ABSTRACT

OBJECTIVE@#To investigate the feasibility of a dual-crosslinked injectable hydrogel derived from acellular musclar matrix (AMM) for promoting myoblasts proliferation and myogenic differentiation.@*METHODS@#Firstly, hyaluronic acid was oxidized with NaIO 4 and methylated to prepare methacrylamidated oxidized hyaluronic acid (MOHA). Then, AMM obtained by washing enzymatically treated muscle tissue was aminolyzed to prepare aminated AMM (AAMM). MOHA hydrogel and AAMM were crosslinked using Schiff based reaction and UV radiation to prepare a dual-crosslinked MOHA/AAMM injectable hydrogel. Fourier transform infrared spectroscopy (FTIR) was used to characterize MOHA, AAMM, and MOHA/AAMM hydrogels. The injectability of MOHA/AAMM hydrogel were evaluated by manual injection, and the gelation performance was assessed by UV crosslinking. The rheological properties and Young's modulus of the hydrogel were examined through mechanical tests. The degradation rate of the hydrogel was assessed by immersing it in PBS. The active components of the hydrogel were verified using immunofluorescence staining and ELISA assay kits. The promotion of cell proliferation by the hydrogel was tested using live/dead staining and cell counting kit 8 (CCK-8) assays after co-culturing with C2C12 myoblasts for 9 days. The effect of the hydrogel on myogenic differentiation was evaluated by immunofluorescence staining and real time quantitative polymerase chain reaction (RT-qPCR).@*RESULTS@#FTIR spectra confirmed the successful preparation of MOHA/AAMM hydrogel. The hydrogel exhibited good injectability and gelation ability. Compared to MOHA hydrogel, MOHA/AAMM hydrogel exhibited higher viscosity and Young's modulus, a reduced degradation rate, and contained a higher amount of collagen (including collagen type Ⅰ and collagen type Ⅲ) as well as bioactive factors (including epidermal growth factor, fibroblast growth factor 2, vascular endothelial growth factor, and insulin-like growth factor 1). The live/dead cell staining and CCK-8 assay indicated that with prolonged incubation time, there was a significant increase in viable cells and a decrease in dead cells in the C2C12 myoblasts within the MOHA/AAMM hydrogel. Compared with MOHA hydrogel, the difference was significant at each time point ( P<0.05). Immunofluorescence staining and RT-qPCR analysis demonstrated that the deposition of IGF-1 and expression levels of myogenic-related genes (including Myogenin, Troponin T, and myosin heavy chain) in the MOHA/AAMM group were significantly higher than those in the MOHA group ( P<0.05).@*CONCLUSION@#The MOHA/AAMM hydrogel prepared based on AMM can promote myoblasts proliferation and myogenic differentiation, providing a novel dual-crosslinked injectable hydrogel for muscle tissue engineering.

ABSTRACT

RESUMEN Existe una variedad de tumores epiteliales, mesoteliales y de tejidos blandos que pueden desarrollarse en la red testicular y paratesticular. Se presenta el caso clínico de paciente con tumoración paratesticular derecha: Miofibroblastoma Mamario Paratesticular. Importante recalcar la variedad de origen embrionario para el desarrollo de estos tumores, así como el diagnóstico oportuno para el tratamiento y seguimiento resolutivo.


ABSTRACT There is a variety of epithelial, mesothelial, and soft tissue tumors that can develop in the testicular and paratesticular area. We present the case of a patient with a right paratesticular tumor that was diagnosed as paratesticular mammary myofibroblastoma. It is important to emphasize the embryonic origin variety for the development of these tumors, as well as having a timely diagnosis for treatment and decisive follow-up.

ABSTRACT

Objective To explore the influence of muscle and fat distribution on sit-up ability of female college students. Methods A total of 1560 female students majoring in non-physical education in a Guangxi University were randomly selected. Body composition was measured by the bioresistance antibody component meter. According to the National Physical Health Test Standard, the height, weight and sit-ups of female college students were measured. The students were divided into four groups according to the score of sit-ups:0, P<0.05). Compared with other parameters, the correlation coefficient between trunk muscle mass and sit-ups was the largest. Conclusion The distribution of fat and muscle has different effects on sit-up ability. Female college students' sit-up ability is greatly affected by subcutaneous fat and trunk muscle, and has nothing to do with visceral fat.

