ABSTRACT
Leukopenia is a common clinical disease with diverse pathogenesis. Leukopenia is mostly caused by drugs, especially anti-tumor drugs. For the treatment of leukopenia, Chinese materia medica can play an ability of comprehensive adjusting control from multi-angle, multi-dimension, and multi-target. As a good Chinese medicine for tonifying blood and replenishing qi, Lvjiao Buxue Granules has the advantages of high curative effect, convenient to use, and easy to be accepted by patients. It has been reported in clinical application for elevating leukocytes. This present review will focus on the research progress on the elevating leukocytes effects and mechanisms of Lvjiao Buxue Granules based on complicated Chinese herbal formula, herb-pair, and single herb, so as to provide a reference for expanding the clinical indications and rational use of the drug.

Objective To investigate the positive effect and possible mechanism of Lvjiao Buxue Granules on the leukopenia induced by cyclophosphamide in mice using 1H-NMR metabolomics approach. Methods Cyclophosphamide was used to establish the leucopenia mice, and the content of white blood cells, red blood cells, hemoglobin, platelets and other routine blood indexes were measured. The changes of endogenous metabolites in serum were analyzed by 1H-NMR metabolomics to investigate the regulation effect of Lvjiao Buxue Granules on serum endogenous metabolites in mice with leukopenia and the regulation mechanism of metabolic pathway. Results Compared with the control group, the contents of white blood cells, red blood cells, hemoglobin, and platelets in the model group were decreased significantly (P < 0.01), while the level of white blood cells and platelets in the treatment group were increased significantly (P < 0.05), and Lvjiao Buxue Granules can call back the level of potential differences metabolites. Conclusion Lvjiao Buxue Granules can improve leukopenia by regulating the immune function, energy metabolism, and amino acid metabolism.

Objective: To explore the change of endogenous metabolites in serum of rats with erythronoclastic anemia and the intervention of Lvjiao Buxue Granules by using 1H-NMR metabonomics coupled with multivariate statistical analysis. Methods: Acetylphenylhydrazine was used to establish the rat model of erythronoclastic anemia and after a week the rats except control and model groups were given to Lvjiao Buxue Granules (8 g/kg) separately once daily for two weeks. While the rats in the control and model groups were ig given equivalent distilled water respectively. The endogenous metabolites in serum from all of rats were analyzed by 1H-NMR coupled with multivariate statistical analysis. Results: Compared with the control group, the levels of lipids, lactic acid, and acetone in serum of rats in the model group increased while the levels of alanine, valine, creatinine, phosphocholine, glycerophosphocholine, oxidation of three methyl ammonium, glycine, and arginine were decreased. The levels of these endogenous metabolites with significant difference were reversed to normal by ig asminstration of Lvjiao Buxue Granules. Conclusion: The mechanism of Lvjiao Buxue Granules on tonifying blood is involved in the pathway of energy metabolomic, lipid metabolism, and intestinal bacteria metabolism.
