ABSTRACT

Objecttive To observe the repaired effect of distinct source olfactory ensheathing cells (OECs) on spinal cord injury (SCI) rats. Methods These OECs were dissociated from olfactory bulb and olfactory mucosa of SD rats and transplanted to the injuried region of spinal cord injury rats. The function of nerve, motor evoked potential of hind legs and the histopathlogical diversities of injuried spinal cord were observed. Results The OECs grafts into the SCI area could survive longer time. The BBB scale, incubation stage of EP and histopathologic manifestations showed that the group with transplanted OECs regained more improvement in hindlimb than the control group. Conclusion The OECs of two sources have the same ability to regain and improve the axonal function which can promote axons regeneration of SCI.

ABSTRACT

[Objective]To investigave the feasibility of commensalism of stem cells and olfactory ensheathing cells and to study the preliminary results of graft of stem cells and olfactory ensheathing cells for the treatment of spinal cord injury.[Method]Under the condition of sterility, the stem cells and olfactory ensheathing cells were collected from the embryon of induced labor for 4 to 6 months and the patients. The stem cells and olfactory ensheathing cells were cultured individually until transduced generation, then were cultured together and transplanted into the place where the spinal cord was injuried in the patient.[Result]Eight patients who had the spinal cord injury were treated with the graft of stem cells and olfactory ensheathing cells and were followed up for 1 to 6 months after the operation. One case with the T_(5~7) spinal cord injury got the recovery of superfical and deep sensation of the lower limbs. The other 7 patients had not obvious recovery.[Conclusion]There was no appearance of inhibition when the stem cells and olfactory ensheathing cells were cultured together. The united graft of the stem cells and olfactory ensheathing cells for the treatment of spinal cord injury is safe with the short-term observation.

ABSTRACT

Objective Investigate the morphological features and distribution of olfactory ensheathing cells(Ecs) to study the relationship with CNS regeneration. Methods Luxol fast blue,Mallory special staining methods and TEM were emploed. Results ECs are distributed within the first two layers of olfactory bulb(OB) and olfactory epithelium along the olfactory nerve. The majority of cells are flattened with extended cytoplasm, although some are bipolar or tripolar with long and thin processes.NGFRp75 immunocytochemical reactive are sequentially expressed by ECs in the first two layers of OB and olfactory epithelium.TEM showed that the ECs possessed an irregularly shaped nucleus with a prominent nucleolus, and the mesaxon of each ECs encloses densely packed bundles of unmyelinated axons.Conclusion\ The ECs are a type of macroglia exclusively located in the first two layers of OB and olfactory epithelium, the morphology of EC is flattened with extended cytoplasm and the mesaxon of each ECs encloses densely packed bundles of unmyelinated axons.
