ABSTRACT

@#Objective To develop and verify a double antibody sandwich ELISA detection method for the determination of ovalbumin(OVA),in order to determine the OVA content in influenza vaccines.Methods The rabbit anti-OVA polyclonal antibody was used as coating antibody and HRP labeled rabbit anti-OVA polyclonal antibody as detection antibody to develop a double antibody sandwich ELISA for OVA.The antibody concentration(2,1,0.5 and 0.25 pg/mL) for coating,enzyme-labeled antibody concentration(0.5,0.25 and 0.125 μg/mL),and kinds of blocking reagent(blocking with1% BSA,blocking with 2% BSA,blocking with 1% BSA and 1% sucrose,and blocking with 2% BSA and 2% sucrose,using nonblocking as control) were optimized,and the Cut-off value was determined as the judgment standard.The developed method was verified for the linear range and detection limit,specificity,repeatability and accuracy.Results The optimum detection conditions were as follows:the concentration of coating antibody was 1 μg/mL,the concentration of enzyme-labeled antibody was 0.25 μg/mL;The blocking reagent was 2% BSA and 2% sucrose;The Cut-off value was 0.051 66.The linear range of the method was 5~0.313 ng/mL,and the detection limit was 0.078 ng/mL;It did not react with influenza virus,bovine serum albumin(BSA) and Vero cell supernatant;The intraplate and interplate coefficient of variation(CV) were between 2.562%~13.887% and 4.000%~16.497% respectively;The coincidence rate between the results of this method and the Germany SERAMUN OVA quantitative test kit ranged from 93.79% to 107.05%.Conclusion The developed OVA double antibody sandwich ELISA has good specificity,repeatability,linear range and accuracy with convenience and lower cost,which might be used for the quantitative detection of OVA.

ABSTRACT

To improve the specific recognition and presentation of virus-like particle (VLPs), and to develop immune-targeted VLPs vaccine, the gene fragment encoding OVA₂₅₇₋₂₆₄ peptide was inserted into the VP3 gene of foot-and-mouth disease virus (FMDV) between the 171th and 172th amino acids (aa) or 173th and 174th aa by reverse PCR. The recombinant proteins were expressed by using Escherichia coli and assembled into chimeric VLP (VLP(OVA)) in vitro after purification. The VLP(OVA) was measured by dynamic light scattering and transmission electron microscopy. The recombinant protein and the assembled VLPs were evaluated by Western blotting, enzyme-linked immunosorbent assay and laser scanning confocal microscopy to confirm the insertion of OVA₂₅₇₋₂₆₄ peptide into VP3 and its location. The results show that insertion of OVA₂₅₇₋₂₆₄ into the 173th and 174th aa of FMDV VP3 did not affect the assembly of VLPs. The VLP(OVA) in size was larger than VLPs, and the OVA₂₅₇₋₂₆₄ peptide was located on the surface of VLP(OVA).

Resumen Los Objetos Virtuales de Aprendizaje han generado cambios positivos en la enseñanza, disponibilidad y accesibilidad al conocimiento; a diferencia del modelo tradicional de aprendizaje, tienen como propósito ofrecer mecanismos más flexibles y de múltiple consulta para consolidar un contenido más amplio del conocimiento. El hemograma es un examen de laboratorio de uso diario en medicina; su interpretación exige un entendimiento preciso y sólido para la toma de decisiones. Este artículo describe la experiencia y las fases para construir un objeto virtual de aprendizaje en hematología, basado en el modelo ADDIE (Análisis, Diseño, Desarrollo, Implementación y Evaluación), Crear un objeto virtual de aprendizaje exige el cumplimiento metódico de procesos para garantizar que el resultado se ajuste a los requerimientos de las herramientas de aprendizaje. Mejorar el entendimiento del hemograma es importante en la formación y fomento de competencias para la toma de decisiones asertivas y la racionalidad en la solicitud de estudios diagnósticos; no obstante, para probar la efectividad del OVA como estrategia de aprendizaje se requiere de una prueba piloto.