ABSTRACT

Objective:To investigate the efficacy of anaplastic lymphoma kinase-tyrosine kinase inhibitors (ALK-TKI) in treatment of inflammatory myofibroblastic tumor (IMT).Methods:The clinicopathological data of one recurrent abdominal IMT patient in Renmin Hospital of Wuhan University in 2018 were retrospectively analyzed. The clinicopathological and molecular characteristics, ALK-TKI treatment efficacy and prognosis of 41 patients with IMT reported in the literature from January 2010 to August 2020 were systematically reviewed.Results:This patient with abdominal IMT in Renmin Hospital of Wuhan University was a 27-year-old female who relapsed 2 months after surgery. Chemotherapy combined with bevacizumab was ineffective. After oral administration of crizotinib, the condition resolved after 1 month, and complete remission (CR) was achieved after 29 months. The median age of onset of 41 IMT cases reported in the literature was 22 years old (0-61 years old), of which 32 cases (78.0%) had multiple organ involvement, all of which had recurrence or metastasis. There were 38 cases of ALK mutation and 3 cases of TFG-ROS1 fusion gene-positive. Thirty-four patients treated with crizotinib in the first-line treatment of ALK-TKI, and the median resistance time of crizotinib was 8 months (2-48 months). The total clinical benefit rate of ALK-TKI was 85.3% (29/34), and 20 patients achieved CR. The median time for the first CR was 11 months (4-36 months), and the median duration time of medication for CR patients was 19.5 months (2-60 months). The median progression-free survival (PFS) time of 24 patients who underwent surgery and/or chemotherapy and radiotherapy was 4 months (1-45 months); after progression, ALK-TKI treatment was performed, and the median PFS time was 14 months (3-62 months).Conclusions:IMT is a true neoplasm with characteristics of recurrence and metastasis. Reasonable combination of ALK-TKI with surgery, radiotherapy and chemotherapy can improve the prognosis of IMT patients.

ABSTRACT

BACKGROUND: Bone marrow mesenchymal stem cells can be induced into myocardial tissue-like structure in vitro. Myocardial tissue lysates from different parts of the myocardium can be used to construct differential microenvironments. Few studies have investigated the targeted induction efficiency of bone marrow mesenchymal stem cells and the expression of related genes. OBJECTIVE: To investigate the effects of lysates from different parts of the myocardium on the differentiation of bone marrow mesenchymal stem cells into myocardial tissue-like structures and to assess the correlation between the expressions of related genes during induction and myocardial tissue engineering. METHODS: Passage 3 bone marrow mesenchymal stem cells from Sprague-Dawley rats were cultured. Whole heart tissue lysate, atrial muscle tissue lysate, and ventricular muscle tissue lysate were used to construct different microenvironments to induce bone marrow mesenchymal stem cells to differentiate into cardiomyocyte-like cells. The morphological changes and ultrastructure of cells were observed with an inverted phase contrast microscope and transmission electron microscopy. Expression of α-actin and cTnI was detected by immunofluorescence staining. qRT-PCR was used to detect the expression of upstream molecules ANP, HCN4 and downstream molecules MLC-2v in the cAMP/PKA signaling pathway after induction. RESULTS AND CONCLUSION: (1) Each induction cell group followed similar morphological changes: Cardiomyocyte-like cells were capable of autonomic beats, rhythmic contractions and relaxations; under the transmission electron microscope, there were a large number of arranged myofilaments; immunofluorescent staining showed positive expression of α-actin and cTnI. (2) The whole heart tissue lysate was able to induce the differentiation of bone marrow mesenchymal stem cells into rice grain-sized myocardial tissue-like structures with collagen fibers. (3) Atrial muscle tissue lysate could induce bone marrow mesenchymal stem cells to differentiate into atrial muscle-like cells. Autonomic beats appeared earlier, but the beat frequency and duration were shorter. ANP and HCN4 were highly expressed in atrial myocytes. (4) Ventricular muscle tissue lysate induced bone marrow mesenchymal stem cells to differentiate into ventricular muscle-like cells with no secretory granules, and MLC-2v was highly expressed in ventricular muscle-like cells. (5) To conclude, the whole heart tissue lysate can induce bone marrow mesenchymal stem cells to form myocardial tissue-like structures. Atrial muscle tissue lysate can induce bone marrow mesenchymal stem cells to differentiate into atrial muscle-like cells. Ventricular muscle tissue lysate can induce bone marrow mesenchymal stem cells to differentiate into ventricular muscle-like cells. By constructing a specific microenvironment, bone marrow mesenchymal stem cells can be differentiated to cardiomyocytes in different parts, providing laboratory data for the construction of myocardial tissue engineering.

