipitagao: 141,3 ± 26,2 mg/dL; LDL-c por fórmula de Friedewald: 110,1 ± 35,4 mg/dL). Da mesma maneira se viu afetada a proporgao de indivíduos classificados conforme seu risco coronariano. É necessário comparar as técnicas aplicadas neste estudo com a quantificagao beta para avaliar qual é que tem maior nível de exatidao.

ABSTRACT

La deficiencia de la vitamina D está estimada hasta en 50 por ciento. Se ha encontrado relación entre niveles bajos de vitamina D y los componentes del síndrome metabólico: hipertensión, triglicéridos plasmáticos altos y defectos del metabolismo de la insulina, aunque causa solamente cambios mínimos en los niveles de las lipoproteínas de baja y alta densidad. Existe evidencia que la administración de vitamina D induce estabilización del endotelio y disminución de la inflamación arterial. Parece apropiado considerar un nivel bajo de vitamina D como un posible factor de riesgo cardiovascular pero es imprescindible sopesar el beneficio potencial de la terapia con suplemento de vitamina D, ya que no existen estudios aleatorizados con resultados definitivos