Abstract The virtual learning objects have generated positive changes in the teaching process, availability and accessibility to knowledge; unlike of the traditional learning model, it has as a purpose, to offer more flexible mechanisms and allow multiple queries to consolidate a broader content of knowledge. The complete blood cell count is a laboratory test of daily use in medicine, its interpretation demands a precise and solid understanding for the decision making. This article has the finality to describe the experience and the phases for the construction of an virtual learning object in hematology based in the ADDIE model, as follows: phase I: analysis, phase II: design, phase III: development, phase IV: implementation, phase V: evaluation. The setting-up of a virtual learning object demands the methodical accomplishment of process to assure the adjustment of the results to the requirements of the learning tools. The improvement in the understanding of the blood cell count has importance in the impact as a study element, as well as in the encouragement in the competences for make assertive decisions and the rationality in the request of diagnosis studies, however to prove the effectiveness as a learning strategy a pilot test is required.


Resumo Os Objetos Virtuais de Aprendizagem (OVA) geraram mudanças positivas no ensino, disponibilidade e acessibilidade ao conhecimento; ao contrário do modelo de aprendizagem tradicional, eles visam oferecer mecanismos de consulta mais flexíveis e múltiplos para consolidar um conteúdo mais amplo de conhecimento. O hemograma completo é um teste laboratorial para uso diário em medicina; sua intepretação requer um entendimento preciso e solido para tomada de decisões. Este artigo descreve a experiência e as fases para construir um OVA em hematologia, com base no modelo ADDIE (Análise, Projeto, Desenvolvimento, Implementação e Avaliação). Criar um OVA requer conformidade metódica com processos para garantir que o resultado atenda aos requisitos das ferramentas de aprendizado. Melhorar a compreensão do hemograma é importante no treinamento e na promoção de habilidades para tomar decisões assertivas e racionalidade na solicitação de estudos de diagnósticos; no entanto, para testar e eficácia do OVA como estratégia de aprendizagem, é necessário um teste piloto.

ABSTRACT

The health surveillance through a copro-ovoscopical study was conducted to assess the gastrointestinal parasitic infections in captive canine {jackal (Canis aureus), wolf (Canis lupus), dhole (Cuon alpinus) and hyaena (Hyaena hyaena)} and feline carnivorans {lion (Panthera leo), tiger (Panthera tigris), leopard (Panthera pardus) and jungle cat (Felis chaus)}, maintained at Bannerghatta Biological Park, Bengaluru, Karnataka. A total of 41 fecal samples from apparently normal/healthy captive lions, 35 from tigers, 34 from leopards, 14 from jungle cats, 4 from dholes, 4 from wolves, 4 from jackals and 2 from hyenas were collected over a period of 12 months during 2015-2016 and were screened using classical parasitological techniques including sedimentation and floatation technique followed by microscopic identification of eggs. It revealed the prevalence of ova of 3 (7.31%) Strongyle sp., 8 (19.51%) Ancylostoma sp., 21 (51.21%) Toxocara sp., 16 (39.02%) Toxascaris leonina, 4 (9.75%) mixed (Ascaris and Ancylostoma sp.) in lions; 19 (54.28%) Toxocara sp., 8 (22.85%) Toxascaris leonina, 6 (17.14%) Ancylostoma sp., 2 (5.71%) oocyst of Eimeria sp., 2 (5.71%) Spirometra sp. in tigers; 13 (38.23%) Toxocara sp., 6 (17.64%) Ancylostoma sp. in leopards; 7 (50%) Isospora sp. and 2 (14.28%) Toxocara sp. in jungle cats; 2 (50%) Ancylostoma sp. in jackals; 2 (50%) Toxocara canis and 1 (25%) Ancylostoma sp. in wolves; 2 (50%) Toxocara canis in dholes; while ova of only 1 (50%) Ancylostoma sp. could be observed in hyaena. This preliminary data may be useful for health management of the said species in captivity.

