ABSTRACT

Objective To investigate the expression of MAD2L1 in lung adenocarcinoma and its effect on the prognosis and immune microenvironment of patients. Methods The difference of MAD2L1 expression in lung adenocarcinoma tissue and normal lung tissue was analyzed by TCGA and GEO database. Survival analysis was carried out to evaluate the prognostic significance of MAD2L1 gene expression in lung adenocarcinoma patients. StarBase database was used to construct miRNA-MAD2L1 regulatory network of lung adenocarcinoma. The relation between the expression of MAD2L1 and immune cell infiltration in lung adenocarcinoma was analyzed by TIMER database. Results The expression of MAD2L1 was up-regulated in lung adenocarcinoma, and the high expression of MAD2L1 was significantly correlated with pathological stage and lymph node metastasis of lung adenocarcinoma. The patients with high expression of MAD2L1 had a poor prognosis. miR-101-3p/MAD2L1 axis was identified as the most potential upstream regulation pathway of MAD2L1 in lung adenocarcinoma. The expression level of MAD2L1 was significantly correlated with tumor immune cell infiltration and immune checkpoint expression. Conclusions MAD2L1 is highly expressed in lung adenocarcinoma, which is related to poor prognosis and tumor immune infiltration. MAD2L1 can be used as a potential target for the treatment of lung adenocarcinoma.

ABSTRACT

Objective To investigate the effect of SP600125 on the proliferation, cell cycle, apoptosis and invasion of human cervical cancer HeLa cells. Methods CCK-8 method was used to detect the proliferation of HeLa cells treated with different concentrations of SP600125 at different time points. The 20 μmol/L of SP600125 was determined for subsequent experiments. Cell proliferation ability was detected using plate clone formation assay; nuclear morphology was observed by DAPI staining; cell cycle and apoptosis were measured by flow cytometry; cell migration and invasion were detected by cell scratch and Transwell methods; the mRNA and protein levels of p53, Mad2L1 and CDC20 were measured by qRT-PCR and Western blot after SP600125 treatment at different time points. Results Compared with control group (0.1%DMSO), cells proliferative activity were reduced by 10, 20, 30, 40 and 50 μmol/L SP600125 treatment for 24h. Compared with control group, the rate of apoptosis was significantly increased in SP600125 treatment groups, and the cell proportion in G2/M phase increased (P < 0.001), while the cell proportion in G0/G1 phases cells was reduced after SP600125 treatment for 24h and 48h (P < 0.001), and the clonal number, migration and invasion ability of HeLa cells also decreased significantly (P < 0.001). qRT-PCR and Western blot results showed a significant decrease in Mad2L1 mRNA and protein expression (P < 0.05) and a significant increase in p53 and CDC20 mRNA and protein expression (P < 0.01). Conclusion SP600125 can induce cell cycle arrest of cervical cancer HeLa cells in G2/M phase by upregulating p53 and CDC20 and downregulating Mad2L1 expression, and promote cell apoptosis to inhibit cell proliferation, migration and invasion.

Resumen El artículo analiza la emergencia de los Estudios Locos como tradición investigativa que propone teorías críticas y metodologías alternativas en la producción de saberes desde las personas que han sido etiquetadas con diagnósticos psiquiátricos. Para los Estudios Locos, los significados en torno a la locura expresan la relación de fuerzas y las formas de opresión que constituyen el campo de la salud mental, por lo tanto, enfatizan el carácter político de las investigaciones que se desarrollan en este ámbito y sostienen un cuestionamiento a la pretensión de objetividad y neutralidad del modelo biomédico. En este marco, mediante una revisión de la literatura académica y la netnografía como método de investigación, se describe el creciente protagonismo de las comunidades que han recibido atención de salud mental en América Latina para expresar sus narrativas en la esfera pública, situándose como agentes de conocimientos y actores políticos. En torno a estas nuevas gramáticas, los Estudios Locos Latinoamericanos se constituyen como una perspectiva de investigación hacia la configuración de otros modos de habitar la locura y articular sus luchas de emancipación social en el escenario regional contemporáneo.


