ABSTRACT

To investigate the effect of Yunkang Oral Liquid on preventing lipopolysaccharide(LPS)-induced abortion and regulating immune tolerance in mice, sixty normal ICR mice were randomly divided into normal group, model group,Yunkang Oral Liquid high, middle and low dose groups and progesterone group.Abortion model was established by tail vain injection of LPS(0.1μg/mouse)on the 7th day of pregnancy.Since the first day of pregnancy, the same volume of distilled water,Yunkang Oral Liquid at the dose of 36,18 and 9 m L·kg~(-1)·d~(-1),and progesterone at the dose of 0.038 g·kg~(-1)·d~(-1)were given in corresponding groups. The mice were sacrificed at the 9th day of pregnancy, and the embryo loss of each group was calculated. The levels of Th1 type cytokines(TNF-α,IFN-γ)and Th2 type cytokines(IL-4,IL-10)in uterus homogenate were detected by ELISA.HE staining was performed to examine the histopathological changes in the decidua. The expression levels of CD14,F4/80 in macrophages of uterus were detected by immunohistochemistry.Western blot was used to investigate the protein expression of TLR4,MyD88 and NF-κB in uterine decidua. In our study, all Yunkang Oral Liquid groups could significantly reduce the embryo absorption rate of mice, while high dose group can significantly increase the levels of IL-10 and IL-4;both medium and high dose groups can significantly decrease TNF-α,and IFN-γlevelsin the uterus of model mice, reduce the protein expression of NF-κB,MyD88 and TLR4 in uterine decidua tissue.Various treatment groups could reduce the counts of F4/80,CD14 macrophages and decrease expression area in uterine tissue. Our results showed that Yunkang Oral Liquid could prevent LPS-induced abortion, and the mechanism may be associated with inhibiting the TLR4/MyD88/NF-κB signaling pathway and regulating the balance of Th1/Th2 immune factors, which could improve the endometrial receptivity of mice, and promote the development of decidua and implantation of embryo.

ABSTRACT

Objective:To study the mechanism of kidney-replenishing herb in regulating the tolerance of fetal-maternal interface and the effect of IDO on modulating the phenotype of decidual NK cells. Methods: Indoleamine-2,3-dioxygenase ( IDO) expression in villus tissues from normal pregnancies and recurrent spontaneous abortion (RSA) patients was monitored by immunohisto-chemistry (IHC). IDO expression in HTR-8/Svneo cells treated with control or herb serum medium was detected by flow cytometry (FCM). The influence of kidney-replenishing herb on modulating the phenotype of NK cells,CD16 and CD56 expression in peripheral NK cells co-cultured with control or herb serum medium in the presence or absence of 1-methyltryptophan (1-MT) treated HTR-8/Svneo cells were measured by FCM. Results: IDO expression was decreased in villus tissue of RSA patients compared to normal preg-nancies. Herb medium could increase the IDO expression of HTR-8/Svneo. Kidney-replenishing herb could enhance the regulatory function of trophoblasts on NK cells and further induce the immune tolerance at fetal-maternal interface in IDO dependent and independent manners. Conclusion: Kidney-replenishing herb can modulate the phenotype of peripheral NK cell by up-regulating IDO expression in trophoblasts and play a role in the treatment of RSA patients.

ABSTRACT

Objective:To evaluate the quantity and function changes of dendritic cells ( DC) in peripheral blood of patients with repeated implantation failure ( RIF).Methods:30 patients with RIF and 15 normal controls were enrolled in this study, and the peripheral blood was collected during the mid-luteal phase. The percentage of DC subsets and the expression levels of functional molecules were assessed by flow cytometric analysis. Results:Compared with normal controls, the percentage of lin-HLA-DR+DC cells accounting for leukocytes in patients with RIF was not significantly different ( P>0.05).There were also no significant differences in the expression levels of co-stimulatory molecules ( CD80 and CD86) and immune tolerant molecules CD200 on DC cells surfaces between patients with RIF and normal controls ( all P>0.05).In addition, the percentage of CD11c+CD123-mDC accounting for DC cells was significantly increased in patients with RIF (P0.05). Conclusion:The percentage of mDC accounting for DC cells was significantly increased in patients with RIF, which may be one of factors affecting pregnancy outcomes.