ABSTRACT

Abstract Filariasis is a major public health problem in tropical countries like India, China, Indonesia, Africa and the Far East. Aspiration cytology often helps in demonstration of microfilaria and its various forms, from uncommon diverse areas and in unusual clinical presentations like subcutaneous swelling caused by W.bancrofti in our cases. In our cases various forms of W.bancrofti were seen on cytology. Microfilria being most common form seen on cytology, adult worm, coiled forms (embryos) and ova seen less often. FNAC can be helpful in diagnosis of symptomatic as well as asymptomatic cases of lymphatic filariasis. During cytological evaluation of tissue fluids and aspirate from lesions of any part of the body, possibility of filariasis must be kept in mind as a possible differential diagnosis, particularly in endemic areas. This will help to give appropriate therapy to asymptomatic patients.

ABSTRACT

Objective To establish a simple animal model of cough variant asthma(CVA)through sensitizing Brown-Norway(BN)rats with ovalbumin(OVA). Methods A total of 36 BN rats were randomly divided into three groups, including the normal control group,the model control group and the montelukast group. BN rats in the model group and the montelukast group were intraperitoneally administered with 2.0 mg of OVA and 100 mg of Al(OH)3,and the same volume of sterile saline was given to the normal group by intraperitoneal injection. Boosting was carried out by intraperitoneal administration with 0.01 mg of OVA and 100 mg of Al(OH)33 weeks later,and the rats in the normal group were injected with the same dose of physiological saline. Three weeks later,the actively sensitized BN rats were challenged with aerosolized OVA for 7 times on alternative days,and the rats in the normal group were treated with sterile saline instead of OVA. At the same time, the montelukast group was given 1.3 mg/kg of montelukast 30 minutes before atomization by intragastric administration once a day for 2 weeks,and the normal group and the model group were given the same volume of water. The tests of cough sensitivity to capsaicin and bronchial responsiveness were performed 24 h after the last administration. Results Compared with the normal group, the times of coughing(P< 0.01)and the lung resistance(RL)(P< 0.05)in the model group were significantly increased,while the lung compliance(Cdyn)was significantly decreased(P< 0.05). There was a significant difference(P < 0.05)in the times of coughing caused by capsaicin between the model group and the montelukast group. Compared with the model group,RLin the montelukast group was decreased significantly(P< 0.05), and Cdynwas increased significantly(P< 0.05). Conclusions This rat model of CVA is similar to a variety of clinical features of CVA and is easy to operate. Thus it can be used as an effective animal model of CVA.

ABSTRACT

Aim To establish a cell model which stably co-ex-press human kappa opioid receptor (hKOR) and enhanced green fluorescent protein ( EGFP) labeled catalytic domain of cAMP-dependent protein kinase A(PKAcat) fusion protein (PKAcat-EGFP) in Chinese hamster ovary(CHO) cells, laying the foun-dation for the high-throughput screening of hKOR drugs and drug molecular mechanisms in vitro. Methods Hygromycin B resist-ant hKOR recombinant plasmid [ pcDNA3.1/Hygro ( + ) -hKOR] was transfected into CHO cells stably expressing PKA-cat-EGFP by a lipofectin based method. Transfected cells were selected in culture medium containing hygromycin B. The posi-tive clones were selected by PKA redistribution assay. Z’ factor was used for evaluation and validation the reliability of the cell model. PKA redistribution assay and LANCE cAMP 384 Kit were used to test the function of the receptors in selected clone. Results CHO-PKAcat-EGFP/hKOR-13 cell model exhibited stable response in PKA redistribution assay and LANCE cAMP 384 Kit. Treated with 100 nmol·L-1U-50488 for 30 min, the average value of Z’ factor was 0.596, proving the reliability of the cell model. The hKOR expression in cell model remained stable after a few generations. Conclusion The CHO-PKAcat-EGFP/hKOR-13 cell model with stable co-expression of hKOR and PKAcat-EGFP has been successfully established.