Resumo Este artigo analisa a emergência dos Estudos loucos como uma tradição investigativa que propõe teorias críticas e metodologias alternativas para a produção do conhecimento de pessoas que foram rotuladas com diagnósticos psiquiátricos. Para Estudos loucos, os significados em torno da loucura expressam a relação de forças e as formas de opressão que constituem o campo da saúde mental, portanto, enfatizam o caráter político das pesquisas que se desenvolvem neste campo e sustentam um questionamento da reivindicação de objetividade e neutralidade do modelo biomédico. Nesse quadro, por meio de uma revisão da literatura acadêmica e da netnografia como método de pesquisa, descreve-se o crescente papel das comunidades que receberam atenção em saúde mental na América Latina para elaborar suas experiências no campo acadêmico e expressar suas narrativas em público. esfera, posicionando-se como agentes do conhecimento e atores políticos. Em torno dessas novas gramáticas, propõe-se o surgimento dos Estudos loucos da América Latina como disciplina emergente na configuração de outras formas de habitar a loucura e articular suas lutas pela emancipação social no cenário regional contemporâneo.


Abstract This article analyzes the emergence of Mad Studies as an investigative tradition that proposes critical theories and alternative methodologies in the production of knowledge from people who have been labeled with psychiatric diagnoses. For Mad Studies, the meanings around Madness express the relationship of forces and the forms of oppression that constitute the field of mental health, therefore, they emphasize the political nature of the research that is developed in this field and sustain a questioning of the claim of objectivity and neutrality of the biomedical model. In this framework, through a review of the academic literature and netnography as a research method, the growing role of the communities that have received mental health care in Latin America to express their narratives in the public sphere is described, positioning themselves as agents of knowledge and political actors. Around these new grammars, Latin American Mad Studies are constituted as a research perspective towards the configuration of other ways of inhabiting madness and articulating their struggles for social emancipation in the contemporary regional scenario.

ABSTRACT

Abstract This study assessed mad honey use in alternative treatments. The universe of this descriptive study was patients admitted to the pulmonary disease clinic located in the Ordu province of the Black Sea region between 15 December 2014 and 15 February 2015. We did not use a sampling method and patients who agreed to participate were included in the study (n=353). In order to collect the data, we used a questionnaire prepared by the researchers. In this study, 77% of the participants stated that mad honey was beneficial to health, 44.5% used mad honey, and 53.5% consumed it daily. Furthermore, 28.7% used mad honey for asthma, 6.4% for cough, 12.1% for gastrointestinal diseases, and 3.2% for hypertension. There was a significant relationship between the gender, family structure, age, and chronic disease status of participants and the status of consuming mad honey (p<0.05). In this study, participants used mad honey for asthma, gastrointestinal diseases, hypertension, and cough. Health professionals should provide training and counseling on the health effects and risks of mad honey to improve public health.

Throughout the next few lines, this research will seek to bring reality and theory together, trying to establish more organic relations with the way of producing scientific knowledge about the field of collective mental health, taking as a possible bridge to this, the insurgency. The choice of the theme (insanity), the problem of research (insurgencies in Parade), the methodology (cartography), gives clues that the movement, in the composition with the street, offers a (re)discover with body, art and culture, as devices of production of resistance to a prescribed life.

Filamin A and 14-3-3-σ are closely associated with the development of breast cancer.However,the exact relationship between them is still unknown.The present study aimed to examine the interaction of filamin A with 14-3-3-σ in the invasion and migration of breast cancer.RNA interference technology was employed to silence filamin A in MDA-MB-231 cells.Real-time PCR and Western blotting were used to detect the expression of filamin A and 14-3-3-σ at mRNA and protein levels,respectively.Double immunofluorescence was applied to show their colocalization morphologically.Wound healing assay and Trans-well assay were used to testify the migration and invasion of MDA-MB-231 cells in filamin A-silenced cells.The results showed that silencing filamin A significantly increased the mRNA and protein levels of 14-3-3σ.In addition,double immunofluorescence displayed that filamin A and 14-3-3σ were predominantly colocalized in the cytoplasm of MDA-MB-231 cells.Silencing filamin A led to the enhanced fluorescence of 14-3-3σ.Furthermore,cell functional experiments showed that silencing filamin A inhibited the migration and invasion of MDA-MB-231 cells in vitro.In conclusion,silencing filamin A may inhibit the invasion and migration of breast cancer cells by upregulating 14-3-3σ.

