ABSTRACT

ABSTRACT: In a rural property of the Midwestern Minas Gerais, Brazil, three 2.5 to 3-year-old female bovines (7/8 Holstein x 1/8 Gir) weighing approximately 380kg died after showing brownish colored mucosa of conjunctiva and vaginal vestibule, gray-bluish tongue, dyspnea, sialorrhea, tympany, and progression to sternal decubitus and death. In the macroscopic evaluation of one of the bovines, dark (chocolate-colored) blood of difficult coagulation, and intensely cherry-red skeletal and cardiac musculature were observed. Beyond that were observed brownish lungs, kidneys, liver, and encephalon, and all the stomach compartments were dilated due to large amounts of gas. In addition, the carcass exhaled a nitrous odor. The diphenylamine test and the nitrate dosage in the forage (Pennisetum purpureum, "elephant grass", Napier grass cultivar) used in feeding the bovines showed a high concentration of nitrate, allowing the diagnosis of nitrate/nitrite intoxication. The main factors leading to the poisoning were attributed to excessive fertilization of grass with bovine manure, the occurrence of a drought period followed by a rainy season, lack of adaptation of the bovines to a nitrate-rich diet, and a diet composed almost exclusively of elephant grass. This is the first report of spontaneous nitrate/nitrite poisoning in cattle in southeastern Brazil.


RESUMO: Em uma propriedade rural do Centro-Oeste de Minas Gerais, três fêmeas bovinas 7/8 Holandês e 1/8 Gir, com idade de 2 anos e 6 meses a 3 anos e peso de aproximadamente 380kg vieram a óbito após manifestação clínica de mucosas conjuntivais e do vestíbulo vaginal amarronzadas, língua azul-acinzentada, dispneia, sialorreia, timpanismo e progressão para decúbito esternal e morte. Na necropsia de um dos animais, foram observados sangue escuro (cor de chocolate) e de difícil coagulação, músculos esqueléticos e cardíaco intensamente vermelho-cereja, compartimentos gástricos distendidos por grande quantidade de gases e pulmões, rins, fígado e encéfalo amarronzados. Além disso, o cadáver exalava odor nitroso. O teste de difenilamina e a dosagem de nitrato na forragem (Pennisetum purpureum, "capim-elefante", cultivar Napier) oferecida aos animais revelaram concentração elevada de nitrato na planta, com diagnóstico de intoxicação por nitrato/nitrito. Os principais fatores que levaram à intoxicação foram atribuídos à adubação excessiva do capim com esterco bovino; à ocorrência de um período de estiagem seguido por um período chuvoso; à falta de adaptação dos animais à alimentação rica em nitrato; e à dieta composta, quase exclusivamente, por capim-elefante. Este trabalho é o primeiro relato de intoxicação espontânea por nitrato/nitrito em bovinos no sudeste do Brasil.

ABSTRACT

Descrevem-se os dados epidemiológicos, sinais clínicos e lesões da intoxicação espontânea e experimental por nitrato e nitrito em bovinos alimentados com pastagens de Avena sativa (aveia) e/ou Lolium spp (azevém). A enfermidade ocorre em diferentes regiões do Estado de Santa Catarina, quando as pastagens tem crescimento exuberante, após receberem quantidades excessivas de adubo químico e/ou orgânico, principalmente quando ocorrem condições climáticas de seca e posteriormente chuva. Os animais em contato com essas pastagens desenvolvem rapidamente mucosas de coloração marrom, taquipnéia, andar cambaleante, micção frequente, timpanismo, decúbito lateral e morte em poucos minutos, ou recuperação algumas horas após. Na necropsia de quatro animais que adoeceram espontaneamente, as principais alterações encontradas foram a coloração marrom das mucosas, a cor escura do sangue (cor de chocolate) e a coloração vermelho intensa da musculatura esquelética e do miocárdio esquerdo. A reprodução experimental da doença foi realizada em sete bovinos, com pastagens de quatro propriedades onde ocorreu adoença. Aveia e/ou azevém verdes e sob a forma de feno foram administradas aos bovinos. Destes bovinos quatro morreram, dois adoeceram e se recuperam, um naturalmente e outro com a aplicação de azul de metileno a 1%, na dose de 2mg/kg/peso vivo, e um bovino não apresentou alterações. Os sinais clínicos observados e as lesões nos animais que adoeceram e morreram foram idênticos aos casos naturais. Alterações microscópicas não foram observadas nos casos naturais e experimentais. O teste da difenilamina nas amostras de pastagens onde ocorreram os surtos resultou positivo em todas as propriedades. A análise bromatológica realizada em amostras coletadas de várias propriedades em que ocorreram surtos revelou de 0,30 a 3,36% de nitrato na matéria seca. A enfermidade caracterizou-se principalmente por respiração ofegante, sangue de coloração escura, mucosas de coloração marrom e morte rápida dos bovinos e está relacionada á ingestão de pastagens de aveia e/ou azevém super adubados, que acumularam alto teor de nitrato, após um período de chuvas precedido de seca.