ABSTRACT

El artículo es el resultado de una investigación realizada entre los años 2015-2016, cuyo objetivo principal es mejorar el proceso de aprendizaje del concepto sustancia en estudiantes del grado 5to, mediante el diseño e implementación de una hipermedia educativa. El enfoque metodológico de la investigación es mixto, se realizó con 33 estudiantes de grado quinto. La metodología consta de tres momentos de análisis, en el primer momento se realizó un cuestionario y una entrevista para identificar las ideas previas de los estudiantes, y las dificultades en la enseñanza; en el segundo momento se hizo la descripción de las sesiones desarrolladas con los estudiantes desde la descripción hasta la implementación de la hipermedia; en el tercer momento se analizaron los resultados obtenidos durante y posterior a la implementación de la hipermedia, a partir de una matriz o rejilla previamente elaborada. Como resultado se evidenciaron aspectos positivos tanto en el aprendizaje como en las actitudes de los estudiantes, tales como el interés y la motivación que despierta en ellos este tipo de herramientas, permitiéndoles reconocer diferenciar y construir definiciones muy cercanas a la realidad, lo que facilito el avance y apropiación de otros conceptos, como a su vez la importancia de la apropiación del uso e implementación de lasTIC por parte de los docentes en el aula de clase.


This article is the result of research conducted between 2015-2016, whose main objective is to improve the learning process of the concept substance in the 5th grade students through the design and implementation of an educational hypermedia. The methodological approach of the research is mixed, was performed with 33 fifth graders. The methodology consists of 3 phases of analysis, the first time a questionnaire and an interview was conducted to identify the previous ideas of students, and difficulties in teaching; the second time the description of the sessions with students from the description to implementation of hypermedia was made; the third time the results obtained during and after the implementation of hypermedia from a matrix or grid made out previously analyzed. As a result positives were evident both in learning and attitudes of students, such as interest and motivation aroused in the student such tools, allowing them to recognize, differentiate and build definitions very close to reality, which facilitated the advancement and appropriation of other concepts, as in turn the importance of appropriation of use and implementation of ICT by teachers in the classroom.


O artigo é o resultado de uma pesquisa realizada entre 20l5-20l6, cujo objetivo principal é melhorar os alunos da 5a série processo de aprendizagem conceito substância através da concepção e implementação de uma hipermídia educacional. A abordagem metodológica da pesquisa é misturado, foi realizado com 33 estudantes na quinta série. A metodologia consiste em três etapas de análise, em primeiro lugar um questionário e uma entrevista foi realizada para identificar as ideias anteriores de estudantes, e dificuldades no ensino; a segunda vez foi a descrição das sessões desenvolvidas com os alunos a partir da descrição de aplicação da hipermídia; na terceira vez e os resultados obtidos durante hipermídias postimplementation, a partir de uma matriz preparada previamente ou grade analisados. Como resultado positivos foram evidentes tanto na aprendizagem e atitudes dos alunos, como o interesse ea motivação despertou neles essas ferramentas, permitindo-lhes reconhecer diferenciar e construir definições muito próximo da realidade, facilitando progresso e apropriação de outros conceitos, como, por sua vez a importância da apropriação de uso e implementação das TIC pelos professores em sala de aula.

ABSTRACT

Objective To investigate the immunopotentiating effects of cultivated Cistanche deser-ticola (C.deserticola) crude polysaccharides (CCDCP) as an adjuvant on the model antigen ovalbumin (OVA). Methods Low,medium and high doses of CCDCP in combination with OVA were intramuscularly injected twice into ICR mice at an interval of two weeks,respectively. Aluminum adjuvant was used to set up positive control group. Levels of IgG,IgG1 and IgG2a antibodies were detected by ELISA. Splenocyte prolif-eration was detected by MTT assay. Growth conditions of the immunized mice were observed. Results IgG level was significantly increased in the high dose group 7 days after the first immunization(P<0.05),espe-cially on the 21st and 28th days (P<0.01) as compared with that of the aluminum adjuvant group. High dose of CCDCP in combination with OVA significantly up-regulated the levels of IgG1 and IgG2a in mice as compared with immunization with OVA alone (P<0.05). Moreover, IgG2a level in mice immunized with high dose of CCDCP and OVA was higher than that of the aluminum adjuvant group(P<0.05). Splenocyte proliferation was significantly enhanced in the medium and low dose groups in comparison with that of the OVA group (P<0.05) and the aluminum adjuvant group (P<0.01). No significant difference in mouse body weight was observed in different groups(P>0.05). Conclusion CCDCP as an adjuvant of OVA pro-tein vaccine can enhance Th1 and Th2 immune responses,especially the early antibody production and Th1 immune response. These results will provide some information for further studies of CCDCP as a vaccine ad-juvant.