ABSTRACT

Objective: To observe the effect of phlegm and blood stasis on the expressions of sirtuin 3(SIRT3)protein and urate transporter 1(URAT1) mRNA in skeletal muscle of diabetic rats with gout. Method: The 40 healthy rats, excepting the normal group, the remaining groups were fed with high-fat diet combined with low-dose streptozotocin solution (40 mg·kg-1) once a day, with blood glucose "16.7 mmol·L-1" as the criterion for the diabetes model. After 4 days, the 5% sodium urate solution was injected into the joint cavity once to induce the gout model. After the successful modeling, the Biling group (10 g·kg-1), the indomethacin group (5 mg·kg-1) and the pioglitazone group (10 mg·kg-1) continued to be administered for 21 days. The normal group and the model group were given the same amount of normal saline. The expression of SIRT3 protein in skeletal muscle tissue was determined by Western blot, URAT1 mRNA expression in bone tissue was detected by quantitative real-time fluorescence polymerase chain reaction(Real-time PCR),and blood was collected to measure blood glucose (GLU), blood uric acid (UA) and C-reactive protein (CRP). Result: Compared with the normal group, GLU, UA and CRP in the model group were significantly increased (PPPPPPPPConclusion: Biling Qutong prescription with effects in purging turbidity, detoxifying and dredging collaterals can significantly reduce the content of serum inflammatory factor CRP, significantly increase the protein expression of SIRT3 in skeletal muscle tissue of model rats, lower the content of URAT1 mRNA, reduce the blood glucose and blood uric acid levels in diabetic gout rats, and protect joints.

Objective To investigate the secreted protein acidic and rich in cysteine (SPARC) expression in the smooth muscle tissue of ob/ob mice.Methods Study time:February 2017 to June 2017.Six ob/ob mice and 6 littermates aged 10 weeks were selected for the trial.The mesenteric artery smooth muscle tissues were collected.qRT -PCR was used to detect SPARC mRNA expression in smooth muscle tissue.The protein expression of SPARC was assayed by Western blot.Results The SPARC mRNA [(1.73 ± 0.65)] and protein [(1.73 ± 0.65)] in smooth muscle tissue of ob/ob mice presented higher expression compared with those in the littermates [(1.00 ± 0.31),(1.00 ± 0.33),t =8.437,5.533,all P < 0.05].Conclusion The higher expression of SPARC in smooth muscles tissue of ob/ob mice may be involved in diabetes combined with atherosclerosis.

AIMS: To investigate the effects of resistance exercise and fish oil intake on muscle morphology in Wistar rats. METHODS: Forty-eight animals that performed resistance exercise were initially divided into two groups. One group did not take fish oil and the other group took fish oil. The animals of the second group underwent training and took fish oil for eight weeks. At the end of the last resistance exercise session, the 48 rats were organized into six subgroups of eight each, according to the time gap (12, 24 or 48 hours) elapsed until the gastrocnemius muscle withdrawal procedure. At each established time after the last resistance exercise session, the gastrocnemius muscle was removed for morphological analysis. RESULTS: Skeletal muscle cells of the animals that did not receive fish oil presented higher scores of edema, especially those from the groups that had their muscles withdrawn at 24 and 48 hours of time gap. As for the group that took fish oil, we observed a smaller amount of inflammatory infiltrate and reduced areas of necrosis compared to animals that exercised without the use of fish oil, at all post-exercise time gaps. CONCLUSIONS: Fish oil intake attenuated morphological changes in muscle tissue after high-intensity exercises