Epidemiological data, clinical and pathological findings of the spontaneous andexperimental poisoning by nitrate and nitrite in cattle fed oats (Avena sativa) and/or ryegrass (Lolium spp.), diphenylamine test, and the nitrate content in the samples of the pasture where the outbreaks occurred, are described. The disease occurs in different regions of the State of Santa Catarina, in which pastures have exuberant growth, after receiving excessive amounts of chemical and/or organic fertilizer, mainly when raining occurs after a period of dry wheather. The animals grazing these pastures quickly develop brown mucosa, tachypnea, staggering gait, frequent urination, bloating, lateral recumbency and death in few minutes or hours. At necropsy of four animals that died spontaneously, the main lesions found were brown mucosa, dark color of the blood (chocolate), intense red color of the skeletal muscles and of the left part of the myocardium. The experimental reproduction of the disease was performed in seven cattle, with pastures from four farms where the disease occurred. The animals were fed with fresh oats and ryegrass and/or hay of it. Four animals died, two became ill and recovered, and one was treated with 2mg/kg per body weight of methylene blue 1%, and one cattle did not show changes. Clinical signs and lesions of the diseased animals that died were similar to natural cases. Microscopic changes were not observed in spontaneous and experimental poisoning. The diphenylamine test was positive in all the farms where the outbreaks occurred. The chemical analysis performed on samples of the pastures from several farms, in which outbreaks of the disease occurred, ranged from 0.30to 3.36% of nitrate in the dry matter. The disease is associated with the ingestion of oats and/or ryegrass pastures heavily fertilized, which accumulates high levels of nitrate after a period of rain and is characterized by rapid breathing, dark-colored blood, brown mucous and rapid death.

ABSTRACT

PURPOSE: To investigate gastric juice nitrate/nitrite concentration according to mucosal surface pH extent (area) of gastric corpus intimately contacting the gastric juice. MATERIALS AND METHODS: We included ninety-nine patients with dyspepsia. To evaluate gastric mucosal surface pH and its extent, gastric chromosocpy was performed by spraying phenol red dye on the corpus mucosa and estimating the extent of area with color changed. Nitrate/nitrite concentrations and pH of gastric juice were measured by ELISA and pH meter, respectively. Silver staining was done to histologically confirm the presence of Helicobacter pylori. RESULTS: Intragastric nitrate/nitrite concentrations in patients, showing phenol red staining mucosa were higher than those of unstaining mucosa (p=0.001): the more extensive in the area of phenol red staining area of corpus, the higher gastric juice pH found (r=0.692, p<0.001). Furthermore, the intragastric nitrate/nitrite concentrations correlated positively with gastric juice pH (r=0.481, p<0.001). CONCLUSION: The changes of mucosal surface pH and its extent in gastric corpus might affect either pH or nitrate/nitrite level of gastric juice.

ABSTRACT

Changes in nitrate and nitrite contents (leaves and stem) of Amaranthus gangeticus (AG) and Amaranthus paniculatus (AP), resulting from blanching, storage time (0-4 days), storage temperature (0 and 4ºC), and reheating were analysed. Results showed that fresh AG (1859 ± 7.07 mg/kg) had higher nitrite content than AP (1262 ± 2.12 mg/kg). Nitrites content was 506 ± 2 and 825 ± 3.5 mg/kg for AG and AP, respectively. Reheating and storage times significantly increased the conversion of nitrate to nitrite in AG and AP. Storage at 0 and 4oC exhibited a significant change (P < 0.05) in nitrate and nitrite contents for both samples. Higher nitrite content was found in AP when stored at 4oC and 0oC. The present study indicated that storage time and temperature affected the nitrite contents in blanched AG and AP when stored in low temperatures. Apart from that reheating was also found to increase the formation of nitrite.

ABSTRACT

Daily dietary intakes of nitrate, nitrite and vitamin C were investigated in Chanle county, Fujian province, where gastric cancer was very common, with a marked geographic variation in mortality. The average daily intake of nitrate of residents in Zhanggang village (high risk area) was 132.75 mg, which was significantly higher than that (84.65 mg) in Shouzhan village (moderate risk area), but similar to that (113.12 mg) in Meihua village (low risk area). However, nitrite intake in Zhanggang village was greatly higher than that in Shouzhan and Meihua villages (3.36 mg vs 0.21 and 0.37 mg, respectively). The average daily intake of vitamin C in Shouzhan village was 56.37 mg, significantly lower than that in Zhanggang village (123.09 mg) and in Meihua village (105.90 mg). The molar ratio of vitamin C intake to nitrite in the three villages was 3.22, 3.30 and 4.33, respectively. It was invesely associated with the mortality of gastric cancer. The results suggested that nitrate and nitrite might be etiological factors of gastric cancer in the county.