ABSTRACT

Asthma is characterized by chronic inflammation, goblet cell hyperplasia, the aberrant production of the Th2 cytokines, and eosinophil infiltration into the lungs. In this study, we examined the effects of baicalein, wogonin, and Scutellaria baicalensis ethanol extract on ovalbumin (OVA)-induced asthma by evaluating Th1/Th2 cytokine levels, histopathologic analysis, and compound 48/80-induced systemic anaphylaxis and mast cell activation, focusing on the histamine release from rat peritoneal mast cells. Baicalein, wogonin, and S. baicalensis ethanol extract also decreased the number of inflammatory cells especially eosinophils and downregulated peribronchial and perivascular inflammation in the lungs of mice challenged by OVA. Baicalein, wogonin, and S. baicalensis ethanol extract significantly reduced the levels of tumor necrosis factor α, interleukin (IL)-1β, IL-4, IL-5 and the production of OVA-specific IgE and IgG1, and upregulated the level of interferon-γ and OVA-specific IgG2a. In addition, oral administration of baicalein, wogonin, and S. baicalensis ethanol extract inhibited compound 48/80-induced systemic anaphylaxis and plasma histamine release in mice. Moreover, baicalein, wogonin, and S. baicalensis ethanol extract suppressed compound 48/80-induced mast cell degranulation and histamine release from rat peritoneal mast cells. Conclusively, baicalein and wogonin as major flavonoids of S. baicalensis may have therapeutic potential for allergic asthma through modulation of Th1/Th2 cytokine imbalance and histamine release from mast cells.

ABSTRACT

Estudios realizados sobre el tema de valoración de objetos virtuales de aprendizaje han aportado instrumentos que en gran medida han sido pensados para atender la generalidad de aspectos estructurales y no han profundizado en aspectos metodológicos, ni han atendido a particularidades intrínsecas de los conocimientos propios de una disciplina, lo que sugirió la necesidad de proponer e investigar sobre la definición de las características de un instrumento de valoración, atendiendo a las particularidades de las matemáticas y de sus contenidos. Para consolidar la construcción del instrumento de valoración de objetos virtuales de aprendizaje, se llegó a la conclusión que este debería atender a la relación entre el contenido, la pedagogía y la tecnología, aspectos que se profundizan en el marco teórico de la investigación. Un instrumento de carácter didáctico-conceptual, que abarca varias dimensiones y cuenta con unas orientaciones generales para los temas del área de matemáticas, en específico para la enseñanza de las fracciones, proporcionando un espacio de participación al profesor para la construcción del instrumento como tal. El instrumento de valoración fue validado en varias etapas, donde fueron partícipes profesores de matemáticas de diferentes niveles educativos. Como resultado de este proceso investigativo surgió un Instrumento nombrado "Valoración de Objetos Virtuales de Aprendizaje para la enseñanza de las fracciones", que pretendió llenar el vacío estructural y de contenido identificado en los demás instrumentos existentes.


Studies about the subject of valuation of virtual learning objects have contributed with instruments which are largely designed to serve the generality of structural aspects and they have not gone deeply in methodological aspects or they have attended intrinsic peculiarities of discipline-specific knowledge, which suggested the need to propose and investigate on the definition of the characteristics of a measurement instrument attending to the particularities of mathematics and their contents. To strengthen the construction of the instrument for the evaluation of virtual learning objects it was reached the conclusion that this should address the relationship between the content, pedagogy and technology, aspects that deepens in the theoretical framework of research. An instrument of didactic-conceptual character which encompasses several dimensions, and has general guidelines for subjects in the area of mathematics, in particular for the teaching of fractions, providing a space for participation to the Teacher for the construction of the instrument as such. The assessment instrument was validated in several stages, where they were involved math teachers from different educational levels. As a result of this research process emerged an instrument named "Valuation ofVirtual Learning Objects for Teaching Fractions", which sought to fill the empty structural and content identified in other existing instruments.