OBJETIVOS: Investigar os efeitos do exercício resistido e da ingestão de óleo de peixe na morfologia da fibra muscular em ratos Wistar. MÉTODOS: Quarenta e oito animais realizaram exercício resistido e foram divididos inicialmente em dois grupos. Um dos grupos não recebia óleo de peixe e o outro grupo ingeria o óleo de peixe. Os animais do segundo grupo realizaram o treinamento e ingeriram o óleo de peixe por um período de oito semanas. Ao final da última sessão de exercício resistido os animais foram organizados em seis subgrupos de oito cada, segundo o intervalo de tempo (12, 24 e 48 horas) transcorrido até o procedimento de retirada do músculo gastrocnêmio. Em cada tempo determinado após a última sessão de exercício resistido, o músculo gastrocnêmio foi retirado para análise morfológica. RESULTADOS: As células do músculo esquelético dos animais que não receberam óleo de peixe apresentaram escores maiores de edema, especialmente as dos grupos que tiveram os músculos retirados em 24 e 48 horas. No grupo que ingeriu o óleo de peixe observou-se menor quantidade de infiltrado inflamatório e áreas de necrose reduzidas em comparação com os animais que se exercitavam sem o uso de óleo de peixe, em todos os intervalos de tempo pós-exercício. CONCLUSÕES: A ingestão de óleo de peixe atenuou as alterações morfológicas no tecido muscular após exercícios de alta intensidade.

ABSTRACT Introduction: The prevalence of excess weight/obesity in adolescence has increased, and physical training may be effective in combating this scenario. Objective: To analyze the effect of different intensities of aerobic training on the body composition of obese adolescents undergoing multidisciplinary intervention. Methods: In this study, 107 pubertal (Tanner 3 and 4), obese (BMI=34.72±4.10 kg/m²) adolescents (14.85±1.44 years) were randomly assigned to three groups: high intensity training (HITG - ventilatory threshold I (VTI)); low intensity training (LITG - 20% below the VTI) and control group (CG), without intervention. The adolescents in the HITG and LITG underwent nutritional and psychological (1x/week) and clinical (1 x/month) follow-up and physical training (3x/week) for 12 weeks. Results: Reductions in fat mass (FM) and body fat percentage (BFP) (p< 0.001) and an increase in fat-free mass (FFM) (p<0.001) were observed in all groups. There was a reduction in BMI only in the experimental groups (p<0.001). Conclusion: The effects of multidisciplinary treatment on the body composition of obese adolescents occur independently of the intensity of aerobic physical training. Level of Evidence I; High quality randomized clinical studies with or without statistically significant difference, but with narrow confidence intervals.


RESUMO Introdução: A prevalência de sobrepeso / obesidade na adolescência aumentou, e treinamento físico pode ser eficaz para combater este cenário. Objetivo: Analisar o efeito de diferentes intensidades de treinamento aeróbio sobre a composição corporal de adolescentes obesos, submetidos à intervenção multidisciplinar. Métodos: Neste estudo, 107 adolescentes (14,85±1,44 anos), púberes (Tanner 3 e 4) e obesos (34,72±4,10 kg/m²) foram aleatoriamente alocados em três grupos: treinamento de alta intensidade (GTAI - limiar ventilatório I (LVI)), treinamento de baixa intensidade (GTBI - 20% abaixo do LVI) e grupo controle (GC), sem intervenção. Os adolescentes do GTAI e GTBI receberam acompanhamento nutricional e psicológico (1x/semana), clínico (1x/mês) e treinamento físico (3x/semana) durante 12 semanas. Resultados: Foram observadas reduções na massa gorda (MG) e no percentual de gordura (%G) em todos os grupos (p<0,001) e um aumento na massa livre de gordura (MLG) (p<0,001). Houve redução no IMC apenas nos grupos experimentais (p<0,001). Conclusão: Os efeitos do tratamento multidisciplinar sobre a composição corporal de adolescentes obesos ocorrem independentes da intensidade do treinamento físico aeróbio. Nível de Evidência I; Estudos clínicos randomizado de alta qualidade com ou sem diferença estatisticamente significante, mas com intervalos de confiança estreitos.