ABSTRACT

Objective:To explore combined detection of mad2 with anti-nuclear mitotic spindle apparatus antibody(MSA)and anti-centromere antibody(ACA)and their clinical value for the diagnosis of small cell lung cancer(SCLC).Methods:One hundred and twen-ty SCLC patients,110 non-small cell lung cancer(NSCLC)patients,and 115 pulmonary nodule(PN)patients were enrolled in this study. The expression of mad2 was analyzed by qt-PCR.MSA and ACA were detected by indirect immunofluorescence(IIF)staining.Results:mad2 was overexpressed in SCLC and NSCLC samples(P<0.05).There were significant differences between the results obtained for SCLC and NSCLC samples by qt-PCR(P<0.05).AUC in ROC curve for mad2 expression was 0.799 with an intermediate diagnostic value. In the correlative analysis,the odds ratio of MSA and ACA was 6.94 and 5.60,respectively.In the correlation analysis,Kappa value of mad2 with MSA was 0.49,and Kappa value of mad2 with ACA was 0.42.In the parallel analysis,the sensitivity and specificity was 83.31% and 79.34%,respectively,while the Youden Index was 0.62.Moreover,in the serial analysis,the sensitivity and specificity was 65.32% and 93.35%,respectively,and the Youden Index was 0.59.Conclusions:In comparison with the NSCLC and PN samples,mad2 was overexpressed in SCLC samples.Therefore,mad2 ought to play a critical role in the pathology of SCLC.The combined expression of mad2 with MSA and ACA may contribute to enhancing the sensitivity and specificity of detection;this expression may allow early diag-nosis and clinical diagnosis of SLCC and may be a promising treatment for SCLC.

ABSTRACT

Objective To establish a method for specific gene editing of the second exon of mouse MAD2L1 gene by CRISPR/Cas9, and analyze its off-target effect. Methods The gene editing site for MAD2L1 gene was designed by CHOP?CHOP, and the Cas9?MAD2L1 vector was constructed based on the designed editing site. Cas9?MAD2L1 was then transfected into NIH/3T3 cells and screened with puromycin, followed by observing GFP expression using fluorescence microscopy. The genomic DNA from transfected cells was extracted and a partial fragment of MAD2L1 gene was amplified by PCR. T7E1 analy?sis and Sangger sequencing were used for gene editing and off?target analysis. Results After Cas9?MAD2L1 transfection and puromycin screening, a large number of GFP?expressing cells were observed under the fluorescence microscope. Combined the PCR result with TE71 analysis, the amplified 228 bp PCR products can be digested into 166 bp and 62 bp fragments. The se?quencing result showed that the second exon of MAD2L1 gene was successfully edited, and the off?target effect was undetected in our system. Conclusions The method for specific gene editing of the second exon of mouse MAD2L1 gene by CRISPR/Cas9 is successfully established, and off?target effect of MAD2L1 gene is not detected.

The spindle assembly checkpoint ( SAC) is an important monitoring mechanism to monitor the connection between centromeres and microtubules and to ensure proper chromosome separation in human .Mitotic arrest defective protein(Mad)family,as an important part of SAC,plays a crucial role in the process of mitosis. Mutations or altered expressions of Mad may lead to abnormal separations of chromosomes and play a partial role in tumorigenesis ,poor prognosis and chemotherapy drug resistance .

ABSTRACT

PURPOSE: The aim of this study was to evaluate the clinical characteristics and outcome of patients who presented to the emergency department (ED) with cardiotoxicity caused by ingestion of Himalayan mad honey. METHODS: Medical records of 12 patients who presented to the ED from January 1, 2005 to December 31, 2012 with cardiotoxicity caused by ingestion of Himalayan mad honey were retrospectively reviewed. RESULTS: The mean age of patients was 54.5 years and 58.3% were men. The median amount of mad honey ingested was 30.0 cc, and the mean time from ingestion to onset of symptoms was 39.4 minutes. All patients had hypotension and bradycardia upon arrival in the ED. The initial electrocardiogram showed sinus bradycardia in seven patients, junctional bradycardia in four patients, and atrial fibrillation with slow ventricular response in one patient. Four patients were treated with intravenous normal saline solution only. Eight patients were treated with intravenous normal saline solution and atropine sulfate in a dose ranging from 0.5 to 2.0 mg. Blood pressure and pulse rate returned to normal limits within 24 hours in all patients. CONCLUSION: Our study showed that all patients with cardiotoxicity caused by ingestion of Himalayan mad honey had severe hypotension, bradycardia, and bradyarrythmias, including sinus bradycardia and junctional bradycardia and all patients responded well to conservative treatment, including intravenous normal saline solution and intravenous atropine sulfate.