Estudos sobre o tema da avaliação de objetos virtuais de aprendizagem, têm fornecido ferramentas que foram em grande parte destinados a abordar a generalidade dos aspectos estruturais, e não já mergulhou em aspectos metodológicos, nem tenham frequentado peculiaridades intrínsecas da experiência de uma disciplina; Ele está sugerindo a necessidade de propor e investigar sobre a definição das características de um instrumento de avaliação, com base nas especificidades de matemática e seu conteúdo. Para fortalecer a construção dos objetos virtuais de aprendizagem instrumento de avaliação, concluiu-se que esta deve abordar a relação entre aspectos de conteúdo, pedagogia e tecnologia que aprofundam no referencial teórico da investigação. Um instrumento de natureza didático-conceitual, abrangendo várias dimensões e tem diretrizes gerais para os problemas na área da matemática, especificamente para fracções de ensino, proporcionando um espaço de participação para o professor para a construção do instrumento como tal. A ferramenta de avaliação foi validada em várias etapas, onde os professores de matemática eram participantes de diferentes níveis de ensino. Como resultado deste processo de investigação um instrumento designado "ranking Objetos Virtuais de Aprendizagem para frações de ensino", que visam preencher a estrutura vazia e conteúdos identificados nos outros instrumentos existentes surgiram.

ABSTRACT

Objective To investigate the efficacy of using crude polysaccharides extracted from cultivated Artemisia rupestris L. ( CARCP) in Xinjiang as an immunologic adjuvant for ovalbumin ( OVA) . Methods The mice were subcutaneously immunized twice with OVA vaccine formulated with CARCP. ELISA assay was performed to measure the levels of IgG, IgG1 and IgG2a antibodies. Flow cytometry analy-sis was performed to detect the percentages of different T lymphocyte subsets ( CD3+CD4+ and CD3+CD8+T cells, CD4+CD44+ and CD8+CD44+T cells) in splenocytes as well as the expression of intracellular cyto-kines ( CD4+IFN-γ, CD8+IFN-γ) and CD4+CD25+Foxp3+Treg cells. Results The levels of IgG, IgG1 and IgG2a antibodies in serum, the percentages of CD3+CD4+, CD3+CD8+, CD4+CD44+ and CD8+CD44+T cells as well as the ratio of CD4+/CD8+T lymphocytes increased significantly in mice immunized with OVA in combination with CARCP (P<0. 05). Moreover, CARCP enhanced the expression of CD4+IFN-γand CD8+IFN-γ( P<0. 05 ) , but inhibited the expression of CD 4+CD 2 5+Foxp 3+Treg cells ( P<0. 05 ) . Conclusion As an adjuvant for OVA, CARCP enhanced the humoral and cellular immune responses, especially the T cell immune responses.

ABSTRACT

Paragonimiasis is an important food-borne parasitic zoonosis caused by trematode species of the genus, Paragonimus occurring in many parts of the world except in Australia and Antarctica. In India, it is an emerging parasitic disease, which is endemic in the northeast states where people have a common practice of eating raw or inadequately cooked freshwater crabs. In these states, Paragonimus heterotremus has been identified as the major causative agent of the human paragonimiasis. The most common clinical form of the disease is pulmonary paragonimiasis; however, extra-pulmonary manifestations are not uncommon. Here, we report a case of primary massive unilateral pleural effusion due to paragonimiasis. The diagnosis was confirmed by finding Paragonimus ova in the pleural fluid. The patient was successfully treated with repeated thoracocentesis and a course of praziquantel.

The differential white cell count in BALF was performed, and lung tissue was detected by morpho-logicalanalysis.Results AllofOVA+pertussissensitization,OVA sensitization and pertussis sensitization could deteriorate lung function, increase inflammatory cells and cause pathological change, and OVA + pertussis sensitized rat model had better effect than OVA sensitized and pertussis sensitized rat models. Conclusion OVA+pertussissensitizationandOVAaerosolisa successful rat asthma model.