RESUMEN Introducción: La prevalencia de sobrepeso / obesidad en la adolescencia aumentó, y el entrenamiento físico puede ser eficaz para combatir este escenario. Objetivo: Analizar el efecto de diferentes intensidades de entrenamiento aeróbico sobre la composición corporal de adolescentes obesos sometidos a la intervención multidisciplinaria. Métodos: En este estudio, 107 adolescentes (14,85 ± 1,44 años), púberes (Tanner 3 y 4), obesos (34,72 ± 4,10 kg / m²) fueron aleatoriamente asignados en tres grupos: entrenamiento de alta intensidad (GEAI - umbral de ventilación I (LVI)); El entrenamiento de baja intensidad (GEBI - 20% por debajo del LVI) y el grupo de control (GC), sin intervención. Los adolescentes del GEAI y GEBI recibieron seguimiento nutricional y psicológico (1x/semana), clínico (1x/mes) y entrenamiento físico (3x/ semana) durante 12 semanas. Resultados: Se observaron reducciones en la masa grasa (MG) y en el porcentaje de grasa (% G) en todos los grupos (p<0,001) y un aumento en la masa libre de grasa (MLG) (p<0,001). Se observó una reducción en el IMC sólo en los grupos experimentales (p<0,001). Conclusión: Los efectos del tratamiento multidisciplinario sobre la composición corporal de adolescentes obesos ocurren independientes de la intensidad del entrenamiento físico aeróbico. Nivel de Evidencia I; Estudio clínico aleatorio de alta calidad, con o sin diferencia estadísticamente significativa, pero con estrechos intervalos de confianza.

Objective@#To study the clinicopathologic characteristics, immunophenotype and ALK gene alterations of gastrointestinal inflammatory myofibroblastic tumor.@*Methods@#Clinical data, histological features and immunohistochemical results were analyzed in 7 cases of gastrointestinal inflammatory myofibroblastic tumor at Zhejiang Province Taizhou Hospital from January 2005 to December 2016. ALK gene status was investigated by ALK fluorescence in situ hybridization.@*Results@#There were 4 female and 3 male patients. The age of patients ranged from 1 to 72 years (median age=53 years and mean age=40 years). The tumor was located in stomach (n=4), left hemicolon (n=1), right hemicolon (n=1) and rectum (n=1). Histologically, the tumors consisted of spindle fibroblast and myofibroblast cells growing in bundles with inflammatory infiltration primarily composed of plasma cells and lymphocytes. Immunohistochemical study showed spindle tumor cells were positive for vimentin (7/7), SMA (7/7), but were negative for CD34, CKpan, CD117, DOG1, S-100 and desmin. Two cases expressed ALK protein and fluorescence in-situ hybridization revealed the presence of ALK gene rearrangement in the both cases.@*Conclusions@#Gastrointestinal inflammatory myofibroblastic tumor is a rare neoplasm that is easily misdiagnosed. Its surgical removal is a reliable treatment. ALK may be a potential novel therapeutic target for inflammatory myofibroblastic tumor.

ABSTRACT

Objective To investigate the ultrasound features of hepatic inflammatory myofibroblastic tumor (HIMT).Methods A retrospective analysis was performed for the ultrasound features of 4 patients who visit Shengjing Hospital from February 2007 to July 2017 with pathologically confirmed HIMT,including the location,size,boundary,morphology,internal echo,and blood flow of lesions.Results The patients were aged 2-69 years and there were 2 male and 2 female patients.All 4 patients had single tumor with a maximum diameter of 3.4 -21.2 cm.Two patients had clear boundaries and two had unclear boundaries.Two patients had solid tumor with inhomogeneous low echo and 2 had mixed cystic and solid type;color Doppler flow imaging found blood flow signal in tumor in all patients.Conclusion Ultrasound can clearly reveal the size,internal echo,and blood flow of HIMT,but the ultrasound images of HIMT lack specificity.Pathological examination is still needed for a confirmed diagnosis.

