ABSTRACT

AIM To study the formation rules of baicalin-berberine complex.METHODS Under the conditions of different ratios of baicalin to berberine,heating methods and media (pH,ionic strength),the yield and composition of mixed decoction precipitation were determined by HPLC.RESULTS 0.5 g Baicalin and 1 g berberine produced the highest amount of precipitation.Higher heating temperature,nearly neutral pH (6.8) and glucose addition were beneficial to precipitate generation,in sharp contrast to NaCl addition.CONCLUSION The yield and composition of baicalin-berberine complex are mainly influenced by heating temperature and additives.

ABSTRACT

Objective To compare the content variation of catalpol between single and mixed decocting Rehmanniae Radix and Paeoniae Radix alba; To preliminarily explore the variation rules of compatibility of Rehmanniae Radix and Paeoniae Radix alba. Methods HPLC method was used with Waters Symmetry C18 column (4.6 mm×150 mm, 5 μm), flow rate of 1.0 mL/min, detection wavelength of 210 nm, column temperature at 25 ℃. The mobile phase was acetonitrile-0.01% phosphoric acid in water isocratic elution system. Results Catalpol regression equation was Y=270 054X-57 724, r=0.999 6, showing good linear range among 0.225–7.2 μg. The average recovery rate was 101.15%, RSD=0.39%. The average content of catalpol in the mixed decoction of Rehmanniae Radix and Paeoniae Radix alba was 18.66%more than the single decoction of Rehmanniae Radix. Conclusion The content of catalpol in mixed decoction is higher than the single decoction of Rehmanniae Radix and Paeoniae Radix alba. The mixed decoction is favorable for the dissolving of catalpol from the medicine. The compatibility of Rehmanniae Radix and Paeoniae Radix alba is rational.

ABSTRACT

Objective: To study the chemical constituents from the ethanol extract fraction herbs of Qibai Pingfei Granule (QPG), the difference of mixed decoction and separated decoction of ethanol extract fraction herbs in QPG. Methods: UPLC-Q-TOF-MS was used as the analytic method. The information on quasi-molecular ion, fragment ions, retention time, and references were synthetically analyzed to confirm the constituents in the ethanol extract fraction herbs of QPG. Principal components analysis was applied to process the acquired data, and marked differences between mixed decoction and separated decoction in the ethanol extract fraction herbs of QPG. Results: Twenty-two compounds were identified, and four compounds were tentatively identified among 26 ions. In addition, twelve compounds were determined as markers to distinguish the mixed decoction from the separated decoction of the ethanol extract fraction herbs in QPG. Conclusion: The findings obtained from the study can provide the useful information for the determination of bioactive substances, the perfection of quality standard, and the preparation technology of QPG.

ABSTRACT

Objective: To study the correlation of combinations in different proportions of Angelicae Sinensis Radix and Carthami Flos and explore the regularity in proportion of the two drugs compatibility. Methods: Data mining technology was applied to screening the proportions of Angelicae Sinensis Radix and Carthami Flos. The changes of the contents of ferulic acid, hydroxysafflor yellow A, chlorogenic acid, and coumaric acid in the single and mixed decoction of Angelicae Sinensis Radix and Carthami Flos were determined by HPLC. The experiments of thrombin time and scavenging DPPH were carried out to evaluate the pharmacological activities of compatibility between Angelicae Sinensis Radix and Carthami Flos. Results: The top 10 proportions of Angelicae Sinensis Radix and Carthami Flos were screening. The contents of ferulic acid, hydroxysafflor yellow A, chlorogenic acid, and coumaric acid were higher than the single decoction of Angelicae Sinensis Radix and Carthami Flos. Thrombin time was prolonged and the activity of scavenging DPPH was enhanced compared to the same drug dosage of the single decoction of Angelicae Sinensis Radix and Carthami Flos. Conclusion: Mixed decoction of Angelicae Sinensis Radix and Carthami Flos is rational and good for the stripping of aromatic acids from the drugs, which has some correlation with the change of activities of the compatibility between Angelicae Sinensis Radix and Carthami Flos.

ABSTRACT

AIM: To compare the contents of baicalin,paeoniflorin and glycyrrhetic acid in the seperated and mixed decoction of Huangqin Decoction(Radix Scutellariae,Radix Paeoniae alba,Radix et Rhizoma Glycyrrhizae and Fructus Jujubae). METHODS: The contents of baicalin,paeoniflorin and glycyrrhetic acid were analyzed by HPLC.Chromatographic conditions included: Hypersil BDS C_(18) column and the mobile phase consisted of a mixture of methanol-water-phosphoric acid(47(∶)53(∶)0.2),acetonitrile-water-phosphoric acid(18(∶)82(∶)0.1) and methanol-water-phosphate buffer solution(70(∶)29(∶)1).Baicalin,paeoniflorin and glycyrrhetic acid were detected at 280 nm,230 nm and 250 nm respectively. RESULTS: The linear range of baicalin,paeoniflorin and glycyrrhetic acid were 2.88—72.00 ?g/mL,2.85—22.80 ?g/mL and 4.16—52.00 ?g/mL respectively.The contents of baicalin in the mixed decoction was higher than that in the seperated decoction.The contents of paeoniflorin and glycyrrhetic acid in the mixed decoction were almost as same as that in seperated decoction. CONCLUSION: The contents of active ingredients in the mixed and seperated decoction are different.

ABSTRACT

OBJECTIVE:To study the infuence of mixed decoction of Cortex Phellodendri,Radix Glycyrrhizae and Ramu?lus Loranthi on extraction rate of berberine.METHODS:The extraction amount from Cortex Phellodendri,Radix Glycyrrhizae and Ramulus Loranthi mixed decoction was determined by HPLC.The HPLC conditions were:Hypersil BDS C 18 column;mobile phase,acetonitrile-33mmol/L KH 2 PO 4 -triethylamine(20∶72∶0.1);detecting wavelength,345nm;column temperature25℃;flow fate1.0ml/min.RESULTS:Decoction of Cortex Phellodendri,Radix Glycyrrhizae and Ramulus Loranthi in combi?nation obviously decreased the extraction rate of berberine.CONCLUSION:Cortex Phellodendri should be extracted separately in formulating extract technic for preparations containing above-mentioned herbs.