ABSTRACT

BACKGROUND: Mitotic arrest deficiency protein 2 (MAD2) is a key component of spindle assembly checkpoint function, which mediates cell apoptosis through microtubule kinetics. Aberrant expression of MAD2 is believed to be associated with the development of chromosome instability. MAD2 also has a signihicant role in cellular drug resistance to taxane chemotherapeutic agents. METHODS: Expression of MAD2 and p53 was investigated using immunohistochemistry in 85 cases of ovarian carcinomas. Clinicopathological data including progression-free survival were analyzed. RESULTS: A significant (p=.035) association was observed between the grade of serous carcinoma and the expression level of MAD2. While low-grade serous carcinoma showed a low-level expression of MAD2, high-grade serous carcinoma showed a high-level expression of MAD2. We also determined that low-level expression of MAD2 was associated with reduced progression-free survival (PFS) (p=.016) in high-grade serous carcinoma. CONCLUSIONS: MAD2 expression in ovarian carcinoma is related to the grade of serous carcinoma and PFS of high-grade serous carcinoma. Expression level of MAD2 detected by immunohistochemistry may serve as an indicator in predicting the response of microtubule-interfering chemotherapeutic agents.

ABSTRACT

Honey is produced by bees from nectar collected from nearby flowers. Sometimes, honey produced from the Rhododendron species is contaminated by Grayanotoxin (GTX) in Nepal and other countries. There have been reports of GTX intoxication, also known as 'mad honey disease', from honey produced in countries other than Korea. The importation of wild honey has been prohibited by the Korean Food and Drug Administration since 2005, yet it is still distributed within Korea by the occasional tourist. We report a case of GTX intoxication from contaminated honey which included the symptoms of nausea, vomiting, general weakness, dizziness, blurred vision, hypotension and sinus bradycardia. By means of infusion with normal saline and atropine sulfate, the patient's condition fully recovered within 8 hours of hospital admission, and she was discharged without any complications.

ABSTRACT

Prion diseases are known as transmissible spongiform encephalopathies (TSE), a group of rare, rapidly progressive, and fatal neurologic diseases. The agents responsible for human and animal prion diseases are abnormal proteins (prion or proteinaceous infectious particle) that can trigger chain reactions causing normal proteins in the brain to change to the abnormal protein. These abnormal proteins are resistant to enzymatic breakdown, and they accumulate in the brain, leading to damage. All have long incubation periods followed by chronic neurological disease and fatal outcomes, have similar pathology limited to the CNS include convulsions, dementia, ataxia (balance and coordination dysfunction), and behavioral changes, and are experimentally transmissible to some other species.

ABSTRACT

O artigo resulta de uma pesquisa que objetivou compreender os sentidos sobre a loucura e o louco que atravessam canções brasileiras contemporâneas. As considerações de Michael Foucault sobre a História da Loucura e as de Mary Jane Spink sobre práticas discursivas e a produção de sentidos formaram as bases teóricas do estudo. A metodologia consistiu na realização de uma cartografia da circulação de sentidos sobre o louco e a loucura em trinta canções reunidas pelo motor de busca Google. Os resultados da cartografia indicam a polifonia do discurso literomusical e apontam uma paisagem discursiva heterogênea onde se destacam cinco zonas de sentido sobre a loucura e o louco. Conclui-se que estes signos estão em disputa no dia-a-dia, sendo que alguns sentidos circulantes nas canções remetem a modos instituídos de lidar com essa situação, enquanto outros podem constituir vetores de subjetivações fugidias às normalizações cotidianas. Assim, ao realçar algumas conformações simbólicas que perpassam o imaginário social, o artigo chama a atenção para a relevância de o campo multidisciplinar da saúde mental escutar as vozes sociais sobre as várias loucuras, perscrutando suas condições de produção e suas possíveis ressonâncias.


This article is the result of research associated with the representation of madness and the insane in contemporary Brazilian songs. Michel Foucault's considerations about the history of madness and those of Mary Jane Spink about discursive practices and the production of meaning formed the theoretical base for the study. The methodology consisted in mapping the circulation of words and meanings about madness in thirty songs collected by Google's search engine. The results of the mapping reveal the polyphony of the literary/musical discourse, and a heterogeneous discursive panorama in which five zones of meaning about madness and the insane stand out. Thus, it can be concluded that these signs are prevalent on a day-to-day basis, Some of the meanings in the songs refer to institutionalized ways of dealing with madness, others constitute modes of subjectivity that flee from routine treatment. By delineating symbolic formations that permeate social imagery, this article brings systematic attention to popular representations of various forms of madness of relevance to multidisciplinary fields in mental health, discussing their appearance in popular music and their possible repercussions.