Objective To analyze the CT and MRI features of splenic inflammatory myofibroblastoma (SIMF).Methods The CT and MRI images of 6 patients with SIMF were retrospectively analyzed.Results 5 cases underwent CT examination;2 cases did MRI.Six cases of SIMF were all single lesions;the average diameter was (3.9 ± 0.9) cm;the lesions were round or gourdshaped;most have clear boundaries with uneven scan density.On the MRI,the parenchymal part was slightly low signal on T1WI,inhomogeneous hypointensity on T2WI,or high signal on T2WI.After CT or MRI enhancement,lesions were progressively enhanced.Correct preoperative diagnosis was established in only 1 patient.Conclusion In this study,all SIMF cases present as single lesion.The circular appearance and clear boundaries are in common.Asymptotic intensification with map-unenhanced regions is the main enhancement feature.

OBJECTIVE: To report various anatomic locations and clinical characteristics of pathologically proven myofibroblastoma in Koran patients. METHODS: Pathologic reports of patients who underwent surgeries at two centers between April 2003 and March 2016 were retrieved from the electronic medical record system of the hospital. Pathologic reports were included after performing a search using the keyword “myofibroblastoma”. RESULTS: The cohort consisted of 11 subjects and included eight female and three male individuals. The patients' ages ranged from 9 to 66 years. Tumors were located in the vagina in three patients and presented in the breast in seven patients. One case presented with an abdominal mass. The tumors ranged in mean size from 4.0 to 53.0 mm. Despite a relatively long-term follow-up, no case had evidence of tumor recurrence. CONCLUSION: We evaluated the various anatomic locations of pathologically proven myofibroblastoma in Korean patients. As an extremely rare tumor, physicians should pay special attention to differential diagnosis. Surgical resection is the preferred method for a cure, and the recurrence rate is extremely low.

Objective@#To explore the clinical characteristics, diagnosis and treatment of pulmonary inflammatory myofibroblastic tumor in children.@*Methods@#The clinical data of 13 patients with pulmonary inflammatory myofibroblastic tumor between October 2009 and October 2015 were retrospectively analyzed, including 9 boys and 4 girls aged 1 to 12 years 4 months (mean 4 years 9 months). Seven patients underwent total tumor resection, 4 had lobectomy, 1 had partial tumor resection and 1 had biopsy. The follow-up time ranged from 2 months to 6 years and 2 months.@*Results@#Among the 13 patients, 9 were cured. Two patients lived with tumor. One of them had tumor which shrinked gradually and the other had stable disease. One patient with multiple lesions encountered recurrence after 3 operations and developed suspicious hepatic metastasis. One died during the operation because of airway mismanagement.@*Conclusions@#Pulmonary inflammatory myofibroblastic tumor in children has rather good prognosis while a few cases have the potential of recurrence and canceration. Surgery is the primary therapeutic approach. Close post-operative follow up is necessary.

ABSTRACT

Hormonal fluctuations during the different estrous cycle are a well-recognized cause of insulin resistance in bitches, and little is known about insulin receptor binding or post-binding defects associated with insulin resistance in dogs. To evaluate insulin binding characteristics in muscle tissue of bitches during the estrous cycle, 17 owned bitches were used in the study (six in anestrus, five in estrus, and six in diestrus). An intravenous glucose tolerance test (IVGTT) was performed in all patients by means of injection of 1mL/kg of a glucose 50% solution (500mg/kg), with blood sample collection for glucose determination at 0, 3, 5, 7, 15, 30, 45 and 60 minutes after glucose infusion. Muscle samples, taken after spaying surgery, were immediately frozen in liquid nitrogen and then stored at -80 ºC until the membranes were prepared by sequential centrifugation after being homogenized. For binding studies, membranes were incubated in the presence of 20,000cpm of human 125I-insulin and in increasing concentrations of unlabeled human regular insulin for cold saturation. The IVGTT showed no differences among bitches during the estrous cycle regarding baseline glycemia or glycemic response after glucose infusion. Two insulin binding sites - high-affinity and low-affinity ones - were detected by Scatchard analysis, and significant statistical differences were observed in the dissociation constant (Kd1) and maximum binding capacity (Bmax1) of the high-affinity binding sites. The Kd1 for the anestrus group (6.54±2.77nM/mg of protein) was smaller (P<0.001) than for the estrus (28.54±6.94nM/mg of protein) and diestrus (15.56±3.88nM/mg of protein) groups. Bmax1 in the estrus (0.83±0.42nM/mg of protein) and diestrus (1.24±0.24nM/mg of protein) groups were also higher (P<0.001) than the values observed in anestrus (0.35±0.06nM/mg of protein). These results indicate modulation of insulin binding characteristics during different phases of the estrous cycle in dogs, showing that muscle insulin binding affinity for its receptor is reduced during estrus and diestrus. However, this poor hormone-receptor affinity is compensated for by a greater total binding capacity, once there is no difference in patients' glycemic response after an intravenous glucose load.(AU)


