ABSTRACT

Objective To establish an UV spectrophotometry method for the determination of total flavonoids in Oxytropis falcata Bunge. Methods Using rutin as comparison,three coloration methods were used to find the optimal assay method, optimize the color conditions and take a systematic methodological investigation. Results The best color development method is aluminum chloride-sodium acetate color method. Using the test product without adding color reagent as a reference, 0.3 mol/L aluminum chloride solution 3.0 ml, 1.0 mol/L sodium acetate 4.0 ml, placed for 16 min, the sample was detected at 277 nm wavelength by AlCl3-CH3COONa reaction by using rutin as reference. The rutin content had a good liner relationship in the range of 8.8 to 44 μg/ml (r=0.9996), and the average recovery rate was 100.91% with RSD of 2.426%. Conclusion This method is simple, rapid, accurate and sensitive and can be used as a method for the determination of total flavonoids from Oxytropis falcata Bunge.

ABSTRACT

Objective: To break the dormancy of Oxytropis falcata seeds and improve the seed vigor. Methods: The physical and chemical methods were used to break the hard seeds, and the exogenous chemicals were used to improve the seed vigor. Results: The 1000-seed weight was 1.239 g, and the seed hardiness rate was as high as 90.67%. After 8 min of treatment with concentrated sulfuric acid, the seeds were activated by CTK + KNO3 combination. The germination rate was up to 82.7%, the germination potential was up to 68.0%, and the germination time was 2 d earlier than the control group. Conclusion: After treated with concentrated sulfuric acid for 8 min, treated with 170 mg/L CTK for 12 h, then soaked in 2% KNO3 for 12 h, the germination rate and uniformity of seeds can be greatly improved.

ABSTRACT

Objective To investigate the antibacterial activity of the alkaloids extract from Oxytropis falcata Bunge against 9 common pathogens in vitro. Methods Alkaloids was extracted from Oxytropis falcata Bunge. The agar plate dilution method was adopted to evaluate the activity against bacteria of 4 Gram-negative test strains and 5 Gram-positive test strains, while the MIC and MBC were determined by tube double dilution method. Results The alkaloids extract from Oxytropis falcata Bunge had the antibacterial activity against 9 pathogens and there was a good dose-response relationship between sensitivity and concentration. Antibacterial activities against bacillus subtilis and stentrophmomas maltophilia showed stronger than other tested bacteria, the MIC and MBC were 0.95 mg/mL and 1.91 mg/mL, respectively. In other tested bacteria, MIC and MBC were ranged in 1.91-7.63 mg/mL. Conclusion The alkaloids extract showed a broad spectrum of a stronger antibacterial activity against all tested pathogens. Gram-positive strains were more sensitive than Gram-negative strains to the inhibitory effects of the alkaloids extract.

ABSTRACT

Objective To observe the anti-inflammatory effects of rhamnocitrin in Oxytropis falcata Bunge;To provide experimental evidence for searching the anti-inflammatory active component of Oxytropis falcata Bunge. Methods Kunming mice or Wistar rats were randomly divided into model group, naproxen group and rhamnocitrin high-, medium-, low-dose groups, and corresponding medicines were administered by gavage for 7 days. Animal inflammatory models were established respectively by the mice ear swelling induced by dimethylbenzene, the mice abdominal capillary permeability induced by acetic acid, the rat paw edema created by injection of albumen and the rat granuloma induced by cotton pellet wool. The effects of rhamnocitrin on acute and chronic inflammatory animal models were observed. Results Different doses of rhamnocitrin groups could reduce the animal ear swelling, abdominal capillary permeability, paw edema and granuloma. Compared with the model group, there were obvious differences in rhamnocitrin groups (P<0.05). High dose rhamnocitrin group and naproxen group showed relatively strong inhibition effects on swelling, with statistical significance (P<0.05). Conclusion Rhamnocitrin has anti-inflammatory effects and is an important biological component of Oxytropis falcata Bunge.