ABSTRACT

Objective:To determine the immunological effects of a new type adjuvant CTA 1-DD/IgG.Methods:Intranasal im-munization of BALB/c mice with OVA in combination with adjuvant CTA 1-DD or CTA1-DD/IgG.Blood samples were subjected to OVA-specific IgG Abs detection and IgG subclass profiles assessment using ELISA.Splenic plasma cells secreting anti-OVA Abs were detected by ELISPOT.Results:CTA1-DD/IgG showed stronger activity in inducing OVA-specific IgG Abs and splenic Abs-secreting cells compared with CTA1-DD.The enhancement of IgG1,IgG2a and IgG2b Abs were observed,and the ratio IgG2a/IgG1 were >1 in both groups.Conclusion: CTA1-DD/IgG can exert enhanced adjuvant effects compared with CTA 1-DD.Mixed IgG subclasses are induced,indicating a more balanced Th1/Th2 response.

ABSTRACT

Immunostimulants are susbstances that stimuli the response of effector cells to activate the immune response such as antigen uptake, cytokine release or antibody response. These substances can increase resistence to infection by different types of microorganisms, reducing dependence of antibiotics used in livestock animals. Recent reports have demonstrated the positive effect of Propionibacterium acnes (P. acnes) to control animal diseases. In this study, we evaluated the effect of the non-specific immunostimulant P. acnes on immunological functions and growth performance in goat kids. Twenty five goat kids served as control group (A) and another 25 animals received P. acnes being the experimental group (B). Kids were challenged with ovalbumin (OVA) to assess humoral immunity. To assess in vivo cell immunity, delayed type hypersensitivity (DTH) test with phytohemagglutinin (PHA) was used, clinical signs and body weight were recorded each week until 9 weeks of age when the experiment ended. Blood samples were obtained to analyze serum proteins fractions and anti-OVA specific antibodies. No clinical signs of disease and no differences (p>0.05) on body weight between groups were recorded (7.32±0.81 kg in group A, 7.13±0.65 kg in group B). Goat kids from group B had more total protein (59.8±5g/l) and albumin levels (32.8±3.3g/l) than goat kids from group A (56.6±5.7 g/l, 29.6±3.9 g/l respectively) (p<0.05). DTH response in goat kids from group B on day 42 was higher (p<0.05) than group A. At day 63, goat kids from group receiving P. acnes had higher percentage (85.4) of anti-OVA IgM titers (p<0.05) than control group (57.7). In conclusion, the results showed that oral administration of P. acnes to goat kids improved some aspects of the immune system of the animals and it could be used to control goat diseases.

En este artículo se analizan con parámetros de composición 58 puestas de trucha arcoiris obtenidas, manejadas y monitoreadas individualmente en desempeño desde la fertilización hasta finalizar la etapa de reabsorción de vesícula. Además de los valores reproductivos, el contenido de ácidos grasos en las ovas se determinó en fresco, mediante cromatografía de gases. En el perfil medio se destacan los ácidos palmítico (C16), oleico (C18:1n-9) y docosahexaenoico (C22:6n-3) como los más representativos, con casi el 60% del total y, en general, en concentraciones estables entre las hembras. Tanto para cada ácido determinado como para el conjunto de contenidos integrados de las series n-3 y n-6, y los colectivos de saturados (SAF), monoinsaturados (MUFA) y poliinsaturados (PUFA), se define un patrón de composición similar al reportado para la especie en otros esquemas de manejo y bajo regímenes nutricionales variables. This article analyzes, with composition parameters, 58 rainbow trout egg-layings obtained, handled and monitored individually in performance from their fertilization until the end of the gallbladder reabsortion phase. Aside from the reproductive values, the content of fatty acids in the eggs was determined in fresh conditions through gas chromatography. The average profile highlights the palmitic acid (C16), oleic acid (C18:1n-9) and docosahexaenoic acid (C22:6n-3), as the most representative, with almost 60% of the total and, in general, in stable concentrations among females. Both for each determined acid and for the suite of integrated series n-3 and n-6, and the groups of saturated (SAF), monounstaturated (MUFA) and polyunsaturated (PUFA), a pattern of similar composition to the one reported is defined for the species in other management schemes and under varying nutritional regimes. Além dos valores reprodutivos, o conteúdo de ácidos graxos nas ovas foi determinado em fresco, mediante cromatografia de gases. No perfil médio destacam-se os ácidos palmíticos (C16), oleico (C18:1n-9) e docosaexaenoico (C22:6n-3), como os mais representativos, com quase 60% do total e, em geral, em concentrações estáveis entre as fêmeas. ABSTRACT