As flutuações hormonais durante as diferentes fases do ciclo estral são uma causa importante de resistência insulínica em fêmeas caninas, e poucas informações são conhecidas sobre defeitos na ligação da insulina ao seu receptor, ou defeitos pós-receptor associados com resistência à insulina em cães. Para avaliar as características da ligação insulina-receptor no tecido muscular de cadelas durante o ciclo estral, dezessete pacientes foram utilizadas no estudo (seis em anestro, cinco em estro e seis em diestro). Um teste de tolerância à glicose intravenosa (IVGTT) foi realizado em todas as pacientes por meio da infusão de 1mL/kg de uma solução de glicose 50% (500mg/kg), com coletas de sangue para determinação de glicemia nos tempos 0, 3, 5, 7, 15, 30, 45 e 60 minutos da injeção de glicose. Amostras de tecido muscular foram coletadas durante ovariohisterectomia, imediatamente congeladas em nitrogênio líquido, e posteriormente armazenadas a -80°C até a preparação das membranas por meio de homogeneização e centrifugação sequencial. Para os experimentos de ligação hormônio-receptor, as membranas foram incubadas na presença de 20.000cpm de 125I-insulina humana, e concentrações crescentes de insulina regular humana não marcada para saturação fria. O IVGTT não mostrou diferenças entre as pacientes em diferentes fases do ciclo estral com relação a glicemia basal, ou na resposta glicêmica após infusão de glicose nos tempos estudados. Dois sítios de ligação da insulina, um de alta-afinidade, e outro de baixa afinidade, foram detectados pela análise de Scatchard, e diferenças significativas foram detectadas na constante de dissociação (Kd1) e capacidade de ligação máxima (Bmax1) dos sítios de ligação de alta-afinidade. O Kd1 para o grupo anestro (6,54±2,77nM/mg de proteína) foi menor (P<0,001) que os Kd1 dos grupos estro (28,54±6,94 nM/mg de proteína) e diestro (15,56±3,88nM/mg de proteína). Os Bmax1 dos grupos estro (0,83±0,42nM/mg de proteína) e diestro (1,24±0,24nM/mg de proteína) também foram maiores que os valores encontrados no grupo anestro (0,35±0,06nM/mg de proteína). Estes resultados demonstram uma modulação das características de ligação da insulina nas diferentes fases do ciclo estral em cães, evidenciando uma menor afinidade de ligação da insulina ao seu receptor no tecido muscular durante o estro e diestro. Contudo, esta menor afinidade de ligação hormônio-receptor é compensada por uma maior capacidade de ligação, o que fica também evidenciado pela ausência de diferenças na resposta glicêmica das pacientes após um desafio com glicose por via endovenosa.(AU)

ABSTRACT

Objective To retrospectively analyze the clinical data of 3 middle-aged and elderly cases with inflammatory myofibroblastic tumor (IMT) of the kidney,in order to improve its diagnosis and treatment.Methods The records of 3 patients who were diagnosed as IMT of the kidney by pathology in our hospital from Jan 2005 to Jun 2014 were reviewed to identify the characteristics including age,gender,clinical manifestation,tumor imaging and pathological features,treatment and prognosis.Results The mean age of the patients was 60 years.One patient underwent partial nephrectorny,1 patient underwent radical nephrectomy,and another patient received nephroureterectomy with bladder cuff excision.Patients were followed up for 8 months to 10 years,and no tumor recurrence was found.Conclusions IMT of the kidney is the tumor of low malignant potential with a good prognosis.Surgery is the first choice for the diagnosis and treatment.

