ABSTRACT

Objective To investigate the diagnostic value of Bcl-2, miR-451 and Th17 cells in esophageal cancer and their relation with recurrence. Methods We selected 101 patients with esophageal cancer as the experimental group and 95 healthy patients as the control group. The correlation between the clinicopathological characteristics and the level of each peripheral blood index was analyzed. The ROC curve was used to analyze the diagnostic value of each peripheral blood index. Multiple linear regression analysis was used to analyze the relation between each index and tumor recurrence. Results Peripheral blood Bcl-2, miR-451 and Th17 cells in the experimental group were higher than those in the control group (all P < 0.05); differentiation degree, clinical stage, tumor diameter and lymph node metastasis were positively correlated with the levels of Bcl-2, miR-451, and Th17 cells (all P < 0.05). Bcl-2, miR-451 and Th17 cells in relapsed patients were higher than those in non-relapsed patients (all P < 0.05); after controlling other factors such as differentiation degree, clinical stage, tumor size and lymph node metastasis, the levels of Bcl-2, miR-451 and Th17 cells were significantly correlated with recurrence (all P < 0.05). Conclusion Bcl-2, miR-451 and Th17 cells in peripheral blood of esophageal cancer patients are abnormally expressed and their expression are closely related to differentiation degree, clinical stage and recurrence. The combined detection could provide an objective basis for clinical diagnosis and prognosis assessment.

ABSTRACT

AIM:To investigate the effect of microRNA(miR)-451 by targeting proteasome subunit βtype 8 (Psmb8)on the inflammatory responses in mouse glomerular mesangial cells(MCs)under high-and low-glucose condi-tions.METHODS:The expression levels of miR-451,IL-18 mRNA and TNF-αmRNA were detected by qPCR.The pro-tein expression levels of IL-18,TNF-αand Psmb8 were determined by Western blot when miR-451 was over-expressed and down-expressed in the MCs.Moreover, the expression of IL-18 and TNF-αwas detected when Psmb8 was silenced by si-Psmb8 in MCs.RESULTS:The expression of miR-451 was significantly decreased in the MCs treated with high glucose compared with low glucose group(P<0.01).However,the expression of Psmb8 was increased in high glucose group as compared with low glucose group(P<0.01).Moreover, the expression levels of Psmb8, IL-18 and TNF-αwere signifi-cantly decreased when miR-451 was over-expressed in high glucose group(P<0.01).Additionally,the expression levels of IL-18 and TNF-αwere significantly reduced when Psmb8 was silenced in the MCs under high glucose condition.CON-CLUSION:miR-451 reduces the expression of inflammatory factors via targeting Psmb8 in the MCs under high glucose condition.Therefore,miR-451 may play a role in inflammation of diabetic nephropathy.

ABSTRACT

Aberrant expression of microRNAs (miRNAs) has been shown to be involved in early observations of depression. The aim of this study was to determine if serum levels of miRNA-451a, miRNA-34a-5p, and miRNA-221-3p can serve as indicators of disease progression or therapeutic efficacy in depression. We collected data from 84 depressed patients and 78 control volunteers recruited from the medical staff at the West China Hospital. Depression severity was rated using the 24-item Hamilton Depression Scale (HAMD). Serum miRNA-451a, miRNA-34a-5p, and miRNA-221-3p levels were determined in samples from the depressed patients before and 8 weeks after antidepressant treatment as well as in samples from controls. Compared with the controls, the patients had lower miRNA-451a levels, higher miRNA-34a-5p and miRNA-221-3p levels, and increased HAMD scores whether they underwent antidepressant treatment or not. Eight weeks after antidepressant treatment, the patients exhibited increased miRNA-451a levels, decreased miRNA-34a-5p and miRNA-221-3p levels, and reduced HAMD scores. The serum level of miRNA-451a was negatively correlated with HAMD scores of the patients, while the serum levels of miRNA-34a-5p and miRNA-221-3p were positively correlated with HAMD scores whether the patients underwent antidepressant treatment or not. Paroxetine was markedly effective in 50 patients who also displayed an increased level of miRNA-451a but reduced levels of miRNA-34a-5p and miRNA-221-3p. In contrast, paroxetine was moderately effective or ineffective in 34 patients. In conclusion, depressed patients had lower serum miRNA-451a but higher serum miRNA-34a-5p and miRNA-221-3p, and these miRNAs are potential predictors of the efficacy of antidepressants.

ABSTRACT

Objective To explore the expression level and clinical significance of microRNA‐451 (miR‐451) in the plasma of patients with diffuse large B lymphoma (DLBCL) .Methods We enrolled 27 patients with DLBCL and 27 healthy controls for the study .The plasma level of miR‐451 was detected by reverse transcription polymerase chain reaction (RT‐PCR) .Then we analyzed the relationship between the plasma level of miR‐451 and pathological characteristics of DLBCL .Results The plasma level of miR‐451 in the patients with DLBCL was significantly lower than in healthy controls [(0 .41 ± 0 .15) vs .(0 .64 ± 0 .21)] (P0 .05) ,but with Ann Arbor tumor stage and IPI score ( P< 0 .05 ) .Conclusion The plasma level of miR‐451 decreases in patients with DLBCL ,which indicates that miR‐451 may participate in the pathology of DLBCL .