Objective To elucidate potential roles of tyrosine kinase 2 (Tyk2) in the generation and maintenance of Ag-specific CD8+ T cells. Methods We followed the fate of OVA-specific CD8 + T cells in Tyk2-deficient ( Tyk2 -/- ) mice after infection with recombinant OVA-expressing BCG ( rBCGOVA ). Because the immunostimulatory BCG-derived peptides recognized by CD8 + T cells have not been defined, and the OVA is definite peptide for specific CD8 + T cells that has been accepted widely, therefore we examine the kinetics of the OVA-Ag-specific CD8 + T cell response after rBCG-OVA infection in mice.Tyk2-/- and wild type(Tyk2+/+ ) mice were inoculated with rBCG-OVA by intra-trachea( i. t. ), after the examination of bacterial growth in the lung and spleen, the population of CD8 + T cells were detected by FACS analysis, the epitope-specific CD8 + T cells were followed with tetrameric H-2Kb molecule folding with OVA257 264 peptide, and the kinetics of Ag-specific CD8 + Tc1 cells were detected by intracellular IFN-γ production in response to OVA257-264 peptide by cytokine FACS analysis. Results After rBCG-OVA challenge,the bacteria number in spleen and lung of Tyk2 -/- mice were significantly larger than those in Tyk2 +/+ mice on days 14, 21 and 49. Almost as same as that in Tyk2+/+ mice, the size of epitope-specific CD8+ T cella with OVA257-264/Kb-tetramer-positive and the CD8 +Tc1 (T eytotoxic 1 )cells positive for intracellular IFN-γ could proliferate to its peak on day 21, then contract and maintain to the memory phase in spleen and lung of Tyk2-/- mice, but the population of CD8+ T cells in spleen and lung of Tyk2 -/- mice were significantly smaller than those in Tyk2+/+ mice on days 21 and 49, the number of epitope-specific CD8+ T cells in spleen and lung of Tyk2 -/- mice were significantly decreased and the frequency of CD8 + Tc1 cells in spleen and lung of Tyk2 -/- mice significantly reduced on day 21,49 and 70 after rBCG-OVA infection. So correspond with the larger number of bacteria in Tyk2-/- mice than those in Tyk2 +/+ mice, the expansion of OVA257-264-specific CD8 + T cells and CD8+ Tc1 response were attenuated in Tyk2 -/- mice following rBCG-OVA infection. Conclusion These results suggest that the lack of Tyk2 signaling impairs the proliferation and difference of effector CD8 + T cell to rBCG-OVA infection and at least, is partly responsible for the susceptible to the rBCG-OVA infection.

ABSTRACT

Objective To investigate the effect of Schistosoma japonicum ova on the expression of intestinal NOD2/CARD15 in the mice induced by 2,4,6-trinitrobenzesulfonic acid (TNBS). Methods Mice (n=50) in the experiment were randomly allocated into 3 groups: control group (n=10), TNBS+saline (n=20) and TNBS+Schistosoma japonicum ova (n=20). TNBS enema (100 mg/kg) was applied to the two TNBS groups in order to establish a colonitis model. Schistosoma japonicum ova was administered i.p. on the 14~(th) and 3~(rd) day before the instillation of haptenating agent. All mice were killed on the 7~(th) day after colitis induction. The transcription level of NOD2 in colon tissues was measured by Real time PCR, and the expression of NOD2 protein was measured by Western blot. Results The transcription and protein levels of NOD2 in TNBS-induced mice increased statistically compared with those of the normal group (P<0.05). Compared with the TNBS-induced mice, Schistosoma japonicum ova-treated ones exhibited a statistical reduction of gene and protein expression (P<0.05). Conclusions TNBS-treated mice exhibited a statistical increased expression of NOD2/CARD15, Schistosoma japonicum ova treatment reduced the severity of experimental colitis through down-regulating NOD2/CARD15.