La obtención de DNA humano en buena cantidad y alta pureza a partir de tejido muscular fijado en Formaldehido no tamponado es de suma importancia en la amplificación por la Reacción en Cadena de la Polimerasa (PCR) para su aplicación en estudios de identificación y filiación genética. En este estudio se evaluó la eficiencia del Kit QIAampR DNA FFPE TISSUE y una modificación del mismo basado en lavados con PBS y el tiempo de digestión con proteinasa K, frente a la cantidad y calidad de este ácido nucléico. Las diferencias fueron significativas entre los tiempos de acción con la proteinasa K (PK) en relación a la cantidad y en la pureza producto de los lavados del tejido muscular previa extracción. Estos resultados proporcionan una pauta para el diseño de experimentos de acuerdo con el efecto de la fijación, optimizando recursos humanos e insumos.


Obtaining human DNA in large quantity and high purity from muscle tissue fixed in formaldehyde unbuffered is of utmost importance in amplification by Chain Reaction (PCR) for application in identification studies and genetic affiliation. In this study, the efficiency of DNA FFPE TISSUE QIAampR Kit and a modification thereof based washes with PBS and the time of digestion with proteinase K, compared to the amount and quality of the nucleic acid were evaluated. Differences were significant between the exposure times with proteinase K (PK) in relation to the quantity and purity of the product prior washings muscle tissue extraction. These results provide a guideline for the design of experiments according to the effect of fixing, optimizing human resources and inputs.

Neste estudo, a eficácia do kit QIAampR DNA FFPE TISSUE e uma modificação do processo por lavagens com PBS e o tempo de digestão com proteinase K, em comparação com a quantidade e a qualidade do ácido nucléico desta base. As diferenças foram significativas entre os tempos de exposição com proteinase K em relação à quantidade e pureza produto das lavagens do tecido muscular extraída previamente. Estes resultados fornecem uma diretriz para o desenho de experimentos de acordo com o efeito de fixação, otimizando recursos humanos e insumos.

ABSTRACT

The present study was carried out to determine the total carbohydrate dynamics in the non infected and infected muscle samples of Nemipterus japonicus, harboured with pathogenic Klebsiella pneumoniae and Vibrio vulnificus. About twenty seven fish collected from the coast off Visakhapatnam, identified with lesions were undergone microbial analysis to isolate the incriminating pathogenic bacteria. The total muscle carbohydrate levels were analysed by Dubois method. The estimated total carbohydrates in the infected fish were found significantly higher as compared to the non infected fish. Thus it can be concluded that an increase of carbohydrate concentrations in infected fish may cause harmful physiological effects, reducing the immune response of the fish to the microbial organisms.

BACKGROUND: Low-intensity pulsed ultrasound (LIPUS) has been shown to stimulate tissue metabolism and accelerate muscle healing. However, the optimal parameters in the use of LIPUS are still not clear. OBJECTIVE: The aim of this study was to analyze the effects of LIPUS on muscle healing in rats subjected to a cryolesion. METHOD: Twenty rats were divided into the following groups: an injured control group (CG) and an injured treated group (TG). Both groups were divided into 2 sub-groups (n=5 each) that were sacrificed 7 and 13 days post-surgery. Treatments were started 24 hours after the surgical procedure and consisted of 3 or 6 sessions. After euthanasia, the muscles were submitted to standard histological procedures. RESULTS: Qualitative analyses were based on morphological assessments of the muscle. The histopathological analysis on day 7 revealed that the muscles in the CG and the TG presented an intense inflammatory infiltrate, a large necrotic area and a disorganized tissue structure. After 13 days, both the CG and the TG had granulation tissue and newly formed fibers. The TG presented a more organized tissue structure. The quantitative analysis of collagen indicated similar findings among the groups, although the qualitative analysis revealed a better organization of collagen fibers in the TG at 13 days. The immunohistochemical analysis indicated that, at both time points, the expression of cyclooxygenase-2 was upregulated in the TG compared to the CG. CONCLUSIONS: LIPUS used as a treatment for muscle injury induced a more organized tissue structure at the site of the injury and stimulated the expression of COX-2 and the formation of new muscle fibers. .