ABSTRACT

PURPOSE: The purpose of this study was to compare the marginal fit of three-unit zirconia fixed dental prostheses (FDPs) fabricated using CAD/CAM and MAD/MAM system. MATERIALS AND METHODS: Dentiform maxillary central and lateral incisor were prepared for 3-unit FDP and fixed in yellow stone. This model was duplicated to epoxy resin die. On the resin die, fifteen 3-unit FDPs were fabricated. Metal-ceramic group was three-unit metal-ceramic FDPs, Everest(R) group was zirconia three-unit FDPs fabricated using the Everest(R) system (Kavo Dental GmbH, Biberach, Germany) and Rainbow(TM) group was zirconia three-unit FDPs fabricated using the Rainbow(TM) system (Dentium Co. Inc., Seoul, South Korea). They were cemented to resin dies with adhesive resin cement. After removing pontics, each retainers were separated and observed under measuring machine (Presize 440C) and analyzed through one-way ANOVA and Duncan test (alpha= .05). RESULTS: Mean values and standard deviations of marginal gap dimensions in each group for three-unit FDPs were 78.5 +/- 11.05 microm for the metal-ceramic group, 59.30 +/- 11.63 microm for the Everest(R) group and 70.34 +/- 13.98 microm for the Rainbow(TM) group. CONCLUSION: 1. The Everest(R) group in comparison with metal-ceramic group showed better marginal fit, which had significant differences (P.05). 3. The mean marginal gap values between Rainbow(TM) group and metal-ceramic group did not showed significant differences (P>.05). 4. The mean marginal gaps of each group were within clinically acceptable range (120 microm).

ABSTRACT

In this article, the status of spindle assembly checkpoint and the alteration of its major component, Mad2 protein level were examined in A2780 and SKOV3 ovarian cancer cell lines. Recombinant eukaryotic expression plasmid pEGFP-Mad2 was transfected into paclitaxel-resistant SKOV3 cells and Mad2 protein was knocked down by Mad2-specific siRNA in paclitaxel-sensitive A2780 cells. Then the expression level of Mad2 gene was detected by Western blotting. Flow cytometry revealed that SKOV3 cells were not fully arrested in G2/M phase in contrast to A2780 cells in the presence of paclitaxel. However, paclitaxel sensitivity assay showed that sensitivity to paclitaxel was reversed after the transfection in both cell lines in terms of number of cells arrested at G2/M phase and the expression of Bcl-2 was significantly changed. These results suggest that weakened spindle checkpoint with reduced expression of Mad2 is associated with resistance to paclitaxel in ovarian cells and Bcl-2 may be involved in this process.

ABSTRACT

PURPOSE: The purpose of this study was to evaluate the fit of zirconia core using MAD/MAM system comparing to that of conventional metal-ceramic and CAD/CAM system. MATERIALS AND METHODS: Duplicating the prepared resin tooth, 50 improved stone dies were fabricated. These dies are classified as a group of 5 to create the core. The groups were composed of metal-ceramic, Cercon(R), Ceramill(R), Rainbow(TM), and Zirkonzhan(R). Each core was cemented to stone die, and then, absolute marginal discrepancy was measured with microscope at a magnification of x50. Statistical analysis was done with one-way ANOVA test and Tukey's HSD test. RESULTS: The mean absolute marginal discrepancy for metal-ceramic was 51.97 +/- 23.38 micrometer, for Cercon(R) was 62.16 +/- 25.88 micrometer, for Ceramill(R) was 67.64 +/- 40.38 micrometer, for Rainbow(TM) was 125.07 +/- 42.19 micrometer, and for Zirkonzhan(R) was 105 +/- 44.61 micrometer. CONCLUSION: 1. Fit of margin was identified as in the order of metal-ceramic, Cercon(R), Ceramill(R), Zirkonzhan(R), and Rainbow(TM). 2. Absolute marginal discrepancy of the zirconia core that designed by MAD/MAM system had significant differences in order of Ceramill(R), Zirkonzhan(R), and Rainbow(TM). 3. The mean absolute marginal discrepancy between Cercon(R) and Ceramill(R) did not show significant differences.
